Description
Oppfinnelsen vedrører jodtyroninimmunogenkonjugat og jodtyroninderivat for fremstilling av jodtyroninimmunogen-konjugatet. Slike antistoffer er nyttige ved immunologiske analyser for å bestemme jodtyroniner i biologiske væsker.
Jodtyroninene har den følgende generelle formel:

Den kvantitative konsentrasjonsbestemmelsen av de forskjellige jodtyroninene, særlig hormonene T-3 og T-4, i serum og av metningsgraden av de jodtyroninbindende stedene ipå. bærerprdteinet tyroidbindende globulin (TBG) er verdi-fulle hjelpemidler ved diagnosen av tyroidsykdommer.
Det er fremstilt derivater av jodtyroniner på mange forskjellige måter ved å koble dem til immunogene bærermaterialer med det formålet å fremstille immunogenkonjugater. Antistoffer fremstilt mot slike konjugater brukes i immunologiske analyser for å bestemme jodtyroniner. Det er også blitt fremstilt forskjellige jodtyroniner i den hensikt å tilkoble en ønsket markørrest, hvorved man fikk et merket konjugat som er nyttig i slike immunologiske analyser.
Den vanlige brukte teknikken for å fremstille jodtyronin-immunogenkon jugat er omfatter å koble jodtyroninet direkte over dets tilgjengelige amino- og karboksylsyregrupper til motsatte amidbundne karboksylsyre- og aminogrupper på bærermaterialet i nærvær av karbodiimid. Det oppnåes en blanding av konjugater og følgelig erholdes det en blanding av anti-jodtyroninantistoffer. Se Gharib et al, J. Clin. Endocrinol 33:509 (1971).
US patentskrift nr. 4.171.432 beskriver sammenkoblingen av et ribonukleotid med jodtyroniner over karboksylgruppen.
US patentskrift nr. 4.040.907 beskriver sammenkoblingen av enzymer med jodtyroninderivater hvor substituenten sitter i fenol-hydroksylgruppen.
Fremstillingen av aminoakryltyroksinderivater hvor substituenten sitter i aminogruppen til tyroksin er beskrevet i Endocrinol. 89:606-609 (1971).
Oppfinnelsen vedrører jodtyroninimmunogenkonjugat som
er karakterisert ved at det har formelen
hvor bærer er et immunogent protein eller polypeptid, R' er karboksyl eller bis-imidiat,n er et helt tall fra 2-6,
p er i gjennomsnitt fra 1-50, R er hydrogen eller alkyl

som inneholder 1-6 karbonatomer, og 3 og 3 er,uavhengig av hverandre hydrogen eller jod. Når brogruppen R' er en binding, er jodtyroninderivatet koblet direkte til bærermaterialet, f.eks. ved amidbindinger mellom aminogruppen i jodtyroninderivatet og karboksylsyregrupper i bæreren,
som i et slikt tilfelle vanligvis er et protein eller polypeptid. Når brogruppen R' er noe annet enn en enkelt binding, kan den omfatte en lang rekke strukturer, f.eks. resten av bifunksjonelle sammenbindende midler som kobler aminogruppen i jodtyroninderivatet til aminogrupper i bæreren som igjen vanligvis er et protein eller polypeptid.
Den foreliggende oppfinnelse er også rettet mot et jodtyroninderivat med for fremstilling av jodtyroninimmunogen-konjugatet, og som er karakterisert ved at det har formelen

Figurene 1 og la utgjør et flytdiagram for synteseveien illustrert i eksemplene ved fremstillingen av jodtyroninderivater som er sammenkoblet med immuno-gene bærere til de immunogene konjugatene ifølge foreliggende oppfinnelse.
Jodtyroninderivatene fremstilles vanligvis på den måten
som er illustrert i diagrammet på tegningsfigurene 1 og la. Et co-aminoalkylaldehyddietylacetal (1) beskyttes som N-trifluoracetatet (2) ved behandling med etyltrifluoraee-tat og trietylamin i et passende oppløsningsmiddel, f.eks. etanol. Det resulterende N-trifluoracetylalkanaldiacetal hydrolyseres til alkylaldehydet og omsettes med en jod-tyroninetylester (3) og natriumcyanborhydrid i et passende oppløsningsmiddel, f.eks. etanol, hvorved man får N-(u-N-trifluoracetylaminoalkyl)jodtyroninetylestereiv (4). Alkalisk hydrolyse girde nye N-(aminoalkyl)jodtyroninderivatene (6). Alternativt kan N-trifluoracetylgruppen fjernes selektivt ved tilbakeløpskoking i saltsyremettet etanol hvorved man får dihydrokloridet av (5) som har bedre oppl.øselighets-egenskaper i organisk oppløsningsmiddel.
Lengden av den rettkjedede alkyIdelen av N-aminoalkylside-kjeden kan variere fra 2 til 12 karbonatomer ved passende utvelgelse av cj-aminoalkylaldehyddietylacetal-utgangsmaterialet. På samme måte vil alkylestergruppen, dersom den er tilstede, variere i overensstemmelse med det utvalgte jodtyraniinalkylester-utgangsmaterialet og . kan være rett-kjedet eller forgrenet og inneholde mellom 1 og 6 karbonatomer, f.eks. metyl, etyl, n-propyl, iso-propyl, n-butyl, sek.-butyl, tert.-butyl og så videre.
N-(aminoalkyl)jodtyroninesterderivatene (5) omdannes til de tilsvarende syrederivatene (6) ved behandling med base. Ester- og syrederivatene av jodtyronin kan bindes ved hjelp av vanlige peptidkondensasjonsreaksjoner til immunogene bærerltiaterialer som inneholder karboksylgruppe, hvorved man får immunogene konjugater med formelen:

Kondensasjonsreaksjonene for peptid som er tilgjengelig for å utføre den direkte sammenbinding av jodtyroninderivatet til en bærer som inneholder karboksylgruppen, er velkjent og omfatter uten begrensninger karbo-diimidreaksjonen (Aherne et al, Brit. J.Clin.Pharm. 3:56
(1976) og Science 144:1344(1974)), den blandede anhydrid-reaksjonen [Erlanger et 'al, Methods in Immunology and Immunochemistry,, ed. Williams and Chase, Academic Press (New York 1967) p': 149] samt syreazid og aktiv ester-reaksjonene [Kopple, Peptides and Amino Acids, W.A. Benjamin, Inc. (New. York 1966).]. Se også Clin. Alternativt kan ester- og syrederivater av jodtyronin kobles gjennom bruken av sammenbindende reagenser som danner en binding i den ene enden med aminogruppen i jodtyroninderivatet og en binding i den andre enden med en passende funksjonell gruppe som er tilstede i bærer-
materialet. For eksempel éir bifunksjonelle koblings-
reagenser velkjente for sammenkobling av aminderivater til
aminmakromolekyler, deriblandt bis-imidaterr bis-isocya-..
nater og glutaraldehyd [Immunochem. 6:53(1969)]. Andre nyttige sammenkoblingsreaksjoner er grundig diskutert i litteraturen, for eksempel i den ovenfor nevnte Kopple-monografen; Lowe and Dean, Affinity Chromatography,
John Wiley & Sons (New York 1974); Means and Feeney, Chemical Modification of Proteins, Holden-Day (San Francisco 1971); og Glazer et al, Chemical Modification of Proteins, Elsevier (New York 1975).
Sammenkobling av jodtyroninderivatet med en protein- eller polypeptidbærer ved bruken av de bis-imidat bifunksjonelle reagensene er en fremgangsmåte hvor de resulter-
ende immunogenkonjutjater vil omfatte en sammenbindende gruppe R<1> med formelen:
hvor m er et helt tall fra 1 til 10 og bærermaterialet er tilkoblet gjennom en aminogruppe. De bis-imidatsammen-koblede reagensene vil vanligvis ha formelen:

hvor m er som definert ovenfor og R og R , som kan være like eller forskjellige, men som vanligvis er like, er alkyl, fortrinnsvis lavere alkyl (dvs. at de har 1-4 karbonatomer) slik som metyl, etyl, n-propyl, iso-propyl osv. Særlig foretrukne bis-imidater er dimetylalkylimidater, spesielt dimetyladipimidat. Bis-imidatene er vanligvis tilgjengelige i handelen, eller kan fremstilles av fagmenn ved hjelp av kjente fremgangsmåter [Hunter and Ludwig, J. Am.Chem. Soc. 84:3491 (1962)]. Bis-imidatene vil vanligvis
bli tilveiebragt i en passende saltform som etter oppløs-ning i de vandige reaksjonsmedia, gir de positivt ladede bis-imidatartene. Tilsvarende vil isolering av det immunogene konjugatet fra vandige media, slik som ved løsningsmiddelinndampning eller utfelling, gi saltformer av bis-imidatene hvor de motsvarende anionene til de protoniserte iminogruppene tas fra tilgjengelige anioner i mediene.
Man lar sammenkoblingsreaksjonen skje i vandig oppløsning under milde betingelser, f.eks. ved en pH mellom ca. 7 og ca. 10, vanligvis mellom 8 og 9, og ved temperaturer mellom ca. 0°C og ca. 40°C, vanligvis mellom 20°C og 30°C. Vanligvis tillates de amino-funksjonaliserte jodtyroninderivatene, bis-imidatet og det ønskede protein- eller polypeptidbærermaterialet i rekkefølge med en kort inkuberingsperiode for omsetning mellom jodtyroninderivatet og bis-imidatet på mellom 1 og 30 minutter, fulgt av tilsetning av proteinet eller polypeptidet og en annen inkubasjonsperiode som varer mellom 10 minutter og 4 timer.
Størrelsen p i formlene ovenfor angir antallet jodtyronin-rester som er konjugert til bæreren, dvs. den epitopiske densitet til immunogenet, og vil vanligvis være i gjennomsnitt fra en til ca. 50, som oftest fra 1 til ca. 20. Optimale epitopiske densiteter, idet man tar i betraktning lettheten og reproduserbarheten av syntese av immunogenet og antistoffrespons, er mellom ca. 2 og ca. 15, vanligvis mellom 4 og 10.
Det immunogene bærermaterialet kan velges blant en hvilken som helst av de vanlige kjente som har tilgjengelige funksjonelle grupper for sammenkobling med jodtyronin amino-derivatene. I de fleste tilfelle er bæreren et protein eller polypeptid, selv om andre materialer slik som karbo-hydrater, polysakkarider, lipopolysakkarider, nukleinsyrer og lignende med tilfredsstillende størrelse og immuniserende egenskaper kan også brukes. Immunogene proteiner og polypeptider vil som oftest ha molekylvekter mellom 4.000 og 10.000.000, fortrinnsvis større enn 15.000,
og vanligvis større enn 50.000. Generelt vil proteiner tatt fra en dyreart være immunogene når de innføres i blod-strømmen til en annen art. Særlig nyttige proteiner er albuminer, globuliner, enzymer, hemocyaniner, gluteliner, proteiner med bestemte ikke-proteinbestanddeler, f.eks. glykoproteiner og lignende. Albuminene og globulinene med molekylvekt mellom 30.000 og 200.000 er særlig fore-trukket. Ytterligere henvisninger med hensyn til tek-nikkens stand når det gjelder alminnelig kjente immuno-
gene bærermaterialer og teknikker for å koble haptener til disse, kan gjøres til de følgende litteratursteder: Parker, Radioimmunoassay of Biologically Active Compounds, Prentice-Hall (Englewood Cliffs, New Jersey USA, 1976); Butler, J. Fremstilling av bestemte antistoffer ved å bruke de foreliggende immunogenkonjugater kan følge enhver alminnelig kjent teknikk. En lang rekke litteratursteder som beskriver de grunnleggende trekkene ved å indusere antistoff dannelse , er tilgjengelige, f.eks. kan det henvises til Parker, Radioimmunoassay of Biologically Active Compounds, Prentice-Hall (Englewood Cliffs, New Jersey USA, 1976). Vanligvis injiseres et vertsdyr slik som en kanin, gjet,
mus, marsvin eller hest på ett eller flere forskjellige steder med immunogenkonjugatet, vanligvis i blanding med et hjelpestoff. Ytterligere injeksjoner gjøres på det samme sted eller andre steder etter regelmessige eller uregelmessige tidsintervaller, idet det tas blodprøver for å fastslå antistoffnivået inntil det er bestemt at optimalt nivå er nådd. Vertsdyret tappes for blod inntil man har fått et passende volum med spesifikt antiserum. Når det
er ønskelig, kan rensningstrinn utføres for å fjerne uønsket materiale, slik som ikke-spesifikke antistoffer før antiserumet ansees som egnet for bruk til å utføre aktuelle analyser.
Antistoffene kan også erholdes ved hjelp av somatiske cellehybridiseringsteknikker, idet slike antistoffer vanligvis refereres til som monoklonale antistoffer. Fremstilling av N-aminoalkyl-jodtyroninderivater og deres 5i3SYie2£ere Reaksjonsrekkefølgen for fremstillingen av disse jodtyroninderivater er vist i diagrammet på tegningsfigurene 1
og la.
4- N-( trifluoracetyl) aminobutyraldehyddietylacetal ( 2), n=4 Til en blanding av 17,74 gram 90% 4-aminobutyraldehyddietylacetal (1) [0,1 mol] og 22,1 ml trietylamin (0,16 mol) i 100 ml tørr etanol ved 0°C under argongass ble det dråpevis tilsatt 21,3 g etyltrifluoracetat (0,16 mol) i løpet av 15 minutter. Blandingen ble hensatt over natten for opp-varming til værelsetemperatur. Reaksjonsvolumet ble konsentrert under vakuum (badtemperatur <40°C), oppløst i eter og vasket med vann og saltvannsoppløsning. Tørking (Na2S0^), filtrering og inndampning av oppløsningsmiddel under vakuum ga 25,3 g av en brun olje. Fraksjons-destillering ved 104-105°C (0,01 mm Hg) ga produktet (2), n=4, i form av en farveløs olje (96%, 24,6 g).

4-N-(trifluoracetyl)aminobutyraldehyddietylacetal (2), n=4[3,097 g, 12 millimol (mmol)] ble omrørt i en 36 ml blanding av tetrahydrofuran/eddiksyre/vann (1:1:1) i 48 timer under argongass. Blandingen ble inndampet under vakuum til en olje som ble tilsatt til 10,09 g (12 mmol)
1 2 tyroksinetylesterhydroklorid (3), 0 =0 =1, i 275 ml absolutt etanol under argongass. Natriumcyanborhydrid [422 mg, 6,8 mmol] ble tilsatt til blandingen som ble
omrørt ved værelsetemperatur i 24 timer. Blandingen ble filtrert og oppkonsentrert under vakuum til et skum som ble oppløst i 40 ml 20% etylacetat-diklormetan, filtrert og hensatt over natten. Det fremkom hvite krystaller av produktet som ble samlet opp, vasket med diklormetan og tørket, hvorved man fikk 4,32 g (37%), smeltepunkt 202-204°C.

Modervæsken ble oppkonsentrert under vakuum, oppløst i
10 ml etylacetat og kromatografert på Waters Prep 500 HPLC (Waters Associates, Inc., Milford, MA) ved å bruke en Prep-Pak-500 5,7 ID x 30 cm silikagelpatron (Waters Associates, Inc., Milford, MA) som en stasjonær fase og 20% etylacetat-diklormetan som den mobile fase.
Fra kolonnen ble det først eluert en olje som ble utfelt som et hvitt pulver (1,1 g) fra diklormetan-heksan, smeltepunkt 64-68°C med dekomponering. Den ble identifi-sert som N,N-bis-[4-N-(trifluoracetyl)aminobutyl]tyroksin etylester på grunnlag av de følgende analyttiske data (7% utbytte).

n=4, 3 =3 =1, (2,5 g, 2,6 mmol) ble oppløst i 100 ml vannfri etanol. Oppløsningen ble oppvarmet til tilbake-løspkoking mens den ble behandlet med saltsyre i gassform 1 7 timer. Blandingen ble oppkonsentrert under vakuum og resten utfelt to ganger fra etanol-eter hvorved man fikk 2,115 g av et hvitaktig pulver (79% utbytte) som ved analyse ble funnet å være bishydrokloriddietanolatet (5), n=4, 3<1>=3<2>=I.

N-( 4- aminobutyl) tyroksin ( 6), n=4, 3 1 =3 2=1•
N-[4-N-(trifluoracetyl)aminobutyl]tyroksinetylester (4), n=4,3 1 =3 2=1, 2,0 g (2,06 mmol) ble oppløst i 20 ml tetra-hydrofuran under argongass og behandlet med 4,1 ml av en 2 N natriumhydroksydoppløsning. Blandingen ble omrørt over natten ved 50°C. Reaksjonsblandingen ble så forhånds-adsorbert på en liten mengde med Silicar CC-7 (kiselsyre) og tilført toppen av en kolonne med 175 g Silicar CC-7 R pakket med den nedre fasen av en 2:1:1 kloroform-metanol-ammoniumhydroksydblanding. Kolonnen ble i rekkefølge eluert med 3 liter av- hver av de nedre fasene av en 2:1:1 og 1:1:1 kloroform-metanol-ammoniumhydroksydblanding. Det hvitaktige pulveret som ble erholdt, ble oppløst i etanol og 5 ml 2N natriumhydroksyd, behandlet med Norit p, filtrert gjennom celitt, og ble utfelt med eddiksyre.
Det ble erholdt 1,5 g (88% utbytte) av konjugatet (6),

Hydrokloridet ble omrørt ved å omrøre 100 mg av produktet
i 6 ml 6N saltsyre i 2 timer ved tilbakeløpskoking under argongass. Filtrering og tørking ved 55°C (0,1 mm Hg) i 17 timer ga 82 mg av produktet (79% utbytte).

Eksempel 2
Fremstilling av antistoffer mot tyroksin ved å bruke bis-i5?i^at_sammenkoblet_immuno2en 83 mg N-(4-aminobutyl)tyroksin (eksempel 1) ble tilsatt til 3,0 ml 0,1 M natriumkarbonat og 35^,ul 0,1 N natriumhydroksyd ble tilsatt. Dimetyladipimidatdihydroklorid (90 mg) ble tilsatt og en utfelling dannet. Utfellingen oppløstes etter tilsats av 70^ul 10 N natriumhydroksyd. Da denne reaksjonsblandingen hadde stått ved værelsetemperatur i 3 til 5 minutter, ble den tilsatt dråpevis til en omrørt oppløsning som inneholdt 200 mg serumalbumin fra okse i
50 ml 0,2N natriumhydroksyd. Denne blandingen ble hensatt ved værelsetemperatur i 2,5 timer og deretter ble pH justert til 7,0 med 5 N saltsyre. Utfellingen som fremkom ble fjernet ved sentrifugering og supernatanten ble
konsentrert til 5-7 ml ved trykk dialyse.
Konsentratet ble kromatografert på en 2,5 x 45 cm kolonne med Sephadex G 25 (grov) ekvilibrert med 0,1 M natriumfosfat, pH 7,0, inneholdende 0,02% natriumacid. Ca. 12 ml fraksjoner ble samlet opp og fraksjonene 9-12 ble slått sammen.
To tiendedels milliliter av væsken ble fortynnet i 0,8 ml av 0,1 N natriumhydroksyd og det optiske absorbsjons-spektrum fra 260 til 360 nanometere ble målt. Et absorb-sjonsmaksimum fremkom ved 328 nm. Absorbansene ved 280 og 328 nm ble brukt til å beregne en innlemming av 6,8 mol N-(4-aminobutyl)tyroksin pr. mol serum albumin fra okse.
Væsken med konjugatet av N-(4-aminobutyl)tyroksin og serum albumin fra okse ble fortynnet til 0,6 mg konjugat pr. ml. For bruk ved den første immunisering, ble denne oppløsning blandet med et like stort volum Freud<1>s fullstendige hjelpestoffer. 1/10 ml av blandingen ble injisert subkutant i hver pote til en kanin og 0,6 ml ble injisert subkutant i ryggen til kaninen. Tilleggsimmuniseringer ble admini-strert til ryggen 3, 7, 11 og 15 uker senere. Ved disse immuniseringer ble det brukt Freunds ufullstendige hjelpestoffer. Blod ble tappet i den 16. uke og serum ble samlet opp ved sentrifugering.
Ved å kombinere reagenser i den rekkefølge som er gjengitt
i tabell A, ble det fremstilt antistoffbindende reaksjonsblandinger for titrering av tyroksinantistoffene. Det 125I-merkede tyroksin ga ca. 630.000 tellinger pr. minutt (cpm) pr. ml når det ble målt på et "Gammacord" krystall-scintillasjonsinstrument. Alle reagensene ble fremstilt i 0,1 M natriumsfosfatbuffer, pH 7,0. Antistoffet ble inku-125
bert med I-tyroksinet i 2 til 3 timer ved værelsetemperatur før 50% polyetylenglykol ble tilsatt. Etterat poly-etylenglykolet var tilsatt og iblandet, ble uoppløselige
proteiner sedimentert ved sentrifugering og supernatanten ble dekantert. Radioaktiviteten i pelleten ble målt.
Eksempel 3
5ådioa^tiy_immun^lo2isk_analYse_ay_ tyroksin Reaksjonsblandinger for konkurrerende binding ble fremstilt med forskjellige nivåer av tyroksin ved å kombinere reagenser i den rekkefølge som er angitt i tabell B (det anvendte antiserum var frembragt mot det bis-imidatsammenkoblede immunogenet ifølge eksempel 2).

Oppløsningen av I-tyroksin ga ca. 450.000 tellinger pr. minutt (cpm) pr. ml. Reaksjonsblandingene ble inkubert ved værelsetemperatur i 2 til 3 timer før 50% polyetylenglykol-oppløsningen ble tilsatt. De uoppløselige proteinene ble samlet opp som redegjort for ovenfor, og radioaktiviteten ble målt.
Resultatene viser at ettersom nivået av tyroksin økte, avtok mengden <125>I-tyroksin bundet til antistoff.
Eksempel 4
Immunologisk analyse av tyroksin uten bruk av radioaktive isotoper
Reaksjonsblandinger ble fremstilt ved å kombinere NADPH, buffer, tyroksinantistoff (eksempel 2) og forskjellige nivåer av tyroksin i de mengder og konsentrasjoner som er angitt i tabell C, og inkubert ved værelsetemperatur i minst 30 sekunder. Deretter ble 100^,ul av 0,16 ^,uM metotreksat-aminobutyltyroksin konjugat [fremstilt fra N-(4-aminobutyl)tyroksinetylester-bis-hydroklorid (5),
n=4, P 1 =0 2=1, se ovenfor, som beskrevet i søkerens U.S. patentsøknad nr. 318.028, tilsatt og blandingen ble inkubert ved værelsetemperatur i 2 minutter. Etter den andre inku-beringen ble dihydrofolatreduktase tilsatt og blandingen ble inkubert i ytterligere 5 minutter ved 37°C. Reaksjonen ble startet opp ved å tilsette dihydrofolat og en start-absorbans ved 340 nm ble målt. Reaksjonen ble fortsatt i 20 minutter ved 37°C og en sluttavlesing av absorbancen ble målt. Resultatene i tabell C viser at enzymaktiviteten (AA) avtok etter hvert som tyroksinnivået økte.
Eksempel 5
Fremstilling av antistoffer mot tyroksin ved å bruke direkte sammenkgblet_immunogen
Et immunogen ble fremstilt ved å sammenkoble den primære aminogruppen i N-(4-aminobutyl)tyroksin til karboksylgruppene av serumalbumin fra okse. Proteinet (151 mg oppløst i 3 ml vann) ble avkjølt i et isbad. Etylesteren av N-(4-aminobutyl) tyroksin (39 mg) ble oppløst i 5 ml vann og tilsatt til proteinoppløsningen. pH-en var 4,3. l-etyl-3-(3-dimetyl-aminopropyl)-karbodiimid (202 mg oppløst i 2 ml vann) ble avkjølt og tilsatt dråpevis til albumin, N-(4-aminobytyl)-tyroksinoppløsningen. pH var 5,1 og ble justert til 4,6 med 0,1 M HC1.
Reaksjonen ble fortsatt i 5 timer ved 4°C. Deretter ble reaksjonsblandingen overført til en 3 x 50 cm kolonne med Sephadex G-50 (fin) ekvilibrert med 50 mM natriumacetatbuffer, pH 5,0. 4 ml fraksjoner ble fanget opp og den først eluerte topp av materialet med absorbanc ved-'.'280 nm ble slått sammen og dialysert mot 0,15 M natriumklorid.
Dette immunogen hadde 6,0 mol tyroksinrester pr. mol serumalbumin fra okse.
Esterfunksjonen i tyroksinrestene ble hydrolysert med alkali.
25 ml av det dialyserte immunogen ble kombinert med 0,25 ml
1 M natriumhydroksyd og man fikk en pH på 12. Denne blanding ble hensatt ved værelsetemperatur i 18 timer. Deretter ble oppløsningen justert til pH 7,5 med 1 M HC1. 4 ml immunogen (1 mg/ml) ble kombinert med 10 ml Freuds fullstendige hjelpestoffer og 2 ml saltvannsoppløsning. Kani-ner ble immunisert subkutant med 2 ml av denne blanding. 3 uker senere ble de på nytt immunisert med den samme blandingen fremstilt med ufullstendig Freuds hjelpestoffer.
Tilleggsimmuniseringene ble gjentatt hver fjerde uke. Blodprøver ble tatt en uke etter tilleggsimmuniseringene. Antiserum med passende titere ble holdt 4 måneder etter
den første immuniseringen.
Eksempel 6

De følgende reagenser ble fremstilt:
Buffer: 0,1 M natriumfosfat, pH 7,0, inneholdende 0,1%
(vekt/volum) serumalbumin fra okse og 0,1%
(vekt/volum) natriumacid.
Tyroksin: 200 ng tyroksin/ml av 0,1 M natriumfosfatbufferen,
pH 7,0.
Antiserum mot tyroksin: Antiserumet (fra eksempel 5) ble fortynnet i 0,1 M natriumfosfatbuffer, pH 7,0, slik at 20 ml aliquoter tilsatt under analysen
ga antiserumnivåer som angitt i tabellen nedenfor. Sammensatt reagens: 0,1 M natriumfosfatbuffer, pH 7,0,
inneholdende 0,1% (vekt/volum) natriumacid,
0,125 M glukose, 2,5 mM 2-hydroksy-3,5-diklor-benzensulfonat, 1,25% (vekt/volum) serumalbumin
fra okse og 23,8^,ug pepperrotperoksydase/ml. Apoglukoseoksydasereagens: 0,1 M natriumfosfatbuffer,
pH 7,0, 6,0^uM apoglukoseoksydase-bindende steder (se US patentskrift nr. 4.268.631), 12 mM 4-amino-antipyrin, 0,1% (vekt/volum) natriumacid, 400^ul antiserum mot glukoseoksydase/ml og 30%(volum /volum)
glycerol.
FAD-tyroksin: 0,1 M natriumfosfatbuffer, pH 7,0, inneholdende 40 nM flavinadenindinukleotidkonjugat. Konjugatet ble fremstilt som beskrevet i U.S. patent nr. 4.171.432.
To serier med cuvetter (12 i hver) ble betegnet A og B.
Buffer (0,23 ml) ble tilsatt til settene A og B. 50 mikro-liter av tyroksinreagensen ble tilsatt til hver cuvette i sett B.
Antiserum ble tilsatt til cuvettene i hvert sett slik at nivåene som er angitt i tabellen nedenunder ble nådd. Kuvettene ble hensatt ved omgivelsestemperatur i minst
5 minutter.
En og seks tiendedel milliliter av det sammensatte reagens ble tilsatt til hver cuvette. Deretter ble 50^ul av FAD-tyroksin tilsatt og umiddelbart etterfulgt av 50^ul Apoglukoseoksydasereagens. Reaksjonsblandingene ble inkubert ved 37°C i 15 minutter og deretter ble absorbansen ved 520 nm målt.
Man fikk de følgende resultater:
Resultatene fra sett A viser at glukoseoksydaseaktiviteten avtok etterhvert som antistoffnivået økte, hvilket indikerer at antistoffet inaktiverte flavinadenindinukleotid-tyroksinkonjugatet. I nærvær av tyroksin, sett B, var absorbansene høyere, hvilket indikerer at tyroksin og konjugatet konkurrerte om antistoffbindings steder.
