Fremgangsmåte for fremstilling av antelmintisk virksomme benzimidazolderivater.
Nærværende oppfinnelse vedrører en fremgangsmåte for fremstilling av en antelmintisk virksom forbindelse med formel
hvor er en rettkjedet eller forgrenet lavere alkyl med 1-6 karbonatomer og Y er rettkjedet eller forgrenet lavere alkyl eller alkoksy med 3-6 karbonatomer.
De mest fordelaktige forbindelser er de med formel I, hvor Y er 5-lavere alkyl med 1-6 karbonatomer, 5-lavere alkoksy med 2-6 karbonatomer eller 5-lavere alkyl-tio, og Rj er lavere alkyl med 1-6 karbonatomer.
En hittil ukjent forbindelse med formel I med eksepsjonell virkningsgrad er 5(6)-n-butyl-2-karbometoksyaminobenzimidazol, som oppviser utmerket aktivitet overfor springorm hos mus i en dose på 10 mg pr. kg, overfor viktige fårenematoder i en dose på 5 mg pr. kg og overfor vandrende stadier av Ascaris suum hos mus og svin i en dose på 0, 1% og mindre av føden.
Andre forbindelser med utmerket virkningsgrad er 5-n-amyl-2-karbometoksy-aminobenzimidazol, 2-karbometoksyamino-5-n-propyl-benzimidazol, 5-n-butoksy-2-karbometoksyaminobenzimidazol og 5-n-propoksy-2-karbometoksyaminobenzimidazol.
Fremgangsmåten ifølge oppfinnelsen karakteriseres ved at et S-lavere alkylpseudotiourinstoffsulfat i nærvær av en base behandles med en eller to ekvivalenter av en forbindelse med formelen

Forbindelsene med formel I, hvor Y er lavere alkyl, kan fremstilles ved å gå ut fra den ønskede lavere alkylbenzol. F.eks. nitreres en alkylbenzol i nærvær av eddiksyre eller svovelsyre til dannelse av o- og p-nitroalkylbenzol. Det fraskilte
para-mellomprodukt reduseres med tinnklorid til dannelse av den tilsvarende p-aminoalkylbenzol etterfulgt av nitrering i mineralsurt medium med amylnitrat eller konsentrert salpetersyre til dannelse av en o-nitro-p-alkylanilin. Sistnevnte mellomprodukt reduseres igjen med tinnklorid til dannelse av et 4-lavere alkyl substituert o-fenylendiamin. Diaminet omdannes ved den ovenfor omtalte thiourinstoffsulfatprosess til den ønskede 5-lavere alkyl substituerte benzimidazol-2-carbaminsyreester.
Ved å gå ut fra den fraskilte o-nitrobenzol og foreta reduksjon og ytterligere nitrering etterfulgt av reduksjon fås på lignende måte 3-lavere alkyl substituert o-fenylendiamin, som omdannes til den tilsvarende 4-substituerte benzimidazol.
Forbindelsene med formel I, hvor Y er dialkylamino, kan fremstilles ved å gå ut fra en dialkylaminobenzol og følge den ovenfor beskrevne rekkefølge av trinn til dannelse av den dialkylaminosubstituerte benzimidazol-2-carbaminsyreester.
Forbindelsene med formel I, hvor Y er alkylamino, fremstilles ved å gå ut fra 3 ,4-dinitroanilin. Dinitroforbindelsen behandles med f .eks. butyrylklorid til dannelse av l-butyramido-3 ,4-dinitrobenzol, som reduseres med lithiumaluminium-hydrid til dannelse av l-butylamino-3 ,4-diaminobenzol. Triaminobenzolen omdannes til den tilsvarende benzimidazol-2-carbaminsyreester ved den ovenfor omtalte thiourinstoffmetode.
Forbindelsene med formel I, hvor Y er alkoxy, fremstilles ved å gå ut fra 4-hydroxyacetanilid. Anilidet behandles med detønskede alkylbromid og et alkalimetallhydroxyd til dannelse av det tilsvarende p-alkoxyacetanilid etter fremgangsmåten ifølge Buu-Hoi m.fl. , J. Chem. Soc. , 1955, 1573.
Den substituerte forbindelse nitreres med rød rykende salpetersyre, mens den er suspendert i iseddik og eddiksyreanhydrid ved ca. 0°C. En modifisert fremgangsmåte ved høyere temperatur (60°C) innebærer anvendelse av konsentrert salpetersyre og vandig eddiksyre. Det fremkomne o-nitro-p-alkoxyacetanilid oppsamles og omkrystalliseres fra methanol. Dette disubstituerte acetanilid blir så de-acylert ved oppvarmning under tilbakeløpsavkjøling med et alkalimetallhydroxyd i ethanol, idet det disubstituerte anilin utvinnes av ansyret vann. Det disubstituerte anilin hydreres så ved 3 ,50 - 5 ,60 kg/cm^ i benzol ved fjerning av oppløsningsmidlet ved destillasjon, hvilket gir det tilsvarende diamin. Dette diamin omdannes ved thio-urinstoffsulfatprosessen til den ønskede lavere alkoxysubstituerte benzimidazol-2-carbaminsyreester.
Forbindelsene med formel I, hvor den ene eller begge av Y og Z er lavere alkyl, kan fremstilles ved å gå ut fra den ønskede mono- eller di-lavere alkylbenzol. F.eks. nitreres en alkylbenzol i nærvær av eddiksyre til dannelse av p-nitroalkylbenzol. Dette mellomprodukt reduseres med tinnklorid til dannelse av den tilsvarende p-aminoalkylbenzol etterfulgt av nitrering i mineral surt medium med amylnitrat til dannelse av et o-nitro-p-alkylanilin. Sistnevnte reduseres igjen med tinnklorid til dannelse av lavere alkyl substituert o-fenylendiamin. Diaminet omdannes ved den ovenfor omtalte thiourinstoffsulfatprosess til denønskede lavere alkyl substituerte benzimidazol- 2- karbaminsyr eester.
Forbindelsene med formel I, hvor Y og Z er alkoxy, fremstilles ved å gå ut fra o-dihydroxybenzol. Benzolen behandles med detønskede alkylbromid og et alkalimetallhydroxyd i ethanol til dannelse av den tilsvarende o-dialkoxybenzol. Den substituerte forbindelse nitreres med salpetersyre, mens den er suspendert i eddiksyre til dannelse av 1,2-dialkoxy-4,5-dinitrobenzol (J. Proe. Roy. Soc. , N.S. Wales, 71, 103 - 11 (1938)) etterfulgt av hydrering til dannelse av det tilsvarende substituerte diamin. Diaminet omdannes som tidligere beskrevet til en 5 ,6-dialkoxy-benzimidazol- 2- carbaminsyreester.
Benzimidazolylcarbamatene med formel I har vist seg å ha nyttige anthelmin-tiske egenskaper, d.v.s. en bredspektret aktivitet overfor parasitter hos varmblodige dyr, deri innbefattet både utviklede og uutviklede parasitiske former. I særdeles-het har disse forbindelser vist seg å utvise en kraftig virkning overfor forskjellige helmintiske infeksjoner i tarmområdet hos dyr av økonomisk betydning og samtidig en lav systemisk toksisitet overfor vertsdyret.
Dyr med lav vekt behandles med enhetsdoser, som ikke ligger høyere enn noen få milligram, derimot dyr med høy legemsvekt, slik som drøvtyggere, krever forholdsvis større enhetsdoser, som går opp til flere gram. Fortrinnsvis administreres en enkelt dose daglig til hver dyreart beregnet på vekten av arten.
Mengden av den administrerende bestanddel avhenger av vekten av vertsdyret, men vil i regelen være mellom ca. 1 mg pr. kg og 500 mg pr. kg vekt daglig.
Overfor lam, som var naturlig infiserte med forskjellige gastrointestinale nematoder, ble tre av forbindelsene med formel I , hver spesielt avprøvet i de an-førte doser i mg pr. kg legemsvekt (B. W) i en enkelt dose på 1% i ledningsvann med de resultater, som er vist i nedenstående tabell.
De således fremstilte preparater administreres i regelen oralt til en infisert vert 1 til 5 ganger daglig for å gi en anthelmintisk virkning.
De følgende eksempler illustrerer synteser, som kan anvendes til fremstilling av preparater ifølge oppfinnelsen.
EKSEMPEL 1.
Fremstilling av 5- n- butyl- 2- carbomethoxyaminobenzimidenazol.
Under anvendelse av en 1000 ml trehalset rundbunnet kolbe utstyrt med meka-nisk omrører, skilletrakt og termometer omrøres 13 ,9 g (0,05 mol) 2-methyl-2-thio-pseudourinstoffsulfat i ca. 10 ml vann i et isbad. 9 ,45 g methylklorformiat (0 ,1 mol) tilsettes på en gang. Blandingen omrøres ved 0°C i 10 minutter, deretter tilsettes i alt 19 ml 25% natriumhydroxyd i løpet av 10 minutter, mens temperaturen holdes under 20°C.
På dette punkt er pH-verdien ca. 8 og forblir på denne verdi etter 5 minutters omrøring. Det tilsettes 10 ml eddiksyre, hvilken gir en pH-verdi på ca. 5, og temperaturen holdes på ca. 20°C,
En oppløsning av 4-n-butyl-o-fenylendiamin i ethanol fremstilt av 11,9 g (0,05 mol) hydroklorid tilsettes reaksjonsblandingen.
Skilletrakten erstattes med et avkjølingsrør, som er forbundet med tre låser for methylmerkaptan, hvorav den ene er tom, og to inneholder 10% vandig NaOH.
Det tilføres meget langsomt varme til den omrørte blanding, og gass utvikles kon-stant. Etterhånden som temperaturen langsomt stiger, tilsettes ethanol for å regu-lere skumningen. Den samlede tilbakeløpsavkjølingstid er ca. 3/4 time. Reaksjonsblandingen avkjøles til værelsetemperatur og står til henstand weekenden over.
Det oppsamles et lysebrunt fast stoff, som vaskes med vann, og derpå suspen-deres i 50% vandig ethanol og oppsamles. Det faste stoff tørkes på en porøs plate i en ovn.
Produktet omkrystalliseres fra 1200 til 1400 ml 30-ethanol pluss 150 ml vann og står til henstand i kjøleskap natten over. Produktet oppsamles og vaskes to ganger med 20% vandig ethanol.
Produktet omkrystalliseres så igjen med 20% vandig ethanol. Det tørkes i vakuum over PgOg til dannelse av det rene produkt, smeltepunkt 225 til 227°C under dekom pon e rin g.
Strukturen bekreftes ved elementæranalyse og spektraldata.
EKSEMPEL 2.
Fremstilling av 2- carbomethoxyamino- 5- n- propylbenzimidazol.
Mellomproduktet 4-n-propylnitrobenzol omdannes til 4-n-propylanilin ved behandling med stanno-klorid dihydrat i alkoholisk medium. 2-nitro-4-propylanilin fremstilles av foregående forbindelse ved behandling med tertiær butanol og amylnitrat. Foregående 2-nitro-forbindelse reduseres med stanno-klorid i alkoholisk medium til dannelse av 4-n-propyl-o-fenylendiamin.
Under anvendelse av fremgangsmåten, som er beskrevet i eksempel 1, blir 8 ,2 g 4-n-propyl-o-fenylendiamin, 7 ,8 g methylklorformiat og 15 ,2 g 2-methyl-2-thiopseudourinstoffsulfat bragt til å reagere i vandig medium til dannelse av 10 ,25 g rått produkt, smeltepunkt 234 til 235°C. To omkrystallisasjoner av 85% vandig ethanol gir urene hvite krystaller, som tørkes i vakuum over P^CX til dannelse av 7,7 g rent produkt, smeltepunkt 239 - 240,5 oC under dekomponering, hvis struktur ble bekreftet ved elementæranalyse og spektraldata.
EKSEMPEL 3.
Fremstilling av 5- n- amyl- 2- carbomethoxyaminobenzimidazol.
Under anvendelse av fremgangsmåten i eksempel 2 fremstilles 4-n-amyl-o-fenylendiamin ut fra 4-n-amylanilin ved nitrering og reduksjon til diaminet.
12,0 g 2~methyl-2-thiopseudourinstoffsulfat bringes til å reagere med 6,15 g methylklorformiat og 7 ,7 g 4-n-amyl-o-fenylendiamin til dannelse av 8 ,5 g rått produkt, smeltepunkt 215 - 2i8°C under dekomponering.
To omkrystallisasjoner av 85% vandig ethanol etterfulgt av tørking i vakuum over P C0 i 0 O,. gir 7,0 g rent produkt, smeltepunkt 228 til 230°C under dekomponering, hvis struktur blir bekreftet ved elementæranalyse og spaktraldata.
EKSEMPEL 4.
Fremstilling av 2- carbomethoxyamino- 5- n- hexylbenzimidazol.
Under anvendelse av fremgangsmåten, som er beskrevet i de foregående eksempler, fremstilles 4-n-hexyl-o-fenylendiamin ut fra n-hexylbenzol, først ved nitrering i rekkefølge, derpå reduksjon til dannelse av 4-n-hexylanilin. Langsom addisjon til det substituerte anilin av eddiksyreanhydrid etterfulgt av 70% HNO^gir 2-nitro-4-n-hexylacetanilid.
Det fraskilte faste anilid (110 g) anbringes i 250 cm o konsentrert saltsyre etterfulgt av tilsetning av 480 cm^ ethanol og derpå av behandling med 480 cm^ stanno-klorid-dihydrat i 604 cm konsentrert HC1. Reaksjonsblandingen nøytraliseres og ekstraheres med ether, hvilket gir det ønskede 4-n-hexyl-o-fenylendiamin.
Til en vandig oppløsning av 36,3 g urinstoffsulfat tilsettes 18 ,7 g methylklorformiat etterfulgt av tilsetning av 26,1 g av det i det foregående fremstilte diamin i vandig ethanoloppløsning, hvilket gir 22,0 g av et rått produkt.
To omkrystallisasjoner av 85% vandig ethanol gir et renset produkt av lyse-brune små plater (12 ,5 g) med smeltepunkt 221,5 til 223 ,0°C under dekomponering, hvis struktur blir bekreftet ved elementæranalyse og spektraldata.
EKSEMPEL 5.
Fremstilling av 5- n- propoxy- 2- carbomethoxyaminobenzimidazol.
4-propoxy-l ,2-diaminobenzol fremstilles på følgende måte:
151 g 4-n-hydroxyacetanilid, 750 ml 2B ethanol og 56 g kaliumhydroxyd sam-menblandes, hvoretter det på en gang tilsettes 125 g n-propylbromid, mens reaksjonsblandingen holdes under en nitrogenatmosfære. Blandingen oppvarmes under tilbake-løpskjøling i 3 timer, avkjøles så til 0°C, hvorpå det tilsettes 1 liter vann og et fløte-farget rått produkt oppsamles og lutttørkes. Etter tørking i vakuum ved værelsetemperatur fås en tørr 4-propoxyforbindelse, smeltepunkt 118 - 120°C.
4-propoxy-2-nitroacetanilid fremstilles av ovenstående mellomprodukt (46,3 g) ved blanding og oppvarmning på dampbad med 60 ml iseddik, 20 ml eddiksyreanhydrid
og 50 ml vann. Omrøringen fortsettes ved værelsetemperatur, inntil krystaller be-gynner å danne seg ved ca. 40°C. Deretter tilsettes 20 ml konsentrert salpetersyre, og temperaturen stiger og holdes ved ca. 60 til 65°C i 10 minutter ved hjelp av et isbad. Det avkjøles langsomt over en periode på 15 minutter til 25°C under dannelse av et gult utfelt produkt ved ca. 35°G , som oppsamles og tørkes i vakuum ved værelsetemperatur. 2-nitroacetanilid, smeltepunkt 88 til 91°C , omkrystalliseres fra 50% ethanol.
Acetanilidet deacyleres til dannelse av det tilsvarende anilin med Claisen's alkali (Org. Syn. Col. , bind III, side 663). Reaksjonen utføres under en nitrogenatmosfære, mens det oppvarmes under tilbakeløpskjøling i 20 minutter. Etter av-sluttet tilbakeløpsavkjølingsperiode avkjøles den mørke oppløsning, som dannes, og det oppsamles et rødt fast produkt (4-propoxy-2-nitroanilin) , som tørkes i vakuum ved værelsetemperatur natten over.
4-propoxy-o-fenylendiamin fremstilles ved reduksjon av ovenstående nitro-anilin med tinnklorid i HC1 oppløsning. Nitroanilinet tilsettes porsjonsvis, idet temperaturen holdes ved 100 til 105°C i 30 minutter. Reaksjonsoppløsningen for-tynnes med 200 ml vann og avkjøles til 25°C og gjøres så sterkt basisk med 40% natriumhydroxyd. Det dannes et brunt fast stoff, som avkjøles og anbringes i vakuum ved værelsetemperatur natten over. Mellomproduktet gjennoppløses i ether, og det fremkomne organiske lag fraskilles og tørkes over natriumsulfat. Etter filtrering avdestilleres etheren, og derved fås det ønskede diamin.
Under anvendelse av den metode, som er beskrevet i eksempel 1, fremstilles det ønskede benzimidazolprodukt ved å behandle 55 ,7 g 2-methyl-2-thiopseudourin-stoffsulfat med 37 ,8 g methylklorformiat og 33 ,2 g av den i det foregående fremstilte substituerte o-diaminobenzol til dannelse av et rått produkt. Omkrystallisasjon fra dimethylsulfoxyd og derpå vasking med 2B ethanol gir et rent produkt, som tørkes i vakuum over P2°5 • nvilket gir 11,85 g produkt med smeltepunkt 230 til 230 ,5°C , hvis struktur blir bekreftet ved elementæranalyse og spektraldata.
EKSEMPEL 6.
Fremstilling av 5- n- butoxy- 2- carbomethoxyaminobenzimidazol.
Under anvendelse av fremgangsmåten, som er beskrevet i foregående eksempel fremstilles 4-n-butoxy-o-fenylendiamin ut fra p-hydroxyacetanilid ved alkylering, nitrering, deacylering og reduksjon til 4-n-butoxy-o-fenylendiamin. 55,7 g 2-methyl-2-thiopseudourinstof f sulfat bringes til å reagere med 37 ,8 g methylklorformiat og 36,0 g av ovenstående diamin til dannelse av det rå produkt. Det rå produkt oppløses i dimethylsulfoxyd ved 100°C , filtreres og får lov til å avkjøle til værelsetemperatur. Produktet oppsamles, vaskes med 2B ethanol og lufttørkes til dannelsé av 24 ,7 g rent produkt med smeltepunkt 224 til 225°C , hvis struktur blir bekreftet ved elementæranalyse og spektraldata.
EKSEMPEL 7.
Fremstilling av 5- substituerte 2- carbalkoxyaminobenzimidazoler.
Når følgende substituerte o-fenylendiaminer anvendes i stedet for o-fenylen-
diamin ved fremgangsmåten i nedenstående eksempel, fås de nedenfor anførte pro-
dukter .
EKSEMPEL 8.
Fremstilling av 2- substituerte aminobenzimidazoler.
Når følgende halogenformiater anvendes i stedet for methylklorformiat ved fremgangsmåten i nedenstående eksempel, fås de nedenfor anførte produkter.
Claims (2)
1. Fremgangsmåte for fremstilling av antelmintisk virksomme benzimidazolderivater med formel
hvor R^ er rettkjedet eller forgrenet lavere alkyl med 1-6 karbonatomer og Y er rettkjedet eller forgrenet lavere alkyl eller alkoksy med 3-6 karbonatomer ,karakterisert vedat et S-lavere alkylpseudotiourinstoffsulfat i nærvær av en base behandles med en eller to ekvivalenter av en forbindelse med formelen hvor R^har foran angitte betydning,
og reaksjonsproduktet kondenseres med et substituert o-fenylendiamin med formelen hvor Y har foran angitte betydning,
i nærvær av en syre.
2. Fremgangsmåte ifølge krav 1 for fremstilling av 5(6)-n-butyl-2-karbometoksy-aminobenzimidazol,karakterisert vedat 2-metyl-2-tiopseudourinstoffsulfat behandles i nærvær av en base med metylklorformiat, hvoretter det kondenseres med 4-n- butyl- o- f enylendiamin.
