Antibacteriële voersamenstellingen en werkwijze voor de bereiding daarvan.
De onderhavige uitvinding heeft betrekking op een nieuwe voersamenstelling die een antibacterieel systeem bevat naast de voedingsbestanddelen zelf in het bijzonder.heeft de onderhavige uitvinding betrekking op diervoersamensteHingen 5 die genoemd antibacterieel systeem bevatten.De onderhavige uitvinding heeft ook betrekking op de bereiding van de voer-samenstelling en op het gebruik daarvan in farmaceutische produkten die het antibacteriële systeem van de samenstelling bevatten.
10 De onderhavige uitvinding heeft betrekking op een voersamenstelling die een antibacterieel systeem bevat, waarbij genoemd systeem een antibacteriële werking uitoefent in het maagdarmkanaal.De onderhavige uitvinding heeft ook betrek king op voersamenstellingen, die een gunstige invloed op de 15 groei hebben.
Het is welbekend, dat huisdieren en in het bijzonder jonge dieren, onderhevig zijn aan ernstige maagdarm infekties - die in hoofdzaak bërusten op infekties door E.Coli en verschillende species Salmonella.Tot nu toe bekende werkwijzen 20 om deze infekties uit te sluiten behelzen daarbij ofwel de toediening van antibiotica met het risiko van de vorming van tegen antibioticum resistente bacteriesoorten, of door het uitvoeren van een vaccinatie, hetgeen tot hoge kosten leidt.
Er is ook een samenstelling bekend, die lactoperoxi-25 dase bevat, die het mogelijk maakt de maagdarm infekties te verminderen en tevens een verhoging van de gewichtstoename mogelijk maakt; er is echter vastgesteld, dat tijdens de eerste levensweken van de jonge dieren de diarree betrekkelijk vaak optreedt.
30 Er is ook voorgesteld de lactoperoxidase concentra tie te verhogen, maar zonder een aanmerkelijke vermindering van de diarree te verkrijgen.
Er bestaat derhalve een behoefte om een antibacte-rieel systeem beschikbaar te hebben, dat het mogelijk maakt 35 darminfekties, in het bijzonder gedurende de eerste levensweken van de dieren, te verminderen, terwijl het een verbetering van de gewichtstoename toelaat.

Een doel van de onderhavige uitvinding is een anti-bacterieel systeem, dat een uitwerking heeft op de maagdarm infekties.

Een ander doel van de onderhavige uitvinding is een 5 diervoersamenstelling, die genoemd antibacterieel systeem bevat.
De aanvrager heeft nu gevonden, dat het gezamenlijk gebruik van lactoperoxidase en lactoferrine een synergistische werking heeft om deze bacteriële infekties te 10 voorkomen en/of te doen verdwijnen, als beide eiwitten aanwezig zijn in het voer van de jonge dieren.
De voersamenstelling volgens de onderhavige uitvinding omvat naast de voedingsbestanddelen een antibacterieel systeem, dat gevormd wordt door tenminste lactoperoxidase en 15 lactoferrine, thiocyanaat en een voorloper.
De werkwijze volgens de onderhavige uitvinding voor het bereiden van de voersamenstelling, die een antibacterieel systeem bevat, dat in het maagdarmkanaal geaktiveerd wordt door middel van thiocyanaat samen met een voorloper, be- 20 staat uit het toevoegen van het antibacteriële systeem aan bekende voersamenstellingen, waarbij genoemd systeem gevormd wordt door tenminste lactoperoxidase en lactoferrine.
Er is gevonden, dat het gelijktijdige gebruik van lactoferrine en lactoperoxidase, gebruikt in hoeveelheden, 25 die overeenkomen met ongeveer 90 mg mengsel van beide enzymen per kg voersamenstelling, een onverwachte antibacteriële werking verschaft in vergelijking met een systeem dat ofwel lactoperoxidase alleen of lactoferrine alleen bevat.
Er is gevonden, dat het aantal diarree-gevallen on-30 geveer een derde was dan indien lactoferrine alleen gebruikt werd en ongeveer de helft was dan bij gebruik van lactoperoxidase alleen.
Er is ook gevonden, dat het gelijktijdige gebruik van lactoferrine en lactoperoxidase in voersamenstellingen 35 een gunstige invloed heeft op de gewichtstoename van de dieren.
Deze invloed schijnt onafhankelijk te zijn ..van de anti-diarree werking. Zo is er vastgesteld dat de werking belangrijker is dan die waargenomen als elk enzym alleen 40 gebruikt wordt.

De hoeveelheden lactoferrine en lactoperoxidase, die in de voersamenstellingen aanwezig zijn, kunnen binnen ruime grenzen variëren; ze hangen af van de gebruikte voersamens telling. In het algemeen wordt lactoperoxidase gebruikt 5 in een hoeveelheid gelegen tussen 2 en 10 mg/1 van het melk-surrogaat, terwijl lactoferrine gebruikt wordt in een hoeveelheid, die ligt tussen 40 en 120 mg/1 en bij voorkeur tussen 70 en 80 mg/1 van het melksurrogaat.
Het in de voersamenstellingen aanwezige lactoperoxi-10 dase moet worden geaktiveerd door middel van een systeem, dat thiocyanaat en een H202 voorloper bevat.In het algemeen wordt thiocyanaat toegevoegd in een hoeveelheid van tenminste 16 ppm, betrokken op de voersamenstelling, welke hoeveelheid berekend wordt als NaSCN.
15 De #2®2 voorl°Per wordt zodanig toegevoegd, dat hij een voldoende hoeveelheid H202 vormt om de 16 ppm NaSCN te oxideren.In het algemeen worden de H202 voorlopers, die gebruikt kunnen worden, gekozen uit de groep die het ascorbine-zuur-koper systeem, het glucose-glucoseoxidase systeem of 20 nog wateroplosbare peroxiden, zoals alkalische percarbonaten en andere analogen, bevat.
Volgens de werkwijze volgens de uitvinding wordt het thiocyanaat toegevoegd in een hoeveelheid gelegen tussen 10 en 100 mg/1 van het melksurrogaat.Thiocyanaat en H202 voor-25 loper worden toegevoegd in een zodanige hoeveelheid, dat er van 10 tot 100 ppm OSCN gevormd worden in een tijdbestek van 30 minuten.
De hoeveelheden van de hierboven vermelde verschillende bestanddelen zijn zodanig, dat het thiocyanaat gebruikt 30 wordt in een zodanige hoeveelheid, dat zijn concentratie in het darmkanaal tenminste 0,1 mmol bedraagt, de H202 voorloper wordt gebruikt in een zodanige hoeveelheid, dat zijn concentratie in het darmkanaal, uitgedrukt in H202, tenminste 0,1 mmol bedraagt, en het lactoperoxidase-lactoferrine mengsel, 35 genomen onder een gezuiverde vorm en/of de vorm van een in melk omgezet produkt, dat het mengsel bevat, wordt gebruikt in een zodanige hoeveelheid, dat zijn concentratie in het darmkanaal tenminste 45 ng/liter melksurrogaat bedraagt.
Als H202 voorloper wordt bij voorkeur het glucose-40 glucoseoxidase systeem gebruikt.Genoemd systeem wordt in de 35021 13 - 4 - voersamenstelling gebracht in een hoeveelheid van 10 mg/1. Andere H202 voorl°PersYstemen kunnen echter ook gebruikt worden, zoals natriumpercarbonaat en andere analogen.De vakman kent de hoeveelheden te gebruiken voorloper om de gewens 5 te hoeveelheden H202 te verkrijgen.
De onderhavige uitvinding heeft ook betrekking op een werkwijze voor een verbeterde bereiding van eetbaar vlees, volgens welke kalveren, varkens, schapen, konijnen, bisons, bekende voedingsbestanddelen ontvangen samen met een 10 antibacterieel systeem, dat een mengsel bevat van lactofer-rine en lactoperoxidase, thiocyanaat en een H202 voorloper, waarbij het thiocyanaat toegevoegd wordt in een zodanige hoeveelheid, dat zijn concentratie in het darmkanaal tenminste 0,1 mmol bedraagt, de H202 voorloper wordt toegevoegd in 15 een zodanige hoeveelheid, dat zijn concentratie in het darmkanaal, berekend als H202, tenminste 0,1 mmol bedraagt, het lactoferrine-lactoperoxidase mengsel wordt toegevoegd in een zodanige hoeveelheid, dat zijn concentratie in het darmkanaal tenminste 45 mg'liter melksurrogaat bedraagt.
20 Een ander doel van de onderhavige uitvinding is een antibacterieel systeem, dat een lactoferrine-lactoperoxi dase mengsel, een thiocyanaat en een Η2<02 voorloper bevat.
Een ander doel van de onderhavige uitvinding is een werkwijze voor het behandelen van maagdarm infekties bij 25 zoogdieren, met inbegrip van de mens, welke werkwijze bestaat uit het oraal toedienen van een therapeutisch werkzame hoeveelheid van het hierboven beschreven antibacteriële systeem.
Nog een ander doel van de onderhavige uitvinding 30 zijn farmaceutische preparaten die het hierboven genoemde antibacteriële systeem bevatten; genoemd systeem kan gebruikt worden op elke farmaceutisch geschikte drager.
Als voorbeelden van behandelde dieren kunnen genoemd worden vafkens, kalveren en andere huisdieren, zoals 35 katten en honden.
De benodigde hoeveelheden lactoperoxidase aktivato-ren, die gebruikt kunnen worden in het darmkanaal worden goed beschreven in Belgisch Octrooischrift nr.852.195.
In de onderhavige uitvinding*: waarin een antibacte-40 rieel systeem dat lactoferrine en lactoperoxidase gebruikt 850 21-1 0 ? - 5 - wordt gebruikt, zijn de boven genoemde hoeveelheden nog geschikt. Er zij aan herinnerd, dat in het geval van lactoper-oxidase ëën eenheid ervan de hoeveelheid lactoperoxidase voorstelt, die 1 mg pyrogalline vormt in 20 seconden, uit-5 gaande van pyrogallol, bij pH 6,0 en bij een temperatuur van 20°C.
Ook zij er aan herinnerd, dat LD^q van thiocyanaat 484 mg/kg lichaamsgewicht is bij intraveneuze injektie bij muizen, terwijl hij 764 mg/kg lichaamsgewicht is bij orale 10 toediening aan ratten.
Bij klinisch gebruik worden ge bestanddelen volgens de onderhavige uitvinding gewoonlijk oraal of rectaal toegediend in de vorm van een farmaceutisch preparaat, dat het antibacteriële systeem volgens de uitvinding bevat, afgezet 15 op een geschikte farmaceutische drager.
In het algemeen ligt de hoeveelheid aktieve bestand delen tussen 0,1 en 99 gew.% van het preparaat, en gunstig tussen 2 en 50 gew.% in preparaten, die oraal toegediend moe ten worden.
20 Bij de bereiding van de farmaceutische preparaten, die een antibacterieel systeem volgens de onderhavige uitvin ding bevatten in de vorm van doseringseenheden voor orale toediening, kunnen de bestanddelen gemengd worden met een welbekende vaste poedervormige drager, zoals lactose, saccha 25- rose, sorbital, mannital en andere analogen.Indien overtrokken tabletten gewenst worden, kan het bovenstaande geval worden overtrokken met een oplossing van een polymeer, dat oplost of permeabel is in het darmkanaal.
Doseringseenheden voor rectale toediening kunnen 30 bereid worden in de vorm van zetpillen, die de aktieve bestanddelen bevatten in eem engsel met een neutrale vetbasis of ze kunnen bereid worden in de vorm van gelatine rectale capsules, die de aktieve stof bevat vermengd met een plantaardige olie of paraffine olie.
35 Farmaceutische preparaten die een antibacterieel systeem volgens de uitvinding bevatten zijn bedoeld om gebruikt te worden bij de behandeling van bacteriële infekties in het maagdarmkanaal, veroorzaakt door Shigella, Salmonella E.Coli, Vibero Colera, Pseudomonas (Ps pyocyanae), Staphylo-40 coccus (Staph.albus, aureus), Streptococcus (Strep.viridous, 0 2110 -6-
Strep.faccalis, B-streptococcus), Proteus.
De volgende voorbeelden dienen ter nadere toelichting van de uitvinding.
Voorbeeld 1 5 Een melksurrogaat werd bereid uit de volgende be standdelen: melkpoeder : 60 kg lactoserum : 14,15 kg vet : 17 kg 10 glucose : 5 kg ethoxyquine : 0,125 kg cholinechloride s 0,025 kg mineralen en vitaminen: 2,5 kg emulgatoren.
15 De concentraties van lactoferrine en lactoperoxidase in het melkpoeder waren respektievelijk 61 g en 77 g per 100 kg poeder.
Als aktivator voor lactoperoxidase werden 12,3 g natriumthiocyanaat samen met 61 g glucoseoxidase (71.000 U/g) 20 toegevoegd.
De bestanddelen werden grondig gemengd in een menger Water werd aan het mengsel toegevoegd om een Γ3% waterige oplossing te verkrijgen.Genoemde oplossing bevatte 10 mg lactoperoxidase en 80 mg lactoferrine per liter melksurrogaat.
25 Genoemd melksurrogaat werd aan kalveren toegediend in een hoeveelheid van 3 1/dag in de eerste week, 4 1/dag de tweede week, 6 1/dag de: derde week en 8 1/dag de vierde week.
Ter vergelijking werden dezelfde hoeveelheden van verschillende mengsels aan kalveren gegeven, waarbij genoemde 30 mengsels noch lactoferrine noch lactoperoxidase (mengsel A), bevatten of alleen maar 10 mg/1 lactoperoxidase bevatten (mengsel B), of alleen maar 80 mg/1 lactoferrine bevatten (mengsel C).
De antibateriële werking werd beoordeeld door het 35 aantal gevallen van diarree te meten, terwijl de voedingswaarde bepaald werd door de gewichtstoename van de kalveren te meten.
Kalveren werden gedurende een tijdsbestek van vijf weken behandeld.

In het onderhavige geval is het synergistische effekt karakteristiek.Opgemerkt wordt dat een toename van 68% ten opzichte van de kontrole verkregen wordt, terwijl het 20 maximum dat verkregen wordt bij gebruik van de produkten alleen 57% en 32% is met respektievelijk lactoperoxidase en lactoferrine.
Voorbeeld 2
Het in Voorbeeld 1 beschreven mengsel is gebruikt en 25 toegediend aan kalveren gedurende 10 weken in een hoeveelheid van 8 1/dag vanaf de vierde week.De concentratie aan vaste stof werd geleidelijk verhoogd van 13 tot 25 gew.% tussen de 5de week en de 10de week.
Ter vergelijking werd een mengsel dat geen lactofer-30 rine of lactoperoxidase bevatte, toegediend.
In de 10de week trad geen diarree meer op met het mengsel volgens de uitvinding, terwijl dat niet het geval is met de kontrole.
Er is bovendien vastgesteld dat met het mengsel vol-35 gens de uitvinding een gewichtstoename van 865 g/dag wordt verkregen tegen slechts 755 g/dag met de kontrole.
Voorbeeld 3
Het in Voorbeeld 1 beschreven mengsel werd toegediend aan schapen in een hoeveelheid van 0,5 1/dag gedurende 40 de eerste week, 1 1/dag gedurende de tweede week en 2 1/dag daarna.Er werden dergelijke resultaten als die verkregen met $502110
Claims (13)
1. Werkwijze voor de bereiding van levensmiddelen of diervoer, dat een antibacterieel systeem bevat, dat geakti-veerd wordt in het maagdarmkanaal door middel van een thio- 5 cyanaat en een H202 voorloper, met het kenmerk, dat de werkwijze bestaat uit het toevoegen van het antibacte-riële systeem aan bekende voedingssamenstellingen, waarbij genoemd systeem tenminste lactoperoxidase en lactoferrine bevat. 10
2. Werkwijze volgens conclusie 1, m e t het kenmerk , dat een antibacterieel systeem gebruikt wordt dat is 'samengesteld uit lactoperoxidase, gebruikt in een hoeveelheid die overeenkomt met 2 tot 20 mg/1 melksurrogaat 15 en lactoferrine in een hoeveelheid die overeenkomt jnet 40 tot 120 mg/1 melkpoeder.
3. Werkwijze volgens conclusie 2,met het ken merk, dat lactoperoxidase gebruikt wordt in een hoevee1- 20 heid die overeenkomt met 8 tot 12 mg/1 melksurrogaat, en lactoferrine gebruikt wordt in een hoeveelheid die overeenkomt met 70 tot 80 mg/1 melksurrogaat.
4. Werkwijze volgens conclusie l,met het ken 25m e r k , dat de H202 voorloper.gekozen wordt uit de groep die het glucose-glucoseoxidase systeem, het ascorbinezuur-koper systeem, de alkalipercarbonateïi, de aardalkaliperoxiden, en het carbamideperoxide omvat.
5. Werkwijze volgens conclusie 1, m e t het ken merk, dat het aktivator systeem en de H202 voorloper van het antibacteriële systeem worden toegevoegd in een zodanige hoeveelheid dat van 10 tot 100 ppm OSCN gevormd worden in een tijdsbestek van 30 minuten. 35

Levensmiddel of voersamenstelling, die een bekende 8502110 - 9 - voedingssamenstelling bevat en een antibacterieel systeem, dat geaktiveerd wordt in het maagdarmkanaal door middel van een thiocyanaat en een H202 voorloper, met het kenmerk , dat het antibacteriële systeem bestaat uit 5 tenminste lactoferrine en lactoperoxidase.
7. Levensmiddel of voersamenstelling volgens conclusie 6, met het kenmerk, dat het antibacteriële systeem bestaat uit lactoperoxidase, dat gebruikt wordt in 10 een hoeveelheid van 2 tot 20 mg/1 melksurrogaat en lactoferrine, dat gebruikt wordt in een hoeveelheid van 40 tot 120 mg/1 melksurrogaat.
8. Levensmiddel of voersamenstelling volgens conclusie 15 7,met het kenmerk, dat lactoperoxidase gebruikt wordt in een hoeveelheid van 8 tot 12 mg/1 melksurrogaat en lactoferrine gebruikt wordt in een hoeveelheid van 70 tot 80 mg/1 melksurrogaat.
9. Levensmiddel of voersamenstelling volgens conclusie 8,met het kenmerk, dat het antibacteriële systeem, dat bestaat uit het mengsel van lactoferrine en lactoperoxidase, gebruikt wordt in een hoeveelheid van tenminste 45 mg/1 melksuroogaat. 25

Levensmiddel of voersamenstelling volgens conclusie 6, met het kenmerk, dat de H202 voorloper van het antibacteriële systeem gekozen wordt uit de groep die het glucose-glucoseoxidase systeem, het ascorninezuur-koper 30 sysreem, de alkalipercarbonaten, de aardalkalioxiden en de carbamideperoxiden omvat.
11. Levensmiddel of voersamenstelling volgens conclusie 10,met het kenmerk, dat het glucoseoxidase 35 wordt toegevoegd in een concentratie van 10 mg/1 melksurrogaat.
12. Antibacterieel systeem dat oraal wordt toegediend in het maagdarmkanaal, geaktiveerd door thiocyanaat en een
40 H202 voorloper, met het kenmerk, dat het lac- 8592110 Λ' - 10 - toperoxidase en lactoferrine bevat.
13. Werkwijze voor het behandelen van bacterj.ële infek-ties bij zoogdieren, met inbegrip van de mens, waarbij een 5 therapeutische hoeveelheid van een antibacterieel systeem, geaktiveerd door een thiocyanaat en een voorloper wordt toegediend, met het kenmerk, dat het systeem tenminste lactoferrine en lactoperoxidase bevat.
