Thioalkanoylcamitinen, werkwijze ter bereiding ervan en rauco-lytische farmaceutieche samenstellingen, die deze caraitinen bevatten.
De onderhavige uitvinding heeft betrekking op een nieuwe klasse thioalkanoylcamitinen met de algemene formule 1, waarin R een rechte of vertakte alkyleengroep met 2 tot 6 koolstof-atomen Is, 5 een rechte of vertakte kleine alkylgroep met 1 tot 4 koolstof- atomen is en X' een halogeenanion is.
let is duidelijk dat afhankelijk van het feit of B een rechte of vertakte groep is, de thioalkanoylgroep R^OS- in de 10 verbindingen met formule 1 hetzij eindstandig hetzij niet-eindstan-dig geplaatst is.
De verbindingen met de algemene formule 1 zijn begiftigd met een opmerkelijke mucolytische en anti-hoestactiviteit en lage toxiciteit, zoals hierna zal worden getoond. Derhalve heeft de 15 onderhavige uitvinding eveneens betrekking op farmaceutische samenstellingen, die een mucolytische en anti-hoestactiviteit bezitten, gekenmerkt door de aanwezigheid als werkzaam bestanddeel van ten minste één van de verbindingen met formule 1 tezamen met de gebruikelijke in de farmaceutische technologie toegepaste ver-20 snijdingsmiddelen.
Tenslotte heeft de uitvinding betrekking op een werkwijze ter bereiding van thioalkanoylcamitinen met formule 1.
Deze werkwijze wordt in zijn meest algemene vorm gekenmerkt door omzetting van thiozuur met de algemene formule 2, waarin de 25 hiervoor vermelde betekenis heeft, (a) wanneer in de verbindingen met formule 1 de thioalkanoylgroep R^COS- in de eindstandige positie is, met een verbinding met de algemene formule 3, waarin n een geheel getal tussen 0 en 4 is volgens het reactieschema 30 met fig. 1, of (b) wanneer in de verbindingen met formule 1 de thioalkanoylgroep R^GOS- niet in de eindstandige positie is, met een verbinding met de algemene formule 4, waarin / ."1 m en m gehele getallen tussen 0 en 3 zijn en 35 R„ een kleine alkylgroep met 1-4 koolstofatomen is volgens het 8204638 - :f r η - 2 - reactieschema met fig. 2.
De groep R wordt hij voorkeur gekozen uit de groep bestaande uit ethyleen, trimethyleen, tetramethyleen, ethyl-ethyleen, propyleen, 1-methyltrimethyleen en 2-methyltrimethyleen.
5 De groepen R^ en R^ worden bij voorkeur gekozen uit de groep bestaande uit methyl, ethyl, propyl, isopropyl en butyl.
De reactie tussen de verbindingen met formules 2 en 3 en tussen 2 en 4 heeft zeer gemakkelijk plaats. Het is voldoende de reagentia met elkaar in aanraking te brengen zelfs bij afwezigheid 10 van oplosmiddelen en suspendeermiddelen en het reactiemengsel enkele uren tot enkele dagen te roeren bij een temperatuur die ligt tussen ongeveer kamertemperatuur en 50°C. Het verdient de voorkeur een overmaat van het thiozuur met formule 2 met betrekking tot de verbinding?met formule 3 en 4 te gebruiken. Be overmaat varieert 15 van ongeveer 3*1 (molair) wanneer de reactie wordt uitgevoerd bij aanwezigheid van oplosmiddelen tot 30s1 (molair) bij afwezigheid van oplosmiddelen.
De eventuele oplosmiddelen en suspendeermiddelen en de afscheidings- en zuiveringsmethoden zijn die, die gewoonlijk bij 20 de organische syntheses worden gebruikt.
De volgende niet beperkende voorbeelden lichten de werkwijze voor de bereiding van enkele verbindingen van de uitvinding en hun chemisch-fysische eigenschappen toe.
Voorbeeld I
25 5-thioaoetylT>ropionylcamitine (S3? 405) (zie reactievergelijking mét fig. 3) (1) Bereiding van acryloylcamitine.
Aan een oplossing van 4 S broompropionylcarnitine in 40 ml water werden 100 cm IR 45 Amberlite hars geactiveerd in de 0H-30 vorm toegevoegd. Het reactiemengsel werd 2 uren geroerd, daarna gefiltreerd en gelyofiliseerd.
De aldus verkregen grondstof werd gezuiverd door chroma-tografie met silicagel gebufferd met 1,5% Ha^HPO^ en elueren met methanol. Het eluaat werd met HC1 behandeld en vervolgens gelyofili-35 seerdj 1,5 g zuiver produkt werd verkregen. Rendement 42 %.

(2) Bereiding van 3-thioacetylpropionyloarnitine.
Acryloyloaraitine inwendig zout (0,008 mol) verkregen bij de voorafgaande trap (1) werd in 40 ml absolute ethanol ge-3 suspendeerd. Aan de verkregen suspensie werd 0,0238 mol thioazijn- zuur toegevoegd. Be suspensie werd 3 uren geroerd, vervolgens met watervrije diethylether geprecipiteerd en 1 nacht in een koelkast bewaard. Een olie sloeg neer, die door decanteren werd afgescheiden en herhaalde malen werd gezuiverd door de olie in ethanol op te 10 lossen en met diethylether te precipiteren. Be aldus verkregen grondstof werd met water opgenomen, met geconcentreerd HC1 bij 0°C aangezuurd en vervolgens gelyofiliseerd. 1,9 g Frodukt werd verkregen. Rendement 75 %· MR B20 6 5,6 (1H, m, -CH-); 3,8 (21, m, Ί^^Ν-ΟΗ^ ); 15 5,3 (91, *, (0Hj)5-V ); 3.0 - 2,6 (6a, a, -CHgCOOH, -COCJ-Ogj-S-)! # 2.3 (3H, e, -C0CH3).
Voorbeeld II
5-thioacetyloentanoTrl-camitine fST 412) 20 (zie reaotieverlijking met fig. 4) (1) Bereiding van allylacetyl-camitine.
Aan een oplossing van camitinehydrochloride (9 g; 0,045 mol) in 15 om5 trifluorazijnzuur werd 0,13 mol allylacetyl-chloride toegevoegd. Be verkregen oplossing werd 4 uren op 45°C 25 gehouden. Vervolgens werd aceton aan de oplossing toegevoegd, werd niet-omgezette camitine afgescheiden en werd diethylether toegevoegd, waarbij een neerslag werd verkregen. Be aldus verkregen grondstof werd door precipitatie uit isopropanol-diethylether gezuiverd. 9,5 g Frodukt werd verkregen. Rendement 66 %.

( 4 - A - (2) Bereiding van 5-thioacetylpentanoylcarnitine.
22,4 g (0,38 mol) thioazijnzuur werden aan het allyl-acetylcarnitine (4 g; 0,013 mol) van de voorafgaande trap (1) toegevoegd. Be verkregen oplossig werd 1 nacht op 40°C gehouden.
5 Vervolgens werd diethylether aan de oplossing toegevoegd en werd het aldus gevormde neerslag door decanteren afgescheiden. Het neerslag werd in water opgelost en driemaal met diethylether geëxtraheerd. Be waterige fase werd geconcentreerd en met aceton gewassen, waarbij 3»9 g produkt verkregen werd. Rendement 84 %· 10 MR B20 £ 5,6 (1Ξ, m, -CH- ); 3,7 (2H, m, JZ^rN-CHg-); Ó + 3,2 (9H, s, (CH3)3-H-) ; 2.7 (6H, m, -CHgCOOH; OCOCHg- ; CÏÏ2S)j 2.4 (3H, s, -C0CH3); 1,6 im, m, -COCHgCHgCiyjHg S-).

3-thioacetylbutyrvlcarnitine (ST 406) (zie reactievergelijking met fig. 5) (1) Bereiding van crotonoylcarnitine.
8 cm^ (O,08 mol) Crotonoylchloride werden toegevoegd aan 20 een oplossing van carnitinehycLrochloride (8 g; 0,04 mol) in tri-fluorazijnzuur. Het reactiemengsel werd 1 nacht bij 50°C geroerd. Vervolgens werd diethylether aan het mengsel toegevoegd en ontstond een neerslag. Het neerslag werd afgefiltreerd en als zodanig bij de volgende reactie gebruikt.

30 15 cm^ (0,20 mol) Thioazijnzuur werden aan 3,5 g (0,01 mol) crotonoylcamitine van de voorafgaande trap (1) toegevoegd.
Het mengsel werd 4 dagen bij kamertemperatuur geroerd. Vervolgens werd diethylether aan het reactiemengsel toegevoegd. Een olie werd 8204638 * lr" + - 5 - . . ; * verkregen die gezuiverd werd door de olie in ethanol op te lossen en deze opnieuw met diethylether neer te slaan. Se preoipitatie werd driemaal herhaald. Vervolgens werd het neerslag in water opgelost en driemaal met diethylether geëxtraheerd. Se waterige oplos-5 sing werd gelyofiliseerd. 2,6 g Produkt werd verkregen. Rendement 70 %.

Se acute toxiciteit, de slijmlosmakende en mucolytische activiteiten en het effekt op de ciliaire beweeglijkheid van de verbindingen met de formule 1 weden onderzocht.
15 Acute toxiciteit
Be LB^q van de verbindingen met de algemene formule 1 met de methode volgens Veil bepaald ("Tables for convenient calculation of median effective dose (LB^q or EB^Q) and instructions in their use", Biometrics, 249-253, 1952) door e.p. toediening 20 bij muizen is in tabel A getoond.

Sli.imlosmakende activiteit
Be proeven werde'h uitgevoerd bij mannelijke konijnen met 30 een gewicht van 2-3 kg, geanesthetiseerd met ethylurethan volgens de methode van Perry c.s. (J.Pharm.Exp.Ter. 21» (1941), 65).
Be geanesthetiseerde dieren, met de kop ria-a-r beneden vastgemaakt op een operatietafel onder een hoek van 60° hadden een

- 6 - canule opgenomen in hun trachea. Elke canule was verbonden met een toevoer-inrichting, die een gestadige stroomsnelheid van voorverwarmde lucht (36-38°C) met een constante vochtigheid (80 %) leverde. Aan het laagste einde van elke canule was een geijkte cilinder 5 bevestigd, waarin de bronchiale secretieprodukten werden verzameld. Alle dieren ademden spontaan en zij regelden dientengevolge zelf de luchttoename die geschikt was voor een normale ademhaling. Na 1 uur volgend op de canule-opname ontvingen de dieren oraal (door een maagbuis) de verbindingen met de algemene formule 1 opgelost 10 in gedestilleerd water bij doseringen tussen 20 en 40 mg. Elke dosering van geneesmiddel werd aan 5 dieren toegediend. De controle-dieren (8) ontvingen alleen water. De hoeveelheid secretie werd na 1, 2 en 4 uren vanaf de toediening bepaald. De resultaten samengevat in tabel B laten zien, dat de verbindingen met de algemene 15 formule 1 geen slijmlosmakende activiteit uitoefenen.
Mucolytisohe activiteit
De proeven werden in vitro uitgevoerd onder toepassing van de methode beschreven door Morandini c.s. (iotta contro la tuber-colosi 47, (1977), Nr. 4) Een thrombo-elastograaf werd gebruikt 20 om de variaties teweeggebracht door de verbindingen met de algemene formule 1 en acetylcysteine op de rheologische eigenschappen van menselijk sputum te volgen. De resultaten daarvan, samengevat in tabel C, laten zien dat de verbindingen met formule 1 een grotere afname van dichtheid van menselijk sputum teweegbrachten dan die 25 isweeggebracht door acetylcysteine.
Effekt on de ciliaire activiteit
Het vermogen van de verbindingen met formule 1 de ciliaire beweeglijkheid te beïnvloeden werd bestudeerd door met de microscoop de ciliaire beweging van trachee-ringen van de rat 30 gedompeld in oplossingen van de proefverbindingen waar te nemen.
Met deze techniek is het mogelijk, in verband met de concentratie van de verbinding en de contacttijd, de ciliaire kering bewegingsblol^ veroorzaakt door de proefverbindingen te onderzoeken, die verband houdt met de slijmopruiming uit ciliair epithelium.
35 In de vorm van oplossingen toe te passen stoffen moeten de voorafgaande blokkering niet doen plaatshebben in minder dan vijftien minuten vanaf het contact.

Zoals experimenteel aangetoond modificeren de verbin-30 dingen van de onderhavige uitvinding de Theologische eigenschappen van sputum aanzienlijk. Bij het nagaan van de verkregen resultaten wordt een afname in sputumdichtheid bij de hogere doseringen (of lagere verdunningen) en bij de kleinere doseringen (of hogere ver- 8204638 4 % * - 8 - dunningen) constant hoger dan die teweeggebracht door acetylcysteine vastgesteld. Anderzijds vergroot geen van de verbindingen de bronchiale secretie en is evenmin in staat de ciliaire beweging van het epithelium van tracheeringpreparaten in tijdsintervallen 5 korter dan die toegelaten, te blokkeren.
De verbindingen van de onderhavige uitvinding zijn therapeutisch geschikt voor de behandeling van ziekten van het ademha-lingskanaal. De patiënten, die daaraan behoefte hebben, zullen oraal of parenteraal een therapeutisch werkzame hoeveelheid van een 10 verbinding met de algemene formule 1 toegediend krijgen.
De oraal of parenteraal toegediende dosering van de verbinding met de algemene formule 1 zal in het algemeen gelegen zijn tussen ongeveer 15 en ongeveer 70 mg/kg lichaamsgewicht/dag, hoewel grotere of kleinere doseringen kunnen worden toegediend 15 door de behandelende arts rekening houdende met de leeftijd, het gewicht en de algemene toestand van de patiënt, onder toepassing van een gedegen professionele beoordeling.
In de praktijk worden de verbindingen oraal of parenteraal toegediend in elk van de gebruikelijke farmaceutische vormen, 20 die bereid worden volgens gebruikelijke voor de deskundige op het gebied van de farmaceutische technologie bekende methoden. Tot deze vormen behoren vaste en vloeibare orale eenheidsdoserings-vormen zoals tabletten, capsules, oplossingen, siropen en dergelijke alsmede injecteerbare vormen, zoals steriele oplossingen voor 25 ampullen en flesjes. Hierna worden enkele niet beperkende voorbeelden van samenstellingen geschikt voor orale of parenterale toediening gegeven.

Claims (6)
1. Thioalkanoy1carnitinen met de algemene formule 1, waarin R een rechte of vertakte alkyleengroep met 2-6 koolstofatomen is, R^ een rechte of vertakte lage alkylgroep met 1-4 koolstofatomen is, 5 en X*”een halogeenanion is.

5· Werkwijze ter bereiding van de thioalkanoylcaraitinen 10 met formule 1 volgens conclusie 1, m e t het kenme rk, dat men een thiozuur met de algemene formule 2, waarin R^ de hiervoor vermelde betekenis heeft omzet (a) wanneer in de verbindingen met formule 1 de thioalkanoylgroep R^COS- in de eindstandige positie is, met een verbinding met 15 de algemene formule 3 , waarin n een geheel getal tussen 0 en 4 is volgens het reactieschema met figuur 1 of (b) wanneer in de verbindingen met formule 1 de thioalkanoylgroep R^COS- in de niet-eindstandige positie is, met een verbinding met de algemene formule 4» waarin m en m1 gehele getallen van 2 20. tot 3 zijn, R een kleine alkylgroep met 1-4 koolstofatomen is volgens de reactieverlijking met figuur 2.
