Werkwijze voor de vergisting van cellulose tot ethanol en koolzuur.
Betrekkelijk weinig anaerobe microörganismen zijn geïsoleerd en beschreven die onder thermofiele en extreem thermofiele omstandigheden nog op koolhydraten groeien ("glycoly-tisch" zijn) en daaruit ethanol maken. Voorbeelden van behoorlijk 5 beschreven glycolytische anaerobe bacteriën die onder thermofiele tot extreem thermofiele temperaturen op een geschikt kweekmedium groeien behoren alle tot het geslacht Clostridium, en daaronder Cl. thermoaceticum, Cl. tartarivorum, Cl. thermosaccharolyticum,
Cl. thermocellim, Cl. thermocellulaseum en Cl. thermohydrosulfurieum. 10 Stammen van deze laatste soort zijn geïsoleerd en beschreven door J. Wiegel en L.G. Ljungdahl (zie üitstreksel I 75 in het verslag van de Jaarvergadering van de Amerikaanse Vereniging voor Microbiologie te Las Vegas, Nevada (1978). Een nieuw type van Cl. therme-hydrosulfuricum, namelijk E 100-69, werd door F. Hollaus en U.Sleytr 15 uit het sap van een Oostenrijkse suikerfabriek geïsoleerd; zie Arch. Microbiol. 86 (1972) 129-146.
Naast deze bekende soorten zijn twee geen sporen vormende stammen geïsoleerd en beschreven, die tot een nieuwe soort en zelfs tot een nieuw geslacht gerekend moeten worden.
20 Deze nieuw ontdekte thermofiele anaeroben zijn als zuivere culture geïsoleerd en hebben de naam Thermoanaerobacter ethanolicus gekregen. Een zuivere subcultuur, aangeduid als JW 200, is onder no.
ATCC 31550 gedeponeerd bij de American Type Culture Collection te Rockville, Maryland. Tijdens het isoleren, zuiveren en volledig be-25 schrijven van deze nieuw ontdekte soort, die niet een Clostridium is, werd gevonden dat de nieuwe soort een efficiënte producent van ethanol uit diverse koolhydraten is, in het bijzonder uitgaande van de gewone mono- en disacchariden.
Zoals uit de nog volgende beschrijving blijkt 8000338 2 kan T. ethanolicus ethanol maken uit diverse koolhydraten, maar het is een kenmerk van deze anaerobe dat het geen cellulose aantast.
Het is bekend dat het in 1950 geïsoleerde 5 anaerobe microörganisme Clostridium thermocellum bij hoge temperaturen cellulose vergist tot waterstof, koolzuur, ethanol, formiaat, acetaat en lactaat, en in mindere mate tot dicarbonzuren. Zie R.H. McBee, "The anaerobic thermophilic cellulolytic bacteria" in Bacteriological Reviews (1950) 51-63. Maar nu is ook waargenomen 10 dat sommige produkten die bij de inwerking van het cellulase-sys-teem ontstaan, zoals cellobiose en glucose, de neiging hebben de groei van Cl. thermocellum te remmen, waardoor er minder cellulase ontstaat. Andere mogelijkerwijs in het vergistingsmedium aanwezige suikers zoals lactose kunnen het cellulase ook remmen. Hoewel Cl.
15 thermocellum deze suikers omzet is de groei daarop langzaam en is de opbrengst aan ethanol laag. Dus zal Cl. thermocellum in biologisch zuivere vorm, hoewel het cellulose afbreekt, de ontstaande suikers niet efficient omzetten, en in feite wordt de vergisting geremd door de ophoping van dit tussenprodukt.
20 Een nieuw geïsoleerde, voor Cl. thermocellum representatieve stam, door deuitvinders JW 20 genoemd, is onder no. ATCC 31549 gedeponeerd bij de eerder genoemde American Type Culture Collection.
Deze soort kan men natuurlijk gebruiken voor 25 de omzetting van diverse koolhydraten, waaronder cellulose, in ethanol en andere nuttige produkten, maar dat moet onder thermofiele omstandigheden (dus bij hoge temperaturen) gebeuren. De bekende vergisting van suiker tot ethanol door gist (diverse soorten Saccharomyces) moet, zoals bekend, bij temperaturen beneden 37°C 30 onder half aerobe omstandigheden gebeuren, en verder moet men zorgvuldig waken tegen besmetting met schadelijke bacteriën, schimmels en gisten.
Volgens de uitvinding vergist men cellulose doordat men Thermoanaerobacter ethanolicus en Clostridium thermo-35 cellum in gemengde culture onder anaerobe, thermofiele omstandigheden kweekt op een medium dat cellulose bevat, totdat een winbare 8000338

3 hoeveelheid ethanol ontstaan is.
In een gemengde culture vergisten deze twee soorten samen cellulose efficient tot winbare hoeveelheden ethanol.

In deze beschrijving en in de conclusies worden met "thermofiele 5 omstandigheden" het kweken bij temperaturen tussen 45° en 70°C bedoeld, en met "extreem thermofiele omstandigheden" het kweken bij temperaturen boven 70°C.
Hoewel het niet de bedoeling is de strekking van deze uitvinding door een of andere theorie te beperken zal uit 10 beschrijving en het voorbeeld blijken dat het koppelen van T.etha-nolicus en Cl. thermocellum de afbraak van cellulose aanzienlijk versterkt, de ophoping van tussenprodukten voorkomt, en de vergis-ting naar de vorming van ethanol verschuift.
Het microörganisme T. ethanolicus werd gevon-15 den in en geïsoleerduit monsters modder uit hete bronnen van het
Yellowstone National Park. Eén stam JW-201, werd uit een zure bron geïsoleerd, de Dragon Mouth, met een pH van ongeveer 5,5, en de tweede stam, JW-200, uit een alkalische bron, namelijk de White Creek, met een pH van ongeveer 8,8. De twee stammen lijken echter 20 heel veel op elkaar, en ze werden gevonden in gezelschap van de hierboven reeds genoemde thermofiele Clostridium-soorten.
Hoewel de nieuwe stammen anaëroob zijn en ook bij hoge temperaturen koolhydraten kunnen vergisten maken ze geen sporen, zodat ze niet tot het geslacht Clostridium gerekend 25 kunnen worden. Gelet op de morfologie en de metabolische eigenschappen moeten deze nieuwe stammen tot een nieuwe soort en een nieuw geslacht gerekend worden, te weten Thermo-anaerobacter ethanolicus, waarvan ATCC 31550 de type-stam is.
Het isoleren van T. ethanolicus tot een zui-30 vere stam gebeurde met de anaerobe techniek van Hungate (zie
Bacteriol.Rev. 14 (1950) 1-49) zoals gemodificeerd door Bryant and Robinson (zie J. Dairy Science 44: 1446-1456), welke techniek de deskundigen voldoende bekend zal zijn. Het medium voor het aanhouden van deze stammen heeft bij voorkeur de volgende samenstel-35 ling: KH2P04 1,5 g/1; Na2HP04.12H20 4,2 g/1; NH4C1 0,5 g/1;
MgCl2 0,18 g/1; gist-extract (Difco) 2,0 g/1; glucose 8,0 g/1; en 8000338 4 en Wolfe's mineralen-oplossing 5 ml, en dit medium werd ook gebruikt voor het isoleren en verrijken. Dit medium wordt onder anaerobe omstandigheden aangemaakt en onder een inert gas (stikstof of argon) bewaard. De pH ligt tussen 6,8 en 7,8 en is bij voorkeur 7,3; deze 5 wordt bijgesteld met steriel en anaëroob NaOH of HC1. Voorraad-cultures worden aangehouden op hetzelfde medium dat met 2 % agar vastgemaakt is en bij.4°C bewaard wordt. Vloeibare cultures kunnen na toevoegen van een zelfde volume glycerol bij -18°C bewaard worden.
10 Deze nieuwe soort werd met de gebruikelijke microbiologische technieken onderzocht en beschreven: a) Morfologie
Bij 60°C gekweekte cellen zijn beweeglijk; ze tuimelen. Oudere cellen of bij hogere temperaturen (zoals 75°C) 15 gekweekte cellen zijn minder beweeglijk, hoewel ze flagellen hebben. Per cel zijn er I tot 10 flagellen van het vertraagde peritryche type, die tot 80pm lang zijn. De bacteriecellen zijn 4 tot 8pm lang en 0,6 tot 0,9pm dik. Jonge cellen in de logarithmische fase vertonen vaak puntige uiteinden. Aan het eind van de logarith-20 mische groeifase kunnen de cellen uit groeien tot 200pm lengte, en bij het begin van de stationaire fase verdelen die zich dan tot ketens van bacteriën.
b) Sporen.
Sporen worden niet gevormd.
25 c) Andere kenmerken.
Deze bacteriën zijn strikt anaëroob, gram-variabel en katalase-negatief; pyruvaat wordt met het pyruvaat-ferredoxine-reductase-systeem omgezet. Er zijn twee anaerobe ferredoxinen en een rubredoxine aanwezig.
30 d) Vereisten voor groei.
Gist-extract (0,2 %) en een bron van koolstof zijn nodig. Als stikstof-bron zijn ammonium-ionen niet nodig, e) Substraten.
Glucose, andere monosacchariden en disacchari-35 den kunnen omgezet worden. Groei treedt ook op met pyruvaat, pectine, zetmeel en, in geringe mate, op gist-extract. Cellulose wordt 8000338 J t 5 niet aangetast.
f) Fermentatie-produkten.
Deze bacteriën vergisten glucose tot ethanol en koolzuur als voornaamste produkten, en in geringe mate tot ace-5 taat, lactaat en waterstof. Vastgesteld is dat de afbraak van glucose voornamelijk volgens het Emden-Meierhof-schema verloopt.
g) pH-traject: 4,5-9,8; optimale pH: 5,7-8,6.
h) Temperaturen
Minimum 36°C (Td bij 37°C meer dan 100 uur).
10 Optimale temperatuur 68°C (Td bij 68°C 1,5 uur).
Temperatuur-maximum 78°C (Td bij 76°C ongeveer 10 uur) (Td is verdubbelings- of generatie-tijd).
i) Nucleinezuur-samenstelling
Guanósine plus cytosine (g + c) ongeveer 15 38 % van het DNA (bepaald uit het soortelijk gewicht) en 32 % (bepaald uit de denaturatie bij verhitting).
j) Taxonomie.
De nieuwe stammen JW-200 en JW-20I hebben sommige eigenschappen gemeen met Clostridium thermohydrosulfuricum, 20 de enige andere bekende extreem thermofiele, anaerobe en glycoly-tische bacterie-soort. Maar de nieuwe stammen vormen geen sporen, zodat ze buiten’het geslacht Clostridium liggen. Andere eigenschappen zoals hierboven genoemd laten niet toe de nieuwe stammen in te delen bij enig ander geslacht beschreven m de 8 uitgave van 25 Bergey's Manual of Determinative Bacterology ( Williams &
Wilkins Comp., Baltimore, 1974). Daarom worden de nieuwe stammen tot een nieuw geslacht en een nieuwe soort gerekend, genaamd Thermoanaerobacter ethanolicus.
Hoewel glucose het bevoorkeurde substraat is 30 kunnen andere monosacchariden, zoals xylose, ribose, mannose, fructose en galactose, en disacchariden zoals saccharose, lactose, maltose en cellobiose, ook gebruikt worden. Groei treedt ook op op pyruvaat, pectine en zetmeel. Opgemerkt moet worden dat T.ethanolicus gist-extract voor groei nodig heeft.Zonder gist-extract krijgt 35 men in de volgende cultures geen groei meer. Hoewel de groei veel minder is dan in aanwezigheid van glucose kan gist-extract in con- 8000338 6 centraties boven 0,5 % als enige bron van koolstof, stikstof en energie dienen. Zoals in het nog komende voorbeeld zal blijken vergisten zuivere cultures van T. ethanolicus cellulose niet.
Zuivere cultures van T. ethanolicus voor toe-5 passing kan men gemakkelijk verkrijgen op hetzelfde medium als voor isoleren gebruikt werd, waarbij men onder anaerobe omstandigheden bij temperaturen tussen 36° en 78°C incubeert; de optimale temperatuur is ongeveer 68°C, waarbij de verdubbelingstijd ongeveer 90 minuten is. Deze groei is weinig gevoelig voor de pH; deze mag 10 variëren van 4,5 tot 9,8, ligt bij voorkeur tussen 5,7 en 8,6, en is het allerbeste ongeveer 7,3.
Clostridium thermocellum is eerder geïsoleerd, gekweekt en beschreven door meerdere onderzoekers. Behalve naar het eerder genoemde overzicht van McBee kan ook verwezen worden naar: 15 J.L. Een stam van Cl. thermocellum, namelijk JW 20, inmiddels onder no. ATCC 31549 bij de American Type Culture Collection gedeponeerd, werd door uitvinders dezes geïsoleerd uit een baal katoen afkomstig uit Louisiana. Net als met T. ethanolicus 35 werd deze stam Cl. thermocellum in biologisch zuivere vorm afgescheiden met behulp van de door Bryant and Robinson gemodificeerde 80^0338 7 techniek van Hungate. Daarbij werd ook het zelfde medium gebruikt, behalve dat in plaats van glucose nu 10,8 g/1 cellulose gebruikt werd, en dat het gehalte aan gist-extract tot 5,0 g/1 verhoogd was. Het voor aanhouden gebruikte medium moet onder anaerobe om-5 standigheden aangemaakt en onder een inert gas (stikstof of argon) aangehouden worden. De pH van dit medium ligt tussen 6,8 en 7,8, en is bij voorkeur 7,3.
Cl. thermocellum kan gemakkelijk onder anaerobe omstandigheden bij temperaturen tussen 45° en 65°C (optimum tempe-10 ratuur ongeveer 60°C) gekweekt worden op hetzelfde medium als waarop het geïsoleerd werd. De bevoorkeurde pH is ongeveer 7,5.
Hetzelfde cellulosebevattende medium dat voor het isoleren van Cl. thermocellum gebruikt werd is ook heel geschikt voor de gekoppelde vergisting van cellulose door T. etha-15 nolicus en Cl. thermocellum onder anaerobe, thermofiele omstandigheden. Zoals uit het nog komende voorbeeld zal blijken, ontstaat daarbij een belangrijke hoeveelheid ethanol, bij 60°C, pH = 7,5 en onder anaerobe omstandigheden wel 1,46 mol ethanol per verbruikte glucose-eenheid aan cellulose.
20 De uitvinding wordt nu nader toegelicht door het volgende, niet beperkende voorbeeld.
Vergisting van cellulose
Cellulose werd vergist met zuivere cultures van T. ethanolicus en Cl. thermocellum en met een gemengde culture.
25 Deze gebeurden allemaal onder argon bij 60°C en gedurende 168 uur. Men gebruikte 50 ml van het eerder beschreven medium, waarin echter in plaats van het glucose 540 mg cellulose zat, en dat 0,5 % gist-extract bevatte. De begin-pH was 7,5. Bij proef A werd de pH tijdens de vergisting op 7,5 Bijgeregeld, terwijl men bij proef B de pH door de vorming van zuren liet dalen.
Het Blijkt dat de koppeling van Cl.thermocel-5 lum met T. ethanolicus de afbraak en vergisting van cellulose aanzienlijk versnelt en tot een aanzienlijk hogere produktie van ethanol leidt. Ook ziet men dat men betere resultaten krijgt als men de pH tijdens de vergisting bij blijft regelen.

1. Werkwijze voor de vergisting van cellulose tot ethanol en koolzuur, met het kenmerk, dat men Thermoanaerobacter ethanolicus en Clostridium thermocellum in gemengde culture onder 5 anaerobe, thermofiele of extreem thermofiele omstandigheden kweekt op een medium dat cellulose bevat, totdat een winbare hoeveelheid ethanol ontstaan is.
