Abstract

La invencion se refiere al campo de la terapia génica, a la expresion génica y a los vectores para esto usos. En particular, la invencion se refiere a un método para producir unidades de ADN repetitivas grandes, estructurales intactas, especialmente utiles para la clonacion estable del ADN satélite alfa, y para el desarrollo y el uso de un cromosomas sintético o artificial para la expresion génica y la terapia génica en mamíferos, y especialmente en humanos. La invencion permite la construccion controlada de los cromosomas sintéticos o artificiales estables construidos a partir de los segmentos aislados de ADN purificado. Los segmentos mínimos funcionales incluyen preferentemente ADN centromérico, ADN telomérico, y ADN genomico. El cromosoma artificial realiza las funciones cromosomicas esenciales de los cromosomas de origen natural, para permitir que el cromosomas funcione como un vector efectivo para la terapia génica.
