Abstract
Para obtener una planta de cereal transgénico que se transforma establemente, un meristemo de cereal expuesto se somete a bombardeo biolístico con el fin de apuntar a células del meristemo no diferenciadas para su transformacion. Los embriones inmaduros en el proembrion temprano, proembrion medio, proembrion tardío, etapa coleoptilar transicional o temprana se cultivan para bombardeo biolístico. El tejido del meristemo o células destinadas a contribuir al meristemo, se manipulan después con el fin de alargar los sectores transgénicos, ya sea mediante seleccion y/o por realizar una proliferacion del tejido de los brotes o multiples meristemos por sí mismos. La poblacion de los brotes así obtenida, se tamizo después, por medio de un análisis de enriquecimiento no letal, para identificar cualesquiera sectores quiméricos que contribuirán a la transmision de la línea germinal, quimeras periclinales L2, sin sectores, que por definicion, transmitirán a la progenie. El tiempo incrementado en el cultivo, bajo seleccion, incrementa los prospectos para conversiones sectoriales a periclinales, y también selecciona conversiones de L1 a L2 las cuales, mediante un cambio de posicion contribuyen finalmente a la línea germinal. Los sectores transgénicos también se estabilizan durante el paso de formacion de retoños.
