Abstract

La presente invención se refiere a un método para producir un producto de la conjugación enzimática de tres polipéptidos, que comprende la incubación simultánea de i) un primer polipéptido que comprende la secuencia de aminoácido LPXTG (SEC ID NO: 20, en donde X puede ser cualquier residuo de aminoácido), un segundo polipéptido que comprende la secuencia de aminoácido LPXTA (SEC ID NO: 31, en donde X puede ser cualquier residuo de aminoácido), un tercer polipéptido que tiene dos extremos N-terminal por lo cual el polipéptido tiene una oligo-glicina Gm (m = 2 (SEC ID NO: 22), o 3 (SEC ID NO: 23), 0 4 (SEC ID NO: 24), o 5 (SEC ID NO: 25)) secuencias de aminoácidos en su primer extremo N-terminal y una oligo15 alanina Am (m = 2 (SEC ID NO: 26) , o 3 (SEC ID NO: 27) , o 4 (SEC ID NO: 28), o 5 (SEC ID NO: 29)) secuencias de aminoácidos en su segundo extremo N-terminal, un cuarto polipéptido con actividad de sortasa por lo cual el polipéptido se deriva de sortasa A de Staphylococcus aureus, y un quinto polipéptido con actividad de sortasa por lo cual el polipéptido se deriva de sortasa A de Streptococcus pyogenes y la recuperación del conjugado de la mezcla de reacción.
