Abstract
La presente invención se refiere a un método para detectar una variación genética en una muestra genética proveniente de un sujeto, caracterizado porque comprende: poner en contacto un primero y un segundo conjunto de sondas con la muestra genética, donde el primer conjunto de sondas comprende una primera sonda marcadora y una primera sonda trazadora, y el segundo conjunto de sondas comprende una segunda sonda marcador ay una segunda sonda trazadora, hibridar por lo menos algunas partes del primero y el segundo conjunto de sondas con unas primera y segunda regiones de ácido nucleico de interés en las moléculas de nucleótidos de la muestra genética, respectivamente, ligar el primer conjunto de sondas, ligando por lo menos la primer sonda marcadora y la primera sonda trazadora, ligar el segundo conjunto de sondas, ligando por lo menos la segunda sonda marcadora y la segunda sonda marcadora y la segunda sonda trazadora, opcionalmente, amplificar los conjuntos de sondas ligadas, inmovilizar las sondas trazadoras en una posición predeterminada sobre un sustrato, donde las primera y las segundas sondas marcadoras y/o las sondas marcadoras amplificadas de las mismas ligadas a las sondas trazadoras inmovilizada comprende primeros y segundos marcadores, respectivamente, los primero y segundos marcadores son diferentes, los marcadores inmovilizados e pueden resolver ópticamente por tipo de marcador, las primeras y segundas sondas trazadoras inmovilizadas y/o las sondas trazadoras amplificadas de las mismas comprende primeros y segundos trazadores, respectivamente, y llevar a cabo el paso de inmovilización inmovilizando a los trazadores en la posición predeterminada, contar (i) un primer número del primer marcador inmovilizado en el sustrato, y (ii) un segundo número del segundo marcador inmovilizado en el sustrato, y comparar los primeros y segundos números para determinar la variación genética en la muestra genética.
