Abstract
La presente invención se refiere a un método de perfusión para la manufactura de etanercept correctamente plegado, caracterizado porque comprende los siguientes pasos: (a) preparar una mezcla que comprende células capaces de expresar una proteína que comprende etanercept y un medio cultivo adecuado para llevar a cabo tal expresión; (b) en un recipiente de reacción adecuado que contiene la mezcla, causar que las células produzcan la proteína que comprende etanercept; y (c) remover periódica o continuamente medio de cultivo usado de, y agregar medio de cultivo fresco a, el recipiente de reacción, en donde la proteína que comprende etanercept producida en el método comprende al menos 40% en peso 50% en peso o 60% en peso de etanercept correctamente plegado, y en donde el medio de cultivo comprende un medio para célula de ovario de hámster Chino como un medio de alimento base y en donde el medio de cultivo comprende dexametasona, galactosa y N-acetilmanosamina (ManNac).
