Abstract
La presente invención se relaciona con métodos novedosos para la purificación de proteína de fusión Fc-péptido (cuerpos peptídicos) derivada de cuerpos de inclusión después de la expresión procariótica. En particular, se relaciona con métodos cromatográficos de los péptidos de fusión después del replegamiento y dimerización que comprende cromatografías de captura por afinidad, intermedia y de pulido. Esos métodos facilitan la disminución de las impurezas relacionadas con el producto, como variantes de sulfuro o variantes de carga de las proteínas de fusión Fc-péptido en el producto final. Además, la presente invención se relaciona con condiciones específicas y amortiguadores seleccionados que evitan la agregación, precipitación y degradación de las proteínas de fusión Fc-péptido. Finalmente, los métodos de la presente invención dan como resultado una composición farmacéutica formulada o una composición farmacéutica en etapa previa que contiene una proteína de fusión Fc-péptido de alta pureza.
