Claims (1)
- 【特許請求の範囲】 1.bFGFを含むサンプルから精製bFGFを回収する方法であって、該方法は本質的 に以下の工程からなる: (a)bFGF含有溶液を強カチオン交換マトリックスと接触させる工程; (b)該強カチオン交換マトリックスから、多数の画分を、bFGFを含む少なく とも1つの画分で溶出する工程; (c)bFGFを含有する強カチオン交換マトリックス画分を疎水的相互作用マト リックスと接触させる工程; (d)該疎水的相互作用マトリックスから、多数の画分を、bFGFを含む少なく とも1つの画分で溶出する工程; (e)bFGFを含有する疎水的相互作用マトリックス画分を弱カチオン交換マト リックスと接触させる工程; (f)該弱カチオン交換マトリックスから、多数の画分を、bFGFを含む少なく とも1つの画分で溶出する工程;および (g)bFGFを含有する弱カチオン交換マトリックス画分から精製bFGFを回収す る工程。 2.前記強カチオン交換マトリックスがスルホプロピル−アガロース、デキスト ラン、およびアクリルアミドマトリックスらなる群から選択される、請求項1に 記載の方法。 3.前記疎水的相互作用マトリックスが、フェニル、アセチル、および(1〜8 C)アルキルからなる群から選択される官能基に結合しているアガロース、シリ カ、およびポリマー性樹脂からなる群から選択される支持体を含む、請求項1に 記載の方法。 4.前記弱カチオン交換マトリックスがカルボキシル官能基に結合したシリカ支 持体を含む、請求項1に記載の方法。 5.各溶出工程が約2℃と25℃との間の温度で行われる、請求項1に記載の方法 。 6.前記接触工程、工程a、c、およびe、ならびに前記溶出工程、工程b、d 、およびfが約6と8との間のpHで行われる、請求項1に記載の方法。 7.前記精製bFGFがタンパク質アナログを含み、ここで1またはそれ以上のシス テイン残基が中性アミノ酸残基により置換され、該タンパク質アナログが天然bF GFの生物学的活性を示す、請求項1に記載の方法。 8.前記精製bFGFが天然bFGFのアミノ末端欠失アナログを含む、請求項1に記載 の方法。 9.前記官能基がポリアスパラギン酸である、請求項4に記載の方法。
