Abstract

La presente descripción proporciona métodos de alto rendimiento para mutagenizar, seleccionar y dirigir rápidamente microbios candidatos que son capaces de fijar nitrógeno atmosférico en presencia de nitrógeno exógeno. Los métodos utilizan un biosensor microbiano capaz de detectar la presencia/ausencia de amonio y/o glutamina en una composición y señalizar con un indicador fluorescente. La presente descripción utiliza además ensayos de detección visual rápida capaces de procesar miles de microbios candidatos. Los métodos y el biosensor descritos se pueden usar para identificar bacterias mutantes con capacidades de fijación de nitrógeno mejoradas. También se describen bacterias mutantes con capacidades de fijación de nitrógeno mejoradas, así como métodos para utilizar estas nuevas bacterias para proporcionar nitrógeno fijo a una planta.
