Abstract
Se proporcionan métodos y composiciones relacionados con la administración intracelular de proteínas de edición de genes. La invención se refiere a composiciones y métodos para transportar polipéptidos de edición de genes, tales como Cas9 o Cas12, a una célula ex vivo o in vivo. La invención incluye un agente de edición activa de genes dirigida (TAGE) que incluye un resto de unión a la membrana celular extracelular, p. ej., un polipéptido de unión a antígeno, un péptido de penetración celular (CPP), un ligando o combinaciones de estos, que se une específicamente a una molécula unida a la membrana celular extracelular (p. ej., una molécula de la superficie celular) y un polipéptido modificador dirigido al sitio que reconoce una secuencia de ácido nucleico.
