Abstract
La proteína conocida como factor de crecimiento epidérmico humano 2 (HER2) se expresa en las células de cáncer de mama y su expresión se correlaciona con un crecimiento tumoral agresivo. La presente invención proporciona anticuerpos humanos de longitud completa (IgG) novedosos, que se unen al HER2 humano (anticuerpos monoespecíficos) o a la APLP2 (anticuerpos monoespecíficos). La presente invención también proporciona anticuerpos biespecíficos (bsAb) novedosos, que se unen tanto al HER2 como a la APLP2 y median la internalización y degradación de HER2 a través del complejo de APLP2, en presencia de tumores que expresan HER2. Se describen moléculas de unión al antígeno biespecíficas y ADC que comprenden un primer dominio de unión al antígeno que se une específicamente a APLP2 humana y un segundo dominio de unión al antígeno que se une específicamente a HER2 humano. Los ADC biespecíficos descritos son capaces de inhibir el crecimiento de ciertos tumores que expresan HER2 y pueden ser útiles para el tratamiento del cáncer de mama y trastornos en los que se desee y/o resulte terapéuticamente beneficioso dirigir un agente terapéutico hacia una célula tumoral que exprese HER2.
