Abstract

Se describe un proceso mejorado, de costo efectivo y breve para la purificación del polisacárido capsular de S. pneumoniae, Streptococcus del Grupo B, 1-1. influenzas, S. typhi y N. meningitidis. El proceso incluye un cóctel de tratamiento con enzima, filtración de flujo tangencial y purificación de cromatografia multimodal. Para bacterias Gram-negativas, el proceso de remoción de endotoxina involucra la resina endotrap HD. Este proceso acortado logró la pureza requerida a través de WHO/EP/BP para el uso en la preparación de vacunas humanas, con pasos simples y un rendimiento más alto según comparado con procesos convencionales. Los pasos del proceso evitan el uso de solventes orgánicos tales como, por ejemplo, alcohol, fenol y ultracentrifugación que de otra manera son costosos y consumidores de tiempo para realizar y/o peligros de salud para uso comercial. Este proceso descrito también es simple, eficiente, no tóxico, fácil de escalar y ambientalmente favorable.
