Abstract
Las regiones V de inmunoglobulina específica de antígeno se identifican a partir de una colección de ácidos nucleicos ampliada utilizando una reacción de cadena de polimerasa que usa cebadores directos específicos de secuencia líder. El uso de cebadores de secuencia líder permite que todas las secuencias de la región V se amplíen (que incluyen aquellas con mutaciones del terminal 5´ extensivo) sin la pérdida de la secuencia del segmento génico V 5´ original. Estas colecciones pueden monitorearse para regiones V específicas de antígeno que utilizan células eucariotas diseñadas para expresar los ácidos nucleicos codificadores de la región V ampliada o utilizando técnicas de expresión de fago bacteriano. En este último caso, se produce una segunda colección de región V utilizando un conjunto de cebadores de región V 5´ mayor que el convencional. Los errores de secuencia introducidos en los productos de ampliación mediante este método se corrigen utilizando información de secuencia obtenida en los productos amplificados por los cebadores de la región V para monitorear la colección creada utilizando los cebadores de secuencia líder. La información de secuencia de aminoácidos de fragmentos de inmunoglobulinas del donante puede utilizarse para ayudar a identificar ácidos nucleicos que codifican las cadenas pesada y liviana de anticuerpos del donante así como también para diseñar cebadores con el objeto de amplificar dichos ácidos nucleicos.
