Abstract
La invención ofrece un método para la elución continua de un producto biológico macromolecular biofarmacéutico proveniente de más de una columna de cromatografía, que comprende los siguientes pasos: (a) disponer una corriente de producto por una entrada, (b) cargar simultáneamente n columnas de cromatografía frontal con la corriente de producto en una zona de carga con un tiempo de carga tB, donde una corriente de salida de las n columnas de cromatografía frontal es distribuida simultáneamente entre al menos n-1 columnas de cromatografía de recuperación en una zona de recuperación, donde n es entre 1 y 5, y donde las n columnas de cromatografía frontal presentan en un momento dado, un tiempo de carga tB de duración diferente, de entre 0 y L1, entre L1 y L2, y hasta de entre Ln-1 y Ln, donde cada columna de cromatografía frontal presenta una tiempo de carga total de Ln, caracterizado porque, después de un tiempo de cambio ts constante (Switch Time) de Ln - Ln-1, periódicamente la n columna de cromatografía frontal sale de la zona de carga y una columna de cromatografía de recuperación de la zona de recuperación entra en la zona de carga, (c) lavado de la columna cargada de producto del paso b) con al menos una solución amortiguadora de lavado, (d) elución del producto de la columna lavada del paso c) con un tiempo de elución tE, que es = 80% que el tiempo de cambio tS (Switch-Time), y porque al menos una columna de cromatografía se encuentra en el paso de elución (d) de manera continua más del 80% del tiempo operativo total tG.
