Abstract

La presente invención proporciona un método para determinar la presencia de uno o más biomarcadores de polipéptido en una muestra, en donde el método comprende los pasos de: (a) someter la muestra a un análisis espectrométrico de masa (MS) y registrar el índice del tiempo de retención y masa correspondiente de cada señal detectada; (b) correlacionar la masa que corresponde a cada señal con una base de datos de referencia de masas de biomarcador para formar una correlación entre cada señal y un biomarcador de referencia, y desechar las señales cuyas masas no se correlacionen con una masa de biomarcador de referencia; (c) almacenar las señales cuyas masas se correlacionen con un biomarcador de referencia; (d) confirmar la correlación entre cada señal almacenada y un biomarcador de referencia, mediante el acoplamiento del espectro MS de cada señal con el espectro MS del biomarcador de referencia en la base de datos utilizando una medida de similitud, para definir un conjunto de señales de correlación positiva; (d) medir la intensidad de cada señal de correlación positiva y registrar su intensidad de señal absoluta o su intensidad de señal relativa utilizando una función de discriminación; (e) aplicar un valor de umbral a los valores de calificación obtenidos de la función de discriminación para determinar la presencia o ausencia del biomarcador.
