Abstract
La presente invención está dirigida a un método para. determinare mutaciones de una secuencia de aminoácido en un polipéptido producido, que comprende los siguientes pasaos de a) proporcionar una muestra de un polipéptido producido, b) incubar el polipéptido en la muestra con una proteasa, c) llevar a cabo un análisis bidimensional utilizando cromatografía de fase inversa acoplada con una espectroscopía de masas de alta resolución (FT-ICR/FT-orbitrap) y análisis Ms/MS de los fragmentos de la secuencia de aminoácido de los péptidos, d) evaluación de los datos comparando los grupos de datos LC-MS obtenidos para las muestras paralelamente con el grupo de datos de un muestra de referencia, mediante la búsqueda de diferencias en las intensidades de señal a tiempos de retención dados y por la evaluación de las señales diferenciales con respecto a las mutaciones de la secuencia de aminoácido. La muestra de referencia para la evaluación de los datos (d) puede ser ya sea un estándar bien caracterizado o una de las muestras a ser analizada.
