Abstract

Inventia se refera la biotehnologiile agricole si poate fi utilizata pentru multiplicarea microclonala a vitei-de-vie.Procedeul, conform inventiei, consta în aceea ca se selecteaza lastari cu lungimea de 6…10 cm, se spala, se clatesc în apa curgatoare, se sterilizeaza, se excizeaza apexuri cu marimea de 0,5…1,0 mm, ce contin meristeme cu 2…3 frunzulite primordiale, care se inoculeaza în mediul nutritiv Murashige-Skoog suplimentat cu 5 µM de 6-benzilaminopurina, 0,5 µM de acid naftalinacetic, 2% de zaharoza, 0,7% de agar, având pH-ul 5,7; dupa 2…3 saptamâni de la inoculare apexurile se iradiaza în regim periodic, timp de 20…25 min, cu unde milimetrice de intensitate joasa cu lungimea de unda de 5,6 mm si frecventa de 53,8 GHz, dupa ce regenerantii prolifereaza si se dezvolta ei se transfera pe mediul Murashige-Skoog, se subcultiva pe parcursul a 6…7 cicluri si se efectueaza microbutasirea lor in vitro, regenerantii cu 3…4 internoduri se transfera pe mediul Murashige-Skoog suplimentat cu 0,4 µM de acid naftalinacetic, apoi regenerantii cu sistem radicular bine dezvoltat se transfera în amestec de sol si turba, iar ulterior pe substrat de sol.
