Abstract
본 발명은 많은 그룹 B Streptococcus균주들, 특히 혈청형Ⅲ인 것들의 감염에 대한 보호면역성을 제공하는 Rib으로 표시되는 새로운 단백질 및 그것의 소단편, 다중체 또는 변이체에 관한 것이다.
본 발명은 이 단백질의 정제 방법, 매우 특이적인 항체의 제조방법, 시약키트, 이 단백질을 암호화 하는 DNA서열, 이 단백질 또는 그것의 단편이나 변이체를 함유하는 제약조성물을 포함한다.

제1도는 4가지 주요 혈청형을 나타내는 그룹 B Streptococcus균주로부터 제조한 주출물의 웨턴 블롯(Western blot)분석을 보여준다(타입 Ⅰ a : 균주 A909 : 타입 Ⅰb:SB35:타입Ⅱ :B1284: 타입Ⅲ:BS300. 면역 블롯에서 보여지는 것처럼 타입 Ⅰa 및 Ⅰb의 균주는 알파 및 베타 단백질을 발현하고, 염색된 겔에서 이 단백질의 위치는 화살표로 표시된다.좌측에 나타낸 분자량 마커의 단위는 KD이다.
제5도는 트립신 또는 펩신으로 처린 정제된 알파, 베타, 및 Rib단백질의 SDS-PAGE 분석을 보여준다. 트립신 처리는 pH7.5에서 수행되었고, 펩신처리는 pH4.0에서 수행되었다.SDS-PAGE분석전에 샘플을 중화시켰다. 콘트롤은 트립신 또는 펩신을 포함하는 샘플과 동일한 방식으로 처리했지만 효소를 첨가하지 않았다. 그렇나 처리는 단백질의 분해를 야기하지 않았다. P=펩신 :T=트립신, 좌측에 나타낸 분자량 마커 단위는 KD이다.
Claims (17)
- (a)그룹 B STREPTOCOCCUS균주,특히 혈청형Ⅲ인 것들로부터 얻을 수 있고,b)트립신 및 펩신에 의한 분해에 비교적 저항성이 있고, c)SEQ ID NO : 1에 따른 N-말단 아미노산 서열 또는 그것과 적어도 90% 상동적인 서열을 갖고, 및 d)이 단백질을 발현하는 그룹 B STREPTOCOCCUS균주에 대해서 보호면역성을 제공하는 것을 특징으로 하는 Rib으로 표시되는 정제된 단백질, 및 보호면역성을 제공할 숭 있는 능력을 갖는 천연적으로 존재하거나 인공적으로 변형된 그것의 변이체, 소단편 및 다중체.
- 제1항에 있어서, 상기 균주는 BS 30 또는 BM 110이고 단백질은 SDS-PAGE에 의해 결정할 때 약 95KD의 겉보기 분자량을 갖는 것을 특징으로 하는 단백질.
- Rib단백질을 발현하는 STREPTOCOCCUS그룹 B균주를 배양하고, 배지 및/또는 미생물을 분리학, 세균을 효소 바람직하게는 뮤타노리신으로 분해시키고,프로테아제 억제인자를 선택적으로 첨가하고, 분해된 세균을 상징액으로부터 분리하고, Rib단백질을 상징액으로부터 추출하는 것을 특징으로 하는 Rib단백질의 분리방법.
- 제3항에 있어서, 단백질은 투석, 이온교환 크로마토그피에 의한 분리 및 겔여과에 의해 상징액으로부터 추출되는 것을 측징으로 하는 방법.
- 제1항에서 정의된 Rib단백질 및 그것의 변이체, 소단편 및 다중체에 매우 특이적인 항체.
- Rib단백질에 대한 항체의 검출을 위한 시약키트에 있어서,제1항에서 정의된 Rib단백질 또는 그것의 변이체, 소단편 및 다중체를 함유하는 것을 특징으로 하는 시약키트.
- Rib단백질의 검출을 위한 시약키트에 있어서, 제1항에서 정의된 단백질에 특이적인 항체, 및 선택적으로 표준물로서 Rib단백질 및 그것의 변이체, 소단편 및/또는 다중체를 함유하는 것을 특징으로하는 시약키트.
- 제1항에 정의된 Rib단백질 또는 그것의 변이체, 소단편 또는 다중체를 암호화 하는 DNA서열을 포함하는 벡터.
- 제8항에 있어서, Rib단백질을 암호화하는 벡터의 DNA서열은 수탁번호 DSM 9039를 갖는 파지 람다 Rib 1-3의 대응 DNA서열 중 어느 것과 본질적으로 동일한 것을 특징으로 하는 벡터.
- 제8항에 있어서, Rib 단백질을 암호화하는 벡터의 DNA서열은 수탁번호 DSM 9040을 갖는 파지 람다 Rib 1-5의 대응 DNA서열 중 어느 것과 본질적으로 동일한 것을 특징으로 하는 벡터.
- 제8항에 있어서, Rib 단백질을 암호화하는 벡터의 DNA서열은 수탁번호 DSM 9041을 갖는 파지 람다 Rib 1-7의 대응 DNA서열 중 어느 것과 본질적으로 동일한 것을 특징으로 하는 벡터.
- 수탁번호 DSM 9039을 갖는 파지 람다 Rib 1-3.
- 수탁번호 DSM 9040을 갖는 파지 람다 Rib 1-5.
- 수탁번호 DSM 9041을 갖는 파지 람다 Rib 1-7.
- 제8항에서 정의된 벡터로부터 얻을 수 있는, Rib 단백질 또는 그것의 변이체, 소단편 또는 다중체를 암호화하는 DNA서열.
- 제1항에서 정의된 Rib 단백질 또는 그것의 변이체, 소단편 또는 다중체를 함유하는 제약조성물에서, 상기 조성물이 추가로 제약학적으로 허용가능한 보조약 또는 부형제를 함유하는 것을 특징으로 하는 조성물.
