비엠에스 180048 및 이의 유사체를 대량 생산하는 개선된 방법

Claims (13)

  1. a) 화합물(Ⅲ)을 팔라듐-촉매화된 헤테로환화 단계에서 화합물(Ⅳ)와 반응시켜 2-(R5)3Si-인돌 화합물(Ⅱ)을 얻고; b) 무기산으로 화합물(Ⅱ)의 트리아킬실릴 그룹을 제거하고 염기성화시켜 합성 생성물 일반식(Ⅰ)을 생성시킴을 특징으로 하여, 일반식(Ⅰ)의 유용한 항편두통제를 제조하는 개선된 방법:
    상기식에서, R은 수소, 저급알킬 및 트리플루오로메틸이고; R1은 수소, 저급알킬 및 저급알킬-페닐이며; R2및 R3은 독립적으로 수소 및 메틸중에서 선택되고; R4는 저급알킬이며; R5는 메틸, 에틸, 또는 프로필이고; n은 0 또는 정수 1 또는 2이다.
  2. 제1항에 있어서, R 및 R4가 메틸이고; R1, R2및 R3이 수소이며; n이 1인 방법.
  3. 제1항에 있어서, 일반식(Ⅲ)의 화합물이 a) 화합물(Ⅷ)을 피페라진과 반응시켜 화합물(Ⅴ)을 얻고; b) 화합물(Ⅴ)를 화합물(Ⅶ)과 반응시켜 화합물(Ⅲ)을 얻음으로써 제조되는 방법.
    상기식에서, R4는 저급알킬이며; R5는 메틸, 에틸, 또는 프로필이고; LG는 합성 유기 이탈그룹이다.
  4. 제1항 및 제3항에 있어서, R5가 에틸인 방법.
  5. 제1항에 있어서, 화합물(Ⅲ) 및 (Ⅳ)의 환화가 에탄올중의 팔라듐 아세테이트, 트리페닐프스핀, 염화리튬 및 탄산나트륨으로 수행되는 방법.
  6. 제1항에 있어서, 일반식(Ⅱ)의 2-(R5)3Si-인돌 중간체를 염산으로 처리하여 트리알킬실릴 잔기를 제거함으로써 일반식(Ⅰ)의 생성물로 전환시키는 방법.
  7. 일반식(Ⅱ)의 화합물:
    상기식에서, R은 수소, 저급알킬 및 트리플루오로메틸이고; R1은 수소, 저급알킬 및 저급알킬-페닐이며; R2및 R3은 독립적으로 수소 및 메틸중에서 선택되고; R4는 저급알킬이며; R5는 메틸 또는 에틸이고; n은 0 또는 정수 1 또는 2이다.
  8. 제7항에 있어서, R 및 R4가 메틸이고; R1, R2및 R3이 수소이며; R5가 에틸이고; n이 1인 화합물.
  9. 일반식(Ⅲ)의 화합물:
    상기식엣, R2및 R3은 독립적으로 수소 및 메틸중에서 선택되고; R4는 메틸이며; R5는 메틸 또는 에틸이다.
  10. 제9항에 있어서, R2및 R3이 수소인 화합물.
  11. 제9항에 있어서, R5가 에틸인 화합물.
  12. a) 화합물(Ⅴ)을 화합물(Ⅶ)와 반응시켜 화합물(Ⅲ)을 얻고; b) 화합물(Ⅲ)을 팔라듐-촉매화된 환화반응으로 화합물(Ⅳ)와 반응시켜 화합물(Ⅱ)을 얻고; c) 화합물(Ⅱ)를 무기산으로 탈보호시켜 BMS 180048의 산염을 얻고; d) 염을 물로부터 재결정한 다음 염기성화시켜 BMS 180048을 제조함을 특지응로 하여, BMS 180048을 제조하는 개선된 방법:
    상기식에서, R4는 저급알킬이고; R5는 메틸 또는 에틸이다.
  13. 제12항에 있어서, R5가 에틸인 방법.
