Abstract

고 알칼리성, 위치 비특이적인 리파아제를 Streptomyces 1군의 균주로부터 얻을 수 있다,
Streptomyces 1군 균주는 이제까지 리파이제를 생산하는 것으로 알려지지 않았다,
리파아제 제제는 pH 10에서 최적 활성의 50% 이상인 활성을 갖고 예를들어 세제에 유용하다,

제1도 내지 제7도는 다음 균주 S. griseus LB 501 (DSM 7349), S. griseus LB 502(DSM 7350), S. coelicolor LB 511 (FERM BP-4236), S. coelicolor LB 512 (FERM BP-4237), S. griseus LB 524(DSM 8672), S. coelicolor N 2293(ATCC 23899) 및 S. parvus N 2300 (ATCC 12433)으로부터 유래된 본 발명의 리파아제 제제의 활성에 대한 pH 프로파일을 보여준다. 구체적인 사항은 실시예 6에서 주어진다.

Claims (20)

1)위치 비특이적이고, 2) 40℃에서 20분동안 기질로서의 올리브유 및 유화제로서의 폴리비닐 알콜을 사용한 Ca++없는 분석에서 최적 pH 및 pH 10에서의 두 활성을 측정했을때, pH 10에서 최적 pH에서의 활성의 50% 이상인 활성을 가지며, 및 3)Streptomyces 1군 균주의 배양에 의해 생성된 수 있는 라파이제 제제.
제1항에 있어서, 균주는 S. coelicolor, S. limosus, S. alboviridis, S. griseus, S. parvus, S. setonii 또는 S. nitrosporeus인 것을 특징으로 하는 라파아제 제제.
제2항에 있어서, 균주는 S. coelicolor FERM BP-4236, FERM BP-4237, ATCC 23899, S. limosus ATCC 19778, S. alboviridis ATCC 25425, S. griseus ATCC 23345, DSM 7349, DSM 7350, DSM 8672, S. parvus ATCC 12433, S. setonii ATCC 25497 또는 S. nitrosporeus ATCC 27472인 것을 특징으로 하는 리파아제 제제.
제1항에 있어서, 60분 반응시간동안 기질로서의 올리브유 및 유화제로서의 폴리비닐 알콜을 사용하여 두활성을 측정하고, 세제용액이 선형 알킬 벤젠 설포네이트, 0.35g/ℓ, 알콜 에톡실레이트 0.15g/ℓ, 삼인산나트륨 1.25g/ℓ, 황산나트륨 1.00g/ℓ, 탄산나트륨 0.45g/ℓ 및 메타규산나트륨 0.15g/ℓ으로 구성될 때, pH 10.2인 세제용액에서 pH 10인 디에탄올 아민 완충액에서의 활성의 50%이상인 활성을 갖는 것을 특징으로 하는 리파아제 제제.
제4항에 있어서, 균주는 Streptomyces 아군 1A 또는 1B의 균주인 것을 특징으로 하는 리파아제 제제.
제5항에 있어서, 균주는 S. griseus, S. coelicolor 또는 S. parvus인 것을 특징으로 하는 라파아제 제제.
제6항에 있어서, 균주는 S. griseus DSM 7349, DSM 7350, DSM 8672, S. coelicolor FERM BP-4236, FERM BP-4237, ATCC 23899 또는 S. parvus ATCC 12433인 것을 특징으로 하는 리파아제 제제.
제4항에 있어서, pH 10에서 기질로서의 올리브유 및 유화제로서의 폴리비닐 알콜을 사용하여 두 활성을 측정했을 때 Ca++의 부재하에서 50mM Ca++존재하에서의 활성의 50%이상인 활성을 갖는 것을 특징으로 하는 리파아제 제제.
제8항에 있어서, 균주는 S. griseus DSM 7350인 것을 특징으로하는 리파아제 제제.
전항의 어느 한항에 있어서, 리파이제 제제가 비분말 과립, 안정화된 액체, 슬러리 또는 보호된 효소의 형태로 세제첨가제로서 제공되는 것을 특징으로 하는 리파아제 제제.
1)위치 비특이적이고, 2)40℃에서 20분도안 기질로서의 올리브유 및 유화제로서의 폴리비닐 알콜을 사용하여 측정했을때 pH 9-11의 범위에서 최적 활성을 가지며, 3)Streptomyces 1군 균주에 고유한 세포외의 리파아제와 면역학적으로 동일하거나 부분적으로 동일한 리파아제.
제6항의 리파아제 제제를 생성할 수 있는 Streptomyces griseus 균주.
제12항에 있어서, 상기 리파아제 제제를 생성할 수 있는 S. griseus DSM 7350또는 이들의 돌연변이체나 변이체인 것을 특징으로 하는 Streptomyces griseus 균주.
탄소원, 질소원 및 무기염을 포함하는 적당한 영양배지에서 리파아제 생성균주인 Streptomyces 1군 균주의 배양과 이후 리파아제 제제의 회수로 이루어지는 것을 특징으로 하는 제1항 내지 제10항중 어느 한항에 따른 리파아제 제제의 제조방법.
제14항에 있어서, 균주는 S. coelicolor, S. limosus, S. alboviridis, S. griseus, S. parvus, S. setonii 또는 S. nitrosporeus인 것을 특징으로 하는 방법.
제15항에 있어서, 균주는 S. coelicolor FERM BP-4236, FERM BP-4237, ATCC 23899, S. limosus ATCC 19778, S. alboviridis ATCC 25425, S. griseus ATCC 23345, DSM 7349, DSM 7350, DSM 8672, S. parvus ATCC 12433, S. setonii ATCC 25497 또는 S. nitrosporeus ATCC 27472 또는 이들의 리파이제-생성변이체이거나 돌연변이체인 것을 특징으로 하는 방법.
계면활성제와 제1항 내지 제17항중 어느 한항의 리파이제 제제로 이루어지는 세제 조성물.
제17항에 있어서, 세제빌더를 1∼40% 더 포함하고, 수용액에서 측정했을 때 pH7∼11을 나타내는 것을 특징으로하는 세제 조성물.
제18항에 있어서, 빌더가 인산염 빌더, 제올라이트 또는 시트르산 나트륨인 것을 특징으로 하는 세제 조성물.
제17항 내지 제19항중 어느 한항에 있어서, 프로테아제, 아밀라제, 셀룰라제, 옥시다제 및 퍼옥시다제로 구성되는 군에서 선택된 추가의 세제효소를 더 포함하는 것을 특징으로 하는 세제 조성물.
