Abstract

본 발명은 면역검정, 특히 B12에 대한 검정을 수행하는데 유용한 다음 일반식(*Ⅰ)의 커플링제에 관한 것이다 :

상기식에서, Z 및 Z'는 동일하거나 상이할 수 있고 각각 H2NHN, ICH2
k,m 및 n은 1내지 10의 정수이고, X는 알킬렌 그룹, 사이클로알킬렌 그룹, 알킬사이클로알킬렌 그룹, 2가 방향족 그룹 또는 아미노알킬렌 그룹이며, R 및 R'는 동일하거나 상이할 수 있으며 각각 탄소수 1내지 10의 치환되거나 비치환된 아미노알킬렌 그룹이거나 탄소수 5내지 20의 사이클로알킬렌 또는 알킬사이클로 알킬렌 그룹이다. Z와 Z' 모두가
인 경우에 서로 동일하거나 상이할 수 있는 것을 제외하고는 Z와 Z'는 동일하다.
R-단백질을 함유하는 수성 고유 인자를 정제하는 방법이 기재되어 있다. 당해 방법은 용액 중의 거의 모든 R-단백질을 결합시키기에 충분한 양의 코빈아미드와 용액중의 고유 인자를 결합시키기에 충분한 양의 고유인자 친화성 수지를 고유 인자 용액에 가하고, 결합된 코빈아미드와 수지로부터의 R 단백질을 세척하며, 고유인자를 수지로부터 용출시킨 다음, 용출된 고유 인자를 투석시킴을 포함하다. 정제된 고유 인자는 코빈아미드와의 교차반응도가 0.004% 미만일 수 있으며, 정제된 물질 중의 단백질의 95% 이상이 코발아민을 결합시킬수 있다. 미립자와 정제된 고유 인자를 전술한 커플링제중 하나로부터의 잔기에 의해 접합시킨 접합체 또는 본 명세서내에 기재되어 있고 이러한 미립자와 정제된 고유 인자와의 접합체를 포함하는, 코발아민에 대한 검정을 수행하는 키트가 기재되어 있다. 미립자와 정제된 고유 인자와의 접합체의 사용을 포함하는, 코발아민에 대한 검정을 수행하는 방법이 또한 기재되어 있다.

제1도는 코발아민의 일반식을 나타낸 것이고, 제2도는 본 발명에 따르는 바람직한 화합물 계(family)의 일반식을 나타낸 것이며, 제3도는 제2도의 화합물 계중 바랍직한 말단 그룹의 구조식을 나타낸 것이다.

Claims (32)

  1. 다음 일반(Ⅰ) 또는 (Ⅰ')의 커플링제.
  3. 상기식에서, 일반식(Ⅰ)에서 Z 및 Z'는 동일하고 각각 H2NHN, ICH2또는

  14. 제1항에 있어서, Z가 NHNH2이거나 첨부된 도면의 제3도와 제9도중의 하나의 구조식을 갖는, 제2도의 일반식의 커플링제.
  15. 제4항에 있어서, Z가 NHNH2이거나 제2도의 일반식의 커플링제.
  16. 제4항에 있어서, Z가 첨부된 도면의 제3도의 구조식을 갖는, 제2도의 일반식의 커플링제.
  17. 제4항에 있어서, Z가 첨부된 도면의 제9도의 구조식을 갖는 제2도의 일반식의 커플링제.
  18. 제1항에 있어서, Z'가 첨부된 도면의 제11도의 구조식을 가지 C6H10이 1,4-사이클로헥실인 제10도의 일반식의 커플링제.
  19. 고유 인자로부터 R-단백질을 분리시킴으로써 정제된 고유 인자의 코빈아미드(cobinamide)와의 교차 반응도가 0.004% 미만이 되는 정도로 미립자를 제1항에 따르는 커플링제로부터 유도된 잔기에 의해 상기와 같이 정제된 고유 인자에 접합시킨 방법.
  20. 단백질의 95%이상이 코발아민을 결합시키는 미립자와 제5항에 청구된 바의 고유 인자와의 접합체.
  21. 고유 인자로부터 R-단백질을 분리시킴으로써 정제된 고유 인자의 코빈아미드와의 교차 반응도가 0.004%미만이 되는 정도로 미립자를 제1항에서 청구한 바의 커플링제로부터 유도된 잔기에 의해 상기와 같이 정제된 고유 인자에 접합시킨 접합체, 효소와 정제된 고유 인자에 대한 특이적 결합원을 제1항에서 청구한 바의 커플링제로부터 유도된 잔기에 의해 서로 접합시킨 접합체 및 효소와 반응하여 시그날을 생성하거나 시그날 생성을 유발시킬 수 있는 반응 생성물을 생성시키는 기질 지시제를 포함하는, 코발아민에 대한 검정 수행용 키트.
  22. 제11항에 있어서, 효소가 알칼리성 포스파타제이고 지시제가 MUP인 키트.
  23. 제12항에 있어서, 특이적 결합원이 B12 AMINE인 키트.
  24. 시험 샘플을 변성시키고, 변성된 시험 샘플에 고유 인자로부터 R-단백질을 분리시킴으로써 정제된 고유 인자의 코빈아미드(cobinamide)와의 교차 반응도가 0.004% 미만이 되는 정도로 미립자를 제1항에 따르는 커플링제로부터 유도된 잔기에 의해 상기와 같이 정제된 고유 인자에 접합시킨 접합체를 가하고, 이러한 접합체를 함유하는 변형된 시험 샘플을 배양하며, 이와 같이 배양된 조성물을 고유 인자/미립자 접합체를 보유하는 분리 물질 침착시키고, 분리물질 표면을 세척하여 결합되지 않은 B12를 제거하며, 효소와 정제된 고유 인자에 대한 특이적 결합원과의 접합체를 분리 물질에 가하여 다수의 유리 고유 인자를 결합시키고, 분리 물질을 세척하며, 효소와 반응하여 시그날을 생성하거나 시그날 생성을 유도할 수 있는 반응 생성물을 생성시키는 기질지시제를 가한 유도할 수 있는 반응 생성물을 생성시키는 기질 지시제를 가한 다음, 효소와 지시제간의 반응에 의해 생성된 시그날을 측정하거나 효소와 지시제 간의 반응 생성물을 유발시켜 시그날을 생성시킨 다음 이 시그날을 측정함을 포함하며, 코발아민에 대한 감정을 수행하는 방법.
