제1도는 HTLV-I rex의 뉴클레오티드 및 아미노산 서열[올리고뉴클레오티드에 의해 치환된 아미노산 위치 (1) (87, 88, 89번) 및 (2) (94번)뿐만 아니라, 82, 90, 91및 97번이 표시되어 있다]. 완전한 서열은 189개의 아미노산을 암호화하는 567개의 뉴클레오티드를 함유한다. 제1A도 : HTLV-I rex 유전자내에 삽입된 29개 돌연변이체의 위치, HTLV-I rex 유전자는 189개의 아미노산 단백질을 암호화한다. 특정부위 지향적 돌연변이 유발(site-directed mutagenesis)을 사용하여, 미스센스 치환체를 규정된 아미노산 잔기(박스로 나타냄)에 삽입시키고 rex 유전자내에서의 그의 위치에 따라 명명한다.

- Rex 단백질의 작용인자 활성을 나타내는 야생 타입 Rex 단백질의 펩타이드 영역중에서 적어도 하나의 트랜스우성형 음성 돌연변이에 의해 야생 타입 형태의 Rex 단백질로부터 변형되며 실질적으로 야생 타입 형태의 Rex 단백질의 핵소체 정위 활성을 갖는, HTLV-I Rex 단백질 기능의 트랜스우성형 억제제.
- 실질적으로 야생 타입 HIV-1 Rev의 특이적 결합 기능을 갖는 제1영역 및 야생 타입 HIV-1 Rev의 활성화 기능을 갖지 않으며 하나이상의 돌연변이에 의해 야생 타입 HIV-1 Rev로 부터 변형되는 제2영역을 포함하는 HIV-1 Rev 기능의 트랜스우성형 억제제.
- 실질적으로 야생 타입 Rev의 특이적 결합 기능을 갖는 제1영역을 포함하는, 야생 타입 HIV-1 Rev의 활성화 기능을 갖지 않는, HIV-1 Rev 기능의 트랜스우성형 억제제.
- 적절한 발현 시스템으로부터 상응하는 야생타입 유전자를 단리시키고, 상기 유전자를 적절한 클로닝 시스템에 도입시키며, 목적하는 돌연변이를 상기 유전자내에 유발시키고 목적하는 돌연변이가 일어난 클론으로 부터 생성된 돌연변이에 유전자를 회수함을 포함하는, 제1, 4및 6항중 어느 한 항에 따르는 유전자의 제조방법.
- 적절한 발현 및 증폭 시스템에서 제1, 4및 6항중 어느 한 항에 따르는 돌연변이체 유전자를 발현시키고 증폭시키후 그로부터 생성된 생성물을 회수함을 포함하는 제2, 4및 6항중 어느 한 항에 따르는 단백질의 제조방법.
- 약학적으로 허용되는 담체 또는 희석제와 함께, 제1, 4 및 6항중 어느 한 항에 따르는 유전자, 또는 제2, 4및 6항중 어느 한 항에 따르는 단백질, 또는 그의 기능성 절편 또는 유도체를 목적하는 예방 또는 치료 효과를 달성하기에 적합한 형태로 포함하는 약학 조성물.
- 제1, 4및 6항중 어느 한 항에 따르는 유전자, 또는 2, 4및 6항중 어느 한 항에 따른 단백질, 또는 치료효과를 달성하기에 적합한 형태로 상기 치료가 필요한 환자에게 투여함을 포함하는, 바이러스 또는 그의 기능성 절편 또는 유도체를 목적하는 예방감염의 치료 방법.
- 제8항에 따르는 DNA 단편을 HIV-1, HTLV-I 또는 HTLV-Ⅱ중의 하나를 복제하고 DNA 단편을 발현시켜 HILV-I Rex 기능의 트랜스우성형 억제제를 생성하는 능력을 갖는 세포내에 삽입시킴을 포함하는, HIV-1, HTLV-1 또는 HTLV-Ⅱ바이러스의 복제를 저해하는 방법.
- HIV-1 Rev 기능의 트랜스우성형 억제제를 HIV-1으로 감염시킨 세포내에 삽입시킴을 포함하는, HIV-1 복제를 저해하는 방법.
- 약제로서 사용하기 위한 제1, 4 및 6항중 어느 한 항에 따르는 유전자 또는 제2, 4 및 6항중 어느 한 항에 따르는 단백질 또는 그의 기능성 절편 또는 유도체.
- 제1, 4 및 6항중 어느 한 항에 따르는 유전자 또는 그의 기능성 절편 또는 유도체를, 기능성 형태로 표적 포유동물 세포내로 운반시키는 데 적합한 형태로 함유하는 벡터.
- 제3, 5 또는 7항에 따르는 돌연변이제 Rex 또는 Rev 단백질중의 트랜스우성 영역을 모방할 수 있는 저해제.
- 하기 단계를 포함하는, Rex 단백질의 유전자 활성화 기능의 특이적 저해제를 동정하는 방법:i)Rex단백질에 의해 인식되는 조절 반응 인자 및 적어도 하나의 사용되지 않은 연결 부위를 포함하는 mRNA를 암호화하는 DNA단편, 발현되어 핵으로부터 mRNA의 방출을 유도하는 단백질 생성물을 생성할 수 있는 rex 유전자를 암호화하는 DNA단편, rex 유전자를 암호화하는 DNA 단편, 및 mRNA 를 암호화하고 rex 유전자의 단백질 생성물을 발현시키고 mRNA를 발현시키는 능력을 갖는 DNA 단편에 의해 형질 전환된 숙주 세포를 포함하는 유전자 시스템을 제공하고, ii)상기 유전자 시스템의 세포를 포함하는 배양물을 Rex 단백질의 특이적 저해제로 생각되는 약제가 세포로 침투되도록 하는 조건하에서 상기 약제와 접촉시키며, iii)사용되지 않은 연결 부위를 포함하는 mRNA의 핵으로 부터의 방출에 대한 상기 약제의 영향을 측정하고, iv)연결 부위가 사용된 형태의 mRNA의 핵으로부터의 방출에 대한 상기 약제의 영향을 측정한다(이로써, 연결된 형태의 mRNA 방출에 있어서 어떠한 감소도 없이 사용되지 않은 연결 부위를 포함하는 mRNA 방출의 감소에 의해 상기 약제가 HTLV-I rex 유전자의 활성 또는 rex 유전자 생성물의 활성의 특이적 저해제임을 알 수 있다).
