Abstract
본 발명은 HIV-1에서의 Rev 기능 및/또는 HTLV-Ⅰ 및 HTLV-Ⅱ에서의 Rex 기능을 억제하는 능력을 갖는, HIV-1 rev 유전자 생성물 또는 HTLV-Ⅰ의 rex 유전자 생성물로부터 유도된 바이러스성 유전자 표현형 발현의 트랜스우성형 억제제 및 상응하는 돌연변이 유전자를 기술하고 있다. 일련의 미스센스 및 결실 돌연변이체의 일시적인 유전자 발현 분석법을 사용하였다.
발견된 돌연변이체중 몇몇은 Rev 및 Rex 기능을 모두 저해함으로써 하나이상의 바이러스종에서 활성을 갖는다.
트랜스우성형 바이러스 돌연변이체는 유망한 새로운 부류의 항-바이러스제를 대표한다. 바이러스의 결실 작용, 예를 들면, HIV-1 감염으로부터 세포를 보호하기 위해서 세포내 면역화와 같은 치료 방법에 상기 트랜스우성형 저해제의 세포발현을 사용할 수 있다.

[발명의 분야]
본 발명은 바이러스성 유전자 발현, 더욱 특히 HTLV-Ⅰ의 rex(비리온-단백질 발현의 조절제)유전자 및 레트로바이러스종의 그의 등가물(예를들면, HIV-1의 rev)의 표현형 발현에 관한 것이다.
[발명의 배경]
바이러스, 특히 인간 면역결핍 바이러스 타입 1(HIV-1) 또는 인간 백혈병 바이러스 타입 I(HILV-Ⅰ)과 같은 인간 레트로 바이러스는 매우 중증의 질환에 대한 병원체이다. 이는 HIV-Ⅰ의 경우에 후천성 면역 결핍 증후군(AIDS)이고 HTLV-ⅠT-세포 백혈병(ATL)뿐 아니라 열대성 경련성 경증 페스트(Tropical Spastic Parapestis)로 알려진 비종양 질환이다. HTLV-Ⅱ는 몇몇 경우의 변이체 T-세포 모양세포성 백혈벙에 병인학적으로 관련된다. 2가지 바이러스 그룹은 모두 유전자 발현의 초기 및 말기단계에서, DNA바이러스와 유사하게 그들의 복제 주기가 분할되어 있다. 유전자 발현의 초기상태는 조절 단백질의 발현을 특징으로 하나, 말기상태에서는 구조 단백질이 합성된다.
HTLV-Ⅰ게놈은 Tax로 칭하는 바이러스성 전사의 활성화제를 암호화한다. HIV-Ⅰ에서 Tax의 등가물은 Tat로 칭한다. Tax 및 Tat는 바이러스성 유전자 발현시 주로 레트로바이러스성 LTR(긴 말단 반복단위)에 작용하는 것으로 보인다. 또한, HTLV-Ⅰ은 Rex로 칭하는 바이러스성 구조 유전자 발현의 활성화제를 암호환한다. 기능성 Rex 단백질은 핵으로부터 감염된 세포의 세포질내로 비연결 바이러스 mRNA의 이동을 증가시킨다. 여기에서 이들 mRNA종은 바이러스성 게놈을 구성하며 구조 단백질을 암호화한다. rev 유전자 생성물은 HTLV-Ⅰ시스템에서의 Rex 처럼 HIV-Ⅰ 구조 단백질의 발현에 절대적으로 필요하다.
상기에 대한 기초적인 이유는 rev유전자(및 다른 바이러스종의 그의 등가물)(의 생성물이 감염된 세포에서 바이러스성 mRNA 전사체에 대한 연결 방식의 선택에 뚜렷한 영향을 미치기 때문이다. 이러한 영향은 Rev 및 Rex의 경우에 전사후 조절, 즉 전체길이의 mRNA 전사체의 세포질내로의 이동의 증대에 의해 이루어지며, 이에 의하여 바이러스성 구조 단백질(예를들면, HIV-Ⅰ에 대해 Gag 및 Env)의 발현이 개시되고 조절 단백질의 발현이 동시에 억제되거나(예를들면, Rex에 대해 문헌[M. HIV-Ⅰ에서, 상기 언급한 유도의 선택성은 Rev 반응인자(PRE)로 칭하는 Rev 기능에 필요한 RNA 표적 서열에 기인한다. RRE는 HIV-Ⅰ env 유전자내에 존재하는 거대한 234개의 뉴클레오티드 RNA 2차 구조와 일치한다. HTLV-Ⅰ에서의 동등한 구조를 Rex 반응 인자(RexRE 또는 RRX)로 칭한다. Rev는 서열 특이적 방식으로 RNA의 핵 방출을 조절하는 것으로 알려진 첫번째 단백질인 것으로 생각된다.

조절 단백질의 돌연변이에 의해 야생 타입 기능에 대해 우성의 음성 표현형을 갖는 유전자 생성물이 수득될 수 있다[I. Friedman et al., Nature 335 [1988] 452]. 따라서, 헤르페스 단성 바이러스 트랜스-활성화제 VP16의 과도로 발현된 일부 결실 돌연변이체가 VP16기능을 저해하는 경우, 이에 의하여 정상적으로 자유로운 세포에서 HSV-1의 복제가 방해된다. 레트로바이러스에 관해서, 트랜스우성형 돌연변이체가 또한, 예를들면, HTLV-Ⅱ의 Tax 단백질 [W. Claims (48)
- HIV-1의 야생타입 Rev 단백질의 아미노산 위치 68번 내지 90번에서 하나 이상의 돌연변이를 가지며, HIV-1의 야생타입 rev 유전자의 표현형 발현을 트랜스우성형으로 억제하는 단백질을 암호화하는 돌연변이체 rev 유전자.
- 제 1 항에 따른 돌연변이체 rev 유전자에 의해 암호화된 단백질.
- HIV-1의 야생타입 Rev 단백질의 아미노산 위치 대략 10번 내지 대략 68번을 포함하는 제 1 영역 및 HIV-1의 야생타입 Rev 단백질의 아미노산 위치 대략 68번 내지 대략 90번을 포함하며 하나 이상의 돌연변이를 가짐으로써 변형된 제 2 영역을 포함하며 HIV-1의 야생타입 rev 유전자의 표현형 발현을 트랜스우성형으로 억제하는 돌연변이체 HIV-1 Rev 단백질을 암호화하는 돌연변이체 rev 유전자.
- HTLV의 rex 유전자의 표현형 발현을 트랜스우성형으로 억제하는 단백질 또는 그 단백질을 암호화하는 유전자, 또는 그의 기능성 절편 또는 유도체.

- Rex 단백질의 작용인자 활성을 나타내는 야생타입 Rex 단백질의 펩타이드 영역중에서의 하나 이상의 트렌스우성형 음성 돌연변이에 의해 야생타입 형태의 Rex 단백질로부터 변형되며 실질적으로 야생타입 형태의 Rex 단백질의 핵소체 정위활성을 갖는, HTLV-Ⅰ Rex 단백질 기능의 트랜스우성형 억제제.
- HIV-1의 야생타임 Rev 단백질의 특이적 결합 기능을 실질적으로 갖는 제 1 영역 및 HIV-1의 야생타입 Rev 단백질의 아미노산 위치 대략 68번 내지 대략 90번을 포함하며 하나 이상의 돌연변이를 가짐으로써 변형되고 HIV-1의 야생타임 Rev 단백질의 활성화 기능은 갖지 않은 제 2 영역을 포함하며 HIV-1의 야생타임 rev 유전자의 표현형 발현을 트랜스우성형으로 억제하는 돌연변이체 HIV-1 Rev 단백질을 암호화하는 돌연변이체 rev 유전자.
- 제 1 항에 있어서,HIV-1의 야생타입 Rev 단백질의 아미노산 위치 78번과 79번에서 돌연변이를 가지는 단백질을 암호화하는 돌연변이체 rev 유전자.
- 적절한 발현 시스템으로부터 상응하는 야생타입 유전자를 단리시키고, 단리된 유전자를 클로닝 시스템에 도입시키고, 목적하는 돌연변이를 상기 유전자내에 유발시키고, 목적하는 돌연변이가 일어난 클론으로부터 생성된 돌연변이체 rev 유전자를 회수함을 포함하는, 제 1 항에 따른 돌연변이체 ev 유전자의 제조방법.
- 제 1 항에 따른 돌연변이체 rev 유전자를 적절한 발현 및 증폭 시스템에서 발현 및 증폭시키고, 그로부터 생성되는 생성물을 회수함을 포함하는, 제 1 항에 따른 돌연변이체 rev 유전자에 의해 암호화되는 단백질의 제조방법.
