형질 전환된 대장균으로 부터 인간 α-인터페론의 정제방법

제2도는 S-세파로오즈 컬럼 크로마토그래피 양상,
제3도는 DEAE-Sephacel컬럼에서의 β-메르캅토에탄올농도 구배 용출 양상.

Claims (5)

  1. α-인터페론의 정제 방법에 있어서, 5mM EDTA, 50mM 아세트산 나트륨 완충액(pH 4.6)에 세포(E.coli)를 녹인 후 초음파 처리하여 유전자 재조합 α-인터페론을 불용성으로 추출하는 것이 특징인 α-인터페론의 정제방법.
  2. 제1항에 있어서, 0.6-0.8M 구아니딘-HCl 존재하에 1mM CuSO4를 가하여 재조한 α-인터페론을 정제한 후, 구아니딘-HCl을 제거하는 것을 특징으로 하는 방법.
  3. 제1항에 있어서, 1mM CuSO4를 포함한 상태로 α-인터페론을 Cu-킬레이트컬럼에 결합시켜 이미다졸 농도 구배 용출함을 특징으로 하는 정제방법.
  4. 제1항에 있어서, 재조합 α-인터페론을 S-세파로오즈 컬럼을 이용하여 정제함을 특징으로 하는 방법.
  5. 제1항에 있어서, 재조합 α-인터페론을 DEAE-Sephacel에 결합시켜 β-메르캅토에탄올로 5mM-0mM까지 직선 구배 용출시킴을 특징으로 하여 원래의 디술피드 본드를 이루고 활성이 강한 α-인터페론을 생산하는 방법.
