Claims (9)

  1. 부분적으로 정제된 인체 면역 인터페론을 함유하는 용을 고성능 약 양이온 교환 액체크로마토그라피컬럼상에 통과시키고, 상승되는 농도 구배의 염을 사용하여 칼럼으로 부터 증가된 순도의 인체 면역 인터페론을 함유하는 활성 분획을 용출시킴을 용출시킴을 특징으로 하여, 인체 면역 인터페론의 순도를 증가시키는 방법.
  2. 제1항에 있어서, 상술한 약 양이온 교환액체 크로마토그라피 칼럼이 카복시메틸 실리카인 방법.
  3. 제1항에 있어서, 상술한 칼럼을 pＨ7.5의 인산 칼륨 완충액중의 0 내지 Ｍ 구배의 KCl로 용출시키는 방법.
  4. 부분적으로 정제된 인체 면역 인터페론을 함유하는 용액을 실리카겔 투과 칼럼상에 통과시키고, 저 농도의 염을 함유하는 완충된 용액을 사용하여 칼럼으로 부터 증가된 순도의 인체 면역 인터페론을 함유하는 활성 분획을 용출시킴을 특징으로 하여, 인체 면역 인터페론의 순도를 증가시키는 방법.
  5. 제4항에 있어서, 상술한 용출제 용액이 pＨ7.5의 25mＭ 인산 칼륨 완충액 중의 0.3Ｍ KCl인 방법.
  6. a)조 인체 면역 인터페론의 용액을 상술한 인터페론을 흡착하는 친화성 칼럼에 통과시키고, 상술한 칼럼으로 부터 비흡착 불순물을 세척하여 상술한 칼럼으로 부터 인체면역 인터페론을 함유하는 활성 분획을 용출시키고,
    b)단계 a)로부터 모은 활성 분획을 고성능 약 양이온 교환 액체 크로마토그라피칼럼 상에 통과시키고, 상승되는 농도 구배의 염을 사용하여 칼럼으로 부터 증가된 순도의 인체 면역 인터페론을 함유하는 횔성 분획을 용출시키고,
    c)단계 b)로 부터 모은 활성 분획을 실리카겔 투과 컬럼 상에 통과시키고, 저농도의 염을 함유하는 완충된 용액을 사용하여 컬럼으로 부터 증가된 순도의 인체 인터페론을 함유하는 활성 분획을 용출시켜서 경우에 따라
    d) 단계 c)로부터 모은 활성 분획을 단계 b) 또는 단계 c)에 재순환시켜 균질한 인체 인터페론을 수득함을 특징으로 하여, 균질한 인체 면역 인터페론을 제조하는 방법.
  7. 제6항에 있어서, 상술한 공정 단계 a)의 친화성 칼럼이 콘카나발린-Ａ 세파로즈이며, 조 인체 면역 인터페론이 천연에 존재하는 물질이고, 활성 분획을 완충된 α-메틸-Ｄ-만노사이드 용액으로 용출시키는 방법.
  8. 제6항에 있어서, 상술한 단계 b)의 약 양이온 교환 액체 크로마토그라피 칼럼이 카복시메틸 실리카이며, 상술한 칼럼을 pＨ7.5의 인산 칼륨 완충액중의 0내지 /Ｍ 구배의 KCl로 용출시키는 방법.
  9. 제6항에 있어서, 상술한 실리카겔 투과 칼럼을 pＨ7.5의 25mgＭ 인산 칼륨 완충액 중의 0.3 M KCl용액으로 용출시키는 방법.
