제1도에서 제4도까지는 농도를 달리하는 아질산염의 존재하에서의 종속영양적(heterotrophic)생육 이후의 Nitrobacter nov spec T3속의 생육 곡선을 나타낸 도면임.
Claims (9)
- 초기 수용액중에 함유된 질산염을 질화세균에 의해 환원시키고, 이와같이 얻어진 반응생성물인 암모니아와 아질산염을 연속적인 산화공정에 의해 질산염으로 재전환시켜서, N2O나 NO와 같은 기체상의 질소 화합물을 방출시킴을 특징으로 하는 질화세균에 의한 질화 및 질소제거 방법.
- 제1항에 있어서, 질산염 및 아질산염의 환원을 각각 질화세균에 의해 낮은 산소분압에서 수행함을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, 질화/질소제거 겸용방법을 암모니아, 아질산염 및/또는 질산염으로 오명된 천연수 또는 인공수에 대해 본래 위치의 (in situ)균을 공급함으로써 수행함을 특징으로 하는 방법.
- 제3항에 있어서, 상기 세균이 고정되고 이어서 활성탄, 제올라이트, 도자기류 또는 이온 교환기과 같은 구멍이 열린 다공성 지지체상에 건조된 질화세균인 것이 특징인 방법.
- 제4항에 있어서, 이용된 질화세균이 아질산염 산화세균, 예컨대,Nitrobacter속인것이 특징인 방법.
- 제4항에 있어서, 이요된 질화 세균이 아질산염 산화균과 암모니아 산화균인 것이 특징인 방법.
- 선택된 조건하에서 토양 시료로부터 분리되고 단독으로 20~30℃의 온도에서 교반이나 와동시키지 않으면서 종속영양적으로 생육하며 다음의 특징, 즉 (a) 혼합영양 및 무기독립영양적으로 보다는 종속영양적으로 더 잘 생육하고, b) 혼합영양적으로 배양시켰을 때 50㎎ 총 세포단백질/L의 높은 세포수율을 나타내며, c) 미호기적 조건하에서 최소 6개월이라는 장기간의 시간동안 배자에 단독으로 남겨졌을 때도 생존 능력을 과시하며, d) 산소포화 배지에서의 미호기적 종속영양적 생육 후 산소에 내성을 가지며, e) 산소를 공급한지 2-4일 후에 미호기적 생육후 질화 작용을 개시하고, f) 본 발명에 따른 방법에서 우수한 반응성을 나타내며, g ) 분자량이 80±3인 플라스미드를 갖는 것으로 특징지워지는 독일연방공화국 디-3400 괴팅겔에 소재하는 German Collection of Microorganisms는에 기탁번호 제DSM 4153호로 기탁된Nitrobacter속의 미생물.
- 생체내에서 공유적으로 밀집된 원형의 (Covalently colsed circular)DNA이고, 0.5% 아가로스 겔에서 전기영동적인 이동률로 측정했을 때 분자량이 약 80±3MD인 것이 특징인 제7항의Nitrobacter속 미생물로부터 유래한 플라스미드 DNA.
- 제8항에 있어서, 플라스미드 DNA가 제7항의Nitrobacter속의 미생물을 호기적 또는 미호기적 조건하에서 무기독립영양, 혼합영양 또는 종속영양적으로 세포배양시켜 생체내에서 복제시킴으로써 얻을 수 있는 것임을 특징으로 하는 플라스미드 DNA.※ 참고사항:최초출원 내용에 의하여 공개하는 것임.
