리포솜을 이용한 신규의 면역분석방법

제1도는 다층 리포솜내의 스테아릴아민 함량변화에 따른(막내글루코스농도 대 막외글루코스농도의 비)-(450mm에서의 흡광도의 역수)3/2의 그래프를 나타내며[ ●; 스테아릴아민농도 7.3몰%, □; 14몰%, △; 24몰%. ○; 32몰%].
제2도는 면역리포솜에 대한 항혈청의 영향을 나타내며[ ●; 항-BSA토끼혈청, ○; 정상토끼의 혈청].

Claims (8)

  1. 리포솜내에 효소를 봉입한 후 브롬화시안(CNBr)으로 처리하여 리포솜표면에 노출된 효소를 불활성화한 후, 면역기능을 갖도록 그 표면에 항체 혹은 항원을 결합시킨 다음 냉동건조 시켜서 얻어진 면역리포솜.
  2. 리포솜내에 효소를 봉입한후 브롬화시안(CNBr)으로 처리하여 리포솜표면에 노출된 효소를 불활성화한 후 면역기능을 갖도록 그 표면에 항체 혹은 항원을 결합시킨 다음 냉동건조시켜서 면역리포솜을 제조하는 방법.
  3. 세포성 면역반응에 의해 특이하게 일어나는 세포용혈현상과 효소-기질의 특이적 반응을 이용하여, 혈맥, 헐청, 뇨, 타액 등과 같은 생리액이나 기타 미지시료중의 약물, 항원성물질, 알러지원 등을 신속, 간편하게 분석함에 있어서, 1) 리포솜내에 효소로서 알칼린 포스파타아제 또는 퍼옥시다아제를 봉입시킨 후 브롬화시안으로 처리하고, 항체 혹은 항원을 결합시켜 제조한 면역리포솜 ; 2) 기질인 파라-니트로페닐포스레이트 또는 과산화수소 수용액 ; 및 3) 기니픽보체를 각각 관리 포장하였다가, 분석시에 시료와 혼합하여 사용하는 진단용 시약키트.
  4. 제2항에서 면역리포솜을 제조할 때, 스테아릴아민을 사용하여 봉입효율을 증가시키는 방법.
  5. 제2항에서 면역리포솜을 제조할 때, 10몰% 이상의 콜레스테롤을 사용하여 리포솜의 봉입효율을 증가시키는 방법.
  6. 제2항에서 면역리포솜 제조시, 스테아릴아민과 글루타르알데히드틀 이용하여 항체 흑은 항원을 결합시키는 방법.
  7. 제2항에서 면역리포솜 제조시, 포스파티딜세린을 함유한 리포솜표면의 환원에 의해 항체 혹은 항원을 직접 결합시키는 방법.
  8. 제7항에서 환원제로 보로하이드라이드를 사용하는 방법.
