제1도는 인체의 IFN-α1 (pre D), IFN-α2 (pre A), IFN-α1, 2 (preD/A), IFN-ν (pre ν), 그리고 IFN-β (pre β)의 신호용 프레펩티드의 아미노산 결합 서열의 비교도이다. 밑줄을 그은 아미노산은 pre A와 pre의 아미노산 결합서열의 차이를 나타낸다. Dde 1부위는 혼성 pre D/A프레시퀸스제조물에서의 DA및 프레시퀸스의 접합점을 나타냄.
제2도는 여러가지 유전자 삽입물의 표현을 위한 이란적인 매체로서 본 발명에서 사용되는 효모 표현 플라스미드 YEpIPT의 그림으로 제한 부위 및 흥미있는 여러가지 부위를 보여준다.
제3도는 효모에서 IFN-α1의 직접 표현을 위한 플리-플러스(pre-plus) IFN-α1 유전자를 함유한 EcoRI 단면의 구조이다.
Claims (15)
- 효모 유기체에 의하여 표현, 처리 및 분리되고 불필요한 폴리펩티드 프리시퀸스 또는 기타 표현가공물을 동반하지 않는 분절된 형태의, 상기 효모에 대하여 이종성인 단백질제품.
- 제1항에 있어서, 상기 제품이 인체 인터페론인 제품.
- 제1항에 있어서, 상기 제품이 인체의 백혈구 인터페론인 제품.
- 제1항에 있어서, 상기 제품이 인체의 백혈구 인터페론 A 또는 D인 제품.
- 효모 유기체에 대하여 이종성인 단백질을 생성시킬 수 있는 효모 유기체 세포 배지를 구성함에 있어서, 상기 단백질 암호화 DNA용의 이종성 신호 폴리펩티드와 함께 상기 DNA를 기능적으로 잠복시키는 표현 매체로 변이시킨 생효모 세포 및 이 세포를 담지시키고 상기 유기체의 표현, 처리 및 분비 생성물인 단백질을 함유하는 매걸로 구성되는 것이 특징인 효모 유기체에 대하여 이종성인 단백질 생성능을 갖는 효모 유기체 세포 배지.
- 제5항에 있어서, 상기 효모 유기체가 사카로마이세스 세레비지아에인 세포배지.
- 제5항에 있어서, 상기 이종성 신호 폴리펩티드가 상기 단백질에 기원을 둔 신호 및 제2 이종성 신호 폴리펩티드의 혼성체(잡종)인 세포 배지.
- 제5항에 있어서, 실질적으로 생효모 또는 분열 효모 세포가 없고, 효모 유기체의 표현, 처리 및 분비 생성물로서 상기 효모 유기체에 대하여 이종성인 단백질을 함유하는 효모유기체 세포 매질.
- 분절된 형태의 이종성 단백질을 그 매질내에 방출할 수 있는 효모 유기체 세포 배지.
- 제9항에 있어서, 효모 유기체가 사카로마이세스 세리비지아에인 세포배지.
- 효모 유기체에 대하여 이종성인 단백질을 암호화 하는 DNA를 기능적으로 잠복시키는 효모표현매체 YEp1PT
- 제11항에 있어서, 이종성 단백질을 암호화하는 DNA로 번역 해독 구조(골격)내부의 이종성 신호폴리펩티드를 암호화하는 DNA를 추가로 잠복시키는 매체.
- 제12항에 있어서, 상기 이종성 신호 폴리펩티드가 상기 단백질에 기원을 둔 신호 및 제2이종성 신호 폴리펩티드의 혼성체(잡종)인 매체.
- 인체의 백혈구 A단백질로 번역 해독 구조(골격)내부의 인체 백혈구 DA 신호 폴리펩티드를 암호화하는 DNA 를 기능적으로 잠복시키는 효모 표현 매체.
- 단백질 및 이종성 시호 폴리펩티드를 암호화하는 DNA를 기능적으로 잠복시키는 표현 매체로 효모 유기체를 변성시키고, 상기 유기체의 배지를 조제시켜 이 배지를 생장시킨 다음 그 배지 매질로부터 단백질을 회수하는 것이 특징으로 하여 효모에 의한 표현, 처리 분비 생성물인 상기 효모유기체에 이종성인 단백질의 제조방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
