Description
본 발명은 선택된 탄소원(Carbon Source)에서 박테리아의 작용에 의하여 생성되는 신규 헤테로 다당류의 제조방법에 관한 것이다. 더우기 본 발명은 조절된 조건하에서 선택된 탄소원 및 발효 배지 성분을 박테리아 발효함으로서 헤테로 다당류를 제조하는 신규방법에 관한 것이다. 본 발명의 헤테로 다당류는 대부분의 탄수화물 잔사 및 소량의 단백질을 함유하는 고 분자량의 다당류이다. 이것은 때때로 “검(gum)이라고 칭하여 지나, 헤테로 다당류란 용어가 보다 정확하다고 생각된다. 본 발명의 다음 설명에서, 이것을 때때로 헤테로 다당류 60, 또는 S-60이라고 칭한다.
헤테로 다당류가 흑종의 미생물에 의하여 제조될 수 있음은 공지되어 있다. 이와같은 헤테로 다당류 중몇몇은 친수교질로서 작용하며 그의 정도 및 유동성으로 인하여 수성기(aqueous system)용 농후제로서 사용되어 왔다.
기타 공업 기술분야에서, 농후제 현탁제 및/또는 안정제로서 유용한 성질을 가진 신규 헤테로 다당류를 발견할 목적으로 연구가 계속 되어왔다. 본 발명의 목적은 상술한 소망하는 성질을 가진 신규 헤테로 다당류를 제공함에 있으며, 본 발명의 다른 목적은 상술한 신규 화합물을 제조하는 방법을 제공함에 있다. 본 발명의 또 다른 목적은 농후제, 현탁제 또는 안정제로서 본 발명의 신규헤테로 다당류를 함유하는 제제를제공함에 있다. 기타 본 발명의 목적은 본 발명의 후술하는 설명으로부터 명백해질 것이다.
본 발명의 신규 화합물은 광대한 분류학 연구를 기초로하여 지금까지 기술되지 않은 미생몰 [이것은 명명된바 없는 슈도모나스종(Pseudomonas species)이다]을 가지고 적당한 배지를 발효함으로서 얻어진다. 본 발명의 헤테로 다당류를 제조하는데 이용되는 상술한 미생물은 수납번호 ATCC 31,461로 1978년 11월21일 어메리컨 타이프 컬처 콜렉션(American Type Culture Collection)에 무제한으로 영구 보관되어 있다.

이들 분류표 및 설명은 ATCC 31,461과 유사한 형태학적 및 배양특성을 가진 슈도모나스종에 대한 조사하였다. 다음의 고찰은 신규 슈도모나스종의 지정을 정당화 및 필요하게 하여준다.
균주의 설명
1. 세포 형태학적 특성
단일세포, 직선 토는 때때로 만곡, 일반적으로 0.6-0.8×2.0-3㎛, 때때로 끝이 뾰족함, 더 오래 배향하면 보다 커지고 보다 길어지며(0.8-1.0x>3㎛), 특히 제한량의 탄수화물을 가진 배지에서 기형세포 및 다형성이 나타난다. 이에 반하여 세포는 탄수화물을 가진 배지에서 생장시키면 일관된 봉상을 유지하나 대분부의 세포는 다시 커지기 시작하며 장기간 배양하는 동안 다형성이 나타난다. 그램-음성이며 돌기가 없는 폴리-β-하이드록시부티레이트 및 폴리포스페이트 입상이 질소-결핍 배지의 배양시에 특히 나타난다. 폴라 멀티트리코우스(polar Mulritrichous)편모운동에 의하여 움직이며. 1-4개의 편모는 극단부에 삽입되어 있으며, 때때로 서브폴라 삽입이 나타날 수 있다.
2. 집락(集落)의 형태학적 특성
육즙 한천평판에서 소형(직경0.8-1.1mm) 및 대형(직경 3.2-3.5mm)집락이 나타난다. 이들은 황색 카로티노이드 염색된 매끄러우며 둥글고 철형(凸形)내지 침상형이다. 때때로 대형집락은 동심 주름을 가진다. 이들 집락의 표면은 경질이긴 하지만 점착성 구조를 가진것은 아니며, 모든 집락은 루우프(loop)로 미는경우 제거된다. YM한천판에서 단지 한가지 형태의 비교적 대형(약6-7mm직경)의 황색이며 둥글고 매끄러우며 끈적끈적한 철형 집락만이 나타난다. 끈적끈적한 탄성막피가 이들 집락의 표면에 형성되며. 집락의 표면막피 전체는 제거될 수 있다. 제 2 차 성장이 최초 집락의 변두리 주위에 나타난다. 이들 집락의 색은 변두리 보다도 중심을 향하여 더 검은 황색이며 동심적인 색형성이 나타난다. 세포내 황색 카로티노이드색소 이외에 확산성 갈색 색소가 장기간 배양후 자동 산화 결과로서 나타났다. 이 현상은 육즙 한천에서보다 용이하게 알 수 있다. 형광성 색소는 생성되지 않았다.
3. 생리학적 및 생화학적 특성
균주 S-60의 성장 범위는 약20-41℃이다. 4℃에서 성장이 나타나지 않음. 3.0% NaCI 은 성장억제에충분하며, 균주는 pH 5-11에서 성장할 수 있다.
산이 거의 모든 탄수화물로부터 생성되나 다가알코올로부터는 생성되지 않으며, 가스생성이 없다. 우레아제(crease)는 생성될 수 있다. MR, VP, 및 인돌 시험은 모두 음성이었다. 아르긴 디하이드롤라제, 리신 및 오르니틴 데카복실라제는 생성되지 않는다. 산 및 환원은 리트머스유(miIk)에서 나타난다. 지방분해난환반응은 음성이었다. 젤라틴은 약간 가수분해 되지만 카제인, 전분, 알기네이트, 펙틴, 셀룰로우스, 키틴 및 DNA는 가수분해 되지 않는다.
4. 항생물질에 대한 감수성
균주는 가나마이신, 네오마이신. 클로르테트라사이클린, 에로스로마이신에 대하여 매우 감수성이 있으나 스트렙토마이신 및 페니실린에 대하여는 감수성이 없다.
5. 영양적 특성
유기 성장 인자는 필요치 않으며. 암모늄염은 오직 질소원으로서만 작용한다. 최소한 35종의 유기화합물이 이용되었으며, 이들 화함물은 D-리보스, 전분, 2-케토글루코네이트 및 무케이트가 아닌 대부분의 탄수화물이다. 이외에 아세테이트, 카프로에이트, 카프릴레이트, 페라고네이트, 석시네이트, 아젤레이트, L-말레이트, DL-β-하이드록시부티레이트, 피루베이트, 에탄올, n-프로파날, p-하이드록시 벤조에이트. 페닐아세테이트. L-α-알라닌, L-트레오닌. L-루우실. DL-이소루우신, L-아스파르테이트, L-글루타메이트 및 L-티로신이 이용되었다.
6. DVA의 G+C함량
DNA의 평가는 약 68몰%(Tm에 의하여)로 나타났다.

균주 S-60에 이용된 생화학 및 기타 각종시험
헤테로 다당류 S-60은 명명되지 않은 슈도모나스종의 미생물조 배양한 다음의 조절된 조건하에서 적당한 수성 영향배지의 호기성 발효중에 생성된다. 이 배지는 탄소원, 질소원 및 무기염원을 함유라는 통상적인 배지 이다.
일반적으로, 탄수화물(예컨대 글루코스 프락토스, 말토스, 슈크로스, 크실로스 만니톨등)은 영양배지에서 단독적으로 또는 융합 가능한 탄소원으로서 혼합되어 사용될 수 있다. 배지에 사용되는 탄수화물원의 정확한 량은 부분적으로 배지의 기타 성분에 의존하지만, 일반적으로 탄수화물의 량은 배지의 약 2-4 중량%사이이다. 이들 탄소원은 개별적으로 사용되거나 또는 이들 탄소원 몇개가 배지에 혼합될 수 있다. 일반적으로, 많은 단백질성 물질이 질소원으로서 발효 공정에 사용될 수 있다. 적당한 질소원에는 예컨대 효모 가수분해물, 1차효모, 콩가루, 면실가루, 카제인의 가수분해물, 옥수수액, 증류기의 용해물 또는 토마도 반죽등이 있다. 질소원은 단독으로 또는 혼합되어 수성배지의 약 0.05-0.2중량% 범위의 양으로 사용된다.
배지에 혼합할 수 있는 영양 무기염중에는 나트륨, 칼륨, 암모늄, 칼슘, 인산염, 황산염, 염화물, 탄산염등을 줄 수 있는 통상의 염이 있다. 코발트, 망간, 철 및 마그네슘같은 미량 금속이 또한 포함된다.
본 실시예에 기술된 배지는 사용가능한 여러가지 배지중의 예에 불과하며 제한적인 것은 아니다.
발효는 약 25-35℃의 범위에서 수행되나, 최적결과를 위하여는 약 28-32%의 온도에서 발효를 수행하는 것이 바람직하다. 슈도모나스 배지를 성장시키고 다당류 S-60를 제조하기 위한 영양배지의 pH는 약 6-8사이 이다.
신규 다당류 S-60가 표면 및 심부(Submerged) 배양에 의하여 제조되긴 하지만, 심부 상태에서 발효를수행하는 것이 바람직하다.
소규모 발효는 적당한 배지에 상기 배양물을 접종시켜 수행하는 것이 편리하며, 생산 배지에 옮긴 후 발효는 수일동안 진탕기에서 약 30℃의 일정한 온도에서 진행된다.
발효는 1단계 이상의 종자성장 단계를 거쳐 무균 배지 플라스크에서 개시된다. 종자단계시의 영양배지는 적당히 혼합된 탄소와 질소원일 수 있다. 종자 플라스크를 약 30℃에서 1-2일동안, 또는 성장이 만족스러울 때까지 항온실에서 진탕시키고, 이렇게 하여 성장된 일부를 제 2단계 종자 또는 생산 배지에 접종시키는데 사용한다. 중간단계 종자 플라스크가 사용되는 경후 근본적으로 동일한 방법으로 성장시킨다. 즉 최종 종자단계로 부터의 플라스크 내용물의 일부가 생산배지에 접종시키는데 사용된다. 접종된 플라스크를 수일동안 일정한 온도로 진탕시키고. 접종기간 마지막에 플라스크의 내용물은 이소프로판올과 같은 적당한 알코올을 사용하며 침전시켜 회수한다.
대규모 작업시에는 교반기 및 발효 배지를 통기시키기 위한 장치가 장착된 적당한 댕크(tank)에서 발효를 시키는 것이 바람직하다. 본 발명에 따르면, 영양배지는 탱크에 채워져 약 121℃까지의 온도에서 가열에 의하여 멸균된다. 냉각시킨후 멸균된 배지는 생산배지의 이미 성장한 종자로 접종시키고, 발효를 예컨대 2-4일동안 약 30℃로 유지시키며 영양배지를 교반 및/또는 통기시키면서 처리한다. S-60을 제조하는 이와 같은 방법은 대량 제조에 특히 적당하다.
생성물은 이소프로판을과 같은 적당한 알코울로 침전시켜 발효배지로부터 회수한다.
다당류 S-60의 물리화학적 성질
명명되지 않은 슈도모나스종에 의하여 제조된 헤테로 다당류는 주로 탄수화물, 0-글리코사이드 결합된 에스테르와 같은 아세틸 그룹 3-4.5%, 및 단백질10-15%로 구성되어 있다.
S-60다당류의 탄수화물 부분은 우론산(약 12% b.0. wt검) 및 중성(neutral)당 글루코스 및 람노스를함유하고 있다. 람노스 대(對)글루코스의 대략 몰비는 1.5 : 1이다.
4.5%의 아세틸 함량은 S-60검의 0.2% 수용액을 알카리성 하이드록실아민 시약으로 처리한 다음 산성제 2염화철시약으로 처리하여 측정하였다[S. Hestrin (1949) J. Biol. Chem. 180 249-261]
다당류 S-60의 증성 당은 2ml의 2N H2SO4생성물 10mg을 용해시킨 다음에 혼합물을 100℃에서 4시간 동안 가열하여 측정하였다. 얻어진 용액을 냉각시킨 다음 수산화바륨으로 중화시키고, pH는, 고체 이산화탄소로서 S-6으조 조절하였다. 이렇게하여 얻어진 황산바륨의 침전물을 원심분리하여 제거한 다음 상정액은 감압하에 농측시켜 시럽으로 만들었다. 가수분해물중의 당은 210℃에서 80/100메쉬 가스크롬(Gas Chrome)Q상의 OV-225 3중량 %를사용하는Hewlett-Packard Model 5750 크로마토그래피상에서 그의 알드노니트릴 아세테이트 유도체의 기체-액체 크로마토그래피에 의해 시험적으로 확인된다. 당은 확실한 표준품과 비교하여 확인 및 정량한다[J.K. Baird, M.J. Holroyde, 및 D.C. Ellwood (1973) Carbohydr. Res. 27. 464-467].
또한 다당류의 각종 중성당은 용매로서 피리딘 : 에틸아세테이트 : 물(2:5:5)의 상층을 사용하는Whatman N0. 1여지 크로마토그래피에서 강하하는 여지 크로마토그래피를 사용함으로서 특징지워진다. 크로마토그램은 질산은 침적 및 산성 아날린 프탈레이트 분무시약을 사용하며 착색시켰다. 당성분은 표준당으로 공크로마토그래피하여 그리고 아날리프랄레이트 시약으로 비색반응 시킴으로서 확인하였다.
다당류의 우론산 함량은 두 분리방법에 의하여 측정하였다. 한방법에서는 샘플을 19%염산으로 탈카복실화시킨다음 유리된 이산화탄소를 표준 수산화나트륨에 포집하여 역적정법에(back titration)[B.L. Browning(1967) Methods of wood Chemistry Ⅱ. 632-633]및 카바졸 측색계방법 [T. Bitter H.M. Muir (1962)Anal. Biochem. 4 230-334)에 의하여 측정하였다.
여지 전기영동은 상술한 중화된 산수가분해물에 존재하는 우론산의 분리및 시험적인 확인을 위하여 사용하였다. 공지 우론산 표준물을 카막(Camag) 전기영동 여지 제 68-011호에 가하고 카막 모델HVE전기 영동장치를 사용하여 pH 2.7완충액에서 2.0시간동안 전기영동을 수행하였다. 크로마토그램을 공기 건조시킨 다음 분리된 우론산을 알아볼 수 있게 하기 위하여 질산은 침적시약으로 착색시켰다. 두개의 큰 우론산반점 및 하나의 작은 우론산 반점이 발견되었다. 큰 반점중 하나는 글루쿠론산(RGLCA=1.0)과 동일한 이동도로 이동하였으며 다른 큰 반점(RGLCA=0.85) 및 작은 반점(Rglca=0.73)은 보다 이동이 낮았다. 이들동일 조건하에서 공지 우론산의 상대적인 이동성은 다음과 같다. 즉,
천연 S-60의 적외선 스펙트럼은 KBr펠렛(Pellet)의 건조 물질에서 행하였다.
헤테로 다당류는 3.400cm-1, 2,950cm-1, 1,740cm-1및 1,620cm-1의 피이크에서 하이드록실기, 매틸렌기카보닐기 및 카복실산염을 나타낸다. 이것은 염화메틸렌 염료와 상반되며, N,N-디메틸포름 아미드에선 상당히 불용성이며, DMSO 또는 포름아미드에선 가용성이다.
S-60의 샘플의 원소분석 결과는 N-2.00%, C-42.62% H-5.80%이다.
다당류 S-60은 낮은 농도로 물에 용해되면 수성 배지에 점성을 부여한다. 전단에 대한 그의 감수성 및 전반적인 유동학으로 인하여, 다당류 S-60은 수성기에서 농후제, 현탁제 및 안정제로서, 예컨대 직물날염페이스트에 대한 부가제로서 또는 유동성이 낮은 수성제초 조성물, 샐러드 드레싱, 농후푸딩 및 접착제 조성물의 제제에 유용하다. 이것은 가열 및 냉각후에 미약한 탄력성 겔을 형성한다.
탈아세틸화되고 정화되지 않은 S-60
무수 폴리미 또는 발효 육즙(broth)을 높은 pH(예컨대 탄산나트륨 또는 수산화나트륨을 사용하여 pH10까지)및 90-100℃의 높은 온도에서 10-45분동안 가열하면 탈아세틸화가 용이하게 일어난다. 이와같이하여 탈아세틸화된 다당류 S-60은 많은 공업분야 및 식품분야에 유용한 견고한 비-탄성 또는 취성 겔을 형성한다. 탈아세틸화 S-60의 조성은 주로 탄수화물, 약 17%의 단백질 및 0%의 아세틸이다. 탄수화물 부분은 약 13%의 우론산과 몰비가 약 1.5:1인 중성당 람노스 및 글루코스로 구성되어 있다.
견고하며 취성 겔인 성질에 기인한 탈아세틸화 검에 대한 한가지 용도는 견고한 구조로서 주조되고 사용될 수 있으며 향료와 같은 적당한 용액으로 처리한 후 실내 방취제 또는 공기 청신제등으로 응용할 수 있다는 점에 있다. 또한 탈아세틸화 검은 전자 현미경 검사에서 마이크로 톰에 사용하기 위한 겔화제로서 겔전기 영동에 사용될 수 있다. 천연 및 탈아세틸화검은 방사선학에서 바리움용 현탁제로서, 제과제품에서 및 공구제조, 치과 및 범죄학에서 각인 물질로서 또한 사용될 수 있다.
탈아세틸화 S-60의 적외선 스펙트럼은 KBr펠렛형태의 건조물질에서 행하면, 3,400cm-l, 2,950cm-1, 1,740cm-l, 1,650cm-1및 1,610cm-1에서 피이크를 보여준다.
탈아세틸화 S-60의 샘플은 N-2.60%, C-41.89%, H-6.07%의 원소분석치 를 보여준다.
탈아세틸화되고 정화된 S-60
정화되고 탈아세틸화된 S-60의 조성물은 다음과 같다. 즉 약 2%의 단백질, 0%아세틸, 및 탄수화물로 조성되어 있으며, 탄수화물은 약 22%의 우론산 및 몰비가 약 1.5:1인 중성당 람노스 및 글루코스이다.
정화되고 탈아세틸화된 S-60의 적외선 스펙트럼은 KBr펠렛형 건조물질에서 행하면, 3,400cm-1, 2,950cm-l, 및 1,600cm-1에서 피크를 보여준다.
특히 정화된 탈아세틸화 검은 광범위한 각종 배지를 사용하는 각종 임상 및 비-임상 미생물에 대한 미생물 배지에서의 한천대용물로서 유용하다. 한천으로 대치하기에 필요한 정화된 탈아세틸화 검의 농도는 사용되는 배지에 의존하나, 약 0.5-1.25%범위 이내이다. 미생물의 성장 특성은 표준 한천을 기제로한 배지어서의 특성과 아주 유사하다.
본 발명을 실시예에 의거 상세히 설명하면 다음과 같다.
[실시예 1]
혜테로 다당류 S-60을 제조하기 위한 발효공정
A. 배양 유지
명명되지 않은 슈도모가스 유기체, ATCC 31,461은 배양유지에 일상적으로 사용되는 NA 또는 YM한천상에서 아주 잘 성장한다. 배양온도는 30℃이다. 미생물은 2-5일의 배양에 의하여 황색-오랜지색 카로티노이드 색소 및 갈색 분산성 색소를 생성한다.
B. 종자 제조
플라스크 종자를 30℃에서 배양된 YM육즙내에서 제조한다. 새로운 평판 배양물로 접종하면 YM육즙 배양물은 24시간 지나면 양호하게 성장하여 검을 형성한다.
발효 종자배지는 종자 용기로서 1갈론들이 발효기를 사용하는 최종 발효기 배지와 동일하다.
C. 최종 발효기 배지
검의 나트륨-및 칼륨-염 형태는 하술하는 바와같이 상이한 배지에서 제조된다. 이 미생물은 일정한K+요구량을 가지는데 나트륨 발효 배지에 이것을 가해야만 한다. (3% 덱스트로스가 또한 사용될 수 있음)또한 0.2%옥수수 침적액을 두 배지에 가한다.
홀레염은 주석산, 몰리브덴 마그네슘염, COCl3, ZnCl2, CuC12,붕산, 염화망간 및 황산제 1철을 함유하는 미량원소 용액이다.
낮은 칼슘 생성물이 요구되는 경우, 상출한 배지는 탈이온수와 함께 사용된다. 발효는 약 50시간에 완결되며, 발효액 점도(bear viscosity)는 통상 5,000-8,000cps이다.
D. 회수
생성물의 겔화 성질로 인하여, 양호한 섬유형성은 통상적으로, 주위 침전과 함께 일어나지 앓는다. 그러나 본 발명자들은 10-15분동안 90-95℃에서 저온 살균에 의하여 우수한 섬유는 냉각시킴이 없이 발효액의 용량당 99% 이소프로판을 2용량을 사용하여 발효액을 침전시키므로서 얻어질 수 있음을 발견하였다. 1.5%검의 평균 수율은 20L 및 70L발효기에서 3%글루코스를 사용함으로서 얻어진다.
E. 건조
생성물을 회수하며 공기압송(forced-air tray)건 조기에서 1시간 동안 50-55℃에서 건조시킨다.
F. 생성물의 질
K+염의 1%점도는 통상 3,000cps범위이며, 낮은 칼슘 나트륨염은 약 7,000cps이다.
[실시예 2]
헤테로 다당류 S-60과 탈아세틸화 및 정화
모든 용도에 대하여 반드시 필요한 것은 아니지만, 검의 정화는 검이 한천 대용물로서 사용되는 경우 가치가 있다. 정화는 탈아세틸화전(천연 상태로)이나 또는 탈아세틸화후에 성취할 수 있다. 탈아세틸화는 고온 부식제를 사용하여 정화도 고온으로 행해지기 때문에 두 공정은 용이하고 편리하게 결합된다. 탈아세틸화 및 정화는 발효액 또는 무수 폴리머로 달성될 수 있다. 탈아세틸화에 대하여, 발효액이 사용되는 경우, pH를 KOH로 10로 조절하고, 용액을 15분동안 90℃까지 가열하고, pH를 묽은 H2SO4로 조절한 다음 CaCl2를 가하여 농도를 0.2%로 하여 냉각시킨다. 그러한 견고한 취성겔이 얻어진다.
탈아세틸화 및 정화에 대한 일반적인 공정은 다음과 같다.
A. 발효액 또는 검의 2%용액을 90℃까지 가열한다.
B. pH를 KOH로 10로 조절한다.
C. 발효액 또는 용액의 온도를 15분동안 90-95℃로 유지시킨다.
D. PH를 묽은 HCI 또는 H2SO4로 6-8로 조절한다.
E. 슈퍼 에이드(Super Aid)(10g/l)을 여과하고자 하는 물질에 가한다.
F. 이 물질을 136cm2의 면적을 가진 여과장치를 사용하여, 약 6mm 상(bed)의 슈퍼 에이드 및 약 20-30psi의 압력을 가진 압력여과장치(예열된)를 통하여 여과한다.
G. 여액을 즉시이소프로판올로 침전시켜 겔화를 방지한 다음 섬유를 1시간 또는 그 이하의 시간동안 50℃에서 건조시킨다.
탈아세틸화가 필요없는 경우, pH를 상승시키지 않는것 이외에 온도는 90℃로 유지하면서 상기 공정을 반복하고, 용액을 즉시 여과한 다음 회수한다.
정화는 칼륨형태에서 항시 행하여지며, KCI은 필요에 따라 이미 제조된 생성물의 용액에 가할 수 있다.
[실시예 3]
헤테로 다당류 S-60겔화 투성
카라기난 및 한천으로 K+형태 및 Ca++형태의 천연검및 탈아세 틸화검을 비고한 데이타는 다음과 같다.
세균학적 특성 : 겔화온도 33-39℃, 융점 70℃(최소)(Whistler “Industrial Gums”)
최소 겔화 농도
캅파 카라기난 -0.3% 탈아세틸화 S-60 -0.05%
한천 -0.04% (칼슘 겔)
상술한 바로부터 알 수 있는 바와같이 모든 각종 형태의 겔의 경화 및 융점에 대하여 주어진 온도범위가 광범위하다. 한천에서의 편차는 우선적으로 해초형태에 기인하여, 한편 캅파 카라기난에서의 칼륨이온 농도는 겔 특성을 결정한다. 탈아세틸화 S-60의 겔은 주로 탈아세틸화 정도에 의하여 특정지워진다. 단지약간의 탈아세틸화에 의하여 겔은 보다 높은 온도에서 경화되며 탄성이 더 커진다. 사실상 보다 큰 탄성으로부터 보다 큰 취성까지의 광범위한 겔 형태는 탈아세틸화 정도에 의거한다. 겔은 주로 경화 및 융해 온도 사이의 큰 이력 현상 때문에 캅파 카라기난 보다도 한천에 보다 유사한것 같다. 이들은 융해하기 가 곤란하고, 겔-졸전이는 관찰하기가 곤란함을 알아야 한다. 한편 겔화온도는 겔이2-3도내에서 초기 겔화로부터 고체겔로 신속하게 경화하기 때문에 용이하게 한정될 수 있다.
[실시예 4]
비-정화되고 탈아세틸화된 S-60
S-60발효액을 90℃까지 가열하고 pH는 25% KOH를 가하여 10로 조절한다. 온도를 l5분동안 유지시킨 다음 농축 HCI로 중화시킨다. 이 액체를 가열하면서 발효기로부터 배수시킨 다음 99% IPA 2배량으로 회수한다. 탈 아세틸화 S-60의 섬유를 모아 공기압송 건조기에서 1시간동안 55℃에서 건조한 다음 분쇄하여 분말을 만든다.
[실시예 5]
탈아세틸화 S-60의 정화
실시예4에서 제조한 탈아세틸화 헤테로 다당류 S-60을 1시간동안 라이트닌 혼합기(Lightnin mixer)를 사용하여 탈이온수내의 1%농도로 복원한 다음 50℃까지 가열하면서 아티一바린코(Arti-Barinko)상에서 혼합한다. 용액을 온도를 40℃이상 휴지시킨 소르발(Sorvall) RC 2-B 냉각된 원심분리기에서 20분동안 10,000 R.P.M (CSA 헤드)으로 원심분리한다. 상정액을 경사시켜 버린다음 5μ.3μ.1μ.2μ. 및 0.8μ의 구멍을 갖는 겔만(Gelman)형 AN하이드로필릭 아크로포(Acropor) 막(293mm)을 통하여 여과한다. 여과액을 약3-4배량의 99% 이소프로판올에 가하고, 섬유를 모아 55℃의 공기 압송 건조기에서 간단히 건조시킨 다음분쇄하여 분말로 한다. 생성물은 본 발명의 정화된 탈아세틸화 S-60검이다.
[실시예 6]
정화된 탈아세틸화 S-60을 사용하는 한천 대용물
몇몇 상이한 배지를 다음과 같이 제조한다.
모든 배지에 대하여 탈이온수를 사용하였다. 상기 성분을 결합하고(버크스 한천에 대하여는 제외) 121℃ 및 15psi에서 15-20분 동안 고압 멸균하고 55℃까지 냉각시킨다음 무균 페트리 접시에 주입하였다. 버크스 한천의 성분(글루코스 제외, 이것은 별도로 고압 멸균하였다)을 결합한 다음 고압 멸균한후 배지에 가했다. 이들 배양판을 고화시키고, 무균 여부를 조사하기 위하여 24시간동안 실내온도에서 배양한 후, 이들을 다음의 14개의 배양균으로 줄을 그어 접종하였다.

평판을 30℃에서 3-5일동안 배양한 다음 성장에 대한 조사를 했다. S-60으로 만든 배지에선 모든 구조에 대하여 양호한 성장이 관찰되었으며, 한천 대신에 S-60으로 만든 배지사이에서 집락 형태학적 차이점이 전혀 나타나지 않았다. 이들 결과는 S-60이 미생물 배지에서 있어서 한천에 대한 우수한 대체물임을 보여 주었다. BHI 배지 및 TSA를 제외한 S-60을 함유하는 모든 배지에 대한 겔화점(gell point)은 42℃였다. BHI 한천 및 TSA에 대한 겔화점은 52℃였다. 한천은 전형적으조 42-44℃에서 겔화한다.
[실시예 7]
탈아세틸화 S-60을 사용하여 향기로운 주조 겔의 제조
(A) 1.50% 다당류 S-60 4.00% 이소프로판올
0.75% 탄산나트륨 2.00% 에틸렌글리콜
0.025% 메틸 P-하이드록시벤조에이트 88.50% 물
3.00% 장미 향료
천연 다당류 S-60을 탄산나트륨 및 방부제약 혼합한 다음 70℃의 물에 용해시킨다. 용액을 90℃로 더 가열한 다음 10분동안 상기 온도로 유지하여 다당류를 탈아세틸화한다. 60℃까지 냉간시킨 후 용매에 분산된 향료를 가한 다음 혼합물을 통상의 공기후레스녀(freshener) 플라스틱 주형에 넣었다. 혼합물이 38℃까지 냉각되면 겔화가 일어나고, 양호한 향기를 방출하는 견고한 자활-겡이 얻어진다.
(B) 무수 탈아세틸화 다당류 S-60은 묽은 수산화나트륨으로 pH를 10.0으로 조절한 다음 15분 동안 90℃까지 가열함으로써 발효물로부터 제조된다. 용액을 염산으로 pH7로 중화시키고, 2배량의 이소프로판올에 침전시키고, 건조시킨 다음 분쇄한다. 고체 공기 후레스너겔은 다음 방법으로 탈아세틸화 생성물로부터 제조한다. 즉 3.0g의 탈아세틸화 다당류 S-60을 1.5g의 염화칼륨 및 0.15g의 메틸 P-하이드록시벤조에이트 방부제와 혼합한 다음 177ml 물에 가한다. 용액을 90℃까지 가열하여 용해시키고, 60℃까지 냉각시킨다음 6.0g의 바하유 향료, 8.0g의 이소프로판올 및 4.0g의 에틸렌글리콜의 혼합물을 가한다. 용액을 플라스틱 주형에 넣고 주위온도까지 냉각시키면 강한 박하 향기를 가진 견고한 취성겔이 형성된다. 이 겔은 용이하게 주형할 수 없으며 휘어짐이 없이 그의 형상을 그대로 유지한다.
Claims (1)
- 명명되지 않은 슈도모나스 미생물(Pseudomonas organism) ATCC 31,461을 탄소원, 칼륨이온원, 질소원, 미량무기원소 및 물로 구성된 발효 배지에서 40-60시간동안 pH 6-8 및 28-32℃의 온도에서 배양한 다음 이소프로판올과 같은 적당한 저급 알코올로 침전시켜 검(gum)을 회수하는 것으로 구성된, 10-15%의 단백질, 3-4.5%의 아세틸, 및 약12%의 우론산과 몰비 1.5:1의 람노스와 글루코스를 함유한 탄수화물을 함유하며, 피이크(peak)가 3,400cm-1, 2,950cm-1, 1,740cm-1, 1,620cm-1에 나타나는 적외선 스펙트럼 특성을 갖는 헤테로 다당류 S-60을 제조하는 방법.
