도 2는 생균 및 불활화된 백일해 균주의 단백질 불활화 정도를 Transmission Electron Microscope(TEM)으로 측정한 사진이다.
도 3은 도 2의 TEM 사진으로 단백질 불활화 정도를 세포 내 단백질 응집도로 나타낸 것이다.
도 4는 불활화된 백일해 균주 접종에 의한 specific IgG 항체 역가 효능을 나타낸 것이다.
도 5는 불활화된 백일해 균주 접종에 의한 TNF-a 유도 수준을 나타낸 것이다.

- 청구항 1에 있어서,
상기 균주는,
55℃ 내지 70℃에서 30분 내지 90분 동안 처리하여 불활화된 것을 특징으로 하는 백신 조성물.
- 청구항 1에 있어서,
상기 백신 조성물은,
1℃ 내지 65℃의 온도에서 24개월 이상 보관 안정 상태를 유지하는 것을 특징으로 하는 백신 조성물.
- 청구항 1에 있어서,
상기 조성물은,
약제학적으로 허용가능한 담체, 희석제, 보존제, 안정화제, 완충액 및 면역 보조제로 이루어진 군에서 선택된 하나 이상을 더 포함하는 것을 특징으로 하는 백신 조성물.
- 청구항 6에 있어서,
상기 안정화제는,
알라닌, 히스티딘, 아르기닌, 라이신, 이소류신, 발린, 메티오닌, 글리신 및 프롤린으로 구성되는 군으로부터 선택되는 하나 이상인 아미노산 안정화제인 것을 특징으로 하는 백신 조성물.
- 청구항 6에 있어서,
상기 보존제는,
트레할로스(Trehalose), 탈지유(Skim milk), 소르비톨(Sorbitol), 덱스트란(Dextran) 및 아스코르브산(Ascorbic acid)으로 구성되는 군으로부터 선택되는 하나 이상인 것을 특징으로 하는 백신 조성물.
- 청구항 1에 있어서,
상기 백신 조성물은,
복강 내, 점막 내, 근육 내, 진피 내, 피하, 정맥 내, 직장 내, 폐 내, 경막 내, 경구, 경피 또는 비강 내로 투여하기 위한 것을 특징으로 하는 백신 조성물.
- 청구항 1 내지 8 중 어느 한 항의 백신 조성물의 제조방법으로,
상기 불활화된 백일해 균주에, 조성물 총 중량을 기준으로
5 내지 10중량% 트레할로오스, 5 내지 15중량% 스킴밀크, 2 내지 5중량% 소르비톨 및 3 내지 5중량% 덱스트란으로 이루어지는 군에서 선택되는 하나 이상, 및
0.1 내지 10중량%의 아스코르브산;을 더 포함하는 보존제를 첨가하는 단계;
-80℃ 내지 -70℃에서 24시간 내지 48시간 동안 동결시키는 단계; 및
-50 ℃ 내지 -40℃, 15 Pa 내지 30 Pa 조건하에서 48시간 내지 60시간 동안 동결건조하는 단계;를 포함하는 불활화된 백일해균 백신 조성물의 제조방법.
- 청구항 10에 있어서,
상기 불활화시키는 단계는,
백일해 균주를 55℃ 내지 70℃에서 30분 내지 90분 동안 처리하는 것을 특징으로 하는 불활화된 백일해 균주의 제조방법.
- 청구항 10의 제조방법으로 제조한 것을 특징으로 하는 불활화된 백일해 균주.
