Abstract
본 발명은 단백질 자동 발현 시스템에 관한 것으로, 보다 구체적으로는 HdeAB 프로모터를 포함하는 재조합 벡터, 이로부터 형질전환된 숙주세포 및 이를 이용한 목적 단백질 생산 방법에 관한 것이다. 본 발명에 따른 재조합 벡터는 HdeAB 프로모터를 포함함으로써 이를 포함하는 재조합 미생물을 통해 유도제(inducer) 없이 세포성장 정지기에 목적 단백질을 지속적으로 생산할 수 있으므로, 기존 유도제를 요구하는 발현 시스템에 비해 단백질 발현에 소요되는 비용, 시간 및 노동력을 줄일 수 있다.
Description
본 발명은 단백질 자동 발현 시스템에 관한 것으로, 보다 구체적으로는 HdeAB 프로모터를 포함하는 재조합 벡터, 이로부터 형질전환된 숙주세포 및 이를 이용한 목적 단백질 생산 방법에 관한 것이다.
생명공학 분야에서는 인간이 유용하게 사용할 수 있는 단백질이나 대사산물을 생산하기 위해 미생물 또는 세포를 유전자 조작하는 기술이 개발되어 왔다. 이러한 기술은 자연에서 얻는 것에 비해 높은 효율로 재조합 단백질을 생산할 수 있으며, 시간과 비용이 단축될 수 있는 등 여러 장점을 가지고 있어 의약품, 건강기능식품, 식품 등 다양한 분야에서 유용하게 응용되고 있다.
재조합 단백질의 대량 생산을 위해서는 과발현을 유도하는 기술이 중요하다. 주로 대장균에서 IPTG(isopropyl-β-D-thiogalactopyranoside)에 의해 유도되는 T7 프로모터, lac 프로모터, tac 프로모터 및 trc 프로모터 등을 이용한 발현 시스템이 사용되었으며, 대장균 외에 그람음성균, 그람양성균, 효모균, 곰팡이, 동물세포 등에서 과발현을 위한 배양 조건 확립과 함께 생산된 재조합 단백질의 분리를 위한 단백질 정제 기술에 대한 연구가 병행하여 진행되어 왔다. 특히 재조합 단백질의 과발현을 위해서는 강력한 프로모터의 확보가 필수적인데, 이는 대부분 유도 배양을 통해 실시되므로 고농도 배양을 하기에 부적합하였다. 또한, 세포에 유해한 단백질이나 항미생물 펩타이드의 경우 대장균과 같은 미생물에 해를 끼치기 때문에 과량 발현이 어렵고, 회수하는 공정이 복잡하여 회수율이 매우 낮다는 단점이 있다.
이러한 문제점을 해결하기 위하여 유도 배양이 없는 정지기 발현 시스템(stationary expression system)이 새로운 대안으로 제시되었다. 이 시스템은 미생물의 발육 과정에서 배지 중 영양원을 소비하여 증식이 최대치에 도달하여 일정한 속도로 유지되는 지수기(exponential phase)를 지나 영양원이 모두 소비되어 배지의 pH가 변화하고 성장 억제 물질의 생산으로 미생물의 발육 및 증식이 정지되는 정지기(stationary phase)에 특정 프로모터가 활성화되어 이와 인접한 목적 단백질의 유전자를 발현하는 방식으로, 고농도 배양 및 발현된 재조합 단백질의 회수가 용이하여 단백질 생산에 소요되는 시간과 비용을 절약할 수 있다는 이점이 있다. 최근 정지기 발현 시스템에 관해 많은 연구가 진행 중이며, 원핵생물의 정지기에 발현되는 시그마 인자(sigma factor, σ)에 의해 활성화되는 프로모터가 주목받고 있다.
시그마 인자는 원핵생물의 RNA 중합효소를 구성하는 요소에 해당한다. RNA 중합효소는 α2, β, 및 β'로 이루어진 핵심효소(core enzyme)를 포함하며, 이러한 핵심효소에 시그마 인자가 결합한 것을 전효소(holoenzyme)라 한다. 핵심효소는 RNA를 신장시킬 수 있는 능력이 있지만 프로모터를 선택적으로 인식할 수 있는 능력이 없는 반면, 전효소는 프로모터를 선택적으로 인식할 수 있는 능력을 가진다. 이러한 핵심효소와 전효소의 차이는 시그마 인자에 기인하는 것이며, 즉 시그마 인자는 프로모터의 선택성에 관여하여 외부 상황에 따른 미생물의 선택적 유전자 발현에 중요한 역할을 담당한다. 따라서 원핵생물은 다양한 외부 상황에 대응하기 위하여 σ70 (RpoD), σ24 (RpoE), σ28 (RpoF), σ32 (RpoH), σ38 (RpoS), σ54 (RpoN) 등 다양한 종류의 시그마 인자를 보유하고 있으며, 환경적 스트레스에 반응하여 적절한 시그마 인자를 생산하여 결국 외부 상황에 필요한 유전자의 집단적인 발현이 가능하게 되는 것이다. 특히 RpoS는 정지기에 발현되는 시그마 인자로 알려져 있으며, 이러한 시그마 인자에 의해 작동하는 프로모터는 정지기 발현 시스템에 활용될 수 있어, 이에 관한 연구가 절실히 요구되고 있다.
본 발명은 세포성장 정지기에 활성화되는 새로운 프로모터를 포함하는 재조합 벡터를 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 재조합 벡터로 형질전환된 숙주세포를 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 숙주세포를 이용한 목적 단백질 생산 방법을 제공하는 것을 목적으로 한다.
본 발명의 일 양상은 HdeAB 프로모터 및 이에 작동 가능하게 연결된 목적 유전자를 포함하는 재조합 벡터를 제공한다.
본 발명에서 사용된 “프로모터(promoter)”는 이에 작동 가능하게 연결된 유전자의 전사를 허용하고 조절하는 폴리뉴클레오티드 서열을 나타낸다. 상기 프로모터는 RNA 중합효소를 결합시키기 위한 인식 서열 및 전사 개시 부위를 함유한다.
상기 HdeAB 프로모터는 RNA 중합효소와 함께 전사 시작을 조절하는 시그마 인자(sigma factor) RpoS에 의해 활성이 조절되는 여러 RpoS-의존적 프로모터들 중 하나로, 미생물 성장단계 중 RpoS가 발현되는 초기 정지기 동안 HdeAB 프로모터가 활성화되어 이에 연결된 목적 유전자를 지속적으로 발현시킬 수 있다. 프로모터 서열 중 -35 및 -10 위치는 시그마 인자가 결합하는 부위로, RpoS-의존적 프로모터는 해당 위치에 공통염기서열을 가진다. 하지만 HdeAB 프로모터는 기존 RpoS-의존적 프로모터와 -35 및 -10 위치의 서열이 상이하며, 해당 부분의 서열은 오히려 RpoD-의존적 프로모터와 유사성이 높은 것을 특징으로 한다 (도 2 참고). 따라서 HdeAB 프로모터는 기존 RpoS-의존적 프로모터와 다른 활성 또는 강도를 가지며, 이로 인해 기존 RpoS-의존적 프로모터에 비해 높은 수준의 목적 유전자 발현을 나타낼 수 있다.
본 발명의 일 구체예에 따르면, 상기 HdeAB 프로모터는 대장균(Escherichia coli)에서 유래된 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 HdeAB 프로모터는 서열번호 1로 표시되는 염기서열을 포함하는 것일 수 있다 (도 1 참고).
상기 HdeAB 프로모터의 유전자는 서열번호 1로 표시되는 염기서열과 적어도 60% 이상, 70% 이상, 80% 이상, 85% 이상, 90% 이상, 95% 이상, 또는 99% 이상의 상동성을 가지는 뉴클레오티드 서열을 포함하는 폴리뉴클레오티드 서열을 포함할 수 있다. 상기 서열과 상동성을 갖는 서열로서 실질적으로 동일하거나 상응하는 생물학적 활성을 갖는 폴리뉴클레오티드 서열이라면, 일부 서열이 결실, 변형, 치환 또는 부가된 폴리뉴클레오티드 서열을 갖는 경우도 역시 본 출원의 범주에 포함될 수 있다.
여기서 “상동성”은 주어진 뉴클레오티드 서열과 일치하는 정도를 의미하며 백분율로 표시될 수 있다. 본 명세서에서, 주어진 뉴클레오티드 서열과 동일하거나 유사한 활성을 갖는 그의 상동성 서열이 "% 상동성"으로 표시된다. 상기 뉴클레오티드 서열에 대한 상동성은 예를 들면, 문헌에 의한 알고리즘 BLAST (참조: Karlin 및 Altschul, Pro. Natl. Acad. Sci. USA, 90, 5873, 1993) 또는 Pearson에 의한 FASTA (참조: Methods Enzymol., 183, 63, 1990)를 사용하여 결정할 수 있다. 이러한 알고리즘 BLAST에 기초하여, BLASTN이나 BLASTX라고 불리는 프로그램이 개발되어 있다 (참조: http://www.ncbi.nlm.nih.gov).
본 발명에서 사용된 “목적 유전자”는 재조합 벡터에 도입하려는 유전자를 의마하며, 상기 프로모터의 활성에 의해 전사될 수 있다. 이러한 목적 유전자는 바이러스, 박테리아, 효모, 곰팡이, 동물세포 등에서 유래된 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 구체예에 따르면, 상기 목적 유전자는 펩티드, 폴리펩티드, 결합 단백질, 결합 도메인, 부착 단백질, 구조 단백질, 조절 단백질, 신호 전달 단백질, 독소 단백질, 효소, 효소 저해제, 호르몬, 호르몬 유사체, 항체 및 항원으로 이루어진 군에서 선택된 1종 이상을 암호화하는 것일 수 있다.
본 발명의 일 실시예에서는 프로모터의 활성을 시각적으로 관찰하기 위해 형광 단백질 eGFP(enhanced green fluorescent protein)를 암호화하는 유전자를 목적 유전자로서 사용하였으며, eGFP는 동일한 목적을 위해 당업계에 공지된 형광 단백질로 제한 없이 대체 가능하다. 여기서 사용된 eGFP 유전자는 서열번호 8로 표시되는 염기서열을 포함한다.
이러한 HdeAB 프로모터 및 목적 유전자를 포함하는 재조합 벡터는 HdeAB 프로모터의 활성에 의해 목적 단백질을 생산할 수 있다.
본 발명에서 사용된 “벡터”는 적당한 숙주세포에서 목적 단백질을 발현할 수 있는 발현 벡터로서 유전자 삽입물이 발현되도록 작동 가능하게 연결된 필수적인 조절요소를 포함하는 유전자 제조물을 의미한다. 여기서 "작동 가능하게 연결(operably linked)"이란, 일반적 기능을 수행하도록 핵산 발현조절 서열과 목적하는 단백질 또는 RNA를 암호화하는 핵산 서열이 기능적으로 연결(functional linkage)되어 있는 상태를 의미한다. 예를 들어 프로모터와 단백질 또는 RNA를 암호화하는 핵산 서열이 작동 가능하게 연결되어 암호서열의 발현에 영향을 미칠 수 있다. 발현 벡터와의 작동적 연결은 당해 기술분야에서 잘 알려진 유전자 재조합 기술을 이용하여 제조할 수 있으며, 부위-특이적 DNA 절단 및 연결은 당해 기술 분야에서 일반적으로 알려진 효소 등을 사용 할 수 있다. 또한, “조절요소”는 전사를 수행하기 위한 프로모터, 전사를 조절하기 위한 임의의 오퍼레이터 서열, 적합한 mRNA 리보좀 결합 부위를 암호화하는 서열, 및 전사 및 해독의 종결을 조절하는 서열을 포함한다.
본 발명에서 사용된 “재조합 벡터”는 적합한 숙주세포 내로 형질전환된 후, 숙주세포의 게놈과 무관하게 복제 가능하거나 게놈 그 자체에 봉합될 수 있다. 여기서 "적합한 숙주세포"는 벡터가 복제 가능한 것으로서 복제가 개시되는 특정 염기서열인 복제 원점을 포함할 수 있다.
상기 재조합 벡터는 플라스미드 벡터(plasmid vector), 바이러스 벡터(viral vector) 및 코스미드 벡터(cosmid vector)로 이루어진 군으로부터 선택되는 어느 하나일 수 있다.
상기 플라스미드의 경우 원핵생물 또는 진핵세포에 접합(conjugation)에 의한 전달과 복제가 가능한 플라스미드일 수 있다. 예를 들면, 상기 플라스미드는 pBR계, pUC계, pBluescriptII계, pGEM계, pTZ계, pCL계 또는 pET계 플라스미드일 수 있다. 구체적으로는 pDZ, pACYC177, pACYC184, pCL, pECCG117, pUC19, pBR322, pMW118, pCC1BAC, pUC18, pBAD, pJK001, pCM, 또는 pAWP 등일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 구체예에 따르면, 상기 재조합 벡터는 항생제 저항성 유전자를 더 포함하는 것일 수 있다.
본 발명에서 사용된 "항생제 저항성 유전자"는 항생제에 대하여 저항성을 가지는 유전자를 의미한다. 이러한 유전자가 있는 미생물 또는 세포는 해당 항생제를 처리한 환경에서도 생존하므로, 본 발명에서는 서열번호 1의 염기서열을 포함하는 프로모터에 의해 발현이 조절되는 항생제 저항성 유전자를 포함하는 재조합 벡터 또는 플라스미드를 얻는 과정에 선별 마커 (또는 리포터 마커)로 사용된다.
상기 항생제 저항성 유전자는 당업계에 공지된 것이라면 제한 없이 이용될 수 있다.
본 발명의 일 구체예에 따르면, 상기 항생제 저항성 유전자는 암피실린(ampicilin), 테트라사이클린(tetracyclin), 카나마이신(kanamycin), 클로람페니콜(chloroamphenicol), 스트렙토마이신(streptomycin) 및 네오마이신(neomycin)으로 이루어진 군에서 선택된 1종 이상의 항생제에 대한 저항성 유전자인 것일 수 있다.
본 발명의 일 실시예에서는 클로람페니콜에 대한 저항성 유전자 (서열번호 2)를 사용하였다.
또한, 본 발명의 일 구체예에 따르면, 상기 재조합 벡터는 전사 종결 부위를 더 포함하는 것일 수 있다.
본 발명에서 사용된 “전사 종결 부위”는 RNA 중합효소가 전사를 종결하게 하는 부위를 의미하며, Rho 단백질에 의해 전사가 종결될 수 있도록 Rho 단백질이 인식하는 mRNA의 특정 서열 rut을 포함하거나, 또는 mRNA의 3' 말단에서 헤어핀과 같은 2차 구조를 형성하는 G와 C이 풍부한 역반복 서열과 전사종결신호로 작용하는 A 또는 U이 풍부한 서열을 포함할 수 있다.
본 발명의 일 실시예에서는 전사 종결 부위로서 서열번호 5 및 6의 염기서열을 사용하였다.
본 발명의 일 구체예에 따르면, 상기 재조합 백터는 항생제 저항성 유전자, 목적 유전자, 프로모터 및 전사 종결 부위가 순차적으로 작동 가능하게 연결된 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 재조합 벡터는 세포성장 정지기에 프로모터 활성이 유도되는 것일 수 있다.
본 발명에 따른 재조합 벡터는 세포성장 정지기에 발현되는 RpoS와 결합하는 부위를 가지는 HdeAB 프로모터를 포함함으로써 세포 성장기에 발현이 억제되지만 정지기에 들어서면서 유도제(inducer) 없이 목적 유전자가 발현되어 원하는 단백질을 세포내에서 생산할 수 있다.
본 발명의 다른 양상은 전술한 재조합 벡터로 형질전환된 숙주세포를 제공한다.
상기 숙주세포는 생체내 또는 시험관내에서 전술한 재조합 벡터 또는 폴리뉴클레오티드로 형질감염, 형질전환, 또는 감염된 세포를 포함한다. 본 발명의 재조합 벡터를 포함하는 숙주 세포는 재조합 숙주 세포, 재조합 세포 또는 재조합 미생물이다. 여기서 “형질전환”은 목적 단백질을 암호화하는 폴리뉴클레오티드를 포함하는 발현 벡터를 숙주세포 내에 도입하여 숙주세포 내에서 상기 폴리뉴클레오티드가 암호화하는 단백질을 발현할 수 있도록 하는 것을 의미하며, 상기 발현 벡터가 도입된 숙주세포를 형질전환체라 한다.
이러한 숙주세포는 당업계에 공지된 것을 제한 없이 사용할 수 있다.
본 발명의 일 구체예에 따르면, 상기 숙주세포는 대장균인 것일 수 있다.
또한, 본 발명의 다른 양상은 전술한 숙주세포를 배양하는 단계를 포함하는 목적 단백질의 생산 방법을 제공한다.
상기 배양은 당업계에 알려진 적절한 배지와 배양 조건에 따라 이루어질 수 있으며, 통상의 기술자라면 배지 및 배양 조건을 용이하게 조정하여 사용할 수 있다. 구체적으로, 상기 배지는 액체 배지일 수 있으나, 이에 한정되는 것은 아니다. 배양 방법은 예를 들면, 회분식 배양(batch culture), 연속식 배양(continuous culture), 유가식 배양(fed-batch culture) 또는 이들의 조합 배양을 포함할 수 있으나, 이에 한정되는 것은 아니며, 주입 배치(fed batch) 또는 반복 주입 배치 공정(repeated fed batch process)에서 연속식으로 배양할 수 있다.
본 발명의 일 구체예에 따르면, 배지에 다양한 탄소원, 질소원 및 미량원소 성분을 포함할 수 있다. 사용될 수 있는 탄소원으로는 글루코스, 수크로스, 락토스, 프락토스, 말토스, 전분, 셀룰로스와 같은 당 및 탄수화물, 대두유, 해바라기유, 피마자유, 코코넛유 등과 같은 오일 및 지방, 팔미트산, 스테아린산, 리놀레산과 같은 지방산, 글리세롤, 에탄올과 같은 알코올, 아세트산과 같은 유기산이 포함된다. 이들 물질은 개별적으로 또는 혼합물로서 사용될 수 있으나, 이에 한정되는 것은 아니다. 사용될 수 있는 질소원으로는 펩톤, 효모 추출물, 육즙, 맥아 추출물, 옥수수 침지액, 대두밀 및 요소 또는 무기 화합물, 예를 들면 황산 암모늄, 염화암모늄, 인산암모늄, 탄산암모늄 및 질산암모늄이 포함될 수 있다. 질소원 또한 개별적으로 또는 혼합물로서 사용할 수 있으나 이에 한정되는 것은 아니다. 사용될 수 있는 인의 공급원으로는 인산이수소칼륨 또는 인산수소이칼륨 또는 상응하는 나트륨-함유 염이 포함될 수 있으며, 이에 한정되는 것은 아니다. 또한, 배양 배지는 성장에 필요한 황산마그네슘 또는 황산철과 같은 금속염을 함유할 수 있으며, 이에 한정되는 것은 아니다. 그 외에, 아미노산 및 비타민과 같은 필수 성장 물질이 포함될 수 있다. 또한 배양 배지에 적절한 전구체들이 사용될 수 있다. 상기 배지 또는 개별 성분은 배양과정에서 배양액에 적절한 방식에 의해 회분식으로 또는 연속식으로 첨가될 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 일 구체예에 따르면, 배양 중에 수산화암모늄, 수산화칼륨, 암모니아, 인산 및 황산과 같은 화합물을 미생물 배양액에 적절한 방식으로 첨가하여 배양액의 pH를 조정할 수 있다. 또한, 배양 중에 지방산 폴리글리콜 에스테르와 같은 소포제를 사용하여 기포 생성을 억제할 수 있다. 추가적으로, 배양액의 호기 상태를 유지하기 위하여, 배양액 내로 산소 또는 산소-함유 기체 (예, 공기)를 주입할 수 있다. 배양액의 온도는 통상 20℃ 내지 45℃, 예를 들면 25℃ 내지 40℃일 수 있다. 배양기간은 유용물질이 원하는 생산량으로 수득될 때까지 계속될 수 있으며, 예를 들면 10 내지 160 시간일 수 있다.
본 발명의 일 구체예에 따르면, 상기 방법은 세포성장 정지기에 발현된 목적 단백질을 회수하는 단계를 더 포함하는 것일 수 있다.
상기 회수는 당업계에 알려진 적합한 방법을 이용하여 배지로부터 생산된 목적 단백질을 수집 또는 회수할 수 있다. 예를 들면 원심분리, 여과, 추출, 분무, 건조, 증발, 침전, 결정화, 전기영동, 분별용해 (예를 들면, 암모늄 설페이트 침전), 크로마토그래피 (예를 들면, 이온 교환, 친화성, 소수성 및 크기배제) 등의 방법을 사용할 수 있으나, 이에 한정되는 것은 않는다.
본 발명의 일 구체예에 따르면, 상기 목적 단백질을 회수하는 단계는 배양 배지를 원심분리하여 바이오매스를 제거하고 얻어진 상등액을 크로마토그래피를 통하여 분리할 수 있다.

본 발명에 따른 재조합 벡터는 HdeAB 프로모터를 포함함으로써 이를 포함하는 재조합 미생물을 통해 유도제(inducer) 없이 세포성장 정지기에 목적 단백질을 지속적으로 생산할 수 있으므로, 기존 유도제를 요구하는 발현 시스템에 비해 단백질 발현에 소요되는 비용, 시간 및 노동력을 줄일 수 있다.

이하, 첨부된 도면을 참조하며 본 발명을 보다 상세하게 설명한다. 그러나, 이러한 설명은 본 발명의 이해를 돕기 위하여 예시적으로 제시된 것일 뿐, 본 발명의 범위가 이러한 예시적인 설명에 의하여 제한되는 것은 아니다.
실시예 1. HdeAB 프로모터를 포함하는 균주 제작
1-1. 재조합 벡터 제작
HdeAB 프로모터의 발현을 위하여 5개의 DNA 단편을 사용하여 재조합 플라스미드를 제작하였다. 먼저, pACYCDuet-1 벡터 (Novagen)를 주형으로 프라이머 1 및 2를 사용하여 증폭한 p15A 복제 시작점(origin of replicate) 및 클로람페니콜(chloramphenicol) 저항성 유전자를 포함하는 DNA 단편 (서열번호 3)을, 프라이머 3 및 4를 사용하여 증폭한 다중클로닝자리(multiple cloning site)를 포함하는 DNA 단편 (서열번호 4)을 준비하였다. HdeAB 프로모터가 발현하기 전 상위영역(upstream)에서 다른 유전자의 발현을 종결시키 위해 pRS551 벡터 (Microbial Physiology Laboratory, Department of Gene Function and Phenomics. National Institute of Genetics)를 주형으로 프라이머 5 및 6을 사용하여 증폭한 rrnB T1 반복 종결자(repeat terminator) 유전자를 포함하는 DNA 단편 (서열번호 5)을 준비하였다. Escherichia coli K-12 strain K-12 MG1655 choromosme (NCBI Reference Sequence: NC_000913.3)에서 HdeAB 프로모터의 -147 ~ +50 영역 (197 bp)을 주형으로 프라이머 7 및 8을 사용하여 증폭한 DNA 단편 (서열번호 1)을 준비하였다. 마지막으로, HdeAB 프로모터에 의해 발현된 유전자의 전사를 종결시키기 위해 pBAD33 벡터 (Beckwith Lab)를 주형으로 프라이머 9 및 10을 사용하여 증폭한 rrnB T1/T2 종결자 유전자를 포함하는 DNA 단편 (서열번호 6)을 준비하였다. 준비된 5개의 DNA 단편들을 Gibson Assembly 방법으로 결합하여 HdeAB 프로모터를 포함하는 재조합 벡터(이하 'pKMHdeAB 벡터'라 함) (서열번호 7) (도 3)를 제작하였다.
여기서 사용된 프라이머 서열은 하기 표 1과 같다.

1-2. 형질전환체 제작
pKMHdeAB 벡터의 프로모터 발현 유무를 확인하기 위해 목적 유전자로서 eGFP 유전자를 삽입하였다. 먼저, pKMHdeAB 벡터의 다중클로닝자리를 EcoRI 및 PstI 제한효소로 절단한 후 eGFP 유전자 단편 (서열번호 8)을 삽입하였다. 해당 플라스미드는 E. coli MG1655 (Laboratory stock Collection)에 열충격법(heat-shock)으로 형질전환하여 형질전환체를 제작하였다. 형질전환체는 클로람페니콜 20 μg/mL를 포함하는 LB 고체배지에 평판도말하여 static incubator 37℃에서 배양하였다.
실험예 1. HdeAB 프로모터를 포함하는 균주의 eGFP 발현 확인
1-1. 세포 성장기에 따른 eGFP 발현
실시예 1에서 제작된 형질전환체가 세포성장 초기 정지기(early stationary) 동안 목적 유전자의 단백질을 지속적으로 발현하는 HdeAB 프로모터를 포함하는 것을 고려하여, 세포 성장기에 따른 eGFP 발현을 확인하였다. 형질전환체를 LB 액체배지 10 mL (클로람페니콜 20 μg/mL 함유)에 접종하여 37℃, 200 rpm에서 진탕 배양하면서 흡광도(optical density, OD)값에 따라 세포 성장기를 구분하여 OD600 = 0.5, 1.0, 2.0, 2.0 초과 (overnight, O/N)에 해당하는 형질전환체의 배양액 10 mL를 수집한 후 6,000 rpm에서 10분간 원심분리하여 세포만을 수득하였다. 해당 세포에 대해 통상의 방법으로 SDS-PAGE과 웨스턴 블롯을 실시하였다. 여기서 His-tag를 가지는 eGFP를 검출할 수 있는 His 항체 (Invitrogen, MA1-21315-HRP)를 사용하였다.
그 결과, 도 4에 나타낸 바와 같이, 형질전환체가 성장하는 시기 (OD600 = 0.5)에는 eGFP 발현량이 적으나, 초기 정지기에 도달하면 (0.D600 = 1.0 이상) eGFP 발현량이 현저히 증가하는 것을 확인하였다.
1-2. 정제를 통한 eGFP 발현
실시예 1의 형질전환체를 LB 액체배지 1 L (클로람페니콜 20 μg/mL 함유)에 접종하여 37℃, 200 rpm에서 진탕 배양하여 OD600 = 0.5까지 키운 후 6시간 더 배양하였다. HdeAB 프로모터에 의해 유도된 단백질을 정제하기 위해 배양된 세포를 6,000 rpm에서 30분간 원심분리하여 수확하고 용해 버퍼(lysis buffer)에 풀어주었다. 이후 초음파 파쇄기로 세포를 파쇄하고 13,000 rpm에서 10분간 원심분리한 후 상등액을 0.45 μm 필터로 여과하고 FPLC(Fast-protein liquid chromatography)를 통해 Ni-NTA 컬럼에 용출 버퍼(elution buffer) (20 mM Tris pH 7.0, 0.5 M NaCl 및 0.5 M imidazole 함유)를 주입하여 His-tag를 가지는 eGFP를 수확하였다.
그 결과, 도 5에 나타낸 바와 같이, 정제된 eGFP를 포함하는 튜브에서 형광 발현을 확인하였다.
비교예 1. TalA 프로모터를 포함하는 균주 제작
본 발명의 HdeAB 프로모터와 종래 RpoS 의존성 프로모터의 발현 수준을 비교하기 위해 RpoS에 의해 발현이 조절되는 유전자들 중 지수기(exponential phase)보다 정지기(stationary phase)에 높은 수준으로 발현되는 TalA 유전자를 선정하였다 (Somalinga R. V. Vijayakumar, et al. J Bacteriol. 2004. 186(24):8499-507).
TalA 프로모터를 포함하는 균주는 HdeAB 프로모터를 포함하는 DNA 단편 대신 Escherichia coli K-12 strain K-12 MG1655 choromosme (NCBI Reference Sequence: NC_000913.3)에서 TalA 프로모터의 -147 ~ +50 영역 (197 bp)을 주형으로 프라이머 11 및 12를 사용하여 증폭한 DNA 단편 (서열번호 9)을 사용한 것을 제외하고, 실시예 1과 동일한 방법으로 제작된 재조합 벡터 (이하 'pKMTalA 벡터'라 함)를 이용하여 제작되었다.

실험예 2. 프로모터 차이에 따른 eGFP 발현 비교
HdeAB 프로모터와 TalA 프로모터의 발현 차이를 비교하기 위해 실시예 1 및 비교예 1의 균주를 실험예 1-1의 세포 성장기에 따른 eGFP 발현과 동일한 방법으로 eGFP 검출을 실시하였다.
그 결과, 도 7에 나타낸 바와 같이, 동일한 조건에서 두 균주를 배양하였을 때 HdeAB 프로모터가 TalA 프로모터에 비해 eGFP 발현량이 현저히 증가한 것을 확인하였다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.

상기 목적 유전자는 펩티드, 폴리펩티드, 결합 단백질, 결합 도메인, 부착 단백질, 구조 단백질, 조절 단백질, 신호 전달 단백질, 독소 단백질, 효소, 효소 저해제, 호르몬, 호르몬 유사체, 항체 및 항원으로 이루어진 군에서 선택된 1종 이상을 암호화하는 것인 재조합 벡터. - 청구항 1에 있어서,
상기 재조합 벡터는 항생제 저항성 유전자를 더 포함하는 것인 재조합 벡터. - 청구항 1에 있어서,
상기 재조합 벡터는 전사 종결 부위를 더 포함하는 것인 재조합 벡터. - 청구항 1에 있어서,
