Abstract
본 발명은 바실러스 츄린겐시스(Bacillus thuringiensis , Bt) Cry3Aa 프로모터(promoter)의 상위 부위에 UP 엘리먼트(UP element)를 포함하는 프로모터 변이체에 관한 것으로서, 하위(downstream) 목적 유전자의 발현 증가용 프로모터 변이체에 관한 것이다. 보다 상세하게는 Cry3Aa 프로모터의 -35 부위의 상위 영역에 뉴클레오티드 아데닌(A)과 티민(T)이 풍부한 핵산 서열을 첨가함으로써 하위 목적 단백질의 발현을 증가시키는 프로모터 변이체에 관한 것이다. 본 발명의 발현 시스템을 이용하는 경우, 바실러스 균주에서 목적 단백질의 발현을 현저히 증가시킬 수 있어, 단백질, 폴리펩티드 약물과 같은 목적 단백질의 대량 생산을 통해 생명공학, 의학 뿐만 아니라 산업적으로 유용한 단백질의 생산에 효과적으로 사용될 수 있다.
Description
본 발명은 바실러스 츄린겐시스(Bacillus thuringiensis , Bt) Cry3Aa 프로모터(promoter)의 상위 부위에 UP 엘리먼트(UP element)를 포함하는 프로모터 변이체에 관한 것으로서, 하위(downstream) 목적 유전자의 발현 증가용 프로모터 변이체에 관한 것이다. 보다 상세하게는 Cry3Aa 프로모터의 -35 부위의 상위 영역에 뉴클레오티드 아데닌(A)과 티민(T)이 풍부한 핵산 서열을 첨가함으로써 하위 목적 단백질의 발현을 증가시키는 프로모터 변이체에 관한 것이다.
바실러스(Bacillus)는 대장균 및 효모와 함께 산업적으로 유용한 단백질을 생산하는 주요한 숙주로 사용되어져 왔다. 바실러스 균이 단백질 생산 숙주로서의 장점은 (1) 인체에 무해하고 (2) 많은 양의 단백질을 세포 외로 분비할 수 있으며 (3) 내독소를 생산하지 않고 (4) FDA에 의해 GRAS(generally recognized as safe)로 규정되어 있으며 (5) 코돈 치우침(codon bias)이 없고 (6) 유전자 조작, 형질전환 및 대량 배양이 용이하며 (7) 게놈 정보가 알려져 있다(Shumann W. 2007. Adv . Appl. Microbiol . 62: 137-189)는 점이다. 현재 전 세계 산업용 효소의 60% 정도가 바실러스를 숙주균으로 생산되고 있으며(Westers L. 2004. Biochimica et Biophysica Acta 1694: 299-310) 인체에 무해한 GRAS(generally recognized as safe) 미생물이라는 특성으로 인해 식품용 또는 의약용 단백질 생산에 적합하다고 알려져 있다(Schallmey M. 2004. Can . J. Microbiol . 50: 1-17).
고전적으로 유용한 효소 또는 단백질을 대량으로 생산하기 위하여 돌연변이 유도물질 처리, 자외선 또는 방사선 조사 등을 통해 숙주균을 돌연변이 시키는 방법이 사용되어 왔으나, 상기 방법들은 비 선택적 돌연변이로 인해 종종 숙주균의 성장속도가 저하되거나, 배지조성 및 배양조건을 최적화시키기 까다로운 경우가 많다. 또한, 돌연변이 균주의 경우에는 균주 퇴화가 일어나며, 각각의 목적 단백질 생산을 위한 숙주균을 따로 개발해야 하므로 시간과 노력이 많이 소요되는 불편함이 존재하였다. 최근에는 유전공학의 발달로 인해 다양한 발현 시스템이 개발되어 유용한 단백질의 생산이 시도되고 있는데(Shumann W. 2007. Adv . Appl . Microbiol . 62: 137-189), 이 보고에 의하면 IPTG(isopropyl-1-thio-β-D-galactopyranoside) 및 당과 같은 특정 발현 유도 인자를 이용하여 발현을 인위적으로 유도하는 방법, 세포 성장주기 의존성 프로모터에 의한 방법 및 자동유도 발현 등이 개발되었다. 그러나 발현 유도 인자의 단가가 생산성에 영향을 미치거나, 세포 성장기의 많은 양의 단백질이 발현되어 세포성장에 영향을 주며, 고온조건에서 발현이 유도되면 열 충격에 의한 여러 단백질 분해효소의 발현으로 인해 목적 단백질의 안정성에 문제가 생기는 등 여러 문제점이 여전히 남아 있다.
한편, 발현 숙주로 많이 사용되고 있는 대장균의 경우 발현된 단백질이 대부분 세포 내에 존재하기 때문에 물리적, 화학적 또는 효소적 세포 파괴 방법이 필요하다(Witte A and Lubitz W. 1989. Eur . J. Biochem . 180: 393-398). 따라서 목적 단백질의 대량생산을 위해서는 세포 파괴를 위한 저렴한 방법이 필수적으로 요구된다. 바실러스의 경우 세포 성장 정지기 후반에 이르면 자동으로 세포가 파괴되어 세포 내에 있던 단백질이 배지로 노출되는데(Nugroho FA et al., 1999. J. Bacteriol. 181: 6230-6237), 이러한 특징은 세포 내에서 발현된 단백질의 효율적인 회수가 가능하게 한다. 따라서 바실러스를 이용한 단백질 생산에 많은 연구가 진행되고 있으며, 바실러스 서브틸리스(Bacillus subtilis) pstS 프로모터를 이용하거나 단백질 분해효소를 이용하여 단백질을 대량 발현시키고자 하는 많은 시스템 등이 개발되고 있다. 그럼에도 불구하고 현재까지 바실러스를 숙주균으로 생산된 단백질 의약품이 시장진입에 성공한 예가 없는데, 그 이유는 (1) 대장균과 비교하여 여전히 강력한 동시에, 조절 가능한 프로모터가 없고, (2) 많은 단백질 분해효소의 존재로 인해 발현된 단백질의 안정성에 문제가 있으며, (3) 발현된 단백질의 세포외 분비시 세포막에 존재하는 단백질 분해효소에 의한 분해가 일어나기 때문이라고 해석되고 있다(Schallmey M. 2004. Can . J. Microbiol . 50: 1-17). 이러한 문제점을 동시에 해결하기 위하여 본 발명에서는 바실러스 츄린겐시스 Cry3Aa 프로모터의 상위 부위에 UP 엘리먼트를 포함하는 프로모터 변이체를 이용한 단백질 발현 시스템을 개발하고 이를 이용한 목적 단백질의 대량 생산 방법을 개발하고자 하였다.
바실러스 츄린겐시스(Bacillus thuringiensis)는 그람 양성의 간균으로 포자 형성시 세포 전체 중량의 25 내지 30 %에 이르는 양의 독소 단백질 결정체를 생산하는 특징을 갖는다. 바실러스 츄린겐시스로부터 생산된 독소 단백질 결정체는 곤충에 대해 특이한 살충 효과를 나타내어 미생물 살충제 제조시 주요한 활성 성분이 되며 지금까지 약 450여 종에 이르는 독소 단백질 유전자가 분리되어 그 핵산 서열이 밝혀졌다(http://www.lifesci.sussex.ac.kr/home/Neil_Crickmore/Bt/).
Wong 등 (J. Biol . Chem ., 258: 1960-1967, 1983)은 Cry1A 유전자가 두 가지 프로모터, 즉 BtΙ과 BtⅡ에 의해 대수 증식기 이후에 연속적으로 전사가 이루어진다는 사실을 밝혔으며, Brown과 Whiteley는 상기 두 가지 프로모터가 바실러스 서브틸리스의 σE 및 σK와 상동성을 지닌 σ35 및 σ28에 의해서 전사된다는 사실을 보고하였다 (Proc . Natl . Acad . Sci . USA, 85: 4166-4170; J. Bacteriol ., 172: 6682-6688, 1988). 대부분의 독소 단백질 유전자는 유사한 프로모터 구조를 가지고 있으나, 독소 단백질 유전자 중 Cry3A 유전자는 예외적인 경우로서 비록 세포 성장 정지기에서 발현이 유도되지만 포자 형성 기작과는 독립적인 σA 에 의해 전사가 일어나며 프로모터와 구조 유전자 사이에 전사체를 안정화시키는 핵산 서열(stab)이 존재한다(Mol . Microbiol ., 20: 633-643, 1996). 이러한 Cry3Aa 프로모터를 이용하여 목적 단백질의 발현을 유도하는 특허가 등록된 바 있다(대한민국 등록 특허 1280503). 그러나 목적 단백질의 산업적 생산을 위한 경제성 확보를 위해서는 좀 더 강력한 프로모터의 개발이 필요하였다.
UP 엘리먼트는 일부의 미생물 프로모터 -35 부위의 상위 영역에 존재하며 RNA 폴리머라제 알파 서브유닛(RNA polymeras α-subunit)의 C 말단영역(C-terminal domain, CTD)이 결합하는 부위이다. 아데닌(A)과 티민(T)이 풍부한 핵산 서열을 가지고 있다.
이에 본 발명자들은 상기 Cry3Aa 프로모터의 상위 영역에 UP 엘리먼트를 위치시킴으로써 목적 단백질의 대량 발현 시스템을 개발할 수 있을 것이라 착안하고, Cry3Aa 프로모터와 적합한 UP 엘리먼트를 개발하고자 예의 노력한 결과, 여러 종류의 UP 엘리먼트가 Cry3Aa 프로모터의 발현량을 현저히 증가시킬 수 있음을 확인하고 본 발명을 완성하였다.
본 발명의 목적은 바실러스 츄린겐시스(Bacillus thuringiensis) Cry3Aa 프로모터 -35 부위의 상위(upstream) 영역에 UP 엘리먼트(UP element)가 포함된 하위(downstream) 목적 유전자 발현 증가용 프로모터 변이체를 제공하는 것이다.
본 발명의 다른 목적은 상기 프로모터 변이체 서열을 포함하는 벡터를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 프로모터 변이체 서열을 포함하는 벡터를 포함하는 숙주세포를 제공하는 것이다.
본 발명의 또 다른 목적은 (a) 상기 프로모터 변이체 서열을 포함하는 벡터를 제조하는 단계; (b) 상기 단계 (a)에 의해 제조된 벡터를 숙주세포에 도입하는 단계; 및 (c) 상기 단계 (b)의 숙주세포로부터 목적 단백질을 회수하는 단계를 포함하는 목적 단백질의 생산방법을 제공하는 것이다.
상기 목적을 달성하기 위한 하나의 양태로서, 본 발명은 바실러스 츄린겐시스(Bacillus thuringiensis) Cry3Aa 프로모터 상위(upstream) 영역에 UP 엘리먼트(UP element)가 포함된 하위(downstream) 목적 유전자 발현 증가용 프로모터 변이체를 제공한다.
특히, Cry3Aa 프로모터 -35 부위의 상위 영역에 UP 엘리먼트가 포함된 것일 수 있으며, 예를 들어, Cry3Aa 프로모터 -35 부위에 바로 연결되어 UP 엘리먼트가 포함된 하위 목적 유전자 발현 증가용 프로모터 변이체에 관한 것이다.
본 발명의 바실러스 츄린겐시스(Bacillus thuringiensis)는 1901년 일본의 미생물학자 이시와타가 병든 누에(Bombyx mori)로부터 바실러스(Bacillus)를 분리함으로써 시작되었다. 약 10년 후에는 병든 꽃가루나방으로부터 유사한 미생물을 분리했는데(Berliner), 이 미생물의 이름을 바실러스 츄린겐시스라 명명하였으며 그람 양성 박테리아이자 살충 효과를 가진 토양 미생물이다. 본 발명에서 바실러스 츄린겐시스의 예로는 바실러스 츄린겐시스 아종 알레스티(B. thuringiensis sub. alesti), 소토(B. thuringiensis sub. sotto), 모리소니(B. thuringiensis sub. morrisoni), 산디에고(B. thuringiensis sub. sandiego), 담스타디엔시스(B. thuringiensis sub. darmsradiensis), 솜프소니(B. thuringiensis sub. thompsoni), 이스라엘렌시스(B. thuringiensis sub. israelensis), 토치지엔시스(B. thuringiensis sub. tochigiensis), 쿠스타키(B. thuringiensis sub. kurstaki), 카나덴시스(B. thuringiensis sub. canadensis), 다코타(B. thuringiensis sub. dakota), 갤러리에(B. thuringiensis sub. gallerriae), 엔토모시더스(B. thuringiensis sub. entomocidus), 아이자와이(B. thuringiensis sub. aizawai), 또는 오스트리니에 (B. thuringiensis sub. ostriniae)가 있으나, 상기 예에 의해 본 발명의 Cry3Aa 프로모터의 기원이 제한되는 것은 아니다. 또한, 바실러스 츄린겐시스와 같은 종으로 여겨질 정도로 계통적으로 유사하다고 알려져 있는 바실러스 시리우스(Bacillus cereus) 및 바실러스 안스라시스(Bacillus anthracis)와 같은 균주로부터 수득할 수 있음은 당업자에게 자명하다.
본 발명에서 사용되는 용어, "프로모터"란 폴리머라제에 대한 결합부위를 포함하고 프로모터 하위(downstream) 유전자의 mRNA(messenger RNA)로의 전사 개시 활성을 가지는, 암호화 영역의 상위(upstream)의 비해독된 핵산 서열을 말한다. 본 발명에서는 목적 유전자 발현 증가용 프로모터 변이체와 혼용되어 사용될 수 있다. 진핵생물에서는 전사조절인자(transcription factor)라고 하는 단백질들이 프로모터 부분에 결합함으로써 RNA 중합효소를 이끌고 오는 일에 관여한다. 프로모터는 이러한 전사조절인자들이 결합하는 모든 DNA 핵산 서열부위를 지칭한다고 할 수 있다. 그에 비해 원핵생물에서의 프로모터는 대개 RNA 중합효소가 결합하는 전사 시작 지점(Transcription Start Site) 바로 근처의 결합부위로 정의된다.
원핵생물에서의 프로모터는 전사 시작 지점으로부터 앞쪽으로 각각 -10 염기, -35 염기 위치에 있는 떨어져 있는 두 개의 짧은 핵산 서열로 구성되어 있다. -10 염기에 있는 핵산 서열을 프리브노 박스(pribnow box) 또는 -10 엘리먼트(-10 element, -10 부위)라고 부르며 일반적으로는 TATAAT의 6개의 염기로 이루어져 있다. 프리브노 박스는 원핵생물에서 전사를 시작하기 위해 반드시 필요하다. -35 염기에 있는 핵산 서열(-35 부위)은 일반적으로 TTGACA의 6개의 염기로 이루어져 있고 전사가 활발히 일어날 수 있도록 해주는 역할을 한다.
본 발명에서 사용되는 프로모터인 바실러스 츄린겐시스의 Cry3Aa의 프로모터는 천연형에서는 -10 부위에는 "TAAGCT"의 서열을 가지고, -35 부위에는 "TTGCAA"의 서열을 가지는 것으로 알려져 있어, 상기 원핵생물의 일반적인 -10 부위 또는 -35 부위의 서열과는 상이하다.
이에, 본 발명의 프로모터 변이체는 하위 목적 유전자의 발현 증가를 위하여 프로모터의 -35 부위 및/또는 -10 부위를 변이시킬 수 있으며, 예를 들어, -35 부위와 -10 부위를 각각 TTGACA, TATAAT로 변이시킬 수 있다.
본 발명의 구체적인 일 실시예에서는 상기 일반적이지 않은 바실러스 츄린겐시스의 -35 부위를 원핵생물의 일반적인 서열인 "TTGACA"로 변이시킨 프로모터를 사용하였다.
본 발명에서는 Cry3Aa 프로모터의 폴리뉴클레오티드 서열을 조작함으로써 하위(downstream) 유전자의 발현량을 조절할 수 있다. 본 발명은 아데닌(A)과 티민(T)이 풍부한 폴리뉴클레오티드를 상기 Cry3Aa 프로모터 상위 영역에 위치하게 함으로써 단백질의 대량생산할 수 있도록 하였다. 특히, 상기 Cry3Aa 프로모터 상위 영역은 Cry3Aa 프로모터의 -35 부위의 상위 영역일 수 있다.
본 발명의 구체적인 일 실시예에서는 Cry3Aa 프로모터의 상위 영역에 UP 엘리먼트를 삽입하여 본 발명의 Cry3Aa 프로모터 변이체를 제조하였으며, 해당 프로모터 변이체의 유전자 발현 유도 효과를 확인하였다.
본 발명에서 프로모터 변이체는 당업계에 공지된 다양한 방법에 의하여 제조될 수 있으며, 그 예는 에러-유발 PCR(error-prone PCR), DNA 셔플링(DNA shuffling) 방법, 위치-지정 돌연변이(site-directed mutagenesis) 방법 등이 있다.
상위 영역 UP 엘리먼트와 Cry3Aa 프로모터를 포함하며, 본 발명의 프로모터 변이체와 실질적 활성이 동등하거나 유사한 상동성을 갖는 프로모터 또한 본 발명의 목적 유전자 발현 증가용 프로모터 변이체의 범위에 포함될 수 있다.
본 발명의 용어, "상동성"이란 천연형(wild type) 또는 동일 활성을 갖는 변이체의 핵산 서열과의 동일성을 나타내는 것으로, 상동성의 비교는 육안으로나 구입이 용이한 비교 프로그램을 이용하여 2개 이상의 서열 간의 상동성을 백분율(%)로 계산할 수 있다. 상기 프로모터 서열과의 상동성은 본 발명에서 목적하는 유전자 발현을 위한 프로모터 활성을 보유하는 한, 본 발명에서 유래된 핵산 서열과 균등한 것임은 당업자에게 자명하다. 이러한 상동성 서열은 본 발명의 서열번호 9, 서열번호 10, 서열번호 11, 서열번호 12, 서열번호 13, 서열번호 14 및 서열번호 15으로 기재되는 핵산 서열과 바람직하게는 70% 이상일 수 있으며, 보다 바람직하게는 80% 이상, 더욱 바람직하게는 90% 이상, 보다 더욱 바람직하게는 95% 이상, 가장 바람직하게는 98% 이상의 상동성을 갖는 동일한 핵산 서열을 포함한다.
본 발명의 Cry3Aa 프로모터는 목적 단백질의 효율적인 발현을 위하여, 상기 프로모터의 하위(downstream) 유전자 발현에 필요한 프로모터 변이체 서열로 이루어진 구조물을 추가적으로 포함할 수 있다. 상기 구조물은 리더(leader) 및/또는 stab을 포함한다. "리더"란 프로모터에 의해 전사된 mRNA의 암호화 영역의 상위(upstream)의 비해독된 핵산 서열을 말한다. 바람직하게 본 발명의 "리더"는 바실러스 츄린겐시스 Cry3Aa 프로모터와 stab 사이에 존재하고, "핵산 서열(stab)"은 프로모터에 의해 전사된 mRNA의 암호화 영역의 상위(upstream)의 비해독된 핵산 서열 중 mRNA의 안정화에 기여하는 서열로 바실러스 츄린겐시스 Cry3Aa 리더의 하위에 존재하는 핵산 서열이다.
본 발명의 Cry3Aa 프로모터 변이체는 상기 -35 부위의 상위 영역에 UP 엘리먼트가 포함되어 하위 유전자 발현을 증가시킬 수 있다.
즉, 본 발명의 목적 유전자 발현 증가용 프로모터 변이체는 바실러스 츄린겐시스(Bacillus thuringiensis, Bt) Cry3Aa 프로모터 -35 부위가 TTGACA로 돌연변이된 바실러스 츄린겐시스 Cry3Aa 프로모터; 및 -35 부위의 상위 영역에 뉴클레오티드 아데닌(A)과 티민(T)이 풍부한 염기서열을 포함하는 UP 엘리먼트(UP element)가 포함된 하위(downstream) 목적 유전자 발현 증가용 프로모터 변이체일 수 있다. 특히, 본 발명의 하위 목적 유전자 발현 증가용 프로모터 변이체는 서열번호 9, 서열번호 10, 서열번호 11, 서열번호 12, 서열번호 13, 서열번호 14 및 서열번호 15로 구성된 군으로부터 선택되는 어느 하나의 서열로 구성되는 것일 수 있다.
본 발명의 구체적인 일 실시예에서는 바실러스 츄린겐시스(Bacillus thuringiensis, Bt) Cry3Aa 프로모터의 -35 부위를 TTGACA로 돌연변이시키고, -35 부위의 상위 영역에 아데닌과 티민이 풍부한 UP 엘리먼트를 삽입하되, 다양한 UP 엘리먼트 중에서 최적 조합의 프로모터를 제작하고자 하였다.
구체적으로는 Cry3Aa 프로모터와 가장 적합한 UP 엘리먼트를 찾기 위해 축퇴 프라이머(degenerate primer)를 이용하여 UP 엘리먼트 부위에 핵산 서열 다양성을 제공하였고, Cry3Aa 프로모터와 결합했을 때 가장 발현량 증가를 가져오는 엘리먼트를 스크리닝하고자 하였다. 이에 따라 상기 제조된 프로모터 변이체를 포함하는 GFP(Green Fluorescence Protein)를 리포터 단백질로 발현하는 벡터를 바실러스 서브틸리스(Bacillus subtilis)에 형질전환하여 하위 목적 유전자에 해당하는 GFP의 발현량을 확인하였다. 이를 통해 발현능 증가가 우수한 7개의 프로모터를 선별하였다. 선발된 프로모터는 각각 PuryC1(서열번호 9), PuryC3(서열번호 10), PuryC4(서열번호 11), PuryC11(서열번호 12), PuryC13(서열번호 13), PuryC19(서열번호 14) 및 PuryC23(서열번호 15)으로 명명하였다. 상기 선별된 프로모터들은 모두 목적 유전자의 발현량을 30배 이상 증가시키는 것을 확인하였다(도 1).
다른 하나의 양태로서, 본 발명은 상기 프로모터 변이체 서열을 포함하는 벡터에 관한 것이다.
본 발명에서 용어, "벡터(Vector)"란 적당한 숙주세포에서 목적 단백질을 발현할 수 있는 발현 벡터로서, 유전자 삽입물이 발현되도록 작동 가능하게 연결된 필수적인 조절 요소를 포함하는 유전자 작제물을 말한다.
일반적으로 플라스미드 벡터는 염색체 외의 환형 이중가닥 DNA이고, 세포 내에 존재하며 다양한 기능을 한다. 항생물질에 내성을 가진 물질 및 박테리오신(Bacteriocin)을 생산하여 유사한 균주나 종들을 죽이는 저해 물질로 작용하고, 색소 생성, 화합물 분해, 질소 고정과 같은 생리학적 기능을 수행한다. 최고 약 10kb되는 길이의 외래 DNA 절편을 삽입하기 위해서 제한효소 부위를 가지고 있다.
벡터로 사용되는 플라스미드와 박테리오파지의 중대한 단점인 상대적으로 작은 DNA 절편만 삽입할 수 있는 것은 플라스미드와 파지 DNA의 조작된 혼성체인 코스미드(Cosmid)를 사용하여 더 큰 DNA 절편을 클로닝 할 수 있다. 파지 입자 안으로 포장되어 들어가는 코스(cos) 부위가 있고, 세균 숙주에서 복제하는 플라스미드의 복제 시작점과 플라스미드를 선별할 수 있는 유전자를 가지고 있다. 박테리오파지 벡터 같이 시험관 내에서 단백질 외피로 포장되지만, 포장된 DNA가 대장균 숙주세포를 감염한 후에, 그 DNA는 박테리오파지 DNA보다는 플라스미드 형태로 복제하고, 용균되지 않는다. 크기는 2.5kb, 숙주세포를 감염하여 포장된 후에 cos 부위가 37에서 52kb까지 분리되면, 외래 DNA를 삽입체로 수용한다. 일반적으로 35에서 45kb가 코스미드 벡터(Cosmid vector)로 클로닝 될 수 있다.
또한 박테리오파지의 벡터의 일반적인 형태인 λ 파지의 경우는 염기쌍을 이룰 수 있는 12개 뉴클레오티드의 단일 가닥의 상보적인 말단으로 된 점착성 말단(cohesive termini) 혹은 cos를 가지는 50kb의 이중가닥으로 된 야생형 게놈에서 비롯된 것으로 용균성 경로에서 숙주세포는 새로운 바이러스가 복제되고, 자손 바이러스가 방출된 후에 융균된다. 이 형태의 DNA는 전체 52kb 크기에 3kb를 더하거나 그 게놈의 5% 만도 수용할 수 있으며, 외래 DNA를 위한 공간을 만드는 벡터는 비필수적인 DNA 조각이 제거된 형태이다.
본 발명과 관련된 발현 벡터는 Cry3Aa 프로모터와 UP 엘리먼트로 이루어진 Cry3Aa 프로모터 변이체의 벡터로, 플라스미드 벡터(예: pSC101, ColE1, pBR322, pUC8/9, pHC79 및 pUC19 등), 코스미드 벡터, 박테리오파지 벡터(예: λgt4ㆍλB, λ-Charon, λ△z1 및 M13 등), 바이러스 벡터 등을 포함한다.
바이러스 벡터는 레트로바이러스(Retrovirus), 예를 들어 인간면역결핍바이러스(Human immunodeficiency virus, HIV), 생쥐백혈병 바이러스(Murine leukemia virus, MLV), 백혈증 바이러스(Avian sarcoma and leukosis virus, ASLV), 비장괴사바이러스(Spleen necrosis virus, SNV), 라우스육종바이러스(Rous sarcoma virus, RSV), 마우스 유방암 바이러스(Mouse mammary tumor virus, MMTV) 등, 아데노바이러스(Adenovirus), 아데노 관련 바이러스(Adeno-associated virus, AAV), 헤르페스 심플렉스 바이러스(Herpes simplex virus) 등에서 유래한 벡터를 포함하나, 이에 제한되지 않는다.
본 발명에서 "작동 가능하게 연결된(operably linked)"은 일반적 기능을 수행하도록 Cry3Aa 프로모터와 상위 영역의 UP 엘리먼트로 이루어진 Cry3Aa 프로모터 변이체 핵산 서열과 목적하는 단백질을 코딩하는 뉴클레오티드 서열이 기능적으로 연결되어 있는 것을 말한다. 재조합 벡터와의 작동적 연결은 당해 기술분야에서 잘 알려진 유전자 재조합 기술을 이용하여 제조할 수 있으며, 부위-특이적 DNA 절단 및 연결은 당해 기술분야에서 일반적으로 알려진 효소 등을 사용한다.
본 발명에서 "조절 요소"란 단백질을 암호화하는 핵산 서열의 전사, 번역 또는 발현의 증진을 돕거나 이에 영향을 미치는 비해독화된 핵산 서열을 의미한다. 본 발명의 발현벡터는 조절 요소로 본 발명의 프로모터 변이체를 필수적으로 포함하고, 단백질 발현에 영향을 미칠 수 있는 발현 조절 서열, 예를 들어, 개시 코돈, 종결 코돈, 폴리아데닐화 시그널, 인핸서(enhancer), 막 표적화 또는 분비를 위한 신호서열 등을 포함할 수 있다.
폴리아데닐화 시그널은 전사체의 안정성을 증가시키거나 세포질 수송을 용이하게 한다. 인핸서 서열은 프로모터에서 다양한 부위에 위치하여 인핸서 서열이 없을 때의 프로모터에 의한 전사 활성과 비교하여, 전사 활성을 증가시키는 핵산 핵산 서열이다. 신호서열에는 숙주가 에스케리키아(Escherichia) 속 균인 경우에는 PhoA 신호서열, OmpA 신호서열 등이, 숙주가 바실러스 속 균인 경우에는 α-아밀라아제 신호서열, 서브틸리신(Subtilisin) 신호서열 등을, 숙주가 효모인 경우에는 MFα(Mating factor α) 신호서열, SUC2 신호서열 등을, 이용할 수 있으나, 이에 제한되지 않는다.
또한, 벡터는 복제 가능한 발현 벡터인 경우, 복제가 개시되는 특정 핵산 서열인 복제 개시점(replication origin)을 포함할 수 있다.
또한, 벡터는 선택 마커(selection marker)를 포함할 수 있다. 선택 마커는 벡터로 형질 전환된 세포를 선별하기 위한 것으로, 약물 내성, 영양 요구성, 세포 독성제에 대한 내성 또는 표면 단백질의 발현과 같은 선택 가능 표현형을 부여하는 마커들이 사용될 수 있다. 선택제(selective agent)가 처리된 환경에서 선택 마커를 발현하는 세포만 생존하므로 형질 전환된 세포를 선별 가능하다. 선택 마커의 대표적인 예로써, 영양 요구 마커(auxotrophic marker)인 ura4, leu1, his3 등을 들 수 있으나 상기 예에 의해 본 발명에서 사용될 수 있는 마커의 종류가 제한되는 것은 아니다.
본 발명에서 회수되는 목적 단백질의 정제를 용이하게 하기 위하여, 벡터의 제조시 필요에 따라 다른 서열을 추가적으로 포함할 수 있다. 상기 추가적으로 포함될 수 있는 서열은 단백질 정제용 태그 서열일 수 있으며, 예컨대, 글루타티온 S-트랜스퍼라제(Pharmacia, USA), 말토스 결합 단백질(NEB, USA), FLAG(IBI, USA) 및 6x His(hexahistidine; Quiagen, USA) 등이 있고, 가장 바람직하게는 6x His(hexahistidine)일 수 있으나, 상기 예들에 의하여 목적 단백질의 정제를 위하여 필요한 서열의 종류가 제한되는 것은 아니다. 상기 융합 서열이 포함되어 있는 벡터에 의해 발현된 융합 단백질의 경우, 친화성 크로마토그래피에 의해 정제될 수 있다. 예컨대, 클루타티온 S-트랜스퍼라제가 융합된 경우에는 이 효소의 기질인 글루타티온을 이용할 수 있고, 6x His인 경우에는 Ni-NTA His-결합 레진 컬럼(Novagen, USA)을 이용하여 원하는 목적 단백질을 용이하게 회수할 수 있다.
상기 목적 단백질을 코딩하는 핵산 서열은 프로모터 하위(downstream)에 삽입되어 발현된다. 상기 벡터로 삽입 가능한 목적 단백질은 특별히 제한되지 않으며 의학적 목적 또는 산업적 목적의 다양한 단백질을 삽입할 수 있다. 의학, 산업적으로 유용한 목적 단백질에는 호르몬, 호르몬 유사체, 효소, 효소 저해제, 사이토카인, 응고인자, 수송 단백질, 수용체, 수용체의 단편, 조절 단백질, 구조 단백질, 독소 단백질, 전사 인자, 항원, 항체, 항체 단편, 단일클론 항체 등이 있다.
본 발명의 구체적인 일 실시예에서는 상기 변이체 프로모터 서열의 하위에 GFP(Green Fluoresence protein)를 작동 가능하도록 연결하여 벡터를 제조함으로써, 상기 벡터를 포함한 바실러스 균주의 단백질 발현 활성을 측정하였다(도 1). 그 결과, 본 발명의 선발된 프로모터 변이체는 모두 대한민국 등록특허 제1280503호의 개시된 개량된 Cry3Aa 프로모터 P84에 비해 30배 이상 발현양이 증가하였다(도 1 참조).
또한, 실제로 대량생산이 요구되는 목적 단백질의 일 종으로 단백질 분해효소 aprE를 이용하여, 단백질 발현 활성 및 생산된 단백질의 활성을 측정하였다. 그 결과, 본 발명의 프로모터인 PuryC13 및 PuryC23 프로모터를 사용한 경우 P84 프로모터를 사용한 경우에 비해 aprE의 활성이 현저히 증가하였으며, 단백질 발현 또한 현저히 증가하였음을 확인하였다(도 2의 A 및 B 참조).
또 다른 하나의 양태로서 본 발명은 상기 벡터를 포함하는 숙주세포에 관한 것이다.
본 발명에서 사용되는 용어 "숙주세포"란 다른 미생물 또는 유전자를 기생시켜 영양을 공급하는 세포로, 벡터가 숙주세포에 형질전환됨으로써 숙주세포 내에서 다양한 유전적 또는 분자적 영향을 미치게 되는 세포를 의미한다. 숙주세포는 외부 DNA를 받아들일 수 있는 수용성(competence) 상태에서, 벡터와 같은 외부 DNA가 삽입될 수 있는데, 벡터가 숙주세포에 성공적으로 도입되면, 해당 벡터의 유전형질을 숙주세포에 제공하게 된다. 바람직하게 본 발명의 숙주 세포는 그람 양성 균일 수 있으며, 예를 들면 바실러스 서브틸리스(Bacillus subtilis), 바실러스 리케니포미스(Bacillus licheniformis), 바실러스 메가테리움(Bacillus megaterium), 바실러스 시리우스(Bacillus cereus), 바실러스 츄린겐시스(Bacillus thuringiensis), 바실러스 브레비스(Bacillus brevis), 및 락토바실러스(Lactobacillus ) 속 또는 패니바실러스(Panibacillus) 속인 세균 등을 포함하나, 상기 예에 의해 본 발명의 벡터가 형질전환될 수 있는 숙주세포가 제한되는 것은 아니다. 바람직하게 본 발명에서는 바실러스 서브틸리스를 숙주세포로 사용하였고, 더 바람직하게는 바실러스 서브틸리스 168에 본 발명의 벡터를 형질전환시켜 본 발명의 프로모터 활성을 확인한 결과, 목적 유전자 발현이 천연형 프로모터를 사용하는 경우에 비해 증가함을 확인하였다.
숙주 세포에 벡터를 도입하는 방법은 형질전환 방법이 이용될 수 있다. "형질전환"이란 DNA를 숙주로 도입하여 DNA가 염색체의 인자로서 또는 염색체 통합 완성에 의해 복제 가능하게 되는 것으로 외부의 DNA를 세포 내로 도입하여 인위적으로 유전적인 변화를 일으키는 현상을 의미한다. 형질전환 방법은 임의의 형질전환 방법이 사용될 수 있으며, 당업계의 통상적인 방법에 따라 용이하게 수행할 수 있다. 일반적으로 형질전환 방법에는 CaCl2 침전법, CaCl2 방법에 DMSO (dimethyl sulfoxide)라는 환원 물질을 사용함으로써 효율을 높인 하나한(Hanahan) 방법, 전기천공법(electroporation), 인산칼슘 침전법, 원형질 융합법, 실리콘 카바이드 섬유를 이용한 교반법, 아그로박테리아(Agrobacteria) 매개된 형질전환법, 폴리에틸렌글리콜(polyethylene glycol, PEG)를 이용한 형질전환법, 덱스트란 설페이트(Dextran sulfate), 리포펙타민(lipopectamine) 및 건조/억제 매개된 형질전환 방법 등이 있다. 본 발명의 플라스미드를 형질전환시키기 위한 방법은 상기 예들에 국한되지 않으며, 당업계에서는 통상적으로 사용되는 형질전환 방법이 제한 없이 사용될 수 있다.
또 다른 하나의 양태로서 본 발명은 (a) 목적 단백질 유전자와 본 발명의 프로모터 변이체를 포함하는 벡터를 제조하는 단계; (b) 상기 단계 (a)에 의해 제조된 벡터를 숙주세포에 도입하는 단계; 및 (c) 상기 단계 (b)의 숙주세포로부터 목적 단백질을 회수하는 단계를 포함하는 단백질의 생산방법에 관한 것이다.
본 발명의 Cry3Aa 프로모터와 상위 영역의 UP 엘리먼트로 이루어진 Cry3Aa 프로모터 변이체와 목적 단백질 유전자를 포함하는 벡터는 상기 서술한 바와 같이 당업계에서 통상적으로 사용되는 벡터에 제한효소 및 중합효소 연쇄반응법(Polymerase chain reaction, PCR)을 이용하여 제조할 수 있다. 또한 제조된 벡터는 일반적인 형질 전환 방법으로 숙주세포에 도입함으로써 제조될 수 있는데, 본 발명의 구체적인 실시예에서는 자연도입법을 이용하여 바실러스 서브틸리스 168에 도입하여, 단백질 발현량을 측정하였다.
또한, 상기 방법으로 형질전환된 숙주세포는 필요에 따라 당업계에서 통상적으로 사용되는 배양 방법을 통하여 배양될 수 있으며, 사용될 수 있는 배지 및 배양기간은 필요에 따라 당업자에 의해 임의적으로 선택될 수 있다. 바람직하게 본 발명은 형질 전환된 바실러스 균주를 LB(Luria Bertani) 배지에서 24시간 동안 배양하여 목적 단백질의 생산을 유도하였다.
배양을 통해 생산된 목적 단백질을 수득하기 위하여 본 발명은 목적 단백질을 회수하는 단계를 추가적으로 포함할 수 있다. 본 발명에서 목적 단백질은 상기 벡터가 도입된 숙주세포를 배양한 배양물로부터 수득할 수 있으며, 상기 배양물은 숙주세포 자체와 배양액을 모두 포함할 수 있으며, 숙주세포를 배양하는 과정에서 생기는 모든 물질을 포함할 수 있다.
본 발명의 발현 시스템을 이용하는 경우, 바실러스 균주에서 목적 단백질의 발현을 현저히 증가시킬 수 있어, 단백질, 폴리펩티드 약물과 같은 목적 단백질의 대량 생산을 통해 생명공학, 의학 뿐만 아니라 산업적으로 유용한 단백질의 생산에 효과적으로 사용될 수 있다.

-35의 상위 부분에 UP 엘리먼트(UP element)를 포함하는 Cry3Aa 프로모터 변이체(PuryC)를 제작하기 위하여 먼저 P5D 프로모터와 GFP 유전자가 결합된 카세트를 포함하는 플라스미드 pD5D-GFP를 제작하였다.
이를 위해 Cry3Aa 프로모터의 상위(upstream) 부위(Cry3Aa-up)는 플라스미드 pA84(대한민국 등록특허 제1280503호 참조)를 주형으로 프라이머 C3Aa-f1(서열번호 1)과 P83r1(서열번호 2)를 사용하여 PCR로 확보하였다. 프로모터 P5D는 플라스미드 pD52(J.Biotechnol. 2010. 149: 16-20)를 주형으로 프라이머 P5D-f1(서열번호 3)과 stab-r1(서열번호 4)를 사용하여 PCR로 확보하였다. GFP 유전자는 플라스미드 pAD123(Gene 1999. 226: 297-305)을 주형으로 하고 프라이머 5Dgfp-f1(서열번호 5)과 5Dgfp-r1(서열번호 6)를 사용하여 PCR로 확보하였다. 사용한 프라이머는 하기와 같다.

상기 확보된 PCR 산물 Cry3Aa-up, P5D 및 GFP 유전자는 결합 PCR을 통해 서로 연결하여 5D-GFP를 제작하였다. 제작된 5D-GFP는 EcoRΙ과 BsiWΙ으로 절단한 후 플라스미드 pDG1661(Gene 1996. 180: 57-61)의 같은 사이트에 클로닝하여 플라스미드 pD5D-GFP를 완성하였다.

삽입 벡터 제작
Cry3Aa 프로모터와 가장 적합한 UP 엘리먼트를 찾기 위해 축퇴 프라이머(degenerate primer)를 이용하여 UP 엘리먼트 부위에 핵산 서열 다양성을 제공하고 Cry3Aa 프로모터와 결합했을 때 가장 발현량 증가를 가져오는 엘리먼트를 스크리닝하였다.
이를 위해 상기 pD5D-GFP를 주형으로 하고 프라이머 UryCf(서열번호 7)와 GFP-r1(서열번호 8)을 사용하여 PCR을 수행하였다. 사용한 프라이머는 하기와 같다.

이렇게 확보된 프로모터 PuryC는 EcoRΙ과 BsiWΙ으로 절단한 후 상기 플라스미드 pDG1661의 같은 부위에 클로닝하여 pUryC-GFP를 제작하였다.
상기 플라스미드를 자연도입법(Harwood CR and Cutting SM. 1990. p67. Molecular Biological Methods for Bacillus. John Wiley & Sons Ltd.)으로 바실러스 서브틸리스 168(Kunst F. et al. 1997. Nature. 390: 249-256)에 도입한 후 LB 아가(Agar) 배지에서 7주째 강한 형광을 보이는 콜로니(colony)를 선발하였다. 선발된 균주들은 LB 배지에서 24시간 배양한 후 GFP의 발현을 측정하였는데 기존 개량된 Cry3Aa 프로모터인 P84(대한민국 등록특허 제1280503호 참조, 양성 대조군)와 GFP를 포함하고 있는 균주와 발현량을 비교하였다.
구체적으로, 본 실험에서 양성 대조군으로 사용한 P84는 바실러스 츄린겐시스 Cry3Aa 프로모터 -35 부위가 TTGACA로 돌연변이 되고, Cry3Aa 프로모터 -10 부위가 TATAAT로 돌연변이되고; 이에 추가적으로 cry3Aa 구조 유전자의 N-말단의 49개의 뉴클레오티드 서열을 추가로 포함하여 발현 유도 활성이 증진된 개량된 Cry3Aa 프로모터 변이체에 해당한다. 해당 cry3Aa 구조 유전자의 N-말단의 49개의 뉴클레오티드 서열을 추가하는 변이를 통하여 P84는 발현능을 20배 내지 30배 증진시키는 효과를 보였다. 본 발명에서 사용한 벡터들의 구조는 도 3과 같다.
상기 제조한 각 균주들의 발현량 비교 결과를 정리하면 하기 표 1과 같다.

상기 비교 결과 선발된 PuryC 프로모터는 상기 기존 개량된 프로모터 P84에 비해 30배 이상 발현양이 증가하는 것을 확인하였다(도 1 및 표 1). 즉, 기존 개량되어 발현능이 20내 내지 30배 증가된 프로모터 P84에 비교하여서도, 본 발명의 프로모터 변이체가 발현능을 30배 이상 증가시키는 것을 확인한 것이다.
상기 선발된 프로모터는 각각 PuryC1(서열번호 9), PuryC3(서열번호 10), PuryC4(서열번호 11), PuryC11(서열번호 12), PuryC13(서열번호 13), PuryC19(서열번호 14) 및 PuryC23(서열번호 15)으로 명명하였다(도 3).

프로모터를 이용한 단백질 분해효소 발현
상기 실시예 1-2에서 선별된 프로모터 중 대표적으로 PuryC13과 PuryC23을 사용하여 단백질 분해효소 aprE의 발현능을 확인하였다. 이를 위해 프로모터 PuryC13과 PuryC23은 상기 프라이머 C3Aa-f1(서열번호 1)과 stab-r1(서열번호 4)을 이용하여 PCR로 확보하였다.
한편, aprE 유전자는 상기 바실러스 서브틸리스 168의 염색체를 주형으로 하고 프라이머 5DaprE-f1(서열번호 16)과 5DaprE-r1(서열번호 17)을 사용하여 PCR로 확보하였다. 상기 사용한 프라이머는 하기와 같다.

확보된 aprE 유전자는 상기 프로모터의 PCR 산물과 결합 PCR을 수행하여 PuryC13-aprE와 PuryC23-aprE를 확보한 후 EcoRΙ과 BsiWΙ으로 절단하여 상기 pDG1661의 같은 부위에 클로닝하여 플라스미드 pUryC13-aprE 및 pUryC23-aprE를 제작하였다.
상기 제작된 플라스미드들은 자연도입법으로 바실러스 서브틸리스 168에 도입한 후 Lee 등의 방법(J. Biotechnol . 2010. 149: 16-20)으로 단백질 분해효소 활성을 측정하였고 발현 정도를 단백질 전기영동으로 확인하였다. 이를 정리하면 하기 표 2와 같다.

상기 표 2 및 도 2에서 확인할 수 있듯이, 비교 결과 PuryC13 및 PuryC23 프로모터를 사용한 경우 모두 P84 프로모터를 사용한 경우에 비해 aprE의 활성이 현저히 증가하였으며, 단백질 발현 또한 현저히 증가하였음을 확인하였다(도 2의 A 및 B).
상기 결과를 종합하면, Cry3Aa 프로모터의 -35 부위의 상위 영역에 뉴클레오티드 아데닌(A)과 티민(T)이 풍부한 UP 엘리먼트(UP element)를 첨가한 Cry3Aa 프로모터 변이체인 본 발명의 프로모터 PuryC1, PuryC3, PuryC4, PuryC11, PuryC13, PuryC19 및 PuryC23은 바실러스 균주에서 목적 단백질의 발현양을 약 30 배 이상 증가시킬 수 있음을 확인하였다.

- 바실러스 츄린겐시스(Bacillus thuringiensis, Bt) Cry3Aa 프로모터 -35 부위가 TTGACA로 돌연변이된 바실러스 츄린겐시스 Cry3Aa 프로모터; 및 -35 부위의 상위 영역에 UP 엘리먼트(UP element)가 포함된 목적 유전자 발현 증가용 프로모터 변이체로서,
상기 프로모터 변이체는 서열번호 9, 서열번호 10, 서열번호 11, 서열번호 12, 서열번호 13, 서열번호 14 및 서열번호 15로 구성된 군으로부터 선택되는 어느 하나의 서열로 구성되는 것인, 목적 유전자 발현 증가용 프로모터 변이체.
- 삭제
- 제1항의 프로모터 변이체를 포함하는 벡터.
- 제3항에 있어서, 상기 벡터에 목적 유전자를 추가로 포함하는 벡터.
- 제3항에 있어서, 단백질 정제용 태그 서열을 추가로 포함하는 벡터.
- 제3항 내지 제5항 중 어느 한 항의 벡터를 포함하는 숙주세포.
- 제6항에 있어서, 상기 숙주세포는 그람 양성 세균인 숙주세포.
- 제7항에 있어서, 상기 그람 양성 세균은 바실러스 서브틸리스(Bacillus subtilis), 바실러스 리케니포미스(Bacillus licheniformis), 바실러스 메가테리움(Bacillus megaterium), 바실러스 시리우스(Bacillus cereus), 바실러스 츄린겐시스(Bacillus thuringiensis), 바실러스 브레비스(Bacillus brevis), 및 락토바실러스(Lactobacillus ) 속 또는 패니바실러스(Panibacillus) 속인 세균으로 구성된 군으로부터 선택되는 어느 하나의 그람 양성 세균인 숙주세포.
- (a) 제1항의 프로모터 변이체와 목적 단백질 유전자를 포함하는 벡터를 제조하는 단계;
(b) 상기 단계 (a)에 의해 제조된 벡터를 숙주 세포에 도입하는 단계; 및
(c) 상기 숙주세포로부터 목적 단백질을 회수하는 단계를 포함하는 목적 단백질의 생산방법.
- 제9항에 있어서, (d) 컬럼을 이용하여 목적 단백질을 정제하는 단계를 추가로 포함하는 목적 단백질의 생산방법.
- 제9항에 있어서, 상기 목적 단백질은 호르몬, 호르몬 유사체, 효소, 효소 저해제, 사이토카인, 응고인자, 수송 단백질, 수용체, 수용체의 단편, 조절 단백질, 구조 단백질, 독소 단백질, 전사 인자, 항원, 항체, 항체 단편 및 단일클론 항체로 구성된 군으로부터 선택되는 어느 하나 이상의 목적 단백질의 생산방법.
