Abstract
본 발명은 천련자 (Melia toosendan) 추출물, 분획물 또는 이로부터 분리한 리모노이드 화합물을 세놀리틱 제제의 유효성분으로 포함하는 세포의 노화 관련 증상 억제 및 노화 관련 질환을 예방 또는 치료하는 조성물에 관한 것으로, 천련자로부터 얻은 추출물, 분획물 또는 이로부터 분리 정제한 리모노이드 화합물은 노화 표현형 억제 및 감소된 노화성 자가포식 능력 (Autophagy)의 활성화 효과가 우수하게 나타나며, 이는 특발성 폐 섬유증 (Idiopathic Pulmonary Fibrosis, IPF) 등의 노화 관련 질환의 병리학적 진행을 완화시켜 약품, 건강기능식품, 식품 첨가제, 천연 화장품 등에 유용하게 적용될 수 있다.
Description
본 발명은 세놀리틱 활성을 갖는 천련자 (Melia toosendan) 추출물, 분획물 또는 이로부터 분리한 리모노이드계 화합물을 유효성분으로 포함하는 세포의 노화 관련 증상 억제 및 노화 관련 질환을 예방 또는 치료하는 조성물에 관한 것으로, 더욱 상세하게는 천련자로부터 획득한 리모노이드계 화합물로부터 노화성 자가포식 능력 (Autophagy) 감소의 억제 효과를 통하여 특발성 폐 섬유증 (Idiopathic Pulmonary Fibrosis, IPF)의 병리학적 진행을 완화하는 활성을 지닌 천연물로부터 유래한 조성물에 관한 것이다.
유엔 경제사회이사국 (UN DESA)의 2017년 6월 보고서에서 전 세계 60세 이상의 고령화 인구는 9억 6200만 명 (세계인구의 12.3%)에서 2050년에는 2배 이상 증가한 21억 명 (세계인구의 약 22%)으로 예측하고 있다. 2019년 3월에 발표된 우리나라 통계청 “장래인구 특별추계” 자료를 보면 2017년 65세 이상 고령 인구가 707만 명 (13.8%)에서 전 세계에서 고령화가 가장 빠른 속도로 증가하여 2067년에는 65세 이상 인구 비중이 47%까지 증가하여 유소년 인구보다 5.7배 많을 것으로 전망하였다. 총 인구 중에서 생산가능인구 (15~64세)에 대한 고령 인구 (65세 이상)의 백분비로 표시되는 노년부양비는 우리나라에서 2017년 105.1명에서, 2026년 206명, 2056년 502.2명으로 높아져, 2056년부터는 고령인구가 유소년인구보다 5배 이상 많아질 전망이다. 인구의 고령화는 국가 경제적으로도 심각한 문제를 일으키게 되는데 인구 노령화는 전 세계적으로 나타나는 메가트랜드 (megatrends)로서 세계 각국에서 해결방안으로 노화 연구에 관심이 급증하고 있다.
노화가 진행됨에 따라 신체 조직은 스트레스로부터 회복되는 능력이 점진적으로 떨어지며, 전 세계적으로 노인 인구가 증가함에 따라 3개월 이상의 발생 경과와 회복이 어려운 병리적 상태를 가지는 만성 질환 환자도 증가하는 현상이 나타나고 있다. 한국보건사회연구원의 “노인의 건강실태와 정책과제” 연구 보고서에서 의사의 진단을 받은 만성 질환 유병률의 경우, 전체 노인의 89.2%가 만성 질환을 갖고 있으며, 만성 질환을 3개 이상 지닌 경우도 46.2%에 달해 전체 노인이 평균 2.6개의 만성 질환이 있는 것으로 보고하였다. 나이별로는 나이가 많을수록 대체로 만성질환 유병률도 증가해 65∼69세 연령군은 84.0%, 80∼84세 연령군은 94.1%이고, 복합 만성 질환 유병률은 65∼69세 연령군 60.9%에서 80∼84세 연령군 76.5%로 매우 증가한다. 따라서 노화 과정에서 증가하는 만성 질환 유병률의 증가는 노인 인구의 삶의 질을 저하하며 국가적으로는 의료비 부담의 급격한 증가를 수반하여야 하므로 노화관련 만성질환자의 감소는 미래 사회를 위해 국가적으로 해결해야 할 중요한 과제 중 하나이다.
노화의 주요 메커니즘 중 하나인 세포 노화는 텔로미어 단축, 산화 스트레스, DNA 손상 또는 발암 유전자의 비정상적인 활성화와 같은 스트레스 또는 자극의 결과로 발생하는 돌이킬 수 없는 세포주기 정지 상태를 의미한다 (Hayakawa et al., 2015). 노화 세포의 특징으로 배양 및 조직 샘플에서 느린 세포 분열 경향성을 보이고, 관련 베타-갈락토시다아제 (senescence-associated β-galactosidase, SA-β-gal)의 증가를 나타내며, 육안으로 확인하였을 때 세포의 크기가 커지며 평평한 세포 모양을 보인다 (Wang&Dreesen, 2018). 또한, 세포 노화의 지표로 세포 분열을 늦추는 단백질인 사이클린-의존성 인산화 효소 억제인자(cyclin-dependent kinase inhibitors, CDKI), p16INK4A, p21CIP1 및 종양 억제 유전자인 p53의 발현이 증가하여 노화가 진행 시 이들의 증가된 단백질 발현이 확인된다.
이외에도 노화 세포는 성장 인자와 관련된 다수의 가용성 인자 및 IL-1, IL-6, IL-8, 프로테아제, 세포외기질 금속함유 단백질 분해효소 (matrix metalloproteinase, MMP)를 포함하는 노화관련 분비 표현형 (senescence-associated secretory phenotype, SASP)을 주변 환경으로 분비시키며, 이에 따라 국소 조직 환경을 저하하고 다양한 조직 및 기관에서 염증을 유발하여 기능 장애 및 질병에 이바지할 가능성이 크다고 알려져 있다 (Lim, Park, & Kim, 2015).
흥미롭게도, 노화 세포의 30%만 선택적으로 제거하여도 노화 관련 표현형의 개선이 크게 나타나며, 노화 세포로 인한 부담을 감소시킴으로써 관련된 장애와 만성 질환을 개선할 수 있음이 최근 동물실험 및 임상시험에서 밝혀졌다 (Xu, 2018, Justice & Nambiar, 2019).
또한, 노화 세포에서 증가한 p16INK4A 및 베타-갈락토시다아제 단백질이 감소하면 노화 세포에서 보이는 노화 관련 증상을 저해되었으며, 노화된 세포로부터 분비되는 노화 관련 분비 표현형 (SASP)이 감소할 경우도 노화 세포의 부담을 경감시키거나 노화 관련 현상이 개선되었으며, 궁극적으로 노화에서 유발된 건강 상태가 개선되었다. 이러한 노화된 세포를 선택적으로 제거함으로써 주변 세포에 악영향을 미치는 SASP를 억제하고 세포 분열을 멈추게 하는 단백질의 발현을 감소시켜 노화를 지연 및 완화하거나 예방하는 저분자 물질을 세놀리틱 (senolytics)이라 정의한다.
특발성 폐 섬유증 (Idiopathic Pulmonary Fibrosis, IPF)은 일반적으로 50세 이상의 연령에서 발생하고 나이가 들면서 급격한 유병율의 증가가 나타남으로써 최근에는 노화와 관련된 많은 임상 증후군 중 하나로 여겨지며 전 세계적으로 노화 인구의 급격한 증가와 함께 IPF의 발생률과 유병률 모두 증가하고 있다. 특히, IPF는 폐포 상피의 돌이킬 수 없는 파괴로 폐 기능이 점진적으로 감소하여 예후가 좋지 않고 치료 방법이 제한적인 치명적인 폐 질환으로 알려져 있다. IPF는 진단 시점으로부터 약 3년의 평균 생존율을 나타내며 증상발현과 함께 급속하게 악화하는 특징을 갖는다. 그러나, 현재까지 FDA 승인된 약물은 항섬유화제인 닌텐다닙 (Nintedanib)과 피르페니돈 (Pirfenidone)은 생존이나 삶의 질을 향상시키지 않고 폐 기능의 감소를 늦추기만 할 뿐, 질병을 완치시키는 것이 아닌 한계가 있어 새로운 치료제의 개발이 요구되고 있다.
IPF 유래 페포 상피 및 섬유아세포에서는 p16INK4A, p21CIP1 및 노화관련 베타-갈락토시다아제와 같은 노화 마커의 증가가 관찰되며 이러한 현상은 노화가 IPF 질병의 유발과 큰 관련성이 있음을 제시한다 (Gulati, S., 2019). 최근, 세놀리틱 저해물질인 다사티닙 (Dasatinib)과 케르세틴 (Quercetin) 혼합물질이 폐 섬유증을 초래하는 블레오마이신 (Bleomycin)을 처리한 손상 세포 모델 및 방사선을 조사하여 유발된 폐 섬유증 쥐 모델에서 모두 노화된 페포 상피세포를 선택적으로 제거하여 섬유증을 완화하는 데 효과적임이 보고되었다. 이러한 연구 결과는 폐포 상피세포 노화가 폐 섬유증 발달에 기여하며, 세놀리틱 약물이 IPF에서 유망한 치료법으로 가능성을 제시한다 (Schafer, 2018).
식물 유래 물질의 경우 기존의 합성 물질보다 부작용이 낮고 안정성이 높음으로써 목표하는 환자군 대다수가 고연령인 다양한 노화 관련 질병에 대한 유망한 치료제로 인식되고 있다. 한약재로 잘 알려진 천련자는 천련 (Melia toosendan)의 열매로서 동남아시아, 인도, 중국, 호주 등의 아열대에서 열대지방까지 넓게 자라고 있다. 전통의학적으로 천련자는 항균, 구충 작용 및 진통약으로 복통 및 산통에 효과가 있음이 알려져 있으며 시중에서 쉽게 접할 수 있는 한약재 중 하나로 산업적으로 개발하여 새로운 가치를 창출하는데 매우 용이하다.
천련자와 관련된 선행기술 문헌으로서, 한국출원특허 1020200116031에는 천련자 추출물을 포함하는 구강질환 예방 또는 치료용 조성물에 관하여 개시되어 있고, 한국등록특허 10159718700007호에는 위암에 대한 항암 활성을 갖는 천련자 추출물을 유효성분으로 함유하는 조성물에 대하여 개시되었으나, 이들 문헌에는 천련자 추출물에 포함된 화합물을 규명하지 않았으며, 천련자 추출물, 이의 분획물 또는 이로부터 분리된 특정 화합물의 세놀리틱 활성에 관하여 개시된 바가 전혀 없다.
또 다른 비특허문헌 (Xie, F., 2018, Wu, S. B., 2010)에서는 천련자로부터 분리된 단일성분으로 항염, 항알러지 및 항암 효과를 비교하여 개시하고 있으나, 본 특허에서 주장하는 활성과 다른 활성을 개시하고 있어 본 발명과 다르다.

본 발명은 세놀리틱 저해할성을 갖는 천련자 (Melia toosendan) 추출물, 분획물 또는 이로부터 분리한 리모노이드계 화합물을 포함하는 세포의 노화 관련 증상 억제 및 노화 관련 질환을 예방 또는 치료하는 조성물을 제공하는 데 있다. 더욱 상세하게는 천련자로부터 획득한 리모노이드계 화합물로부터 노화성 자가포식 능력 감소를 활성화하는 효과를 통하여 IPF의 병리학적 진행을 완화하는 천련자로부터 유래한 조성물에 제공하는 데 있다. 또한, 본 발명의 목적은 천련자 추출물부터 분리한 화합물의 분리 방법 및 구성 화합물을 구조를 제공하는 데 있다.
본 발명은 천련 (Melia toosendan)의 열매인 천련자의 추출물, 이의 분획물 또는 이로부터 분리된 리모노이드 화합물을 세놀리틱 저해활성을 유효성분으로 함유하는 세포 노화 또는 노화 관련 질환의 예방 또는 치료용 조성물을 제공한다.
상기 천련자 추출물로부터 분리된 리모노이드 화합물은 하기 화학식 1의 트리칠리닌 (Trichilinin, 화합물 1), 멜리아투세닌 P (Meliatoosenin P, 화합물 2), 12-하이드록시,29-엑소아모라스타톤 (12-hydroxy, 29-exo-amoorastatone, 화합물 3), 12-하이드록시,29-엔도아모라스타톤 (12-hydroxy, 29-endo-amoorastatone, 화합물 4), 및 투센다닌 (Toosendanin, 화합물 5)으로 이루어진 군에서 선택되는 1종 이상을 포함한다.

본 발명은 상기 화학식 1의 트리칠리닌 (Trichilinin, 화합물 1), 멜리아투세닌 P (Meliatoosenin P, 화합물 2), 12-하이드록시,29-엑소아모라스타톤 (12-hydroxy, 29-exo-amoorastatone, 화합물 3), 12-하이드록시,29-엔도아모라스타톤 (12-hydroxy, 29-endo-amoorastatone, 화합물 4), 및 투센다닌 (Toosendanin, 화합물 5)으로 이루어진 군에서 선택되는 1종 이상을 포함하는 세포 노화 또는 노화 관련 질환의 예방 또는 치료용 조성물을 제공한다.
상기 천련자 추출물은 건조된 천련자를 물, C1 내지 C4의 저급 알코올, 헥산, 아세톤, 메틸렌 클로라이드, 에틸아세테이트 및 이들의 혼합용매로 구성된 군으로부터 선택된 어느 하나의 용매로 추출한 것일 수 있다. 보다 바람직하게는 상기 C1 내지 C4의 저급 알코올은 에탄올의 30~100% 수용액일 수 있다.
상기 노화 관련 질환은 피부 노화, 근감소증, 특발성 폐 섬유증, 특발성 폐 섬유증, 녹내장, 백내장, 제 2형 당뇨병, 골관절염으로 이루어진 군에서 선택된 1 이상인 것을 특징으로 한다.
또한, 본 발명은 상기 화학식 1의 트리칠리닌 (Trichilinin, 화합물 1), 멜리아투세닌 P (Meliatoosenin P, 화합물 2), 12-하이드록시,29-엑소아모라스타톤 (12-hydroxy, 29-exo-amoorastatone, 화합물 3), 12-하이드록시,29-엔도아모라스타톤 (12-hydroxy, 29-endo-amoorastatone, 화합물 4), 투센다닌 (Toosendanin, 화합물 5)으로 이루어진 군에서 선택되는 1종 이상의 화합물을 포함하는 천련자 추출물 및 이의 분획물을 함유하는 것을 특징으로 하는 세놀리틱 제제의 유효성분으로 함유하는 세포 노화 또는 노화 관련 질환의 예방 또는 개선용 식품 첨가제 및 건강기능식품에 관한 것이다.
또한, 본 발명은 상기 화학식 1의 트리칠리닌 (Trichilinin, 화합물 1), 멜리아투세닌 P (Meliatoosenin P, 화합물 2), 12-하이드록시,29-엑소아모라스타톤 (12-hydroxy, 29-exo-amoorastatone, 화합물 3), 12-하이드록시,29-엔도아모라스타톤 (12-hydroxy, 29-endo-amoorastatone, 화합물 4), 투센다닌 (Toosendanin, 화합물 5)으로 이루어진 군에서 선택되는 1종 이상의 화합물을 포함하는 천련자 추출물 및 이의 분획물을 함유하는 것을 특징으로 하는 세포 노화 또는 노화 관련 질환의 예방 또는 개선용 천연 화장품에 관한 것이다.
상기 세포 노화 또는 노화 관련 질환의 예방 또는 개선용 식품 첨가제ㅏ 건강기능식품 또는 천연 화장품으로 예방 및 개선할 수 있는 것으로 피부 노화, 근감소증, 특발성 폐 섬유증, 특발성 폐 섬유증, 녹내장, 백내장, 제 2형 당뇨병, 골관절염에서 선택된 것을 특징으로 한다.
본 발명의 천련자 추출물은 유기용매 (알코올, 에틸아세테이트, 아세톤, n-헥산, 에틸아세테이트 등)에 의한 추출, n-헥산, 에틸아세테이트, n-부탄올과 물의 분배, 디아이온 HP-20 레진과 같은 흡착수지 사용방법, 칼럼 크로마토그래피에 의한 방법 등 식물체 성분의 분리 추출에 이용되는 공지된 방법을 단독 또는 적합하게 조합하여 용이하게 얻을 수가 있다. 조 추출물은 필요에 따라서 상법에 따라서 더욱 정제할 수 있다.
본 발명의 천련자 추출물은 건조된 천련의 열매를 C1 내지 C4의 저급 알코올로 추출하여 얻는 것이 바람직하며, 상기 C1 내지 C4의 저급 알코올로는 메탄올, 에탄올, 프로판올, 이소프로판올, n-부탄올 등을 이용할 수 있다. 보다 바람직하게는, 본 발명의 천련자 추출물은 천련자를 100% 메탄올 또는 100% 에탄올로 추출한 추출물이거나, 또는, 천련자를 30~100% 메탄올 수용액(또는 메탄올) 또는 에탄올 수용액 (또는 에탄올)의 추출물을 n-부탄올로 분획한 분획물이다.
본 발명의 천련자로부터 분리된 화합물은, 상기 건조된 천련자 추출물을 n-헥산, 에틸아세테이트와 n-부탄올, 물로 분획한 후, 에틸아세테이트 분획물에서 분리하여 얻을 수 있으며, 바람직하게는 천련자를 30~100%의 에탄올 수용액으로 추출한 추출물의 에틸아세테이트 분획물을 크로마토그래피로 분리하여 얻을 수 있다.
상기 크로마토그래피는 실리카겔 컬럼 크로마토그래피 (silica gel column chromatography), LH-20 컬럼 크로마토그래피 (LH-20 column chromatography), RP-18 컬럼 크로마토그래피 (RP-18 column chromatography), 박층 크로마토그래피 (TLC, thin layer chromatography) 및 고성능 액체크로마토그래피 (high performance liquid chromatography) 등이 이용될 수 있다.
본 발명에 따라 천련자로부터 분리된 리모노이드 계 화합물 중에서 주요 화합물인 투센다닌 (화합물 5)는 세놀리틱 효능이 가장 우수하였다. 또한, 이러한 리모노이드 계 화합물은 천련자에 함유하는 주요 화합물 계열들로써 쉽게 분리할 수 있을 뿐만 아니라 안정도도 높으므로 식품, 의약품, 화장품, 건강기능식품으로 유용하게 적용될 수 있다.
한편, 본 발명의 화합물은 당해 기술 분야에서 통상적인 방법에 따라 합성될 수 있으며, 약학적으로 허용 가능한 염으로 제조될 수도 있다.
또한, 본 발명은 천련자 추출물, 이의 분획물 또는 이로부터 분리된 상기 화학식 1의 화합물 군에서 선택되는 1종 이상의 화합물을 함유하는 것을 특징으로 노화 세포의 노화 관련 증상 억제 및 노화 관련 질환을 예방 또는 치료하는 약학 조성물을 제공한다. 상기 추출물 또는 화합물을 포함하는 약학 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 상기 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 본 발명의 상기 추출물 또는 화합물에 적어도 하나 이상의 부형제, 예를 들면, 전분, 탄산칼슘, 수크로스 또는 락토오스, 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜, 폴리에틸렌글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 천련자 추출물, 이의 분획물 또는 이로부터 분리된 화합물을 포함하는 약학 조성물의 투여량은 치료받을 대상의 연령, 성별, 체중, 치료할 특정 질환 또는 병리 상태, 질환 또는 병리 상태의 심각도, 투여경로 및 처방자의 판단에 따라 달라질 것이다. 이러한 인자에 기초한 투여량 결정은 당업자의 수준 내에 있으며, 일반적으로 투여량은 0.001mg/㎏/일 내지 대략 200mg/㎏/일의 범위이다. 더 바람직한 투여량은 0.1mg/㎏/일 내지 50mg/㎏/일이다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명은 천련자 추출물, 이의 분획물 또는 건조된 천련의 열매로부터 분리된 화합물을 포함하는 약학 조성물은 쥐, 가축, 인간 등의 포유동물에 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁 내 경막 또는 뇌혈관 내 주사에 의해 투여될 수 있다. 상기 추출물 또는 이로부터 분리된 화합물은 독성 및 부작용이 거의 없으므로 예방 목적으로 장기간 복용 시에도 안심하고 사용할 수 있는 약제이다.
또한, 본 발명은 천련자 추출물, 이의 분획물 또는 분리된 상기 화학식 1의 화합물 군에서 선택되는 1종 이상의 화합물을 함유하는 것을 특징으로 식품학적으로 허용 가능한 식품보조 첨가제를 포함하는 노화 세포의 노화 관련 증상을 억제 및 노화 관련 질환을 예방 또는 개선용 건강기능식품을 제공한다. 상기 추출물 또는 화합물은 본 발명의 건강기능식품에 0.001~100중량 %로 하여 첨가될 수 있다. 본 발명의 건강기능식품은 정제, 캡슐제, 환제 또는 액제 등의 형태를 포함하며, 본 발명의 추출물 또는 화합물을 첨가할 수 있는 식품으로는, 예를 들어, 각종 식품류, 음료, 껌, 차, 비타민 복합제 등이 있다.
본 발명은 천련자 추출물, 이의 분획물 또는 분리된 상기 화학식 1의 화합물 군에서 선택되는 1종 이상의 화합물을 포함하는 화장품 등에도 사용 가능하며, 특히 노화 세포의 노화 관련 증상 억제 및 노화 관련 질환을 예방 또는 개선용 천연 화장품 등을 제공한다.
본 발명은 세놀리틱 저해활성을 갖는 천련자 추출물, 이의 분획물 또는 분리된 리모노이드 계 화합물을 유효성분으로 함유하는 노화 세포의 노화 관련 증상 억제 및 노화 관련 질환을 예방 또는 치료하는 조성물에 관한 것이다. 상기 추출물, 이의 분획물 또는 분리된 화합물은 노화 세포를 선택적으로 제거하며, 노화의 지표로 사용되는 단백질 p16INK4A의 발현을 감소하고 대표적인 세포주기 정지 유전자인 p16INK4A, p53, p21CIP1의 발현 억제함으로써 우수한 항노화 활성을 갖는 점을 밝혀내어 본 발명을 완성하게 되었다. 또한, 천련자 추출물, 이의 분획물 또는 분리된 화합물들이 섬유화된 세포에서 지표 인자인 α-SMA, N-cadherin, Collagen 1A1 및 4A1의 mRNA 발현량을 감소시킴에 따라 특발성 폐 섬유증을 예방 또는 치료하는 조성물에 관한 것이다. 특히나, 천련자는 전통적으로 한국에서 사용되어왔던 한약재 중 하나로, 천련자로부터 추출 및 분리 정제한 것이므로, 세포 독성이 적고 안전성이 높으며 항노화 활성을 지닌 천연물로 의약품, 건강식품, 식품첨가제, 화장품 등에 유용하게 적용될 수 있다.

1. 천련자로부터 활성 화합물의 용매 추출 조건 확인>
실시예
1-1. 천련자 추출물의 제조
본 발명에서 사용된 건조된 천련자는 경동시장 덕현당한약국(서울 동대문구 고산자로 445)에서 확보하였다. 상기 천련자 4.55g을 각각 20mL의 증류수, 30%[v/v] 에탄올 수용액, 50%[v/v] 에탄올 수용액, 70%[v/v] 에탄올 수용액, 100% 에탄올, 30%[v/v] 메탄올 수용액, 50%[v/v] 메탄올 수용액, 70%[v/v] 메탄올 수용액, 100% 메탄올, n-부탄올, 아세톤, 에틸아세테이트 및 n-헥산을 사용하여 1시간 30분 동안 실온에서 초음파 추출기로 1회에 걸쳐 추출하였다. 이후, 고형분을 제거한 추출액을 감압 농축하여 추출물을 제조한 후, 추출량을 확인하여 그 결과를 표 1에 나타내었다.

리포터 분석
인간 p16INK4A 프로모터 (GeneBank Accession No.NM_000077.4)의 리포터 구축물을 생성하기 위해, 인간 p16INK4A 유전자 (-722 to -180)의 프로모터를 포함하는 430-bp DNA 단편을 293T의 게놈 DNA를 주형으로 하여 PCR 증폭하였다. PCR 증폭에는 nPfu-Forte DNA 중합효소 (Enzynomics, Daejeon, Korea), KpnI 부위를 포함하는 5'-CCCGGTACCGTGGAAGAAAAGGGGAGGAG-3' 및 XhoI 부위를 포함하는 5'-CCCCTCGAGCCGGACTAGGTAGGTGGAGTC-3' 프라이머 쌍을 이용하였으며, KpnI 및 XhoI로 절단된 PCR 산물을 KpnI 및 XhoI로 절단된 pGL3-Basic luciferase report (Promega, Madison, WI, USA)에 클로닝하여 pGL3_p16INK4A 프로모터를 생성하였다. 이후, 재조합체로 대장균을 형질 전환하고 DNA 서열 분석으로 확인하였다.
한국 세포주 은행에서 구입한 A549 세포(웰당 9,000개)를 96웰 플레이트에 배양하였다. 24시간 배양 후, 배양 배지를 제거하고 세포를 p16INK4A 프로모터에 의해 구동되는 파이어플라이 루시페라제를 코딩하는 0.07 μg pGL3-p16INK4A 프로모터 플라스미드 0.03 μg, RSV-베타-갈락토시다제 플라스미드 및 Lipofectamine 2000 (0.2 μL, Invitrogen, Carlsbad, CA, USA) 20 μL Opti-MEM에 함유하여 혼합물로 형질 감염시켰다. 추가로 세포가 배지로 완전히 덮이도록 20 μL의 Opti-MEM을 첨가하고, 형질감염 4시간 후에 배지를 10% FBS가 보충된 RPMI 1640 배지로 교체하고 세포를 밤새 인큐베이션하였다.
이후, 세포를 대조군에 DMSO를 처리하고, 양성 대조군인 나비토클락스(Navitoclax, N) 또는 천련자 추출물, 이의 분획물 또는 이로부터 분리한 화합물을 처리하여 24시간 동안 배양하고 루시페라제 및 베타-갈락토시다제 발현을 분석하였다. 이후, 세포 배양 용해 완충액 (Promega, WI, USA)을 이용하여 세포 용해물을 수집한 후, 파이어플라이 루시페라제 에세이 kit (Promega, WI, USA)를 이용하여 루시퍼라제 리포터 유전자 에세이를 수행하였고, 형질 감염된 세포에서 파이어플라이 루시페라제 활성은 베타갈락토시다아제 활성으로 표준화되었다.

활성 확인
사이클린 의존성 키나제 억제제인 p16INK4A는 노화 세포에서 특이적인 마커이며, 이는 CDK4 및 CDK6 매개 불활성화를 방지함으로써 G1을 세포 주기의 S-기 진행으로 제한한다. 최근 연구에서는 p16INK4A 프로모터의 활성화가 생체 내 노화를 제한하는데 사용될 수 있고, p16INK4A의 비활성화를 통해 조기 노화가 역전될 수 있음이 알려졌다. 또한, p16INK4A의 발현은 특발성 폐 섬유화증(idiopathic pulmonary fibrosis, IPF) 환자의 폐 조직과 마우스의 블레오마이신 유도 IPF 모델 모두에서 상향 조절되는 것으로 보고됨에 따라, 본 발명자들은 비 노화 인간 폐 섬유 아세포 (IMR90) 세포를 사용하여 루시퍼라제 리포터 유전자 분석을 수행하여 천련자 유래 추출물 및 분획물의 세포 노화를 억제하는 효능을 평가하고 p16INK4A의 전사 수준 정도를 측정하였다. 그리고 이를 비교한 그래프를 도 1 및 표 2에 나타내었다.

전사 억제 효과의 우수성 확인
천련자로부터 세놀리틱 물질을 발굴하기 위하여 건조된 천련자를 70% 에탄올로 추출하여 얻은 조 추출물을 증류수에 현탁 하여 현탁액을 만든 후, n-헥산과 에틸아세테이트, n-부탄올 및 물로 순차적 용매 분획을 수행하여 4개의 분획물을 얻어 농축하였다. 일차적으로 천련자 추출물 및 분획물의 세포 독성 여부를 확인하기 위하여 MTT 어세이를 수행하여 세포 생존력 평가를 시행하고 이를 도 1A에 나타내었다. 도 1A에서 보는 바와 같이, 천련자 추출물, 이의 n-헥산, 에틸아세테이트, n-부탄올 및 물 분획물은 각각 20ug/mL에서 세포 독성을 나타내지 않았다.
세포 독성이 없는 농도 (20ug/mL)로 추출물을 처리하여 실시예 2-1의 방법을 통해 p16INK4A 전사 억제 활성을 측정하여 도 1B에 나타내었다. 도 1B에서 보는 바와 같이, 천련자 추출물, 이의 n-헥산, 에틸아세테이트, n-부탄올 및 물 분획물은 대조군 (DMSO 처리군)과 비교하였을 때, 천련자 유래 추출물에서 p16INK4A 전사 수준이 대략 24% 억제되었으며, 에틸아세테이트 분획물에서 전사 수준이 35% 상당히 감소함이 확인되었다.
따라서 p16INK4A 전사 수준을 강력하게 감소시키는 에틸아세테이트 분획물의 활성 기반 물질을 분리하고자 에틸아세테이트 분획물을 7개의 소 분획물로 분획하였다. 하위 7개의 소분획물의 세포 독성을 확인하고 (도 1C), p16INK4A 전사 억제 활성을 측정하여 도 1D에 나타내었다. 도 1D에서 보는 바와 같이, DMSO 대조군과 비교하였을 때, 1번 소분획물에서 p16INK4A 전사 정도가 38% 감소됨을 확인함으로써 저해 활성이 소분획물 1에서 가장 우수함을 확인하였다.

상기 실시예 2에 이어서, 활성이 가장 강한 에틸아세테이트 분획물로부터 유래한 1번 소분획물을 제조한 후, 다양한 크로마토그래피를 실시하여 5개의 활성 물질을 분리하였다.
건조된 천련자 (1.8 kg)을 70% 에탄올로 3회, 실온에서 초음파 추출한 후, 감압 농축하여 얻었고 건조된 추출물 (130 g)은 2.0 L의 물에 현탁시킨 후, 동량의 n-헥산 (11.0g), 에틸아세테이트 (19.5 g), n-부탄올 (25.5g)로 분획하여 소 분획을 실시하였다. 획득한 에틸아세테이트 분획물을 건조하여 메탄올:물 (30:70 - 100:0, v/v)의 이동상으로 중압 액체 크로마토그래피 (MPLC, RP-C18 Flash cartridge. 컬럼크기: 10×250mm, 입자크기: 5μm, 유량: 3ml/min, UV detection: 205 및 254nm)를 실시하여 하위 분획물 7개 (M1 - M7)를 획득하였다. 활성이 가장 강한 하위 분획물 1 (M1)은 메탄올:물 (100:0, v/v)의 이동상으로 세파덱스 LH-20 컬럼 크로마토그래피를 수행하여 하위 분획물 4개 (M1L1-M1L4)을 얻었다. 하위 분획물 M1L2은 메탄올:물(20:80 - 100:0, v/v)의 이동상으로 역상 실리카 겔 컬럼 크로마토그래피을 수행하여 하위 분획물 18개 (M1L2R1-M1L2R18)를 얻었다. 하위 분획물 M1L2R10은 메탄올:물(20:80 - 100:0, v/v)의 이동상으로 세파덱스 LH-20 컬럼 크로마토그래피를 수행하여 하위 분획물 7개 (M1L2R10L1-M1L2R10L7)을 얻었다. 상기의 소 분획물 M1L2R10L5 및 M1L2R10L6은 아세토나이트릴:물 (63:37)의 이동상으로 성능 액체크로마토그래피 (HPLC, Optima Pak C18 column, 컬럼크기: 10×250mm, 입자크기: 5μm, 유량: 3mL/min, UV detection: 205nm)를 실시하여 화합물 3 (6.9mg) 및 화합물 4 (2.0 mg)를 얻었다. 상기의 소 분획물 M1L2R16는 메탄올:물 (100:0, v/v)의 이동상으로 세파덱스 LH-20 컬럼 크로마토그래피를 수행하여 얻어진 7개의 소분획물 (M1L2R16L1-M1L2R16L7)에서 M1L2R16L4 분획물을 아세토나이트릴:물 (50:50 - 65:35, v/v)의 이동상으로 성능 액체크로마토그래피 (HPLC, Optima Pak C18 column, 컬럼크기: 10×250mm, 입자크기: 5μm, 유량: 3mL/min, UV detection: 254nm)를 실시하여 화합물 5 (70.0 mg)를 얻었다.
소분획물 M2는 메탄올:물(100:0, v/v)의 이동상으로 세파덱스 LH-20 컬럼 크로마토그래피를 수행하여 하위 분획물 4개 (M2L1-M2L4)을 얻었다. 하위 분획물 M2L2은 n-헥산:에틸아세테이트 (80:20 - 0:100, v/v)의 이동상으로 실리카겔 컬럼 크로마토그래피을 수행하여 하위 분획물 5개 (M2L2N1-M2L2N5)를 얻었다. 하위분획물 M2L2N5는 아세토나이트릴:물(52:48)의 이동상으로 성능 액체크로마토그래피(HPLC, Optima Pak C18 column, 컬럼크기: 10×250mm, 입자크기: 5μm, 유량: 3mL/min, UV detection: 254nm)를 실시하여 화합물 1 (4.2 mg) 및 화합물 2 (4.5 mg)를 얻었다.

본 발명의 천련자 추출물 및 활성 분획물로부터 분리한 화합물 1 내지 5의 세포 독성 여부를 확인하기 위하여 상기 A549 세포에 화합물 1 내지 5를 20μM 처리하고 MTT 어세이를 수행하여 이를 도 2A에 나타내었다. 도 2A에서 보는 바와 같이, 화합물 1 내지 5은 모두 20μM에서 세포 독성을 나타내지 않았다.
또한, 본 발명의 천련자 추출물 및 활성 분획물로부터 분리한 화합물 1 내지 5의 p16INK4A의 전사 억제 효과를 확인하기 위해, 상기 실시예 2-1와 같은 방법으로 p16INK4A의 전사 수준을 확인하였다. 상기 도 2A에서 확인한 독성이 없는 20 μM 농도로 단일화합물들을 처리하고 그 결과를 도 2B에 나타내었다. 나비토클락스 (N)는 p16INK4A 프로모터 활성의 억제 효과를 결정하기 위한 양성 대조군으로 사용되었다. 도 2B에서 보는 바와 같이, 화합물 1 내지 5 20μM는 노화 지표인 p16INK4A에 대한 저해 활성을 가짐을 확인하였고, 그 중에서도 화합물 5는 p16INK4A 유전자의 발현을 43.5 %로 가장 크게 감소시키는 것을 확인하였다.

4-1. 활성 탐색을 위한 세포 준비
인간 폐 섬유 아세포 (IMR90) 세포주는 ATCC에서 구매하였고 10% 소 태아 혈청 (FBS; Hyclone, GE Healthcare Life Sciences, UK), 100U 페니실린, 100 μg/mL 스트렙토마이신 (Hyclone)이 첨가된 최소 필수 배지 이글(MEM; Welgene, 대전, 한국)에서 5% CO2가 있는 가습 인큐베이터에서 세포를 배양하였다. 세포의 밀집도가 90%에 도달했을 때 일반적으로 계대 번호 17까지 분열하였다. 또한, 인간 폐 상피 세포주 (A549)는 한국 세포주 은행에서 입수하여 10% FBS (Hyclone), 100U 페니실린 및 100μg/mL streptomycin( Hyclone) 및 5% CO2의 가습 인큐베이터에서 세포를 배양하였다.

및 분리된 화합물의 세포 독성 평가
처리되는 천련자 유래 추출물, 분획물 및 분리된 화합물의 세포 생존력을 측정하기 위해 MTT 분석을 수행하였다. IMR90 세포 (웰당 5,000개) 또는 A549 세포(웰당 8000개)를 96웰 플레이트에 배양하였다. 24시간의 배양 후, 시료 비 처리군으로 DMSO 또는 추출물, 분획물 및 분리된 물질을 처리하고 세포를 추가 48시간 또는 72시간 동안 배양하였다. 이후, 배지를 버리고 무 혈청 배지 중 500μg/mL MTT(3-[4,5-디메틸티아졸-2-일]-2,5-디페닐 테트라졸륨 브로마이드) 용액 50 μL를 첨가하였고, 4시간 배양 후 MTT 용액을 제거하여 formazan을 DMSO에 녹인 후 570nm에서 흡광도를 측정하였다.
천련자로부터 얻은 70% 에탄올 조 추출물 및 4개의 분획물 (n-헥산, 에틸아세테이트, n-부탄올, 물 분획물)은 모두 인간 폐 상피 세포주 (A549)에서 20 μg/mL 농도로 처리하였을 때 독성이 없음을 확인하였고 7개의 소 분획물 (M1-7)의 경우, 5 μg/mL, 5개의 단일화합물은 20 μg/mL 농도에서 처리하였을 때 세포에 독성이 없음을 확인하였다.

5-1. 복제 노화 모델 구축
IMR90 세포는 계대 번호 1부터 세포 분열하여 계대 수가 17에 도달했을 때, 세포 분열이 4주 내에 컨플루언시 (confluency)에 도달할 수 없음을 확인하였다. 노화 세포는 외관적으로 일반 세포보다 비대하고 평평하며 불규칙한 형태가 되는 것을 현미경 상에서 확인하였다. 이후 노화 세포 마커로, 노화 관련 β-갈락토시다아제 (SA-β-gal) 염색 및 세포주기 정지 마커 (p16, p21 및 p53)의 상향 조절된 발현 확인함으로써 복제 노화 모델을 구축하여, 계대 번호 8-10을 비 노화 IMR90 세포로, 계대 번호 16-17을 노화 IMR90 세포로 확립하고 이후 실험에 사용하였다.

본 발명의 천련자 추출물로부터 분리된 단일화합물 중 p16INK4A의 억제 활성이 뛰어난 투센다닌 (화합물 5)이 선택적으로 노화 세포를 저해 여부를 확인하기 위해 상기 실시 예 4의 방법을 통해 비 노화 및 노화 IMR90 세포에서 생존력을 평가하였다. 투센다닌은 비 노화 및 노화 IMR90 세포에서 10 내지 500nM의 다양한 농도로 처리하여 평가되었고, 세포 생존율은 DMSO 대조군 웰과 비교하여 시료를 처리한 웰에서 흡광도로 나타내고 그 결과를 도 2C, 2D 및 표 5에 각각 나타내었다.
표 5에서 보는 바와 같이, 비 노화 IMR90 세포의 경우에는 50 nM의 농도에서 세포 독성이 나타나지 않으나 노화 IMR90세포에서는 투센다닌이 50nM의 농도부터 세포 생존률이 70%까지 감소하며 투센다닌의 농도가 증가할수록 비노화 및 노화 세포의 생존률의 차이가 증가함을 확인할 수 있다. 이에 따라, 투센다닌이 선택적으로 노화 세포만을 저해함을 증명함으로써 투센다닌의 세놀리틱 활성을 확인하였다.

-중합 효소 연쇄 반응 측정 실험법
IMR90 세포를 6-웰 또는 12-웰 플레이트에 밀도 80%으로 배양하였다. 처리 후 TRIzol 시약(Invitrogen, USA) 250 μL를 각 웰에 첨가하여 전체 RNA를 추출하였다. 1 μg의 total RNA와 M-MLV Reverse Transcriptase kit(Bioneer, Korea)를 사용하여 추가 cDNA 합성에 사용하였다. 이는 20 μL의 반응을 수행한 후 100 μL로 희석하여 다음 실시간 qPCR (real-time qPCR)을 수행하는데 템플릿으로 사용하였다. Real-time qPCR은 AccuPower® 2X GreenStar™ qPCR Master Mix(Bioneer)를 이용하여 수행되었다. 모든 실험은 세 번 반복 실험되었고, 모든 마커의 발현 수준은 18s 리보솜 RNA의 발현 수준으로 표준화되었다. 표 6에는 사용된 프라이머 서열을 나타내었다.

발현 억제 효능
본 발명의 천련자로부터 분리된 화합물 중 항노화 활성이 가장 높은 투센다닌 (화합물 5)의 노화 관련 세포주기 정지 유전자의 mRNA 발현 억제 효과를 확인하고자 비 노화 및 노화 IMR90 세포주에서 qRT-PCR 분석을 시행하였으며, 그 결과를 도 3에 나타내었다. 대표적인 세포주기 정지 유전자 p16INK4A, p21CIP1, p53은 노화세포에서 발현이 매우 증가하나, 투센다닌을 처리할 경우 대조군인 DMSO가 처리된 군에 비해 노화 세포에서 억제 효과가 매우 크게 나타났으며(도 3A) 특히나 감소 효과는 비 노화 세포에 비하여 특이적으로 노화 세포에서 매우 뛰어남을 확인하였다. 투센다닌이 처리된 비 노화 및 노화 IMR90 세포주에서 노화 관련 세포주기 정지 유전자의 mRNA 발현을 확인한 결과는 하기의 표 7에서 나타내었다.

세포 배양 상층액의 젖산 탈수소효소(LDH)는 제조업체의 지침에 따라 LDH 세포 독성 검출 키트(Takara Bio, USA)를 사용하여 정량화하여 클로로퀸과 투센다닌 (화합물 5) 그룹간의 독성을 평가하였다 (도 4A). 도 4A에서 보는 바와 같이, 클로로퀸과 투센다닌 각각 또는 혼합 처리한 경우 모두에서 세포 독성을 나타내지 않았다.

실험
IMR90 세포를 6웰 또는 12웰 플레이트에 세포 밀도 80% 로 배양하였다. 물질 처리 후 프로테아제 억제제 (Complete Mini, EDTA-free, Roche(Basel, Switzerland)) 및 포스파타제 억제제 (Phosphatase Inhibitor Cocktail 2 &3, Sigma-Aldrich; PMSF, Roche) 혼합물과 함께 RIPA 완충액을 사용하여 단백질을 추출하였다. 전기영동을 위해 샘플당 20 μg의 단백질을 5-15% 겔에 로딩한 다음 폴리비닐리덴 디플루오라이드(PVDF) 멤브레인 [0.45μm, Millipore(Darmstadt, 독일)] 옮겨 실온에서 1시간 동안 5% 탈지유에서 차단한 다음 4℃에서 밤새 1차 항체와 함께 인큐베이션하였다. 2차 항체와 ECL 키트를 사용하여 화학발광 신호를 확보하고, NIH (National Institutes of Health) ImageJ 소프트웨어를 사용하여 농도계 분석을 수행하였다. 1차 항체는 LC3B (Cell Signaling Technology, Beverly, MA) 및 β-액틴 (SantaCruz Biotechnology)을 사용하였다.

유동 분석법을 통한
투센다닌의
자가포식
활성화 작용 확인
노화 세포에서 단백질 분해 시스템 중의 하나인 자가포식 시스템의 유동상태 (flux)를 평가하였다. 일반적으로 LC3-I이 포스파티딜에탄올아민 (phosphatidylethanolamine, PE)과 결합하면서 LC3-II를 형성한다. 이러한 LC3-II는 막으로 들어가 자가포식소체의 형성 및 성숙 촉진하는데 이에 따라 오토파고좀 마커 단백질로 LC3-II가 알려져있다. 하지만, 웨스턴 블럿에서 LC3-II의 증가가 보이면 자가포식체 (autophagosomes)가 더 형성되기 때문에 세포에서 autophagy가 증가했음을 의미할 수 있으나, 또한 자가포식체의 분해를 억제함으로써 증가할 수 있음에 따라 세포가 autophagy를 감소시켰음을 의미할 수도 있다. 이에 따라서 자가포식을 정량화하는 가장 좋은 방법의 하나로 자가포식 유동 상태를 측정하는 것이며, 이는 자가포식체의 형성과 분해를 모니터링을 할 수 있는 실험법 중 하나이다. 이때 플럭스를 측정하려면 하기 식와 같이 리소좀 억제제가 있는 경우와 없는 경우 LC3-II 수준의 차이를 확인해야 한다.

클로로퀸 (Clochicine, CQ)은 리소좀 억제제 (lysosomal inhibitor)이자 자가포식을 억제하는 약물로 LC3-II 분해되는 것을 막아 주는 물질이며, LC3-II를 축적한다. 이에 따라서 클로로퀸을 활용한 Turnover assay는 자가포식과 관련된 단백질 수준을 변화를 명확히 나타낼 수 있다.
도 4B 및 4C에서 보는 바와 같이, 비 노화 IMR90 세포주에서 투센다닌 처리 여부의 차이는 크게 변화가 나타나지 않았으나, 노화 세포에서 투센다닌을 처리하였을 때 전환율이 매우 증가하였다. 따라서 투센다닌은 노화 세포에서 자가포식을 유도하는 물질임을 확인할 수 있었다 (도 4B 및 4C).

본 발명의 천련자로부터 분리된 투센다닌 (화합물 5)의 섬유화 관련 인자의 mRNA 발현 억제 효과를 확인하고자 상기 qRT-PCR 분석을 시행하였으며, 그 결과를 도 5 및 표 8에 나타내었다.
변형 성장 인자인 TGF-β (Transforming growth factor-β)는 세포외 기질의 단백질을 생산하는 유전자를 상향조절하여 세포외 기질의 축적을 매개하는 것으로 세포의 섬유화를 유도할 수 있다. 이에 따라 TGF-β를 처리하여 섬유화가 유도된 세포에서 대표적인 세포의 섬유화 바이오 마커인 α-SMA (α-smooth muscle actin), N-cardherin, 콜라겐1A1 (collagen 1A1), 콜라겐4A1 (collagen 4A1)의 mRNA 발현 억제 활성을 투센다닌 (화합물 5) 및 대조군으로 DMSO가 처리된 군과 비교하였다. 도 5 및 표 8에서 보는 바와 같이, 대조군(DMSO)보다, 투센다닌은 섬유화 관련 유전자의 mRNA 발현이 상당한 억제 효과를 지니고 있음이 확인하였다.

동물모델 제작
폐섬유화 모델을 만들기 위하여 8주령 마우스 (C57BL6/J 수컷20g, 대조군과 비교군)를 이용하여 블레오마이신 (Bleomycin: 2.0 mg/kg)을 식염수 (0.%)에 녹인 후 30㎕의 양으로 기도 내에 투여하였다. 블레오마이신 (Bleomycin)을 투여하여 섬유화를 유도한 다음날부터 격일에 한 번씩 복강 내 주사 (Intraperitoneal injection , IP)를 통해 투센다닌(TSN)을 주입하였다. TSN 용매의 경우 부작용이 적고 수율이 좋은 DMAc (식염수 (0.9%) N,N-Dimethylacetamide, Tween80을 1:1:8 비율)를 사용하였다. 표 9는 분류 군에 따라 실험에 사용된 동물의 마릿수를 나타낸 실험계획표이다. 단, Control: 정상군; TSN (0.1 mg/kg): TSN (0.1 mg/kg)단독처리군; BLM: 블레오마이신 단독처리군; BLM+TSN (0.1 mg/kg): 블레오마이신과 TSN (0.1 mg/kg)의 병용처리군.
TSN의 첫 번째 투여 이후 이틀에 한 번씩 반복 투여를 진행한 후 14일 차에 마우스를 희생시키고, 폐 조직을 적출하여 추가 실험에 이용하였다. 도 6은 실험 일정을 도식화한 모식도이다.

동물모델의 조직학적 검사결과
마우스를 개체별로 희생하여 적출한 폐 조직을 가지고 10% 포르말린 용액 (10% formalin)으로 고정하였다. 조직을 파라핀으로 처리한 뒤 일정한 두께 (4㎛)로 조직 절편을 유리 슬라이드로 제작하였다. 이후 폐섬유화를 확인하는 염색기법인 Masson's trichrome stain을 하여 콜라겐의 침착 (Collagen deposition) 정도를 현미경으로 촬영하여 도 7A에 나타내었다.
Masson's trichrome stain 실험 기법은 조직 구조 안에서 염료 분자의 pH 그리고 염료 투과성의 차이를 이용한 방법이다. 슬라이드 상에 준비된 조직 절편 주변의 파라핀을 제거하기 위해 자일렌 (Xylene)에 담그고 순차적인 탈수과정 (알코올의 농도를 순차적으로 높이는 과정 100, 90, 70, 50%)에 각각 5분씩 순서대로 담갔다. 이후 조직의 활성화를 위하여 0.1M 농도의 시트르산 용액에 조직을 담가서 10분을 끓이고 10분을 식혔다. 이후 세포질의 염색을 높여주기 위해 56℃에서 Bouin’s Solution (원액) 안에 슬라이드를 20분 동안 담그고 20분을 식힌 이후 흐르는 물에 5분간 세척 하였다. 다음으로 핵을 염색하기 위해 Hematoxylin solution 10%에 5분간 염색을 진행한 후 세포질과 근섬유를 염색하는 시약인, Biebric Scarlet-Acid Fuschin을 1분 동안 염색을 하고 흐르는 물에 5분 동안 세척 하였다. 다음으로 Collagen을 탈색시키기 위한 과정인 Phosphotungstic/Phosphomolybdic acid (원액) 그리고 DW를 1:1:2 비율로 섞어서 5분을 방치한 후 콜라겐의 침착 (Collagen deposition)을 확인하는 aniline blue 2.4%시약에 5분 동안 담근다. 이후 1% 아세트산 (Acetic acid)에 1분 동안 반응시킨 뒤에 순차적으로 탈수과정(알코올의 농도를 순차적으로 높이는 과정. 50, 70, 90, 100%)을 진행한 이후 자일렌 (Xylene)에 담근 이후 커버글라스로 봉입하는 과정 (Mounting)을 통하여 최종적으로 고정하였다. 하루 정도 말린 이후에 현미경으로 관찰한 슬라이드의 이미지를 Graphpad Prism의 통계프로그램을 이용하여 콜라겐의 침착 (Collagen deposition)정도를 도식화하여 도 7B에 나타내었다. 도 7A 및 도 7B에서 보는 바와 같이, 블레오마이신에 의하여 유도된 섬유화 마우스에서 TSN (0.1 mg/kg) 처리는 콜라겐의 침착을 50% 까지 감소하였는 것을 확인하였다. 11. 독성실험>
실시예
11-1. 급성독성
본 발명의 70% 에탄올로 추출한 천련자 추출물을 단기간에 과량을 섭취하였을 때의 급성적 (24시간 이내)으로 동물 체내에 미치는 독성을 조사하고, 치사율을 결정하기 위하여 본 실험을 수행하였다. 일반적인 마우스인 ICR 마우스 계통 20마리를 준비하였고, 각 군별로 10마리씩 배정하였다. 대조군에는 30% PEG-400만을 투여하고, 실험군은 상기 천련자 추출물을 1.0 g/㎏의 농도로 각각 경구 투여하였다. 투여 24시간 후에 각각의 치사율을 조사한 결과, 대조군과 1.0g/㎏ 농도의 상기 천련자 추출물을 투여한 실험군에서는 모두 생존하였다.

실험군
및 대조군의 장기 및 조직 독성 실험
장기 독성 실험은 C57BL/6J 생쥐를 대상으로 동물의 각 장기(조직)에 미치는 영향을 조사하기 위하여 본 발명의 70% 에탄올로 추출한 천련자 추출물을 1.0g/㎏의 농도로 투여한 실험군과 용매만을 투여한 대조군의 동물들로부터 8주 후 혈액을 채취하여 GPT(glutamate-pyruvate transferase) 및 BUN(blood urea nitrogen)의 혈액 내 농도를 Select E(Vital Scientific NV, Netherland) 기기를 이용하여 측정하였다. 그 결과, 간독성과 관계있는 것으로 알려진 GPT와 신장 독성과 관계있는 것으로 알려진 BUN의 경우, 대조군과 비교하여 실험군은 별다른 차이를 보이지 않았다. 또한, 각 동물로부터 간과 신장을 절취하여 통상적인 조직 절편 제작과정을 거쳐 광학 현미경으로 조직학적 관찰을 시행하였으며 특이한 이상이 관찰되지 않았다.

본 발명의 70% 에탄올로 추출한 천련자 추출물 20g을 락토오스 175.9g, 감자전분 180g 및 콜로이드성 규산 32g과 혼합하였다. 이 혼합물에 10% 젤라틴 용액을 첨가시킨 후, 분쇄해서 14 메쉬체를 통과시켰다. 이것을 건조시키고 여기에 감자전분 160g, 활석 50g 및 스테아린산 마그네슘 5g을 첨가해서 얻은 혼합물을 정제로 만들었다.
제제예
1-2. 주사제의 제조
본 발명의 70% 에탄올로 추출한 천련자 추출물을 1g, 염화나트륨 0.6g 및 아스코르브산 0.1g을 증류수에 용해시켜서 100ml를 만들었다. 이 용액을 병에 넣고 20℃에서 30분간 가열하여 멸균시켰다.

- 천련자 (Melia toosendan) 추출물 또는 이의 분획물 또는 이로부터 분리한 리모노이드 화합물을 유효성분으로 포함하는 세포 노화 관련 질환 예방 또는 치료 조성물.
- 제1항에 있어서,
상기 천련자 (Melia toosendan) 추출물은 천련자의 50~100% C1 내지 C4의 저급 알코올 수용액 추출물인 것을 특징으로 하는 세포 노화 관련 질환 예방 또는 치료 조성물. - 제1항에 있어서,
상기 분획물은 천련자를 30~100% C1 내지 C4의 저급 알코올 수용액으로 추출한 농축물을 증류수로 현탁한 후, n-헥산, 에틸아세테이트, n-부탄올로 순차적으로 분획하여 얻은 n-헥산, 에틸아세테이트, 또는 n-부탄올 중에서 1종 이상 선택된 유기용매 분획물인 것을 특징으로 하는 세포 노화 관련 질환 예방 또는 치료 조성물. - 제1항에 있어서,
상기 분획물은 천련자를 30~100% C1 내지 C4의 저급 알코올 수용액으로 추출한 농축물을 증류수로 현탁한 후, n-헥산, 에틸아세테이트, n-부탄올로 순차적으로 분획하여 얻은 에틸아세테이트 분획물인 것을 특징으로 하는 세포 노화 관련 질환 예방 또는 치료 조성물. - 제1항에 있어서,
