본 발명은 암세포로의 특이적 단백질 분자 전달용 조성물 및 이를 이용한 암세포의 검출 또는 치료용 제제에 관한 것이다. 본 발명은 세포 투과 펩타이드(Cell penetraing peptide)의 일종인 암세포 호밍 펩타이드(Tumor Homing peptide)와 융합하여 전달하고자 하는 단백질 분자와 복합체를 형성함으로써 세포막에 대한 투과성이 현저하게 개선되어, 효율적인 암세포 특이적 전달 시스템으로 유용하게 이용될 수 있다. 본 발명의 제제는 특히 단백질의 세포 내 투과 효율을 증진시키며, 단백질 전달을 이용한 목적 암세포에서의 우수한 연구 및 검출, 치료 효율로 달성한다.
Description
본 발명은 암세포에 효율적으로 단백질을 전달할 수 있는 기술에 관한 것이며, 이는 단백질의 효율적 암세포 전달을 위한 제제를 만들어서 암세포 기작 연구 및 진단이나 치료법 개발 등에 활용될 수 있다.
암은 남성 다섯 명 중 한 명, 여성 여섯 명 중의 한 명에게 발병하여, 전 세계적으로 매년 약 천 팔백만 명의 환자가 발생한다. 또한 전세계적으로 매년 약 구백 육십만 명의 사망자가 발생하며, 이는 남성 여덟 명 중 한명, 여성 열 한명 중 한 명의 사망자가 발생한다. 이러한 암을 진단하고 치료하는 것은 인류에게 주어진 가장 큰 사명 중의 하나이며, 이를 위해 수많은 연구들이 진행되고 있다.
암을 치료하기 위한 단백질 의약품은 바이오 의약품 분야에서 큰 시장 규모를 이루고 있으며, 암세포의 생장과 증식을 억제하는 단백질을 재조합으로 대량 생산하여 암 세포를 사멸하여 암을 치료하는데 사용할 수 있다.
다양한 종류의 단백질 의약품들은 대부분 세포의 외부에 결합하여 작용하는 경우가 많다. 이는 세포 내부에서 작용하는 단백질들을 세포 내부로 효과적으로 삽입할 수 있는 기술이 부족하기 때문이다. 이로 인하여, 암을 치료하는 효과가 상대적으로 부족하다는 문제가 있다.
또한 세포 연구나 진단 등의 목적을 위해서 리포터 단백질 등을 세포 내부로 전달해야 하는 경우도 다수 존재하기에 고효율 세포 전달기술이 필요하다. 또한 연구용, 진단용, 치료용 펩타이드의 경우에도 역시 세포 내부 전달이 필요한 경우가 존재할 수 있다.
본 발명에서는 세포 내부에서 작용하는 단백질이나 펩타이드를 특정 세포에 대해서 효율적으로 전달할 수 있는 기술을 개발하고자 하였다.
따라서 본 발명의 목적은 암세포 단백질 고효율 전달기술을 제공하는 데 있다.
본 발명의 다른 목적은 암세포 단백질 고효율 전달을 통한 암세포 연구용 조성물을 제공하는 데 있다.
본 발명의 다른 목적은 암세포 단백질 고효율 전달을 통한 암세포 진단용 조성물을 제공하는 데 있다.

본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명, 청구범위 및 도면에 의해 보다 명확하게 된다.
상기 과제를 해결하기 위하여, 본 발명자들은 높은 효율로 형광 단백질 분자를 목적 암세포 내로 전달할 수 있는 암세포 단백질 고효율 전달 기술(A Efficient Delivery Technique of Proteins Targeting to Cancers)을 개발하기 위하여 예의 연구 노력하였다. 그 결과, 전달하고자 하는 형광 단백질에 세포투과 펩타이드 (cell penetrating peptide)의 일종인 암세포 호밍 펩타이드(Tumor homing peptide)를 융합(fusion)하면 암세포막에 대한 투과성이 크게 개선되어 목적 세포로의 단백질 전달 효율이 현저하게 상승함을 발견하였다.
본 발명에 따르면, 하기 실시예에서 보는 바와 같이 형광 단백질 분자 단독으로 사용한 경우에 비해, 각각의 펩타이드를 융합(fusion)한 경우 단백질 분자의 전달의 효율에 있어 유의한 상승적 효과(synergistic effect)를 나타낸다.
각각의 세포 투과 펩타이드는 암세포로의 투과성을 개선하기 위하여 약 5 ~ 15개의 아미노산으로 이루어진 합성 폴리펩타이드로서, 작은 화학 화합물 또는 거대의 핵산 분자를 전달하는 데에 많이 사용되어져 왔으며, 그에 대한 검증도 이미 진행이 되어있다. 이러한 세포 투과 펩타이드(Cell Penetrating Peptide, CPP)의 한 종류인 암 투과 펩타이드(Tumor Homing Peptide)를 단백질 분자에 직접 융합(fusion)함으로써 암세포로의 투과성을 향상시킨다.
본 발명의 일 양태에 따르면, 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide) 및 암세포 내부에서 작용하는 단백질 또는 펩타이드가 융합된 암세포 투과성 단백질 전달용 조성물을 제공한다.
구체적으로, 본 발명의 일 실시형태에 따르면, 상기 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide)는 서열목록 제1 서열: RLYMRYYSPTTRRYG을 갖는 암 세포 투과 펩타이드(Cell Penetrating Peptide, CPP30)인 것을 특징으로 한다.

본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 CPP30 펩타이드는 15개의 아미노산(RLYMRYYSPTTRRYG)으로 구성되어 있으며, 유방암세포를 타겟으로 하는 암 침투 펩타이드(Tumor lineage-homing cell-penetrating peptide)의 일종이다. 이는 mRNA display라는 방법으로 유방암 세포에 투과력이 높은 단백질로 선별되었다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 CREKA 펩타이드는 5개의 아미노산(CREKA)으로 구성되어 있으며, 암세포를 타겟으로 하는 암세포귀환 펩타이드(Tumor Homing Peptide)의 일종이다. 이는 유방암 질환 동물 모델에서 phage display를 통해 선별되었다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 CGKRK 펩타이드는 5개의 아미노산(CGKRK)으로 구성되어 있으며, 유방암을 비롯한 암세포에서 과다 발현되는 단백질 중에 하나인 p32 단백질을 표적으로 하는 암 투과 펩타이드(p32-binding tumor blood-vessels peptide)의 일종이다. 이는 phage display를 통해 선별되었다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 LinTT1R 펩타이드는 9개의 아미노산(AKRGARSTA)으로 구성되어 있으며, 이는 기존의 LinTT1 peptide를 연구에 맞게 서열을 반대로 적용하여 사용하였다. 이 펩타이드는 유방암을 비롯한 암세포에서 과다 발현되는 단백질 중에 하나인 p32 단백질을 표적으로 하는 암 투과 펩타이드(p32-binding tumor penetrating peptide )의 일종이다. 이는 peptide display를 통해 선별되었다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 CTTHWGFTLC 펩타이드는 10개의 아미노산(CTTHWGFTLC)으로 구성되어 있으며, 암세포에서 과다 발현되는 단백질 중에 하나인 Matrix metalloploteinase - 2 / 9 (MMP - 2/MMP - 9) 단백질을 표적으로 하는 암 표적 펩타이드(Tumor blood vessels homing peptide )의 일종이다. 이는 peptide library를 통해 선별되었다.
본 발명의 구체적인 구현예에 따르면, 본 발명에서 사용되는 PL3R 펩타이드는 8개의 아미노산(RVLERGRGA)으로 구성되어 있으며, 이는 기존의 PL3 peptide를 연구에 맞게 서열을 반대로 적용하여 사용하였다. 이 펩타이드는 암세포에서 과다 발현되는 단백질 중에 하나인 Tenascin C fibronectin-type Ⅲ C(TNC-C) 단백질을 표적으로 하는 암 표적 펩타이드(Tumor blood vessels homing peptide )의 일종이다. 이는 peptide library를 통해 선별되었다.
본 발명의 각각의 펩타이드는 전달하고자 하는 단백질 분자와 융합(fusion)하기 위하여 GGGS 라는 아미노산 서열의 링커를 사용하였다. 펩타이드의 길이에 따라서 링커의 길이를 각각 다르게 설정하여 진행하였다.
본 발명의 다른 실시형태는 상기 암세포 투과성 단백질 전달용 조성물을 포함하는 암세포의 연구, 검출 및 치료 중 어느 하나 이상을 수행하는 조성물을 제공한다.
본 발명의 또 다른 실시형태는 상기 암세포 투과성 단백질 전달용 조성물을 세포와 접촉시키는 단계를 포함하는 암세포 투과성 단백질 전달용 조성물을 세포에 전달하는 방법을 제공한다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 암세포로의 단백질 고효율 전달용 조성물 및 이를 이용한 암세포의 연구, 검출 또는 치료용 조성물을 제공한다.

(d) 단순한 구조 변화를 통해 단백질을 세포내로 쉽게 전달할 수 있는 기술은 암세포 외에 다른 질병에서도 단백질 의약품을 고효율로 전달할 수 있을 것으로 보인다.
도 1은 나노복합체의 암세포 투과를 확인하기 위해 유세포 분석기를 이용한 그림이다. 전체 세포의 평균 형광 강도를 강화된 녹색 형광 단백질을 전달한 실험군을 100%로 기준하여 나타낸 그래프이다.

본 명세서에서, 용어“단백질 분자”는 탄소, 수소, 질소, 산소 또는 황으로 구성된 아미노산이 펩타이드(Peptide) 결합에 의해 연결된 약 250개의 아미노산 사이의 짧은 사슬로 이루어진 폴리펩타이드(polypeptide)가 최소 1개 이상으로 구성되어있는 분자이다. 본 발명의 단백질 분자 전달용 조성물은 연구 목적으로 세포에 전달하기 위해 단백질을 전달하는 리서치 툴(research tool)일 수 있고, 질환을 진단하기 위해 진단 단백질을 시료에 전달하는 진단용 조성물(diagnostic composition)일 수 있고, 질환을 치료하기 위해 치료 단백질을 환자에 전달하는 치료용 조성물(therapeutic composition)일 수 있다.
본 발명의 구체적인 구현예에 따르면, 상기 펩타이드는 목적 암세포로의 단백질 고효율 전달용 분자이다. 본 명세서에서 용어“고효율 전달”은 목적되는 세포로 단백질이 고효율로 전달되어, 세포 내에서 활성을 나타냄으로 진단 및 치료에 이용될 수 있는 것을 의미한다.
본 발명의 고효율 전달용 단백질 분자는 형광단백질, 재조합 단백질을 포함하나, 이에 제한되지 않고, 모든 단백질이 이용될 수 있다.
구체적으로는, 본 발명에서 사용되는 고효율로 전달되어질 단백질 분자는 형광 단백질이다.
본 명세서에서 용어 “형광 단백질(fluorescence protein)”은 형광을 나타내는 단백질을 뜻하며, 실험에서 사용되어진 강화된 녹색 형광 단백질(EGFP)은 해파리로부터 기인한 형광 단백질 중의 하나인 야생형 녹색 형광 단백질(GFP)보다 더 높은 강도의 형광을 방출하는 야생형 녹색 형광 단백질(GFP)의 변이체이다.
본 명세서에서 이러한 용어 “재조합 단백질”은 동물, 식물, 미생물 등에서 추출한 유전자를 조작하여 인위적으로 재조합된 유전자를 통해 발현되어진 단백질을 말한다. 이러한 재조합 단백질은 치료를 위해 대장균이나 동물 배양세포에서 배양되어 정제 후, 치료 등에 사용되어지기도 한다.
본 명세서에서 용어“발현시키다”는 단백질을 대장균을 통해 발현되어지는 것을 의미한다.
본 명세서에서, 용어“단백질 전달 시스템(protein delivery system)”는 단백질을 세포 내로 운반하는 모든 수단을 의미하며, 단백질 전달은 단백질의 세포내 침투(transduction)와 동일한 의미를 가진다. 조직 수준에서, 상기 용어 단백질 전달은 단백질의 확산(spread)과 동일한 의미를 가진다. 따라서, 본 발명의 단백질 전달 시스템은 단백질 침투 시스템 및 단백질 확산 시스템으로 기재될 수 있다.
본 발명의 조성물로 전달하고자 하는 단백질 분자 및 세포 투과 펩타이드(Cell penetrating peptide), 암 투과 펩타이드(Tumor Homing peptide)에 대해서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
본 명세서에서 용어“검출”은 질환 또는 질병을 보유하고 있을 때, 가지는 바이오 마커 등을 검출하여 질병의 진단에 사용되는 것을 의미한다.

본 명세서에서 용어“투여”또는“투여하다”는 본 발명의 조성물의 치료적 유효량을 대상체에 직접적으로 투여함으로써 대상체의 체내에서 동일한 양이 형성되도록 하는 것을 말한다.
본 명세서에서 용어“대상체”는 제한없이 인간, 마우스, 래트, 기니아 피그, 개, 고양이, 말, 소, 돼지, 원숭이, 침팬지, 비비 또는 붉은털 원숭이를 포함한다. 구체적으로는, 본 발명의 대상체는 인간이다.
본 발명의 암세포 단백질 고효율 전달용 조성물 및 이를 이용하여 전달하고자 하는 단백질 분자에 대해서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시 예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실험방법
펩타이드 융합(fusion) 단백질 형성
각각의 peptide(CPP30, CREKA, CGKRK, Lintt1R, CTTHWGFTLC, PL3R)가 표지된 프라이머를 제작하여 강화된 녹색 형광 단백질(EGFP)이 포함된 플라스미드에 중합효소연쇄반응을 통해 증폭시킨 후, pGEM-T Easy vector에 삽입 후, Escherichia coli Top10 세포에서 배양하여 플라스미드를 증폭시켰다. 이후 추출한 플라스미드에 정확한 서열이 삽입되었는지 시퀀싱(sequencing)을 통해 확인 후, 최종적으로 pET-23a 플라스미드에 삽입하여 Escherichia coli BL21(DE3)세포에서 IPTG 유도과정을 통해 단백질을 생산하였다. 생산된 단백질은 히스-태그(his-tag) 정제과정을 통해 정제된 후, 1차 증류수에서 투석한 후, 1X phosphate buffered saline에서 냉장보관하며 실험에서 사용되었다.
본 발명의 일 실시형태에 따르면, 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide) 및 암세포 내부에서 작용하는 단백질 또는 펩타이드가 융합된 암세포 투과성 단백질 전달용 조성물을 제공한다.

세포 배양
인간 삼중음성유방암 세포인 MDA-MB-231 세포를 T75 cell culture flasks (SPL Life Science, Korea)에 10% FBS와 1% 페니실린-스트렙토마이신(Life Technologies, CA, USA)이 포함된 DMEM(dulbecco’s modified eagle’s medium, Corning, MA, USA) 8 mL에서 37℃, 5% CO2 조건으로 CO2 incubator (CCL-170B-8-FD; ESCO, Singapore)에서 배양하였다. 계대 배양은 세포의 컨플루언시 80~90%로 3-4일 간격으로 수행하였다.
유세포 분석
MDA-MB-231 세포를 6-웰 플레이트에 5.0 x 105 세포/웰로 총 2 mL씩 분주한 후, 37℃, 5% CO2에서 24시간 동안 배양하였다. 배지를 Opti-MEM(Gibco, USA)으로 교체한 후, 100 μg/ml의 펩타이드가 표지되지 않은 강화된 녹색 형광 단백질(eGFP)와 각 펩타이드를 재조합하여 표지된 강화된 녹색 형광 단백질을 처리하고 24시간 동안 37℃, 5% CO2에서 배양하였다. Dulbecco’s phosphate buffered saline (DPBS)(Sigma-aldrich, Inc)로 세척한 후 각 웰에 트립신-EDTA(Thermo Fisher Scientific, MA, USA)를 첨가하여 세포 부착성을 떨어뜨리고, 배지 2 mL를 첨가하여 300 x g에서 5분 동안 원심분리하였다. 상층액을 제거하고 DPBS로 재부유한 후 동일한 조건에서 원심분리를 2번 반복하였다. 최종적으로 pellet을 차가운 DPBS로 재부유하여 유세포 분석기 전용 튜브에 옮겨 유세포분석기 (CytoFLEX, Beckman, USA)를 이용하여 측정하였다.

펩타이드 부착 단백질 복합체의 세포 내 투과 효율 평가
펩타이드를 통한 형광 단백질의 암세포 전달 효율을 평가하기 위해 형광 단백질로 강화된 녹색 형광 단백질(EGFP)를 사용하였으며, 정량적인 세포 투과율을 확인하기 위하여 유세포 분석기를 사용하여 세포 내에 투과되어진 강화된 녹색 형광 단백질(EGFP)의 형광세기를 비교하였다.
아무것도 전달되지 않은 세포(0.47%)에 비하여 펩타이드가 부착된 단백질의 경우 형광 세기가 증가한 세포들이 측정되어 뚜렷한 이동(shift)가 각각 19.69%(LinTT1R), 24.99%(CREKA), 35.45%(CGKRK), 38.33%(CPP30), 39.01%(PL3R), 41.25%(CTTHWGFTLC)로 분석되었다(도 2).
또한 전체 세포의 평균 형광세기를 상대적인 값으로 나타낸 결과, 펩타이드가 부착되지 않은 강화된 녹색 형광 단백질(EGPF)을 전달한 것을 기준으로 비교할 때 각각 1.35 배 (CREKA), 1.67 배 (CPP30), 2.05 배 (LinTT1R), 2.16 배 (PL3R), 2.17배 (CGKRK), 5.81 배 (CTTHWGFTLC) 이상 전달되어진 것으로 나타났다 (도 1).
두 결과를 종합하였을 때, 기존의 강화된 녹색 형광 단백질만을 전달한 것에 비하여 본 발명의 펩타이드를 단백질에 직접 융합한 전달체가 단백질의 세포내 전달 효율을 증가시킬 수 있음을 확인하였다.
이상, 본 발명의 일 실시예에 대하여 설명하였으나, 해당 기술 분야에서 통상의 지식을 가진 자라면 특허청구범위에 기재된 본 발명의 사상으로부터 벗어나지 않는 범위 내에서, 구성 요소의 부가, 변경, 삭제 또는 추가 등에 의해 본 발명을 다양하게 수정 및 변경시킬 수 있을 것이며, 이 또한 본 발명의 권리범위 내에 포함된다고 할 것이다.

Claims (12)
- 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide) 및 암세포 내부에서 작용하는 단백질 또는 펩타이드가 융합된 암세포 투과성 단백질 전달용 조성물.
- 제1항에 있어서,
상기 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide)는 서열목록 제1 서열: RLYMRYYSPTTRRYG을 갖는 암 세포 투과 펩타이드(Cell Penetrating Peptide, CPP30)인 것을 특징으로 하는 암세포 투과성 단백질 전달용 조성물.
- 제1항에 있어서,
상기 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide)는 서열목록 제2 서열: CREKA을 갖는 암 세포 투과 펩타이드(Cell Penetrating Peptide, CPP)인 것을 특징으로 하는 암세포 투과성 단백질 전달용 조성물.
- 제1항에 있어서,
상기 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide)는 서열목록 제3 서열: CGKRK을 갖는 암 세포 투과 펩타이드(Cell Penetrating Peptide, CPP)인 것을 특징으로 하는 암세포 투과성 단백질 전달용 조성물.

상기 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide)는 서열목록 제4 서열: AKRGARSTA을 갖는 LinTT1R 펩타이드인 것을 특징으로 하는 암세포 투과성 단백질 전달용 조성물.

상기 암 세포 내부에서 작용하는 단백질은 형광 단백질 및 재조합 단백질 중의 어느 하나 이상인 것을 특징으로 하는 암세포 투과성 단백질 전달용 조성물.
- 제8항에 있어서,
상기 형광 단백질은 강화된 녹색 형광 단백질(EGFP)인 것을 특징으로 하는 암세포 투과성 단백질 전달용 조성물.
- 제1항에 있어서,
상기 세포 투과 펩타이드(Cell Penetrating Peptide)인 암세포 호밍 펩타이드(Tumor Homing Peptide) 및 암세포 내부에서 작용하는 단백질 또는 펩타이드는 서열목록 제7 서열: GGGS를 갖는 링커에 의해 서로 연결된 형태인 암세포 투과성 단백질 전달용 조성물.
- 상기 제1항 내지 제10항 중 어느 한 항의 암세포 투과성 단백질 전달용 조성물을 포함하는 암세포의 연구, 검출 및 치료 중 어느 하나 이상을 수행하는 조성물.
- 상기 제1항 내지 제10항 중 어느 한 항의 암세포 투과성 단백질 전달용 조성물을 세포와 접촉시키는 단계를 포함하는 암세포 투과성 단백질 전달용 조성물을 세포에 전달하는 방법.
