Abstract
본 발명은 신경줄기세포의 줄기세포능 증진용 재조합 벡터 등에 관한 것으로서, GNG2의 발현 및/또는 활성이 증가한 신경줄기세포는 자기 생산 및 자기 복제 능력 등이 향상되는 것을 확인하여 완성된 것이다. 구체적으로, 본 발명자들은 마우스 태아로부터 수득한 신경줄기세포에서 GNG2 재조합 벡터를 과발현시킨 결과 신경줄기세포의 신경세포구 형성능이 증가하고, 줄기세포능 마커인 Pax6의 발현이 증가하는 것을 확인하였다. 또한, 마우스 태아의 뇌에 GNG2 재조합 벡터를 도입한 후 뇌에서 신경줄기세포의 분포를 확인한 결과, 대뇌의 뇌실/뇌실하대 구역에 본 발명에 따른 재조합 벡터를 발현하는 세포가 밀집되어 있는 것을 확인하였다. 즉, GNG2의 발현 및/또는 활성 증가에 의한 신경줄기세포의 줄기세포능 증진 효과가 in vitro 및 in vivo 모두에서 확인된 바, 본 발명에 따른 신경줄기세포의 줄기세포능 증진용 재조합 벡터는 외상성 뇌질환 및/또는 신경 퇴행성 질환의 치료 분야에서 다양하게 활용될 것으로 기대된다.
Description
본 발명은 신경줄기세포의 줄기세포능 증진용 조성물, 및 외상성 뇌손상 또는 신경 퇴행성 질환의 예방 또는 치료용 약학적 조성물 등에 관한 것이다.
신경줄기세포 (neural stem cells)는 자기 재생 (self-renewal)이 가능하며 신경계통 세포 (신경세포, 성상교세포, 희소돌기아교세포 등)로의 분화 능력을 가진 줄기세포이다. 포유동물의 전체 생명 과정에서 신경줄기세포의 증식 및 자기 재생은 지속적으로 이루어지며 신경재생 (neurogenesis)에 중요한 역할을 한다.
외상성 뇌손상, 및 신경 퇴행성 질환은 환경 또는 유전적 요인에 의한 신경세포의 퇴화, 결손, 사멸에 의해 신경 기능에 이상이 생기는 질환으로서, 근래에는 특히 고령 인구가 급증하면서 알츠하이머, 치매 등의 신경 퇴행성 질환에 대한 관심이 높아지고 있다. 그러나 신경세포가 과도하게 손상 또는 퇴행된 경우 재생이 불가하므로 상기 질환들의 근본적인 치료가 어려운 실정이며, 뇌손상 또는 퇴행성 뇌질환의 치료를 위해 일반적으로 사용되고 있는 약물요법 또는 외과적 수술법은 손상된 신경세포 뿐만 아니라 정상 세포에까지 영향을 미칠 수 있어 부작용의 위험이 있다.
최근에는 손상되거나 사멸한 신경세포를 외부에서 공급하는 신경세포로 대체하는 세포대체 요법 (cell replacement therapy)이 효과적인 치료법으로 주목 받고 있으며, 그 중에서도 신경계통세포로 분화가 가능한 신경줄기세포를 이용한 치료법이 각광받고 있다. 특히 뇌신경계 조직은 기타 조직과 달리 면역거부반응이 거의 없기 때문에 신경줄기세포를 이식하였을 때 이식된 세포의 장기간 생존을 기대할 수 있다는 이점이 있다. 따라서 신경세포의 손상 또는 퇴행으로 유발되는 신경계 질환의 치료를 위해, 신경줄기세포를 이용한 세포치료제 연구가 활발히 이루어지고 있다.
그러나, 외상성 뇌질환 및 신경 퇴행성 질환의 효과적인 치료를 위해서는 손상된 신경세포를 대체하기 위해 충분한 양의 신경줄기세포를 이식하여야 하는데, 신경줄기세포를 다량으로 획득하기 위한 방법에 대한 연구는 부족한 실정이다. 따라서, 신경줄기세포의 줄기세포능을 향상시킴으로써 신경줄기세포의 증식을 효과적으로 촉진하기 위한 생물학적 인자의 발굴이 시급하다.
본 발명은 상기 문제점을 해결하기 위해 안출된 것으로서, 본 발명자들은 GNG2 (G protein subunit gamma 2)의 발현 및/또는 활성이 증가한 신경줄기세포는 자기 재생 능력이 향상되는 것을 발견하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 GNG2를 코딩하는 폴리뉴클레오티드를 포함하는, 신경줄기세포의 줄기세포능 증진용 재조합 벡터를 제공하는 것이다.
본 발명의 다른 목적은 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 재조합 벡터를 유효성분으로 포함하는 신경줄기세포의 줄기세포능 증진용 조성물 및/또는 키트를 제공하는 것이다.
본 발명의 또 다른 목적은 상기 재조합 벡터를 신경줄기세포에 형질전환 또는 형질도입시키는 단계를 포함하는 신경줄기세포의 줄기세포능 증진 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 재조합벡터 및/또는 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포를 유효성분으로 포함하는 외상성 뇌손상 또는 신경 퇴행성 질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명은 서열번호 1로 표시되는 아미노산 서열을 포함하는 GNG2 (G protein subunit gamma 2)를 코딩하는 폴리뉴클레오티드를 포함하는 신경줄기세포의 줄기세포능 증진용 재조합 벡터를 제공한다.
본 발명의 일 구현예에서, 상기 GNG2를 코딩하는 폴리뉴클레오티드는 서열번호 2로 표시되는 염기서열을 포함할 수 있으나, 이에 한정되지 않는다.
또한, 본 발명은 본 발명에 따른 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포를 제공한다.
본 발명의 일 구현예에서, 상기 형질전환 또는 형질도입된 신경줄기세포는 GNG2의 발현 또는 활성이 증가할 수 있으나, 이에 한정되지 않는다. 바람직하게는 상기 발현 또는 활성이 증가한 GNG2는 서열번호 1로 표시되는 아미노산 서열을 포함하는 것일 수 있다.
또한, 본 발명은 본 발명에 따른 재조합 벡터를 유효성분으로 포함하는 신경줄기세포의 줄기세포능 증진용 조성물을 제공한다.
또한, 본 발명은 본 발명에 따른 재조합 벡터를 유효성분으로 포함하는 신경줄기세포의 줄기세포능 증진용 키트를 제공한다.
또한, 본 발명은 하기 단계를 포함하는 신경줄기세포의 줄기세포능 증진 방법을 제공한다:
(S1) 본 발명에 따른 재조합 벡터를 신경줄기세포에 형질전환 또는 형질도입시키는 단계; 및
(S2) 상기 (S1) 단계에서 재조합 벡터가 도입된 신경줄기세포에서 상기 재조합 벡터를 과발현시키는 단계.
또한, 본 발명은 본 발명에 따른 재조합 벡터, 및 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포로 이루어진 군에서 선택된 하나 이상을 유효성분으로 포함하는 외상성 뇌손상 (traumatic brain injury) 및/또는 신경 퇴행성 질환 (neurodegenerative diseases) 예방 또는 치료용 약학적 조성물을 제공한다.
나아가, 본 발명은 상기 약학적 조성물을 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 외상성 뇌손상 및/또는 신경 퇴행성 질환 예방 또는 치료 방법을 제공한다.
나아가, 본 발명은 외상성 뇌손상 및/또는 신경 퇴행성 질환 치료용 약제 제조를 위한 상기 재조합 벡터, 및/또는 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포의 용도를 제공한다.
나아가, 본 발명은 상기 재조합 벡터, 및/또는 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포의 외상성 뇌손상 또는 신경 퇴행성 질환 및/또는 신경 퇴행성 질환 예방 또는 치료 용도를 제공한다.
본 발명의 일 구현예에서, 상기 신경 퇴행성 질환은 헌팅턴병 (Huntington's disease), 피크병 (Pick's disease), 크로이츠펠트-야콥병 (Creutzfeldt-Jakob disease), 알츠하이머병 (Alzheimer's disease), 근위축성 측삭 경화증 (amyotropic lateral sclerosis; 루게릭병), 파킨슨병 (Parkinson's disease), 당뇨병성 신경병증 (diabetic neuropathy), 폴리글루타민 (polyQ) 질환, 뇌졸중, 파르병 (Fahr disease), 멘케스병 (Menke's disease), 윌슨병 (Wilson's disease), 뇌 허혈 (cerebral ischemia), 프리온 장애 (prion disorder), 치매 (dementia), 피질기저 퇴행증 (corticobasal degeneration), 진행성 핵상 마비 (progressive supranuclear palsy), 다계통 위축증 (multiple system atrophy), 선천성 경직성 반신부전마비 (hereditary spastic paraparesis), 및 척수소뇌 위축증 (spinocerebellar atrophies)으로 이루어진 군에서 선택된 하나 이상일 수 있으나, 이에 한정되지 않는다.
본 발명의 다른 구현예에서, 본 발명에 따른 재조합 벡터는 하기 특징 중 하나 이상을 만족할 수 있으나, 이에 한정되지 않는다:
(a) 신경줄기세포의 신경세포구 형성능을 증가 또는 활성화시킴; 또는
(b) 신경줄기세포의 Pax6 발현 또는 활성을 증가시킴.
본 발명은 신경줄기세포의 줄기세포능 증진용 재조합 벡터 등에 관한 것으로서, GNG2의 발현 및/또는 활성이 증가한 신경줄기세포는 자기 생산 및 자기 복제 능력 등이 향상되는 것을 확인하여 완성된 것이다. 구체적으로, 본 발명자들은 마우스 태아로부터 수득한 신경줄기세포에서 GNG2를 과발현시킨 결과 신경줄기세포의 신경세포구 형성능이 증가하고, 줄기세포능 마커인 Pax6의 발현이 증가하는 것을 확인하였다. 또한, 마우스 태아의 뇌에 GNG2 과발현 벡터를 도입한 후 뇌에서 GNG2의 발현 및/또는 활성이 증가한 신경줄기세포의 분포를 확인한 결과, 대뇌의 뇌실/뇌실하대 구역에 재조합 GNG2를 발현하는 세포가 밀집되어 있는 것을 확인하였다. 즉, GNG2의 발현 및/또는 활성 증가에 의한 신경줄기세포의 줄기세포능 증진 효과가 in vitro 및 in vivo 모두에서 확인된 바, 본 발명에 따른 신경줄기세포의 줄기세포능 증진용 재조합 벡터는 외상성 뇌질환 및/또는 신경 퇴행성 질환의 치료 분야에서 다양하게 활용될 것으로 기대된다.
도 1은 마우스 태아로부터 수득한 일차 신경줄기세포에 본 발명에 따른 GNG2 발현 벡터를 발현시키거나 발현시키지 않은 후 신경줄기세포 배양 배지에서 부유 상태로 배양했을 때 형성된 신경세포구를 촬영한 이미지이다.
도 2는 마우스 태아로부터 수득한 일차 신경줄기세포에 본 발명에 따른 GNG2 발현 벡터를 발현시키거나 발현시키지 않은 후 신경세포구의 형성 정도를 비교한 결과를 나타낸다.

본 발명자들은 신경줄기세포에 본 발명에 따른 GNG2 발현 벡터를 발현시킨 결과, 자기 생성능이 향상됨으로써 신경줄기세포의 증식 능력이 증가하는 것을 확인하여, 본 발명을 완성하였다.
구체적으로, 본 발명의 일 실시예에서는, 마우스 태아로부터 분리한 신경줄기세포에 본 발명에 따른 GNG2 발현 벡터를 과발현시킨 후 부유 상태로 배양한 결과, 음성 대조군에 비해 신경세포구 형성이 증가한 것을 확인하였다 (실시예 1 참조).
본 발명의 다른 실시예에서는, 마우스 태아로부터 분리한 신경줄기세포에 본 발명에 따른 GNG2 발현 벡터를 과발현시킨 후 신경줄기세포의 줄기세포능 마커인 Pax6의 발현을 확인한 결과, 음성 대조군에 비해 Pax6 발현이 증가되어 있는 것을 확인하였다 (실시예 2 참조).
본 발명의 또 다른 실시예에서는, 전기천공법을 이용하여 태아 마우스 뇌의 신경줄기세포에 본 발명에 따른 GNG2 발현 벡터를 과발현시킨 후 대뇌에서의 신경줄기세포의 분포를 확인한 결과, 음성 대조군에 비해 뇌실/뇌실하대 구역에서의 분포가 증가되어 있는 것을 확인하였다 (실시예 3 참조).
이하, 본 발명에 대해 구체적으로 설명한다.
본 발명은 서열번호 1로 표시되는 아미노산 서열을 포함하는 GNG2를 코딩하는 폴리뉴클레오티드를 포함하는 신경줄기세포의 줄기세포능 증진용 재조합 벡터를 제공한다.
본 발명에 있어서, GNG2 (G protein subunit gamma 2)는 GNG2 유전자에 의해 코딩되는 단백질로, 외부 신호에 대한 세포 반응을 매개하는 구아닌 뉴클레오티드-결합 단백질 (Guanine nucleotide-binding proteins; G 단백질)의 감마 (γ) 서브유닛 (subunit) 단백질이다. GNG2 단백질은 GDP의 GTP로의 교체 및 G 단백질 (G protein)-이펙터 (effector) 상호작용을 위한 GTPase의 활성에 필요한 것으로 알려져 있으며, 비정상적 GNG2 단백질과 관련된 질환으로는 우발적 운동실조 (episodic ataxia) 등이 알려져 있다. GNG2 단백질에 관한 정보는 당업계에 공지되어 있으며, 공공 데이터베이스 UniProt에서 서열 정보 등을 확인할 수 있다 (등록번호 P59768).
본 발명에 있어서, “신경줄기세포 (neural stem cells)은 상대적으로 발생이 덜된 미분화된 세포로서, 자기 재생이 가능하며 특히 신경세포, 성상세포, 희소돌기아교세포 등 신경계통 세포로의 분화 능력을 갖는 세포를 의미한다.
본 발명에 있어서, “줄기세포능 (stemness)”은 줄기세포가 자기 재생 (self-renewal), 자기 복제, 또는 증식하는 능력, 및/또는 여러 종류의 세포로 분화할 수 있는 능력 (다분화능)을 모두 포함하는 개념이다. 신경줄기세포의 줄기세포능은 다양한 인자를 통해 확인할 수 있는데, 본 발명은 신경줄기세포의 줄기세포능이 향상될수록 함께 증가하는 것으로 알려진 신경세포구 형성능, Pax6 유전자 발현, 및 뇌실/뇌실하대 구역 (ventricular/subventricular zone)으로의 분포의 측정을 통해 GNG2의 발현 및/또는 활성 증가에 의한 신경줄기세포의 줄기세포능 향상 효과를 확인하였다. 바람직하게는, 본 발명에 있어서 “신경줄기세포의 줄기세포능 증진”이란 “신경줄기세포의 증식 유도”를 의미할 수 있다.
바람직하게는, 본 발명에 있어서 GNG2는 서열번호 1로 표시되는 아미노산 서열을 포함하고, 더욱 바람직하게는 서열번호 1로 표시되는 아미노산 서열로 코딩되나, 서열번호 1의 아미노산 서열과 80% 이상, 더욱 바람직하게는 90% 이상, 더더욱 바람직하게는 95% 이상의 서열 상동성을 가지는 아미노산 서열에 의해 코딩될 수 있다. 예를 들면, 70%, 71%, 72%, 73%, 74%, 75%, 76%, 77%, 78%, 79%, 80%, 81%, 82%, 83%, 84%, 85%, 86%, 87%, 88%, 89%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99%, 100%의 서열 상동성을 갖는 아미노산 서열을 포함한다.
또한, 본 발명에 있어서 GNG2를 코딩하는 폴리뉴클레오티드는, 바람직하게는 서열번호 2로 표시되는 염기서열을 포함하고, 더욱 바람직하게는 서열번호 2로 표시되는 염기서열에 의해 코딩되나, 서열번호 2의 염기서열과 80% 이상, 더욱 바람직하게는 90% 이상, 더더욱 바람직하게는 95% 이상의 서열 상동성을 가지는 염기서열을 포함할 수 있다. 예를 들면, 70%, 71%, 72%, 73%, 74%, 75%, 76%, 77%, 78%, 79%, 80%, 81%, 82%, 83%, 84%, 85%, 86%, 87%, 88%, 89%, 90%, 91%, 92%, 93%, 94%, 95%, 96%, 97%, 98%, 99%, 100%의 서열 상동성을 갖는 염기서열을 포함한다.
본 발명에 있어서 아미노산 서열 또는 염기서열에 대한 "서열 상동성의 %"는 두 개의 최적으로 배열된 서열과 비교 영역을 비교함으로써 확인되며, 비교 영역에서의 아미노산 서열 또는 염기서열의 일부는 두 서열의 최적 배열에 대한 참고 서열(추가 또는 삭제를 포함하지 않음)에 비해 추가 또는 삭제(즉, 갭)를 포함할 수 있다.
본 발명에 있어서 “벡터 (vector)”란 적합한 숙주 내에서 DNA를 발현시킬 수 있는 DNA 제조물을 의미한다. 상기 벡터는 벡터 내에 삽입된 유전자가 발현될 수 있도록 상기 유전자와 작동 가능하게 연결되는 필수적인 조절 요소를 포함할 수 있으며, 구체적으로는 프로모터, 오퍼레이터, 개시코돈, 종결코돈, 폴리아데닐화 시그널, 인핸서 같은 발현 조절 요소, 막 표적화 또는 분비를 위한 신호 서열 또는 리더 서열 등을 포함할 수 있고, 목적에 따라 다양하게 변형 및 제조될 수 있다. 또한, 발현 벡터는 벡터를 함유하는 숙주 세포를 선택하기 위한 선택성 마커를 포함할 수 있으며, 복제 가능한 발현 벡터인 경우 복제 기원을 포함할 수 있다.
벡터는 플라스미드, 파지 입자, 트렌스포존, 또는 간단하게 잠재적 유전체 (genome) 삽입물일 수 있다. 적당한 숙주에 도입되면, 벡터는 숙주의 게놈과 무관하게 자기 복제하고 기능하거나, 또는 일부 경우에 숙주세포의 유전체 내에 통합될 수 있다. 플라스미드가 현재 벡터의 가장 통상적으로 사용되는 형태이므로, 용어 “플라스미드 (plasmid)” 및 “벡터 (vector)”는 때로 상호 교환적으로 사용될 수 있다. 그러나, 본 발명은 당업계에 알려진 또는 알려지게 되는 바와 동등한 기능을 갖는 벡터의 다른 형태를 포함한다.
따라서, 본 발명의 벡터의 구체적인 종류에는 제한이 없으나, 바람직하게는 플라스미드 벡터, 코스미드 벡터, 바이러스 벡터 등에서 선택될 수 있다. 또한, 상기 바이러스 벡터는 바람직하게는 레트로바이러스 벡터 (Retroviral vector), 렌티바이러스 벡터 (Lentiviral vector), HIV (Human immunodeficiency virus) 벡터, MLV (Murineleukemia virus) 벡터, ASLV (Avian sarcoma leukosis virus) 벡터, SNV (Spleen necrosis virus) 벡터, RSV (Rous sarcoma virus) 벡터, MMTV (Mouse mammary tumor virus) 벡터, 아데노바이러스 벡터 (Adenoviral vector), 아데노 관련 바이러스 (Adeno-associated virus; AAV) 벡터, HSV (Herpes simplex virus) 벡터, 및 에피조말 (episomal) 벡터로 이루어진 군에서 선택될 수 있으나, 이에 제한되지 않는다. 바람직하게는, 본 발명에 따른 벡터는 레트로바이러스 벡터 또는 렌티바이러스 벡터이다.
본 발명에 있어서 “프로모터 (promoter)”에는 일례로 사이토메갈로바이러스 (cytomegalovirus) CMV 프로모터, 레트로바이러스 (retrovirus)의 LTR 프로모터, 라우스 육종 바이러스 (rous sarcoma virus) RSV 프로모터, MMT 프로모터, EF-1 알파 프로모터, UB6 프로모터, 치킨 베타-액틴 프로모터, CAG 프로모터, RPE65 프로모터 및 옵신 프로모터 등으로 이루어진 군으로부터 선택될 수 있으나, 이에 한정되지 않는다. 벡터의 프로모터는 구성적 또는 유도성일 수 있다.
또한, 본 발명은 본 발명에 따른 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포를 제공한다.
본 발명에 있어서 “형질전환 (transformation)”은 수용자 세포 (recipient cells)가 외부 유전물질 (특히, 유리 (free) DNA 등)을 받아들임으로써 새로운 유전 형질이 발현되는 것을 총칭하며, “형질도입 (transduction)”은 파지 (phage) 등 바이러스를 매개로 하여 외부 유전물질이 세포에 도입되어 새로운 유전 형질이 발현되는 것을 총칭한다. 따라서, “형질전환 또는 형질도입된 세포”란 분자유전학적 방법으로 외부의 유전자가 세포 내로 주입된 세포로서, 바람직하게는 본 발명에 따른 재조합 벡터가 주입된 신경줄기세포를 의미한다.
신경줄기세포 내로 본 발명의 재조합 벡터를 주입하는 방법에는 제한이 없다. 즉, 본 발명에 따른 형질전환 또는 형질도입된 신경줄기세포는 형질전환 (transformation), 형질감염 (transfection), 또는 형질도입 (transduction)을 통해 본 발명의 재조합 벡터를 신경줄기세포 내로 주입시킴으로써 제조될 수 있으며, 상기 재조합 벡터의 주입은 구체적으로는 입자 총 충격법 (particle gun bombardment), 초음파 처리법 (sonication), 전기천공법 (electroporation), 리포펙션 (lipofection), 마이크로주입 (microinjection), 열 충격법 (heat shock), 화학적 물질 (CaCl2, DMSO, MgCl2, RbCl, 또는 PEG (Polyethylene glycol))-매개 도입 등의 방법을 통해 이루어질 수 있으나, 외부 유전 물질을 세포 내로 주입할 수 있는 방법이라면 제한 없이 이에 포함될 수 있다.
본 발명에 따른 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포는 GNG2의 발현 및/또는 활성이 증가하는 것을 특징으로 한다. 즉, 본 발명의 재조합 벡터가 도입된 신경줄기세포는 상기 재조합 벡터가 도입되지 않은 신경줄기세포에 비해 GNG2 발현 수준 및/또는 GNG2의 활성이 더욱 증가한 것을 특징으로 한다. 또한, 본 발명의 재조합 벡터가 도입된 신경줄기세포는 GNG2의 발현 및/또는 활성이 증가함으로써 줄기세포능이 증가한 것을 특징으로 한다. 바람직하게는, 본 발명의 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포에서 발현 및/또는 활성이 증가한 GNG2는 바람직하게는 서열번호 1로 표시되는 아미노산 서열을 포함하는 것일 수 있다.
또한, 본 발명은 본 발명에 따른 재조합 벡터를 유효성분으로 포함하는, 신경줄기세포의 줄기세포능 증진용 조성물 및/또는 키트를 포함한다.
본 발명에 따른 신경줄기세포의 줄기세포능 증진용 재조합 벡터는 하기 특징 중 하나 이상을 만족할 수 있다: (a) 신경줄기세포의 신경세포구 형성능을 증가 또는 활성화시킴; 또는 (b) 신경줄기세포의 Pax6 발현 또는 활성을 증가시킴.
본 발명에 따른 키트에는 본 발명의 재조합 벡터 외에도 유전 물질을 세포 내로 도입시키기 위해 당업계에서 일반적으로 사용되는 도구, 시약 등이 제한 없이 포함될 수 있다. 상기 도구 또는 시약의 일예로, 적합한 담체, 검출 가능한 신호를 생성할 수 있는 표지 물질, 발색단 (chromophores), 용해제, 세정제, 완충제, 안정화제 등이 포함되나 이에 제한되지 않는다.
또한, 본 발명은 하기 단계를 포함하는 신경줄기세포의 줄기세포능 증진 방법을 제공한다:
(S1) 본 발명에 따른 재조합 벡터를 신경줄기세포에 형질전환 또는 형질도입시키는 단계; 및
(S2) 상기 (S1) 단계에서 재조합 벡터가 도입된 신경줄기세포에서 상기 재조합 벡터를 과발현시키는 단계.
본 발명에 따른 신경줄기세포의 줄기세포능 증진 방법은 in vivo, in vitro, 또는 ex vivo에서 수행되는 것을 특징으로 할 수 있다. 즉, 본 발명에 따른 신경줄기세포의 줄기세포능 증진 방법은 개체로부터 분리된 신경줄기세포를 대상으로 수행되는 것을 특징으로 할 수 있다.
또한, 본 발명은 본 발명에 따른 재조합 벡터, 및 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포로 이루어진 군에서 선택된 하나 이상을 유효성분으로 포함하는, 외상성 뇌손상 또는 신경 퇴행성 질환의 예방 또는 치료용 약학적 조성물을 제공한다. 바람직하게는, 상기 약학적 조성물은 세포치료제 (cell therapeutics)로 활용하기 위한 것일 수 있다. 즉, 본 발명에 따른 약학적 조성물은 외상성 뇌손상 또는 신경 퇴행성 질환의 예방 또는 치료를 위한 세포치료제의 제조를 위한 것일 수 있다.
본 발명에 있어서, “외상성 뇌손상 (traumatic brain injury)”은 뇌의 산소 결핍이나 외상으로 인해 발생하는 뇌 조직 또는 뇌 신경의 손상을 의미한다. 외상성 뇌손상은 초기 외상으로 인한 신경 손상 외에도 이차적으로 신경의 손상 후 발생하는 뇌압 증가, 뇌부종, 지속적 뇌경련, 뇌허혈 등에 의해서도 발생할 수 있다.
본 발명에 있어서, “신경 퇴행성 질환 (neurodegenerative diseases)”은 신경세포에 퇴행성 변화가 나타나면서 신경세포의 기능이 소실되거나 신경세포가 사멸하여 발생하는 뇌 또는 신경 질환을 의미한다. 신경세포의 점진적인 퇴행으로 인해 뇌 기능이 저하되면서 인지 능력 및 기억력 등이 상실될 수 있으며, 운동신경세포가 손상되어 근육 경련 및 근육 약화 등이 일어나기도 한다.
신경 퇴행성 질환의 구체적인 종류에는 제한이 없으나, 일예로 헌팅턴병 (Huntington's disease), 피크병 (Pick's disease), 크로이츠펠트-야콥병 (Creutzfeldt-Jakob disease), 알츠하이머병 (Alzheimer's disease), 근위축성 측삭 경화증 (amyotropic lateral sclerosis; 루게릭병), 파킨슨병 (Parkinson's disease), 당뇨병성 신경병증 (diabetic neuropathy), 폴리글루타민 (polyQ) 질환, 뇌졸중, 파르병 (Fahr disease), 멘케스병 (Menke's disease), 윌슨병 (Wilson's disease), 뇌 허혈 (cerebral ischemia), 프리온 장애 (prion disorder), 치매 (dementia), 피질기저 퇴행증 (corticobasal degeneration), 진행성 핵상 마비 (progressive supranuclear palsy), 다계통 위축증 (multiple system atrophy), 선천성 경직성 반신부전마비 (hereditary spastic paraparesis), 척수소뇌 위축증 (spinocerebellar atrophies) 등으로부터 선택될 수 있다.
본 발명에 있어서 조성물 내의 재조합 벡터 또는 상기 재조합 벡터가 도입된 신경줄기세포의 함량은 질환의 증상, 증상의 진행 정도, 환자의 상태 등에 따라서 적절히 조절 가능하며, 예컨대, 전체 조성물 중량을 기준으로 0.0001 내지 99.9중량%, 또는 0.001 내지 50중량%일 수 있으나, 이에 한정되는 것은 아니다. 상기 함량비는 용매를 제거한 건조량을 기준으로 한 값이다.
본 발명에 따른 약학적 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다. 상기 부형제는 예를 들어, 희석제, 결합제, 붕해제, 활택제, 흡착제, 보습제, 필름-코팅 물질, 및 제어방출첨가제로 이루어진 군으로부터 선택된 하나 이상일 수 있다.
본 발명에 따른 약학적 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 서방형 과립제, 장용과립제, 액제, 점안제, 엘실릭제, 유제, 현탁액제, 주정제, 트로키제, 방향수제, 리모나아데제, 정제, 서방형정제, 장용정제, 설하정, 경질캅셀제, 연질캅셀제, 서방캅셀제, 장용캅셀제, 환제, 틴크제, 연조엑스제, 건조엑스제, 유동엑스제, 주사제, 캡슐제, 관류액, 경고제, 로션제, 파스타제, 분무제, 흡입제, 패취제, 멸균주사용액, 또는에어로졸 등의 외용제 등의 형태로 제형화하여 사용될 수 있으며, 상기 외용제는 크림, 젤, 패치, 분무제, 연고제, 경고제, 로션제, 리니멘트제, 파스타제 또는 카타플라스마제 등의 제형을 가질 수 있다.
본 발명에 따른 약학적 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 올리고당, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
본 발명에 따른 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
본 발명에 따른 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 본 발명이 속하는 기술분야에 통상의 기술자에 의해 용이하게 결정될 수 있다.
본 발명의 약학적 조성물은 개체에게 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구 복용, 피하 주사, 복강 투여, 정맥 주사, 근육 주사, 척수 주위 공간(경막내) 주사, 설하 투여, 볼점막 투여, 직장 내 삽입, 질 내 삽입, 안구 투여, 귀 투여, 비강 투여, 흡입, 입 또는 코를 통한 분무, 피부 투여, 경피 투여 등에 따라 투여될 수 있다.
본 발명의 약학적 조성물은 치료할 질환, 투여 경로, 환자의 연령, 성별, 체중 및 질환의 중등도 등의 여러 관련 인자와 함께 활성성분인 약물의 종류에 따라 결정된다.
본 발명에서 “개체”란 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는 인간 또는 비-인간인 영장류, 생쥐 (mouse), 쥐 (rat), 개, 고양이, 말, 및 소 등의 포유류를 의미한다.
본 발명에서 “투여”란 임의의 적절한 방법으로 개체에게 소정의 본 발명의 조성물을 제공하는 것을 의미한다.
본 발명에서 “예방”이란 목적하는 질환의 발병을 억제하거나 지연시키는 모든 행위를 의미하고, “치료”란 본 발명에 따른 약학적 조성물의 투여에 의해 목적하는 질환과 그에 따른 대사 이상 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미하며, “개선”이란 본 발명에 따른 조성물의 투여에 의해 목적하는 질환과 관련된 파라미터, 예를 들면 증상의 정도를 감소시키는 모든 행위를 의미한다.
본 명세서 및 청구범위에 사용된 용어나 단어는 통상적이거나 사전적인 의미로 한정해서 해석되어서는 아니 되며, 발명자는 그 자신의 발명을 가장 최선의 방법으로 설명하기 위해 용어의 개념을 적절하게 정의할 수 있다는 원칙에 입각하여 본 발명의 기술적 사상에 부합하는 의미와 개념으로 해석되어야만 한다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1. GNG2 증가에 따른 신경줄기세포의 신경세포구 형성능 비교
GNG2 (G protein subunit gamma 2)가 신경줄기세포의 줄기세포능을 강화시키는지 알아보기 위해, 마우스 태아로부터 신경줄기세포를 채취한 후 GNG2 발현 및/또는 활성 증가 여부에 따른 신경세포구 (neurospheres) 형성 정도를 in vitro에서 확인하였다. 신경세포구는 신경줄기세포가 형성하는 고유의 구형 구조로서, 신경줄기세포가 활발히 분열할수록 더 많은 신경세포구가 형성된다.
1-1. 실험방법
일차 신경줄기세포를 채취하기 위해 코아텍으로부터 ICR종의 수컷 마우스와 암컷 마우스를 준비하였다. 수컷과 암컷 마우스를 교배시키고 14일이 지난 후, 펜토바르비탈 소듐 (pentobarbital sodium)을 이용해 모체 마우스를 마취한 후 경추 탈골 한 뒤 자궁에서 태아를 꺼냈다. 태아로부터 뇌를 채취한 후 신경줄기세포 밀집 지역인 뇌실/뇌실하대 구역 (ventricular/subventricular zone)의 조직을 분리하였다. 이어서 파이펫을 이용하여 조직을 단일 세포 단위로 분리 시킨 후, 신경줄기세포 배양 배지 (DMEM/F12, B27 (Invitrogen), Glutamax (Gibco), 1% penicillin/streptomycin (Gibco) 25ng/ml FGF2, 20ng/ml EGF (Pepro Tech))에서 이틀간 배양하였다.
GNG2 발현 증가에 따른 신경세포구 형성을 비교하기 위해, 상기 신경줄기세포 배양 배지에서 배양 중인 일차 신경줄기세포에 GNG2-과발현 레트로바이러스를 감염시켰다. 상기 GNG2-과발현 레트로바이러스는 인간 GNG2의 CDS 서열 (서열번호 2)이 삽입된 렌티바이러스 벡터 (pWPI 플라스미드; Addgene, #12254)를 포함하도록 제조하였다. 플라스미드의 제조, 재조합 벡터의 생산, 및 형질도입은 [Biochem Biophys Res Commun. 2015 Feb 27;458(1):110-6]을 참고하여 수행하였다.
레트로바이러스 감염 후 48시간이 경과되었을 때, 감염된 신경줄기세포들을 모아서 파이펫을 이용해 단일 세포 수준으로 해리시킨 이후 5,000개의 세포를 신경줄기세포 배양 배지와 함께 1주일간 배양하여 신경세포구를 획득하였다. 신경세포구의 관측 및 카운트는 광학현미경 (Eclipse TS100, Nikon)을 사용하였다.
1-2. 실험 결과
상기 실험방법에 따라 마우스로부터 분리한 일차 신경줄기세포에 본 발명에 따른 GNG2-재조합 벡터를 과발현한 후 신경세포구의 형성 정도를 비교한 결과, 재조합 GNG2 단백질을 발현하는 신경줄기세포는 음성 대조군에 비해 약 1.5배 이상의 더 많은 신경세포구를 형성하는 것을 확인할 수 있었다 (도 1 및 도 2). 즉, GNG2의 발현 및/또는 활성이 증가한 신경줄기세포에서 세포분열이 더욱 활발히 일어난 바, 상기 결과는 GNG2가 in vitro에서 신경줄기세포의 줄기세포능을 강화시킨다는 것을 보여준다.
실시예 2. GNG2 증가에 따른 신경줄기세포의 줄기세포 마커 발현 변화 확인
GNG2에 의한 신경줄기세포의 줄기세포능 강화를 검증하기 위해, GNG2의 발현 및/또는 활성 증가에 의해 줄기세포 마커 단백질의 발현이 변화하는지 확인하였다. 줄기세포능의 마커로 신경줄기세포 증식 및 다분화능의 마커인 Pax6 전사인자를 활용하였다.
2-1. 실험 방법
상기 실시예 1과 동일한 방법으로 마우스 태아의 뇌로부터 신경줄기세포를 추출한 후, in vitro 조건에서 본 발명에 따른 GNG2 재조합 벡터를 레트로바이러스를 이용해 과발현시켰다. 이후 웨스턴 블롯을 진행하기 위해 레트로바이러스에 감염된 신경줄기세포들을 프로테아제 (protease) 및 포스파타아제 (phosphatase) 억제제 (Pierce Biotechnology, Rockford, IL)가 포함된 용해 버퍼 (Lysis buffer) (50mM Tris-cl (Georgiachem), 25mM beta-glycerophosphate disodium salt hydrae (Sigma), 25mM NaCl (Daejung), 2% NP-40 (USB Corporation))을 이용하여 용해시키고, 단백질 용액을 채취하였다.
동량 (35μg)의 단백질 용액을 10% (w/v) sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE)로 분리한 뒤 polyvinylidene fluoride (PVDF) 멤브레인 (Merck)에 옮겨주었다. 멤브레인은 5% (w/v) 탈지 우유를 포함한 TBST (150mM NaCl, 10mM Tris-Cl, 0.1% (v/v) Tween 20, pH 8.0) 용액을 이용해 블로킹 (blocking) 한 후, 하기 1차 항체를 처리하여 4℃에서 밤새도록 반응시켰다: 항-Pax6 항체 (Abcam, ab5790), 항-beta-actin 항체 (Santa Cruz Biotechnology, sc-47778). 1차 항체와의 반응을 마친 멤브레인을 TBST 용액을 이용하여 헹구어 준 뒤, 2차 항체인 겨자무과산화효소 (horseradish peroxidase, HRP)-결합된 항-마우스 항체 (Santa Cruz Biotechnology, sc-516102) 혹은 HRP-결합된 항-토끼 항체 (Santa Cruz Biotechnology, sc-2357)가 포함된 용액에 담아 상온에서 4시간 반응시켰다.
항체와의 배양을 마친 단백질 밴드의 시각화는 X-Omat film (Kodak)을 이용해 수행하였으며 컴퓨터 이미지화 및 그래프화는 Image J 및 Graphpad prism 5를 이용하였다.
2-2. 실험 결과
상기 실험방법에 따라 GNG2 재조합 벡터를 과발현한 신경줄기세포의 Pax6 단백질 발현 수준을 확인한 결과, 음성 대조군에 비해 GNG2의 발현 및/또는 활성이 증가한 신경줄기세포에서 Pax6 단백질 발현이 약 4배 증가한 것으로 나타났다 (도 3 및 도 4). 상술한 바와 같이 Pax6은 대표적인 신경줄기세포 마커로서, 줄기세포능이 강화될수록 Pax6의 단백질 발현이 함께 증가한다는 것이 당업계에 잘 알려져 있는 바, 상기 실험 결과는 GNG2가 신경줄기세포의 줄기세포능을 강화시킨다는 사실을 검증하는 것이다.
실시예 3.
In vivo
에서 GNG2 증가에 의한 신경줄기세포의 줄기세포능 강화 확인
GNG2가 신경줄기세포의 줄기세포능에 미치는 영향을 in vivo 상에서도 확인하기 위해, 자궁 내 전기천공법 (in utero electroporation)으로 마우스 태아의 신경줄기세포에 GNG2 재조합 벡터를 과발현시킨 후 신경줄기세포의 뇌실/뇌실하대 구역에서의 분포를 확인하였다. 신경줄기세포는 줄기세포능이 강화될수록 대뇌 피질의 가장 아랫단인 뇌실/뇌실하대 구역 (ventricular/subventricular zone)에 머무르는 경향을 보이고, 줄기세포능이 약화될수록 뇌실/뇌실하대 구역을 벗어나 중간대 (intermediate zone) 또는 cortical plate (피질판)으로 이동하는 경향을 보인다.
3-1. 실험 방법
자궁 내 전기천공법을 수행하기 위해, 임신 13일차의 마우스에 펜토바르비탈 소듐을 복강주사하여 마취시켰다. 복강을 열어 자궁을 모두 꺼낸 후 태아의 측면 뇌실에 0.01% fast green dye (Sigma)가 섞인 GNG2-재조합 벡터 용액 (2 내지 4.0 μg/μl)을 주입하였다. 이후 상기 재조합 벡터를 뇌실/뇌실하대 구역에 있는 신경줄기세포에 전달하기 위해 포셉 형태의 전극 (CUY650P5, 5mm diameter platinum round plates; Nepagene)을 이용하여 1초당 1회의 전류 펄스 (33V, 50 ms pulse followed by 950 ms gap)을 총 5번 흘려주었다. 이후 자궁을 다시 복강에 넣고 환부를 꿰매어 준 후 모체를 회복시켰다.
본 발명에 따른 GNG2 재조합 벡터를 발현하는 신경줄기세포의 변화를 확인하기 위해, 전기천공법을 수행한지 이틀 후 마우스를 경추 탈골 한 후 태아를 꺼내어 뇌를 채취하였으며, 4% 파라포름알데하이드 용액을 이용하여 고정시키고, 30% sucrose 용액을 이용하여 탈수시킨 후 동결절단 (cryosection) 하여 뇌절편을 획득하였다. 면역염색법 (immunostaining)을 위해, 뇌절편을 PBS로 헹구어 준 후 10% FBS와 0.2% Triton X-100이 포함된 PBS를 이용하여 블로킹 (blocking) 과정을 진행하였다. 이후 염색을 위해 해당 절편을 1차 항체인 항-GFP (A-11122, Invitrogen, 1:500) 항체가 포함된 용액으로 4℃에서 밤새도록 반응시켰다. 이어서 PBS를 이용해서 3번 헹구어 준 후 2차 항체 반응을 위해 Alexa Fluor 488 표지자가 달린 2차 항체 용액으로 상온에서 3시간 반응시킨 후 4`,6-diamidino-2-phenylindole (DAPI; Sigma)를 이용하여 핵을 염색하였다. 형광 사진은 Eclipse 80i (Nikon) 현미경을 이용하여 관찰하였다.
3-2. 실험 결과
상기 실험 방법에 따라 마우스 태아의 뇌실에 GNG2 재조합 벡터를 주입한 후 전류를 통해 신경줄기세포가 밀집되어 있는 뇌실/뇌실하대 구역에 전달하고, 면역염색법을 통해 GNG2의 발현 및/또는 활성이 증가한 신경줄기세포의 분포를 확인하였다. 그 결과, 본 발명에 따른 재조합 벡터를 과발현하는 신경줄기세포는 대조군에 비해 뇌실/뇌실하대 구역에 더 많이 밀집되어 있는 것으로 나타났다 (도 5 및 도 6). 상기 결과는 GNG2가 in vitro 뿐만 아니라 in vivo에서도 신경줄기세포의 줄기세포능을 강화시키는 효과가 있음을 보여준다.
이상에서 살펴본 바와 같이, 본 발명자들은 GNG2의 발현 및/또는 활성 증가가 신경줄기세포의 줄기세포능을 증진시키는 효과가 있음을 in vitro 및 in vivo 모두에서 확인하였다. 따라서, 본 발명에 따른 GNG2는 신경줄기세포를 이용한 뇌손상 혹은 퇴행성 뇌질환 등의 치료의 핵심 인자로 활용될 것으로 기대된다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야 한다.

- 서열번호 1로 표시되는 아미노산 서열을 포함하는 GNG2 (G protein subunit gamma 2)를 코딩하는 폴리뉴클레오티드를 포함하는 신경줄기세포의 줄기세포능 증진용 재조합 벡터.
- 제1항에 있어서,
상기 GNG2를 코딩하는 폴리뉴클레오티드는 서열번호 2로 표시되는 염기서열을 포함하는 것을 특징으로 하는, 재조합 벡터.
- 제1항의 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포.
- 제3항에 있어서,
상기 형질전환 또는 형질도입된 신경줄기세포는 GNG2의 발현 또는 활성이 증가하는 것을 특징으로 하는, 형질전환 또는 형질도입된 신경줄기세포.
- 제1항의 재조합 벡터를 유효성분으로 포함하는 신경줄기세포의 줄기세포능 증진용 조성물.
- 제5항에 있어서,
상기 재조합 벡터는 하기 특징 중 하나 이상을 만족하는 것을 특징으로 하는, 신경줄기세포의 줄기세포능 증진용 조성물:
(a) 신경줄기세포의 신경세포구 형성능을 증가 또는 활성화시킴; 또는
(b) 신경줄기세포의 Pax6 발현 또는 활성을 증가시킴.
- 제1항의 재조합 벡터를 유효성분으로 포함하는 신경줄기세포의 줄기세포능 증진용 키트.
- 하기 단계를 포함하는 신경줄기세포의 줄기세포능 증진 방법:
(S1) 제1항의 재조합 벡터를 신경줄기세포에 형질전환 또는 형질도입시키는 단계; 및
(S2) 상기 (S1) 단계에서 재조합 벡터가 도입된 신경줄기세포에서 상기 재조합 벡터를 과발현시키는 단계.
- 제1항의 재조합 벡터, 및 상기 재조합 벡터로 형질전환 또는 형질도입된 신경줄기세포로 이루어진 군에서 선택된 하나 이상을 유효성분으로 포함하는 외상성 뇌손상 (traumatic brain injury) 또는 신경 퇴행성 질환 (neurodegenerative diseases) 예방 또는 치료용 약학적 조성물.
- 제9항에 있어서,
상기 신경 퇴행성 질환은 헌팅턴병 (Huntington's disease), 피크병 (Pick's disease), 크로이츠펠트-야콥병 (Creutzfeldt-Jakob disease), 알츠하이머병 (Alzheimer's disease), 근위축성 측삭 경화증 (amyotropic lateral sclerosis; 루게릭병), 파킨슨병 (Parkinson's disease), 당뇨병성 신경병증 (diabetic neuropathy), 폴리글루타민 (polyQ) 질환, 뇌졸중, 파르병(Fahr disease), 멘케스병 (Menke's disease), 윌슨병 (Wilson's disease), 뇌 허혈 (cerebral ischemia), 프리온 장애 (prion disorder), 치매 (dementia), 피질기저 퇴행증 (corticobasal degeneration), 진행성 핵상 마비 (progressive supranuclear palsy), 다계통 위축증 (multiple system atrophy), 선천성 경직성 반신부전마비 (hereditary spastic paraparesis), 및 척수소뇌 위축증 (spinocerebellar atrophies)으로 이루어진 군에서 선택된 하나 이상인 것을 특징으로 하는, 약학적 조성물.
