Abstract
Description
도 2는 배양 및 팽창된 STRO-1bright MPC의 STRO-1bright 또는 STRO-1dim 자손(progeny)의 유전자 발현 프로파일을 나타낸 것이다. (A) 생체 외(ex vivo) 팽창된 골수 MPC의 단일 세포 현탁액은 트립신/EDTA 처리하여 제조되었다. 세포는 상기 STRO-1 항체로 염색한 다음, 염소-항 뮤린 IgM-FITC(fluorescein isothiocyanate)로 처리하여 나타내었다. 총 세포 RNA는 FACS(fluorescence activated cell sorting)후에 STRO-1dim 또는 STRO-1bright을 발현하는 세포의 정제된 집단(population)으로부터 제조되었다. (B) RNAzolB 추출 방법, 및 표준 방법을 이용하여 총 RNA는 각각의 하위집단(subpopulation)으로부터 분리되었고(isolated) cDNA 합성을 위한 주형(template)으로 사용되었다. 다양한 전사체(transcript)는 전술한 표준 프로토콜(Gronthos et al. J Cell Sci . 116:1827-1835, 2003)을 사용하여 PCR 증폭으로 평가되었다. 본 연구에서 사용된 프라이머 쌍은 표 2에 나타내었다. 증폭 후, 각각의 반응혼합물은 1.5% 아가로스 젤 전기영동으로 분석되었고, EtBR(ethidium bromide) 염색으로 가시화되었다. (C) 각각의 세포 마커에 대한 상대적 유전자 발현은 ImageQant 소프트웨어를 사용으로 하우스-키핑 유전자(house-keeping gene)인 GAPDH의 발현을 참조하여 평가되었다.
도 3은 배양 및 팽창된 STRO-1+ MPC의 STRO-1bright 자손(progeny)은 높은 수준의 SDF-1를 발현하고 STRO-1dim 자손은 그렇지 아니하였다. (A) STRO-1+ BMMNCs의 MACS-분리된 제제(preparation)는 FACS를 이용하여 STRO-1bright 및 STRO-1dim /dull 영역에 따라 STRO-1 서브세트(subsets)로 분할되었다. (B-C) 총 RNA는 각각의 STRO-1 하위집단(subpopulation)으로부터 제조되었고 STRO-1bright 감산 혼성화 라이브러리(subtraction hybridization library)를 구축하는데 사용되었다. STRO-1bright 감산된(subtracted) cDNA로 형질전환된 박테리아 클론으로부터 증폭된 대표 PCR 산물이 도발(blot)된 니트로셀룰로오스 필터(nitrocellulose filter)를 복제하였다. 그 다음, 상기 필터는 [32P]디옥시시티딘 트리포스페이트(deoxycytidine triphosphate, dCTP)-표지 STRO-1bright (B) 또는 STRO-1dim /dull (C) 감산 cDNA로 표지(probe)되었다. 화살표는 인간 SDF-1에 대응하는 cDNA 단편을 함유하는 한 클론(clone)을 나타낸 것이다. (D) 역전사효소(reverse transcriptase(RT))-PCR 분석은 배양 전 갓 MACS/FACS-분리된 BMMNC STRO-1 집단으로부터 제조된 총 RNA에서의 SDF-1 및 글리세르알데히드-3-포스페이트 탈수소효소(glyceraldehyde-3-phosphate dehydrogenase, GAPDH) 전사체의 상대적 발현을 보여준 것이다. bp는 염기쌍을 나타낸다.
도 4는 간질 세포(stromal cell) 및 1차 장기간 배양물(primary long term cultures)에서의 CD146, CXCL12 및 안지오포이에틴 1(Angiopoietin 1)의 발현을 나타내는 일련의 그래프를 도시한 것이다. (A) 아이소타입 대조군(isotype control)과 함께 CD146(MCAM)에 대한 간질 세포주 HS5 및 HS27a의 FACS 분석을 나타낸 것이다. (B) 아이소타입 대조군과 함께 CD146 lo에 대한 첫 번째 계대(passage)에서 1차 LTC의 FACS 분석을 나타낸 것이다. 상기 1차 LTCs는 FACS-에이디드 유동-선별(FACS-aided flow-sorting)에 의해 CD146hi 및 CD146lo 집단으로 선별되었다. hi 및 lo 집단에 대한 대략적인 게이트(approximate gates) 또한 나타내었다. (C) CD146hi 및 CD146lo으로 선별된 9 LTC의 CXCL12의 수준은 RT-PCR로 정량(1000 계수를 곱한 베타-액틴과 비교)되었다. 같은 배양물로부터 쌍의 샘플은 선으로 연결되었다. (D) 동일한 9 LTC의 안지오포이에틴 1(Angiopoietin 1)의 수준은 또한 (1000 계수를 곱한 베타-액틴에 대한) RT-PCR로 정량되었다. 패널 A에서의 실험은 3개의 독립적 실험으로 확인되었고, 패널 B, C 및 D의 실험들은 9개의 독립적 샘플에서 확인되었다. 패널 C 및 D에서의 데이터 세트의 윌콕슨의 매치 쌍 테스트(Wilcoxon's matched pair test)에 의한 통계분석은 두 경우 모두 0.01 미만의 P-값을 나타내었다.
도 5는 콜라겐의 투여에 의한 내피 기능장애를 유도한 다음, 2주 동안의 처리(treated) 및 무처리된(untreated) 양(sheep)의 혈장에서의 IL-10 수준(ng/㎖)의 평균+SEM을 나타낸 것이다.
도 6은 콜라겐의 투여에 의한 내피 기능장애를 유도한 다음, 2주 동안의 처리(treated) 및 무처리된(untreated) 양(sheep)의 혈장에서의 피브리노겐(fibrinogen) 수준(g/L)의 평균+SEM을 나타낸 것이다.
도 7은 콜라겐의 투여에 의한 내피 기능장애를 유도한 다음, 2주 동안의 처리(treated) 및 무처리된(untreated) 양(sheep)의 혈장에서의 악티빈 A(Actvin A) 수준(pg/㎖)의 평균+SEM을 나타낸 것이다.
도 8은 콜라겐의 투여에 의한 내피 기능장애를 유도한 다음, 2주 동안의 처리(treated) 및 무처리된(untreated) 양(sheep)의 혈장에서의 C-반응성 단백질(C-reactive protein) 수준(mg/L)의 평균+SEM을 나타낸 것이다.
도 9는 콜라겐의 투여에 의한 내피 기능장애를 유도한 다음, 2주 동안의 처리(treated) 및 무처리된(untreated) 양(sheep)의 혈장에서의 C-반응성 단백질(C-reactive protein) 수준의 % 변화율을 나타낸 것이다.
도 10은 MPCs("처리군(treated)") 또는 식염수("대조군(control)")가 투여된 양(sheep)으로부터 엔도텔린-1(endothelin-1)에 따른 수축과 브라디키닌(bradykinin)(A), 카바콜(carbachol)(C) 또는 나트륨 니트로프루시드(sodium nitroprusside)(B 및 D)에 따른 뒤이은 이완 후 양(sheep)의 관상동맥(A 및 C) 또는 디지털 동맥(B 및 D)의 이완률(percentage relaxation)을 나타내는 일련의 그래프를 도시한 것이다.

Claims (17)
- STRO-1+ 또는 TNAP+ 간엽 전구세포(mesenchymal precursor cells, MPCs)가 증가된(enriched) 인간세포 집단 또는 이의 자손(progeny)을 포함하는, 인간 치료 대상에 전신 투여하기 위한 혈관 내피 기능장애(vascular endothelial dysfunction)의 질환 치료 또는 예방용 조성물로서, 상기 혈관 내피 기능장애의 질환은 고혈압(hypertension), 관상동맥 질환(coronary artery disease), 만성심부전(chronic heart failure), 말초동맥 질환(peripheral artery disease), 신장병(nephropathy), 및 만성신부전(chronic renal failure)으로 구성된 군에서 선택되는 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 혈관 내피 기능장애의 질환은 신장병인 것을 특징으로 하는 조성물.
- 제2항에 있어서, 상기 신장병은 당뇨병성(diabetic) 신장병인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 1kg당 0.1x106 내지 5x106 STRO-1+ 또는 TNAP+ MPCs 또는 이의 자손을 포함하는 조성물.
- 제1항에 있어서, 상기 집단의 투여량은 일정한 신체 투여량(constant body dose)인 조성물.
- 제5항에 있어서, 상기 집단의 투여량은 환자의 체중과 무관하게 약 1억 내지 3억 STRO-1+ 또는 TNAP+ MPCs 또는 이의 자손을 포함하는 조성물.
- 제6항에 있어서, 상기 집단의 투여량은 환자의 체중과 무관하게 약 1억 5천만 STRO-1+ 또는 TNAP+ MPCs 또는 이의 자손을 포함하는 조성물.
- 제1항에 있어서, 상기 조성물의 투여주기는 주 1회 또는 그 이하인 조성물.
- 제1항에 있어서, 상기 집단은 자가(autogeneic) 또는 타가(allogeneic)인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 집단은 투여하기 전에 배양 팽창(culture expanded)되는 것을 특징으로 하는 조성물.
- 제1항에 있어서, 펜토산 폴리설페이트(pentosan polysulfate, PPS) 또는 이의 약학적으로 허용되는 염을 추가로 포함하는 조성물.
- 제11항에 있어서, 상기 PPS는 펜토산 폴리설페이트의 나트륨 염(sodium salt)인 것을 특징으로 하는 조성물.
- 제12항에 있어서, 상기 펜토산 폴리설페이트의 나트륨 염의 총량은 약 1 내지 100mg인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 혈관 내피 기능장애의 질환은 고혈압인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 혈관 내피 기능장애의 질환은 관상동맥 질환 또는 말초동맥 질환인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 혈관 내피 기능장애의 질환은 만성심부전인 것을 특징으로 하는 조성물.
- 제1항에 있어서, 상기 혈관 내피 기능장애의 질환은 만성신부전인 것을 특징으로 하는 조성물.
