Abstract
본 발명에 따른 압타머는 SARS-CoV-2의 다양한 부위에 특이적으로 결합하고, SARS-CoV-2 단백질에 대해 높은 민감도 및 특이도를 나타내는 것을 확인하였는 바, SARS-CoV-2 의 진단 용도로 적용할 수 있다.

Claims (13)
- 서열번호 1 내지 58의 폴리뉴클레오티드 서열을 포함하는 군으로부터 선택되는 어느 하나 이상의 폴리뉴클레오티드 서열을 포함하는, SARS-CoV-2 표면돌기(spike)에 특이적으로 결합하는, 압타머.
- 제1항에 있어서, 상기 압타머는 이의 5' 말단 및 3' 말단 중 선택되는 어느 하나 이상에 1 내지 30 개의 폴리뉴클레오티드를 추가로 포함하는 것인, 압타머.
- 제1항에 있어서, 상기 압타머는 30 내지 150 개의 뉴클레오티드를 포함하는 것인, 압타머.
- 제1항에 있어서, 상기 압타머는 서열번호 1 내지 58의 폴리뉴클레오티드 서열을 포함하는 군으로부터 선택되는 어느 하나 이상의 폴리뉴클레오티드 서열 또는 이와 상동성이 90% 이상인 폴리뉴클레오티드 서열을 포함하는 것인, 압타머.
- 제1항에 있어서, 상기 압타머는 이의 폴리뉴클레오티드 서열을 이루는 하나 이상의 뉴클레오티드에 하기 중 선택되는 어느 하나 이상의 변형을 포함하는 것인, 압타머:
i) 하나 이상의 뉴클레오티드 중 T 염기의 5번 탄소가 벤질기, 나프틸기, 2-나프틸기, 피롤벤질기 및 트립토판으로 이루어진 군에서 선택되는 어느 하나 이상으로 치환;
ii) 하나 이상의 뉴클레오티드 내 2번 탄소 위치에서 -OH 기가 데옥시(deoxy), 메톡시(methoxy), 아미노(amino) 및 불소(F)로 이루어진 군에서 선택되는 어느 하나 이상으로 치환; 및
iii) 하나 이상의 뉴클레오티드가 메틸포스포네이트, 포스포로티오에이트, LNA(Locked Nucleic Acid), PNA(Peptide Nucleic Acid) 및 HNA(Hexitol Nucleic acid)로 이루어지는 군으로부터 선택되는 어느 하나 이상으로 치환.
- 제1항에 있어서, 상기 압타머는 이의 폴리뉴클레오티드 서열, 이의 5' 말단 및 3' 말단 중 선택되는 어느 하나 이상에 PEG(폴리에틸렌 글리콜), HEG(헥사 에틸렌 글리콜), idT(inverted deoxythymidin, 역-데옥시티미딘), 비오틴, 형광물질, 아민 링커, 티올 링커, 콜레스테롤 및 지방산으로 이루어지는 군으로부터 선택되는 어느 하나 이상이 접합된 것인, 압타머.
- 제1항 내지 제6항 중 어느 한 항의 압타머를 포함하는, SARS-CoV-2 진단용 조성물.
- 제1항 내지 제6항 중 어느 한 항의 압타머 중 선택되는 어느 1종 이상을 포함하는, SARS-CoV-2 진단용 키트.
- 제8항에 있어서, 상기 분리된 생체 시료 내에 존재하는 SARS-CoV-2를 표지하는 압타머는 착색미립자, 발색효소, 형광물질 및 방사선 동위원소로 이루어지는 군으로부터 선택되는 어느 하나 이상과 결합된 것인, 키트.
- 제7항의 조성물을 개체로부터 분리된 생체 시료와 접촉하는 단계를 포함하는, SARS-CoV-2 진단을 위한 정보 제공 방법.
- 제10항에 있어서, 상기 방법은 측면 유동 분석(lateral flow assay, LFA) 유형 테스트, 면역 흡착 분석(Enzyme-Linked Immunosorbent Assay, ELISA) 유형 테스트, 전계 효과 트랜지스터(field-effect transistor, FET) 유형 테스트 및 방사 면역 분석(radioimmunoassay, RIA) 유형 테스트로 이루어지는 군으로부터 선택되는 어느 하나 이상인, 방법.
- 제9항 또는 제10항에 있어서, 상기 생체 시료는 생물학적 샘플, 환경 샘플, 약제학적 샘플, 식품 샘플, 농업 샘플 및 가축 샘플로 이루어지는 군으로부터 선택되는 어느 하나 이상인 것인, 방법.
