본 발명은 암 치료용 펩타이드 단편, 이의 변이체 및 이의 용도에 관한 것이다.
본 발명의 VGLL1의 펩타이드 단편 및/또는 이의 변이체는 암세포의 증식을 억제시키고 암세포의 침윤을 억제시킴으로써 암의 예방 및 치료에 우수한 효과를 가진다.

Description
본 발명은 VGLL1 펩타이드를 포함하는 암 치료용 조성물 및 이를 이용한 암을 치료하는 방법에 관한 것이다.
암은 세계적으로 높은 사망률을 보이고 있으며, 서구 사회에서는 심혈관 질환 다음으로 가장 일반적인 사망 원인이다. 특히, 인구의 고령화, 식생활의 서구화로 인한 고지방식의 섭취의 일반화, 환경 오염 물질의 급격한 증가, 음주량의 증가 등으로 대장암, 유방암, 전립선암 등이 지속적으로 증가하는 추세에 있다. 이러한 실정에서 암의 조기 예방 및 치료를 가능하게 하여 인간 건강의 증진, 건강한 삶의 질 향상 및 인류 보건 증진에 기여할 수 있는 항암 물질의 창출이 절실히 요구되고 있다.
암의 발생의 가장 큰 요인은 유전자에 돌연변이를 일으키는 것이다. 돌연변이를 일으키게 하는 물질을 발암원 (carcinogen)이라고 말하고, 이 발암원에 의해 유전자 변형이 일어나는 단계를 initiation이라고 한다. initiation 단계에서 포함되는 유전자들 중에는 발암억제유전자 (예; p53, PTEN, INK family, RB)들과 돌연변이로 인해서 항상 활성을 갖게 되는 constitutive active 돌연변이 (활성돌연변이체, 예: EGFRmt, RasG12V)가 있다. 정상세포에는 epidermal growth factor가 EGF 수용체 (EGF receptor)에 결합함으로써 활성신호를 만들어 내지만, 활성 돌연변이체들은 수용체에 ligand가 결합하지 않아도 항상 활성신호를 하류 신호전달계에 보내주게 된다. 이 과정에서 생성된 하나의 돌연변이 세포는 몸 안에 있는 면역 방어시스템을 뚫고 종양덩어리로 성장하여야 하는데 이 과정을 promotion이라고 한다. 일반적으로 promotion 과정은 긴 시간이 요구되며, 주로 신호전달계의 활성화로 이루어진다.
많은 암의 경우, 사이토카인을 통한 다양한 신호전달 시스템의 활성화로 암관련 유전자들의 기능이 조절된다. TGF-β(Transforming growth factor-β)는 SMAD, Rho, PP2A 및 암관련 유전자의 신호 전달 경로 조절을 통해 세포 성장 억제, 세포 사멸, 분화 및 상피-간충직 전이 (epithelial-mesenchymal transition; EMT)와 같은 다양한 기능에 관여하고 있다. 동물 모델 및 환자에서 TGF-β가 발현성인자 (pleiotropic factor)로서 다양한 기전과 광범위한 역할에 대한 많은 연구가 수행되고 있다.
초파리 전사인자 coactivator 인 vestigial (vg) 유전자와 구조 유사성을 가진 Vestigial-like (VGLL) 유전자는 TEA 부위에 결합하는 전사인자 TEADs (TEA domain transcription factors)의 cofactor로 작용한다. 위암에서 VGLL 패밀리 단백질의 기능 및 기본 메커니즘이 연구되어 왔다. 위암 환자에서 VGLL3 과발현은 환자의 낮은 생존율과 관련이 있는 것으로 알려져 있다. YAP(Yes-associated protein) 과 경쟁적으로 TEAD 결합하는 VGLL4는 종양 억제제로서 보고되어 있다. VGLL1은 YAP 및 TAZ (transcription coactivator with a PDZ-binding motif)와 구조적으로 상동성이 높아 VGLL1-TEAD 복합체를 형성한다. 우리는 위암에서 발현량이 높고 PIK3CA 또는 PIK3CB이 과발현 되는 환자의 예후와 상관성이 있는 VGLL1을 위암의 악성화와 관련 있는 유전자로 보고하였다. 또한, PI3K-AKT-β-카테닌에 신호전달 기전을 통해 VGLL1의 전사가 조절된다는 것을 밝혔다. 또한, VGLL1-TEAD 복합체는 MMP9 (Matrix metallopeptidase 9)의 발현을 증가시켜 위암의 증식과 전이에 관여함을 확인하였다.
일련의 암세포의 증식 및 전이와 관련하여 VGLL1의 구체적인 작용에 대해서는 알려진 바가 없다. VGLL1과 관련하여, VGLL1을 바이오 마커로 사용하여 폐암에 걸린 환자의 생존기간을 예측하는 방법이나, VGLL1의 발현 증진과 관련된 암 세포 증식 또는 암 전이에 대해 일부 문헌에서 보고된 바가 있다.
이러한 배경 하에, 본 발명자들은 VGLL1의 펩타이드 단편이 MMP9의 발현을 억제할 뿐만 아니라 암세포의 증식을 억제함을 발견하여 암 치료제로써 본 발명을 완성하였다.
본 발명은 서열번호 1의 VGLL1의 펩타이드 서열의 84번째 세린(Serine)을 포함하고, 35개 이하의 아미노산 길이를 가지는 VGLL1 펩타이드 단편 또는 이의 변이체를 제공한다.

본 발명에 따른 조성물은 VGLL1의 펩타이드가 암세포 증식을 억제시키고 암세포의 침윤을 억제시킴으로써 암의 예방, 전이 억제 및 치료에 우수한 효과를 가진다. 본 발명에 따른 조성물은 직접적 치료 효과, 전이 억제 효과뿐만 아니라 항암 보조제로써의 작용을 모두 포함한다.
본 발명에서, "치료" 및 "치료하는"은 질병 또는 건강 관련 상태의 치료 이점을 얻을 목적의 대상체에의 치료제의 투여 또는 적용 또는 대상체에서의 절차 또는 방식의 수행을 지칭한다. 예방", "예방하는" 등은 질환 또는 병태의 발생 또는 재발 가능성을 예방하거나, 저해하거나 또는 감소시키기 위한 접근을 나타낸다. 또한 질환 또는 병태의 개시 또는 재발을 지연시키거나 또는 질환 또는 병태의 발생 또는 재발을 지연시키는 것을 지칭한다.
본 발명에서, "대상체" 및 "환자"는 인간 또는 비 인간, 예컨대 영장류, 포유류, 척추동물을 지칭한다. 특정 실시형태에서, 대상체는 인간이다.

본 발명에서, 항암은, 예를 들어, 암세포의 사멸을 촉진하거나, 암세포에서 아폽토시스를 유도하거나, 암세포의 성장 속도를 감소시키거나, 전이의 유병율 또는 수를 감소시키거나, 종양 크기를 감소시키거나, 종양 성장을 억제하거나, 종양 또는 암 세포로의 혈액 공급을 감소시키거나, 암세포 또는 종양에 대한 면역 반응을 촉진하거나, 암의 진행을 방지 또는 억제하거나, 암을 가진 대상체의 수명을 증가시킴으로써, 대상체의 암 세포/종양에 부정적인 영향을 주는 것을 의미한다.
본 발명에서 "약제학적으로 또는 약리학적으로 허용 가능한"이라는 어구는 적절한 경우, 동물, 예컨대 인간에 투여되는 경우, 부작용, 알레르기 반응 또는 다른 부적당한 반응을 일으키지 않는 분자성 물질 및 조성물을 지칭한다. 본 개시내용에 비추어, 당업자는 항체 또는 추가의 활성 구성요소를 포함하는 약제학적 조성물의 제조를 인식할 것이다. 또한, 동물 (예를 들어, 인간) 투여의 경우, 제조는 FDA 생물 표준 국에서 요구하는 대로, 멸균, 발열성, 일반 안전성 및 순도 표준을 충족해야 함이 이해될 것이다.
본 발명에 따른 펩타이드 단편 또는 이의 변이체는 앞서 언급한 바와 같이 세포 투과성 펩타이드를 더 포함함으로써 인체 내 세포 전달이 보다 더 용이할 수 있다.
본 발명에 사용하기 위한 상기 약학 조성물을 하나 이상의 생리학적으로 허용 가능한 담체 또는 부형제를 사용하여 표준 기법에 의해 제형화할 수 있다. 적합한 약학 담체는 본 발명 및 문헌[Remington: The Science and Practice of Pharmacy, 21st Ed., University of the Sciences in Philadelphia, Lippencott Williams & Wilkins (2005)]에 개시된다.
본 발명의 펩타이드를 임의의 적합한 경로, 예를 들어 흡입을 통해, 국소, 코, 경구, 비경구 또는 직장에 의해 투여하기 위해서 제형화할 수 있다. 따라서, 상기 언급된 약학 조성물의 투여를 피내, 피하, 정맥 내, 근육 내, 비내, 흡입에 의해, 대뇌 내, 기관 내, 동맥 내, 복강 내, 방광 내, 흉막 내, 관상동맥 내, 피하 또는 종양 내 주사에 의해, 주사기 또는 다른 장치를 사용하여 수행할 수 있다. 경피 투여가 또한, 흡입 또는 에어로졸 투여와 같이 고려된다. 정제 및 캡슐을 경구, 직장 또는 질내 투여할 수 있다.
상기 약학 조성물은 통상적으로 약학적으로 허용 가능한 담체, 바람직하게는 수성 담체 중에 용해된 펩타이드, 또는 펩타이드를 포함할 것이다. 펩타이드는 함께 또는 별개로 제공될 수 있다. 다양한 수성 담체, 예를 들어 완충된 염수 등을 사용할 수 있다. 이들 용액은 살균성이며 바람직하지 못한 물질이 일반적으로 없다. 이들 조성물을 통상적인, 널리 공지된 살균 기법에 의해 살균시킬 수 있다. 상기 조성물은 생리학적 조건에 근접하기 위해 요구되는 바와 같은 약학적으로 허용 가능한 보조 물질, 예를 들어 pH 조절 및 완충제, 독성 조절제 등, 예를 들어 나트륨 아세테이트, 염화 나트륨, 염화 칼륨, 염화 칼슘, 나트륨 락테이트 등을 함유할 수도 있다. 이들 제형 중 펩타이드의 농도는 광범위하게 다양할 수 있으며, 선택된 특정한 투여 모드 및 환자의 필요에 따라 주로 유체 부피, 점도, 체중 등을 근거로 선택될 것이다.
본 발명의 약학 조성물은 정맥 내 투여 또는 체강 내 투여를 포함하여 비경구 투여에 적합하다.
본 발명의 펩타이드를 주사, 예를 들어 일시 주사 또는 연속 주입에 의한 비경구 투여를 위해 제형화할 수 있다. 주사용 제형은 단위 투여형, 예를 들어 앰풀 또는 수회-용량 용기 중에, 첨가된 보존제와 함께 존재할 수 있다. 주사 가능한 조성물은 바람직하게는 수성 등장성 용액 또는 현탁액이며, 좌약을 바람직하게는 지방 유화액 또는 현탁액으로부터 제조한다. 상기 조성물을 살균하고/하거나 상기 조성물이 보조제, 예를 들어 보존제, 안정제, 습윤 또는 유화제, 용해 촉진제, 삼투압 조절용 염 및/또는 완충제를 함유할 수 있다. 한편으로, 상기 활성 성분은 사용 전에, 적합한 비히클, 예를 들어 멸균성의 발열원이 없는 물에 의해 조성되는 분말 형태로 존재할 수 있다. 또한, 상기 활성 성분은 또한 다른 치료학적으로 귀중한 물질을 함유할 수도 있다. 상기 조성물들을 통상적인 각각의 혼합, 과립화 또는 코팅 방법에 따라 제조하며, 이들은 약 0.1 내지 75%, 바람직하게는 약 1 내지 50%의 활성 성분을 함유한다.

본 발명은 약학 조성물을 암과 같은 질병 또는 이의 악성 상태를 예방하거나, 치료하거나 또는 억제하기 위한 치료 유효 용량으로 환자에게 투여한다. 상기 약학 조성물을 환자에게서 유효한 치료 또는 진단 반응을 이끌어내기에 충분한 양으로 상기 환자에게 투여한다. 유효한 치료 또는 진단 반응은 상기 병 또는 악성 상태의 증상 또는 합병증을 적어도 부분적으로 저지하거나 늦추는 반응이다. 이를 수행하기에 적합한 양을 "치료학적으로 유효한 양"으로서 정의한다.
투여되는 펩타이드의 투여량은 포유동물의 종, 체중, 연령, 개별적인 조건, 치료하려는 영역의 표면적 및 투여 형태에 따라 다르다. 상기 용량의 크기는 또한 특정 환자에게서 특정 화합물의 투여에 동반되는 임의의 부작용의 존재, 성질 및 정도에 의해 결정될 것이다.
약 50 내지 80㎏의 포유동물, 바람직하게 인간에게 투여하기 위한 단위 투여량은 펩타이드에 대하여 약 1mg/kg 내지 5mg/kg의 양을 함유할 수 있다.
전형적으로, 본 발명 화합물의 투여량은 목적하는 효과를 성취하기에 충분한 투여량이다. 최적의 투여 스케줄을 펩타이드의 측정, 환자의 신체 중의 누적으로부터 계산할 수 있다. 하루에, 일주일에, 한달에 또는 1년에 1회 이상 제공될 수 있다. 당해 분야의 숙련가들은 최적 투여량, 투여 방법 및 반복 비율을 쉽게 결정할 수 있다. 당해 분야의 숙련가는 당해 분야에 공지되고 본 발명에 개시된 확립된 프로토콜에 따라 인간에게 펩타이드의 투여를 위한 최적 투여를 결정할 수 있을 것이다. 그러나, 유효성분의 실제 투여량은 치료하고자 하는 질환, 질환의 중증도, 투여경로, 환자의 체중, 연령 및 성별 등의 여러 관련 인자에 비추어 결정되어야 하는 것으로 이해되어야 하며, 따라서, 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명은 또한, 암의 예방 또는 치료에 사용하기 위한 서열번호 1의 VGLL1의 펩타이드 서열의 84번째 세린(Serine)을 포함하고 35개 이하의 아미노산 길이를 가지는 VGLL1 펩타이드 단편 또는 이의 변이체를 제공한다.
본 발명은 또한, 암의 예방 또는 치료에 사용하기 위한 약제의 제조에 있어서 서열번호 1의 VGLL1의 펩타이드 서열의 84번째 세린(Serine)을 포함하고 35개 이하의 아미노산 길이를 가지는 VGLL1 펩타이드 단편 또는 이의 변이체의 용도를 제공한다.
본 발명은 또한, 치료학적으로 유효한 양의 서열번호 1의 VGLL1의 펩타이드 서열의 84번째 세린(Serine)을 포함하고 35개 이하의 아미노산 길이를 가지는 VGLL1 펩타이드 단편 또는 이의 변이체를 이를 필요로 하는 대상체에게 투여하는 것를 포함하는 암을 치료하는 방법을 제공한다.

이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
본 발명에서는 VGLL1의 펩타이드 단편 또는 이의 변이체가 암세포 증식을 억제시키고 암세포의 침윤을 억제시킴으로써 암의 예방 및 치료에 우수한 효과가 있음을 확인하여 본 발명을 완성하였다. 이하에서는 이에 대해 구체적으로 살핀다.
<실시예 1> TGF-β에 의한 MMP9의 발현 증가에 의한 암세포의 증식 및 전이 확인
실시예 1-1. TGF-β에 의한 MMP9의 발현 확인
위암세포주에서 TGF-β 처리에 따른 MMP9의 발현 변화를 확인하기 위하여, 인간 위암세포주 NUGC3에 TGF-β를 각 시간대별로 처리한 후 세포로부터 mRNA를 추출하고 상보적 DNA를 합성한 후, Matrix metalloproteinase 9(MMP9) 프라이머(표 4)를 사용하여 RT-PCR을 수행하였다.

도 1에서 확인할 수 있는 바와 같이, TGF-β에 의해 6시간 이후부터 MMP9의 발현이 증가하는 것을 확인하였다.
실시예 1-2. MMP9에 의한 암세포 증식 및 침윤 확인
MMP9의 발현이 위암 세포의 증식 및 전이에 미치는 영향을 확인하기 위하여, siMMP9을 처리하여 암세포의 증식 및 침윤에 미치는 영향을 확인하였다.
구체적으로, 위암 세포주 NUGC3를 6웰 플레이트(6-well plate)에 overnight으로 배양한 후, 20nM siSC, siMMP9(서열번호 22)을 리포펙타민 2000과 혼합하여 상기 세포주에 처리하였다. 48시간 후, 세포로부터 mRNA를 추출하고 상보적 DNA를 합성한 후, MMP9 프라이머를 사용하여 RT-PCR을 수행하였다(도 2A).

또한, MMP9의 세포 증식능을 확인하기 위하여, 위암 세포주 NUGC3를 96웰 플레이트(96-well plate)에 배양한 다음날 20nM siSC, siMMP9을 리포펙타민 2000과 혼합하여 상기 세포주에 처리하였다. 48시간 후, 1% 포르말린(formalin) 용액을 사용하여 세포를 고정시켰다. 그 후, 0.4% Sulforhodamine B용액으로 상기 세포를 1시간 동안 염색한 후 아세트산(acetic acid)으로 세척하여 건조시켰다. 그런 다음, 상기 세포를 10mM Tris 용액에 녹이고 엘라이자 플레이트 리더기(ELISA plate reader)를 사용하여 540nm에서 흡광도를 측정하였다 (도 2B).
다음으로, MMP9의 세포 침윤능을 확인하기 위하여, 24웰 플레이트(24-well plate)에 세포배양용 인설트(cell culture insert)를 고정하고 매트리젤(matrigel)을 20배 희석하여 100㎕ 첨가하고 37℃에서 1시간 이상 반응시켰다. 인설트 아래쪽 24웰 플레이트에 10% 혈청이 포함된 배지 700㎕를 첨가하였다. 위 siRNA로 MMP이 녹다운(knockdown)된 세포를 5 x 105 농도가 되도록 혈청이 없는 배지 300 ㎕에 맞추어 인설트에 첨가하였다. 48시간 동안 배양한 후, 인설트를 꺼내서 10% 포르말린 용액에 고정시켰다. 인설트 안쪽에 남아있는 세포를 면봉으로 제거하고 물로 세척하였다. 0.4% Sulforhodamine B 용액으로 세포를 1시간 동안 염색한 후, 아세트산(acetic acid)으로 세척하여 건조시켰다. 그런 다음, 광학현미경으로 사진을 찍고 인설트의 막(membrane)을 잘라낸 후, 10mM Tris 용액에 녹여 엘라이자 플레이트 리더기(ELISA plate reader)를 사용하여 540nm에서 흡광도를 측정하였다(도 2C).
상기 각각의 실험결과를 도 2의 A 내지 C에 나타내었다.
도 2의 A는 siRNA(siMMP9) 처리에 따라, MMP9의 발현이 감소된 결과를 나타낸다. 또한, 도 2의 B는 siRNA(siMMP9) 처리에 따라 MMP9의 발현이 감소됨으로써 세포 생존율이 크게 감소하는 결과를 나타낸다. 또한, 도 2의 C는 siRNA(siMMP9) 처리에 따라 세포의 침윤 특성이 크게 감소한 결과를 나타낸다.
위 결과들에서 확인할 수 있는 바와 같이, siMMP9에 의해 MMP9의 발현이 억제되고, 이에 따라 위암세포주 NUGC3에서 증식 및 침윤능이 크게 감소되는 결과가 확안되었다.
<실시예 2> MMP9의 프로모터 활성 확인 및 이의 mRNA 발현에 VGLL1과 TEAD4의 작용 확인
실시예 2.1 TGF-β에 의한 MMP9의 프로모터 활성 확인 및 VGLL1과 TEAD4의 조절능 확인
NUGC3 인간 위암 세포주에서 MMP9의 프로모터 활성에 VGLL1과 TEAD4가 관여하는지 확인하기 위하여, TGF-β를 처리한 후 MMP9 프로모터 루시퍼레이즈 (luciferase) 활성을 확인하였다.
구체적으로, 위암 세포주 NUGC3를 48웰 플레이트(48-well plate)에 2 x 104 세포/웰 농도로 세포를 배양하였다. 그 다음날, MMP9 유전자가 삽입된 pGL4.17-luciferase 벡터를 리포펙타민 2000을 이용하여 세포에 형질 주입시켰다. 24시간 후, 20nM siSC(서열번호 23: 5′-CCUACGCCACCAAUUUCGUdTdT-3′) siVGLL1(서열번호 24 : 5′-AGCCUAUAAAGACGGAAUGGAAUdTdT-3′), siTEAD4(서열번호 25 : 5′-GACACUACUCUUACCGCAUdTdT-3′), siYAP(서열번호 26 : 5′-CCACCA AGCUAGAUAAAGAAAdTdT-3′)를 리포펙타민 2000과 혼합하여 상기 세포주에 처리하였다. 다음날 TGF-β를 18시간 처리한 후 세포 용해 완충액(passive lysis buffer)을 이용하여 상기 세포를 파쇄하였다. 그런 다음, 상기 세포를 원심분리한 후, 상층액 수득하고, 수득된 상기 상측액을 기질(제품명 dual-luciferase assay kit)과 함께 반응시키면 루시퍼레이즈 효소와 기질 반응으로 빛을 나타냄으로, 이때, 빅터(victor X Light)를 이용하여 상기 세포의 발광 정도를 확인하였다.
그 결과를 도 3에 나타내었다. 도 3에서 확인할 수 있는 바와 같이, siVGLL1과 siTEAD4로 형질 주입시킨 세포는 대조군(siSC)에 비해 TGF-β에 의해 증가된 MMP9 프로모터 활성이 감소되었다. 이러한 결과는 VGLL1과 TEAD4가 MMP9의 프로모터 활성을 조절하는 것을 보여준다.
실시예 2-2. TGF-β에 의한 MMP9의 mRNA 발현에 VGLL1과 TEAD4의 작용 확인
NUGC3 인간 위암 세포주에서 MMP9의 mRNA 발현에 VGLL1과 TEAD4가 관여하는지 확인하기 위하여, TGF-β를 처리한 후 MMP9 mRNA 발현을 RT-PCR로 확인하였다.
구체적으로, 위암 세포주 NUGC3를 6웰 플레이트(6-well plate)에 배양한 다음날 20nM siSC, siVGLL1, siTEAD4, siYAP을 리포펙타민 2000과 혼합하여 상기 세포주에 처리하였다. 다음날 TGF-β를 18시간 처리한 후 세포로부터 mRNA를 추출하고 상보적 DNA를 합성한 후, RT-PCR을 수행하였다.
그 결과를 도 4에 나타내었다. 도 4에 나타낸 바와 같이, siVGLL1과 siTEAD4로 형질 주입시킨 세포는 대조군(siSC)에 비해 TGF-β에 의해 증가된 MMP9 mRNA 발현을 감소시키는 것으로 확인되었다.
<실시예 3> TGF-β에 의한 VGLL1의 세린(Serine) 84번의 인산화 확인
TGF-β에 의해 VGLL1이 인산화 되는지 확인하기 위해 VGLL1 아미노산 서열 50, 84, 124번째 세린(Serine)을 알라닌 (Alanine)으로 돌연변이 시킨 후, VGLL1의 인산화여부를 확인하였다.
구체적으로, VGLL1의 특정 아미노산을 돌연변이 시키기 위해 pcDNA3-HA-VGLL1- 벡터에서 site-direct mutagenesis 제품을 사용하여 50, 84, 124번째 세린을 알라닌으로 바꾸었다.
또한 위암 세포주 NUGC3를 6웰 플레이트(6-well plate)에 배양한 다음날 pcDNA3-HA(EV), pcDNA3-HA-VGLL1(WT), pcDNA3-HA-VGLL1-S50A (S50A 변이), pcDNA3-HA-VGLL1-S84A(S84A 변이), pcDNA3-HA-VGLL1-S124A(S124A 변이)를 터보펙트와 혼합하여 세포주에 형질전환시켰다. 48시간 후 TGF-β를 15분간 처리하고 상기 세포로부터 리파 버퍼(RIPA buffer)를 이용하여 단백질을 추출하였다. 추출된 단백질은 HA 항체가 결합된 아가로즈를 이용하여 면역침전(Immunoprecipitation)시키고 전기영동으로 분리한 후, 이동된 폴리비닐리덴 플루오라이드 막(polyvinylidene fluoride membrane)에서 항체를 이용하여 유전자들의 단백질 발현을 확인하였다.
그 결과를 도 5에 나타내었다. 도 5에서 확인할 수 있는 바와 같이, TGF-β에 의한 VGLL1의 인산화는 WT, S50A, 및 S124A에서 세린/스레오닌 인산화(pSer/Thr) 항체에 의해 검출되었다. 반면 S84A 돌연변이로 형질전환시킨 세포주에서는 VGLL1의 인산화를 확인할 수 없었다. 이는 VGLL1의 세린 84번 위치가 TGF-β에 의해 인산화되는 부위임을 보여주는 결과이다.
<실시예 4> 인산화된 VGLL1과 TEAD4 결합 확인
VGLL1이 전사인자 TEAD4와 결합하는지 확인하기 위해 면역침전법을 수행하였고, VGLL1의 인산화가 중요하게 작용하는지 확인하기 위해 <실시예 3>에서 사용한 벡터를 이용하였다.
구체적으로, GFP-TEAD4 벡터와 pcDNA3-HA(EV), pcDNA3-HA-VGLL1(WT), pcDNA3-HA-VGLL1-S50A (S50A 변이), pcDNA3-HA-VGLL1-S84A(S84A 변이), pcDNA3-HA-VGLL1-S124A(S124A 변이)를 터보펙트와 혼합하여 24시간 배양된 세포주에 형질전환 시켰다. 48시간 후 상기 세포로부터 리파 버퍼를 이용하여 단백질을 추출하고 HA 항체가 결합된 아가로즈를 이용하여 면역침전시키고 전기영동으로 분리한 후, 이동된 폴리비닐리덴 플루오라이드 막(polyvinylidene fluoride membrane)에서 항체를 이용하여 유전자들의 단백질 발현을 확인하였다.
그 결과를 도 6에 나타내었다. 도 6에서 확인할 수 있는 바와 같이, VGLL1과 TEAD4의 결합은 WT, S50A, S124A에서 지에프피(GFP) 항체에 의해 검출되었다. 반면 S84A 돌연변이로 형질전환시킨 세포주에서는 TEAD4의 결합을 확인할 수 없었다. 이는 VGLL1의 세린 84번 위치가 TEAD4와의 결합에 중요한 부위에 해당함을 보여주는 결과이다.
<실시예 5> TGF-β에 의한 MMP9 발현 증가에 VGLL1의 인산화의 작용 확인
NUGC3 인간 위암 세포주에서 MMP9의 단백질 발현에 VGLL1의 인산화가 관여하는지 확인하기 위하여, 상기 <실시예 3>에서 사용한 VGLL1 벡터들을 사용하여 MMP9의 단백질 발현을 Western blot으로 확인하였다.
구체적으로, 위암 세포주 NUGC3를 6웰 플레이트(6-well plate)에 배양한 다음날 pcDNA3-HA(EV), pcDNA3-HA-VGLL1(WT), pcDNA3-HA-VGLL1-S50A (S50A), pcDNA3-HA-VGLL1-S84A(S84A), pcDNA3-HA-VGLL1-S124A(S124A)를 터보펙트와 혼합하여 세포주에 형질전환시켰다. 24시간 후 TGF-β를 18시간 처리하고 상기 세포로부터 리파 버퍼를 이용하여 단백질을 추출하였다. 추출된 단백질은 전기영동으로 분리한 후, 이동된 폴리비닐리덴 플루오라이드 막(polyvinylidene fluoride membrane)에서 MMP9, HA, GAPDH 항체를 이용하여 단백질 발현을 확인하였다.
그 결과를 도 7에 나타내었다. 도 7에서 확인할 수 있는 바와 같이, TGF-β에 의한 MMP9 단백질의 발현은 VGLL1 84번 세린을 알라닌으로 치환시킨 세포주에서 감소되는 것으로 확인되었다. 이는 TGF-β에 의한 VGLL1의 인산화가 MMP9 발현에 중요하다는 것을 나타내며, VGLL1 펩타이드의 84번째 위치의 세린이 중요인자임을 나타낸다.
<실시예 6> S84 VGLL1 펩타이드 단편의 MMP9 발현 억제와 암세포 증식 및 침윤 저해 효과 확인
실시예 6-1. S84 VGLL1 펩타이드 단편은 TGF-β에 의한 MMP9 발현을 감소시킴
인간 위암세포주에서 VGLL1과 TEAD4의 결합을 저해시키는 펩타이드 단편을 처리하는 경우, MMP9의 발현을 저해시킬 수 있는지 확인하기 위해 하기 표 6의 서열을 가지는 펩타이드 단편을 제작하여, 이의 MMP9 발현 억제 효과를 확인하였다.

상기 표 6에서 펩타이드 단편 서열은 VGLL1의 S84를 포함하는 VGLL1의 77 내지 91번째 서열을 제공한다. 구체적으로, 서열번호 27의 서열은 fluorescein moiety (FITC) 와 세포 내 도입에 필요한 HIV TAT transduction domain sequence인 YGRKKRRQRRR를 포함하고 이에 VGLL1의 77 내지 91번의 서열이 연결된 서열이다. 서열번호 28은 VGLL1의 S84을 S84A로 치환한 서열을 나타낸다. 서열번호 12는 Control 서열로 HIV TAT transduction domain sequence를 나타낸다.
상기 합성 펩타이드 단편을 이용하여 실험을 수행하였다. 위암 세포주 NUGC3를 6웰 플레이트(6-well plate)에 배양한 다음날 혈청이 없는 세포배양에서 24시간 배양시켰다. TGF-β와 컨트롤 펩타이드 단편(서열번호 12), VGLL1의 84번 세린을 포함하는 펩타이드 단편(서열번호 27), VGLL1의 84번 알라닌을 포함하는 변이 펩타이드 단편(서열번호 28)을 18시간 처리하였다. 상기 세포로부터 리파 버퍼를 이용하여 단백질을 추출하고 전기영동으로 분리한 후, 이동된 폴리비닐리덴 플루오라이드 막(polyvinylidene fluoride membrane)에서 항체를 이용하여 유전자들의 단백질 발현을 확인하였다.
그 결과를 도 8에 나타내었다. 도 8에서 확인할 수 있는 바와 같이, TGF-β에 증가된MMP9 발현은 VGLL1의 84번 세린을 포함하는 펩타이드 단편 처리에 의해 감소되었다. 반면 VGLL1의 84번 알라닌을 포함하는 변이 펩타이드 단편 서열의 경우 펩타이드 단편의 처리에 의해 어떠한 영향도 받지 않았다. 이러한 결과로부터, 본 발명에 따른 VGLL1의 84번 세린을 포함하는 펩타이드 단편 서열이 암의 치료에 우수한 효과를 나타낼 수 있음을 확인하였다.
실시예 6-2. S84 VGLL1 펩타이드 단편에 의한 암세포 증식 억제 효과 확인
위암세포에서 VGLL1과 TEAD4의 결합을 억제시켜 MMP9발현을 감소시키면 암세포의 증식에도 영향을 줄 수 있기 때문에, S84를 포함하는 VGLL1 펩타이드 단편에 의해 세포 증식이 억제되는지 여부를 확인하였다.
구체적으로, 위암 세포주 NUGC3를 96웰 플레이트(96-well plate)에 배양한 다음날 펩타이드 단편을 처리하고, 실시간 세포 분석 시스템(제품명 Incucyte)을 사용하여 세포 증식을 분석하였다.
그 결과를 도 9에 나타내었다. 도 9에서 확인할 수 있는 바와 같이, S84 VGLL1 펩타이드 단편은 암세포 증식을 억제시키는 효과를 나타내었다. 반면 컨트롤 펩타이드 단편이나 S84의 위치가 변이된 84번 위치에 알라닌을 포함하는 변이 VGLL1 펩타이드 단편은 암세포 증식 억제에 어떠한 영향도 미치지 못하였다.
상기 결과를 통해 본 발명에 따른 S84 VGLL1 펩타이드 단편 서열이 암세포 증식 억제에 우수한 효과를 나타냄을 확인하였다.
<실시예 7> S84 VGLL1 펩타이드 단편에 의한 암세포 침윤 억제 확인
VGLL1 펩타이드 단편에 의한 위암세포 침윤 실험을 실시예 1-2와 유사한 방법을 수행하였다.
위암 세포주 NUGC3를 펩타이드 단편과 함께 인서트(insert)에 넣고 48시간 세포 배양 후, 인서트를 10% 포르말린 용액에 고정하고 아래쪽으로 이동된 세포를 0.4% Sulforhodamine B 용액으로 염색하여 확인하였다.
그 결과를 도 10에 나타내었다. 도 10에서 확인할 수 있는 바와 같이, S84를 포함하는 VGLL1 펩타이드 단편 서열은 암세포 침윤을 억제시키는 효과를 나타내었는 반면, 해당 위치가 변이된 서열의 경우 그러한 효과를 나타내지 못하였다.
<실시예 8> S84 VGLL1 펩타이드 단편 변이체의 암세포 증식 억제 효과 확인
VGLL1 S84 펩타이드 서열 중 주요 아미노산의 돌연변이에 의한 세포 증식 억제 효과를 확인하기 위해 표 7과 같이 펩타이드를 제작하였다.

그 결과를 도 11에 나타내었다. 도 11의 도면 A는 실시간 세포 증식 그래프이며, 도면 B는 4일 후 세포 증식을 컨트롤 펩타이드 100% 기준으로 그린 그래프이다.
도면에서 확인할 수 있는 바와 같이, S84 VGLL1 펩타이드 뿐만 아니라 S84-S, P85A, T87A 등을 처리한 경우 암세포 증식을 억제시키는데 우수한 효과를 나타내었다.
이러한 결과를 바탕으로, 세포 증식 억제 효과가 나타나는 펩타이드의 서열로 S84 서열이 가장 중요한 것을 확인하였다.
위 결과로부터 본 발명에 따른 VGLL1의 S84번의 세린을 포함하는 펩타이드 단편 서열이 암세포의 증식 억제 및 침윤 억제를 통해 암의 치료에 우수한 효과를 나타내는 것을 확인하였다.
<실시예 9> S84 VGLL1 펩타이드 단편의 다양한 암세포 증식 억제 효과 확인
S84 VGLL1 펩타이드 단편의 다양한 암세포 증식 억제 효과를 확인하기 위하여, 서열번호 27의 VGLL1 S84 펩타이드 서열을 다양한 암 세포주에 처리하였다.

Claims (9)
- 서열번호 1의 VGLL1의 펩타이드 서열의 84번째 세린(Serine)을 포함하고, 35개 이하의 아미노산 길이를 가지는 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 1 항에 있어서, 상기 VGLL1 펩타이드 단편 또는 이의 변이체는 6 내지 25개의 아미노산 서열을 가지는 것인 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 1 항에 있어서, 상기 VGLL1 펩타이드 단편 또는 이의 변이체는 서열번호 1의 52번 내지 115번째 사이의 아미노산 서열에서 84번째 세린 서열을 포함하는 것인 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 1 항에 있어서, 상기 VGLL1 펩타이드 단편 또는 이의 변이체는 84번째 세린 및 이에 인접한 6개 내지 20개 아미노산으로 이루어진 것인 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 1 항에 있어서, 상기 VGLL1 펩타이드 단편 또는 이의 변이체는 서열번호 2 내지 9로 이루어진 군으로부터 선택되는 어느 하나인 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 1 항에 있어서, 세포 투과성 펩타이드를 더 포함하는 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 6 항에 있어서, 상기 세포 투과성 펩타이드는 서열번호 12 내지 19로 이루어진 군으로부터 선택되는 어느 하나인 VGLL1 펩타이드 단편 또는 이의 변이체.
- 제 1 항 내지 제 7 항 중 어느 한 항에 따른 VGLL1 펩타이드 단편 또는 이의 변이체를 포함하는 암의 예방 또는 치료용 약제학적 조성물.
- 제 8 항에 있어서, 상기 암은 위암, 간암, 대장암, 폐암, 유방암, 췌장암, 두경부 편평암 세포 암종, 식도암, 난소암, 자궁경부암, 자궁내막암, 중피종, 흑색종, 육종, 골육종, 지방육종, 갑상선암, 유건종 및 혈액암으로 이루어진 군으로부터 선택되는 어느 하나인 암의 예방 또는 치료용 약제학적 조성물.
