인간 중추 신경계의 복잡한 발달을 이해하고 환자의 신경계 및 정신 장애의 메커니즘을 규명함에 있어서의 진보는 기능적인 인간 뇌 조직에 대한 제한된 접근에 의해 크게 제한되어왔다. 설치류 및 기타 포유류에 대한 연구는 신경 발달의 기본 원리에 대한 중요한 통찰력을 제공했지만 영장류의 대뇌의 대규모 확장을 담당하는 세포 및 분자 과정, 그리고 이의 많은 인간 특이적 특징들에 대해서는 거의 알지 못한다. 최근에, 세포 재프로그래밍의 도입으로 패러다임 전환이 구현되었으며, 이 과정에서 최종적으로 분화된 체세포를 인간 유도 만능 줄기 세포 (hiPSC)라 명명하는 만능성 줄기 세포로 전환시킬 수 있다. 이들 hiPSC는 임의의 개체로부터 생성 될 수 있고, 중요하게는 신경 세포를 포함하여 모든 생식 층 유도체로 시험관 내에서 분화되도록 지시될 수 있다.

희소돌기아교세포는 뇌 기능에서 결정적인 역할을 하는 중추 신경계의 신경아교 세포이다. 희소돌기아교세포는 절연층으로 뉴런의 축삭을 감싸는 과정을 확장하여 뉴런들 간 신호 전달을 더 빠르게 할 수 있다. 희소돌기아교세포는 또한 영양 인자를 뉴런에 공급하고 세포외 환경을 완충시켜 지원하는 역할을 한다. 수초화 소실은 뉴런의 사멸 및 신경 기능 장애로 이어질 수 있다. 결과적으로, 많은 인간 질병은 다발성 경화증 또는 백색 물질 소실 질환 (white vanishing matter disease)과 같은 수초화의 소실 또는 감소와 관련이 있다.
건강과 질병의 수초화에 대한 연구는 적절한 모델의 이용가능성에 의해 제한된다. 설치류는 일반적으로 인간 생물학 양상들을 연구하는 대용체로 사용되지만; 그러나, 수초화는 설치류 뇌보다 인간에서 훨씬 더 광범위하다. 또한, 인간 환자의 건강한 뇌 샘플들의 제한된 이용가능성은 인간 뇌에서 수초화를 광범위하게 연구할 수 있는 가능성을 배제한다. 최근에, 시험관내에서 수초화 연구를 위해 인간 배아 및 유도된 만능성 줄기 세포 (hESC, hiPSC)로부터 희소돌기아교세포 전구 세포 (OPC) 및 희소돌기아교세포를 생성하려는 시도가 있었다.
hESC 및 hiPSC 유래 수초화 모델들은 정상 및 질병 상태하에서 희소돌기아교세포 개발 및 수초화 과정을 연구하는데 필수적일 뿐만 아니라, 약제학적 약물 스크리닝에도 적용될 수 있다. 이들 모델은 특정 질병 상태와 관련된 특정 이상을 교정하는 화합물을 스크리닝하고 인간 노출 전에 새로운 치료 화합물 및 화학 물질의 독성을 테스트하기 위해 사용될 수 있다. 특히, 신경독성 분야에서, 뉴런 또는 신경아교 기능의 손상을 평가할 수 있는 분석법은 인간 세포에 대해서는 여전히 부족하다.
따라서, 뉴런의 수초화를 포함하여, 고도로 기능적인 인간 조직을 간략하게 개요하는 시험관내 스크리닝 플랫폼의 개발이 가장 중요하다.

실험
실시예 1
인간 희소돌기아교세포를 생성하고 시험관내 수초화 과정을 모델하기 위한 선행 방법들이 2차원 (2D) 배양물에서 실시된 바 있다. 이들 프로토콜에서, 수초화는 합성 나노필러를 둘러싼 희소돌기아교세포 감싸기 (wrapping) 과정들로서 연구된다. 수초화는 3차원 과정으로서 인간 뇌에서 일어나기 때문에 이들 방법들로는 수초화를 연구할 수 없다. 대안적으로, 인간 줄기 세포-유래된 희소돌기아교세포를 설치류에 이식하였으나, 어느 정도로 생쥐 환경이 인간 희소돌기아교세포 생물학 및 수초화에 영향을 미치는지에 대해서는 공지되어 있지 않으며, 그리고 이들 방법들은 힘이 들고 높은 처리량의 스크리닝을 할 수 없게 한다. 아래에 기재된 방법은 인간 hiPSC 또는 hESC에서만 유래된 3차원 (3D) 배양물에서 시험관내 수초화 희소돌기아교세포를 최초로 생성하여, 시험관내 건강 및 질환 병태에서 인간 수초화를 연구 할 수 있게 한다.

Claims (19)
- 다음을 포함하는, 시험관내 인간 희소돌기아교세포 제조 방법:
만능성 줄기 세포 현탁액 배양물에서 신경 운명을 유도하여, 신경 전구 세포의 구형체를 제공하는 단계;
구형체에서 신경 전구 세포를 분화시켜, 희소돌기아교세포를 포함하는 인간 희소돌기-구형체 (hOS)를 분화시키는 단계;
hOS 구형체를 장기화된 시기 동안 신경 배지에서 유지시켜 수초화 희소돌기아교세포, 뉴런 및 성상세포를 포함하는 배양물을 유도하는 단계. - 청구항 1에 있어서, 상기 세포는 수초 장애와 관련된 최소한 하나의 대립유전자를 포함함을 특징으로 하는 방법.
- 청구항 1-2 중 어느 하나에 있어서, 상기 만능성 줄기 세포는 유도된 만능성 줄기 세포임을 특징으로 하는 방법.
- 청구항 1-3 중 어느 한 항에 있어서, 상기 만능성 줄기 세포 현탁액 배양물은 유효 용량의 BMP 저해제 및 TGFβ 저해제를 포함하는 배지에서 만능성 줄기 세포의 무손상 콜로니들을 배양함으로써 신경 운명으로 유도됨을 특징으로 하는 방법.
- 청구항 1-4 중 어느 한 항에 있어서, 상기 배지는 만능성 줄기 세포를 신경 운명으로 유도함에 효과적인 용량의 도르소몰핀 및 SB-431542를 포함함을 특징으로 하는 방법.
- 청구항 5에 있어서, 상기 현탁액 배양물은 지지세포층이 없음을 특징으로 하는 방법.
- 청구항 6에 있어서, 상기 배지는 유효량의 wnt 저해제를 추가로 포함함을 특징으로 하는 방법.
- 청구항 7에 있어서, 세포들은 신경 전구 세포를 유지함에 효과적인 용량의 FGF2 및 EGF를 포함하는 신경 배지에서의 배양에 의해 신경 전구체로 분화됨을 특징으로 하는 방법.
- 청구항 8에 있어서, 소닉 헤지호그 경로 효현제 및 레티노산으로 또는 소닉 헤지호그 경로 효현제 및 Wnt 저해제로 구형체를 패턴화하는 단계를 추가로 포함함을 특징으로 하는 방법.
- 청구항 9에 있어서, FGF2 및 EGF가 없고, PDGF-AA, IGF-1, HGF, BDNF, NT3, 인슐린, cAMP, T3, 및 비오틴의 유효량을 포함하는 신경 배지에서의 배양에 의해 신경 전구체를 희소돌기아교세포로 분화시키는 단계를 추가로 포함함을 특징으로 하는 방법.
- 청구항 10에 있어서, 성장 인자들은 없으나 아스코르브산, 인슐린, cAMP, T3, 및 비오틴을 포함하는 신경 배지에서 장기 시기 동안 희소돌기-구형체를 유지시키는 단계를 추가로 포함하여 제조됨을 특징으로 하는 방법.
- 청구항 11에 있어서, 유세포 분석, 자기 면역선택법, 또는 면역패닝법에 의해 피질 구로부터 희소돌기아교세포를 단리하는 단계를 추가로 포함함을 특징으로 하는 방법.
- 청구항 12에 있어서, 단리된 희소돌기아교세포는 후보 물질 또는 관심 치료에 노출됨을 특징으로 하는 방법.
- 청구항 13에 있어서, 상기 희소돌기아교세포는 뉴런 또는 뉴런 전구 세포와 조합됨을 특징으로 하는 방법.
- 청구항 9 또는 10의 방법에 의해 수득된 희소돌기아교세포의 모집단.
- 다음을 포함하는, 인간 희소돌기아교세포에 대한 후보 물질의 효과를 결정하는 방법: 후보 물질을 청구항 9 또는 10의 방법에 따라 유도된 인간 만능성 줄기 세포 (hiPSC)로부터 분화된 성상세포 중 하나 또는 이들의 패널과 접촉시키는 단계, 이 때 희소돌기아교세포는 신경 질환과 연관된 돌연변이를 인코드하는 최소한 하나의 대립유전자를 선택적으로 포함하고; 및 형태학적, 유전적 또는 기능적 매개변수들에 대한 상기 물질의 효과를 결정하는 단계.
- 청구항 16에 있어서, 상기 후보 물질은 면역 효과기 세포 또는 면역 조절 물질임을 특징으로 하는 방법.
- 청구항 16에 있어서, 희소돌기아교세포의 패널은 상이한 유전자형을 가지는 최소한 2개의 성상세포를 포함함을 특징으로 하는 방법.
- 청구항 18에 있어서, 희소돌기아교세포의 패널은 상이한 환경 조건하의 희소돌기아교세포를 포함함을 특징으로 하는 방법.
