Abstract
본 발명은 각각 서열번호 1 내지 3의 아미노산 서열을 갖는 중쇄 상보성 결정 영역 1 내지 3, 및 각각 서열번호 4 내지 6의 아미노산 서열을 갖는 경쇄 상보성 결정 영역 1 내지 3을 갖는 중쇄 및 경쇄를 포함하는 항체 또는 이의 항원-결합 단편에 관한 것이다. 또한, 본 발명은 상기 항체 또는 이의 항원-결합 단편을 포함하는 항암 치료제에 관한 것이다.
Description
본원은 본원에 개시내용 전체가 참고로 포함된 미국 가출원 제62/307,690호(발명의 명칭: PODOCALYXIN AND TRA-RELATED ANTIBODY, METHODS FOR PREPARATION AND USES FOR TREATMENT OF AGGRESSIVE AND/OR METASTATIC CANCER; 출원일: 2016년 3월 14일)를 우선권 주장한다.
본 발명은 다능성- 및 배아 줄기 세포-특이적 항체에 관한 것이다. 또한, 본 발명은 포도칼릭신(podocalyxin) 단백질의 미발달 형태에 반응성이고 이에 대한 결합 활성을 갖는 항체에 관한 것이다. 또한, 본 발명은 암 세포를 퇴행시키고 감소시키고 제거하는 활성 성분으로서 항체 또는 이의 항체 단편을 포함하는 암 치료제에 관한 것이다.

본 발명의 단클론 항체(Bstrongomab 또는 CM-918로 지칭됨)를 표준 마우스 하이브리도마 기술을 사용하여 생산하였다. 마우스를 멸균 PBS에 현탁된 갓 채취한 NCCIT 세포로 복강내로 주사하였다. 각각의 마우스는 주사 당 10e6개의 세포를 제공받았다. 2주 간격의 4회의 주사 후에, 면역화된 마우스의 혈청을 ELISA 및 웨스턴 블롯(Western Blot) 분석으로 시험하여 양성 역가를 갖는 마우스를 확인하였다. 이어서, 가장 높은 역가를 마우스는 비장으로부터 B 세포의 채취 또는 NS0 또는 P3X63Ag8.653 마우스 골수종 세포에 대한 융합 2 내지 7일 전에 융합전 부스트 면역화를 제공받았다.

CM-918 항체를 10% 초저 IgG FBS를 함유하는 하이브리도마 배지에서의 고갈을 위해 배양된 단클론 하이브리도마의 상청액으로부터 정제하였다. 상청액을 원심분리한 후에, 45 um 필터에 통과시켜 세포 및 세포 잔해를 제거하였다. CM-918 항체를 사용한 다능성 배아 암종 암 세포 NCCIT(ATCC # CRL2073) 및 인간 배아 줄기 세포(ESI-017)의 막 단백질 샘플의 웨스턴 블롯은 둘 다의 단백질 막 프렙에서 200 내지 240 kDa 분자량에서 단일 확산 밴드를 나타냈으나, 반응성은 TRA-포도칼릭신을 발현하지 않는 비-다능성 세포주 PC3(ATCC # CRL1435) 또는 A172(ATCC # CRL1620)에서 검출되지 않았다. 동일한 블롯 결과가 동일한 웨스턴 블롯에 대해 상업적으로 입수가능한 IgM 항체, 항-TRA-1-60(abcam 16288)을 사용하여 수득되었다. 결과는 CM-918 항체 및 항-TRA-1-60 항체가 유사한 에피토프에 결합함을 확인하였다. 배아 줄기 세포(ESL-017) 및 다능성 NCCIT 배아 암종 세포를 알렉사(Alexa) 647(시그마)과 접합된 CM-918 항체(0.5 ug/ml), 및 항-TRA-1-60 항체(0.5 ug/ml) 및 항-IgM-알렉사 F488 항체(밀리포어)(0.5 ug/ml)로 염색시켰다. 적색 채널에 의한 형광 현미경 이미지는, CM-918 항체가 배아 줄기 세포 및 다능성 배아 암종 세포의 원형질막에 강하게 결합하나 TRA-포도칼릭신을 발현하지 않는 대조군 세포주 PC3(ATCC # CRL1435) 또는 A172(ATCC # CRL1620)에는 결합하지 않음을 나타냈다. 적색 및 녹색 채널로부터의 겹쳐 놓은 형광 이미지는, 줄기 세포의 표면 상에 동일한 에피토프에 대한 둘 다의 항체의 결합을 보여주는 CM-918 및 항-TRA-1-60 항체의 동일한 염색 오버랩을 나타냈다.

4 - 정상 인간 조직 및 인간 암에 대한 CM-918 항체의 면역조직화학(IHC) 염색
CM-918에 의한 정상 조직 및 암의 면역조직화학 염색을 하기에 기재된 바와 같이 수행하였다. 0 내지 3의 등급 매기기 시스템을 사용하였다: 0: 염색이 관찰되지 않음, 1: 국소 염색(< 5%); 2: 중간 염색(< 50%); 3: 강한 염색(> 50%). 염색된 조직 슬라이드를 병리학자가 점수 매겼다. 2 또는 3의 염색 점수가 하기 암 조직에서 관찰되었다: 췌장암: 38개의 샘플 중 22개; 전립선암: 41개의 샘플 중 19개; 유방암: 145개의 샘플 중 45개; 난소암: 30개의 샘플 중 18개; 위암: 8개의 샘플 중 8개; 결장암: 30개의 샘플 중 15개; 폐암: 66개의 샘플 중 32개. 정상 인간 조직에서의 발현은 0의 발현(골수를 포함한 거의 모든 정상 조직에서)에서 적은 국소 발현(췌장, 식도 및 결장의 일부 경우)으로 매우 제한적이었다. 신장은 세관에서 약 1%의 CM-918을 사용한 모든 샘플에서 양의 점수를 가졌다. 2의 점수를 갖는 유일한 정상 조직은 부갑상선이었고, 이는 9개의 샘플 중 2개가 2-염색 점수를 가졌다. 결과는 CM-918이 많은 다양한 암에서 막 염색을 수행하고 정상 조직에서 매우 제한된 염색을 수행함을 보여준다.
모든 조직을 10% 완충된 포르말린에서 24시간 동안 고정시키고 5 μm로 절단하고 양으로 하전된 슬라이드에 두었다. 염색 전에, 슬라이드를 60도에서 밤새 건조하였다. 자일렌(피셔 사이언티픽(Fisher Scientific), 카탈로그 번호 X3P-1GAL)에서 탈파라핀화시킨 후에, 섹션을 100% 에탄올(에키-켐(Eki-Chem), 카탈로그 번호 4085-1GL), 95% 에탄올 및 70% 에탄올(에키-켐, 카탈로그 번호 4089-1GL)을 통해 재수화시켰다. 이어서, 섹션을 1X 디바 디클로커(Diva Decloaker)(바이오케어 메디컬(Biocare Medical), 카탈로그 번호 DV2004LX)에 담그고, 디클로킹(Decloaking) 챔버(바이오케어 메디컬, 모델: NxGen)를 사용하여 쿠킹하였다. 이후, 섹션을 블록살(BLOXALL)(벡터 랩스(Vector labs), 카탈로그 번호 SP-6000) 및 파워비전 유니버셜(PowerVision Universal) IHC 블로킹/딜루션(레이카(Leica), 카탈로그 번호 BD09-15)에 의해 차단하였다. 일차 항체를 PBS에서 2 ug/ml로 각각의 슬라이드에 적용하고 실온에서 1시간 동안 방치하였다. 5 - CM-918-항체-약물-접합체의 세포

CM-919-vc-MMAE 항체 약물 접합체를 생산하였다: PBS 중의 CM-918의 용액을 37℃에서 TCEP로 처리하여 쇄간 다이설파이드 결합을 환원시켰다. MC-vc-PAB-MMAE의 용액(말레이미드도카프로일-발린-시트룰린-p-아미노벤질옥시카본일-모노메틸 아우리스타틴 E)을 반응 혼합물에 첨가하여 MC-vc-PAB-MMAE의 말레이미드 잔기를 CM-918에 연결시켰다. 생성된 항체 약물 접합체(ADC)를 세파덱스(Sephadex) G50 컬럼 상에서 탈염시켜 잔여 미반응 독소를 제거하고 PBS(pH 7.2)로 완충제 교환하였다. 약물 항체 비(DAR)를 248 및 280 nm에서의 UV 흡광도의 비로부터 결정하였다. HIC(소수성 상호작용 크로마토그래피) 및 SEC(크기 배제 크로마토그래피) 분석을 수행하여 DAR 종 분포를 결정하였다.
다능성 NCCIT 세포, 배아 줄기 세포(ESL-017) 및 TRA-포도칼릭신을 발현하지 않는 대조군 PCS 세포를 사용한 세포 생존능 분석을 수행하였다: 표적 세포를 96-웰 플레이트에 3x103 내지 1x104 세포/웰(90 ul)의 밀도로 시딩하고 5% CO2 항온처리기에서 24시간 동안 세포를 항온처리한다. 세포를 항체(웰 당 10 ul)로 하기와 같이 처리한다: 100 nM의 출발 농도 및 반 로그 희석을 사용함, 농도 당 6개의 반복 시험을 수행함, 항체 처리를 수행하지 않은 마지막 웰을 음성 대조군으로서 사용하였다. 세포를 5% CO2 항온처리기에서 24시간 내지 7일 동안 항온처리 한다. 사용 전에 셀티터-글로(CellTiter-Glo, 등록상표) 발광 세포 생존능 분석 시약(프로메가(Promega), 카탈로그 번호 7572)을 실온으로 평형화시킨다. 100 ul의 셀티터-글로(등록상표) 시약을 각각의 웰에 적용하고 내용물을 2분 동안 오비탈 쉐이커에서 혼합하여 세포 용해를 유도한다. 플레이트를 실온에서 10분 동안 항온처리하여 발광 신호를 안정화시킨다. 마이크로플레이트 판독기(필터맥스(Filtermax) F5, 미국 캘리포니아 서니베일 소재 몰레큘러 디바이시즈(Molecular Devices)) 상의 발광을 기록한다.
세포 생존능 분석 결과는 CM-918-vc-MMAE 항체 약물 접합체에 의한 세포의 IC50 사멸은 NCCIT 세포의 경우 2.7 nM이었고, 배아 줄기 세포의 경우 5.6 nM이었고, CM-918-vc MMAE의 100 nM 농도까지 대조군 PC3 세포에 대해 효과가 관찰되지 않았음을 보여주었다. MMAE와 동일하게 접합된 이소타입 대조군 항체는 100 nM 이하의 농도에서 모든 3개의 세포주에 대해 효과를 나타내지 않았다. 결과는, CM-918 항체 약물 접합체가 다능성 배아 암종 암 세포 및 인간 배아 줄기 세포 사멸에서 매우 효과적이나 TRA-포도칼릭신을 발현하지 않는 세포에서는 그렇지 않음을 보여준다.

15마리의 누드(nude) 마우스에게 2x106 NCCIT 다능성 암 줄기 세포를 피하 주사하고, 종양을 200 mm3의 크기로 발달시키고, 이 시점에 마우스를 5마리씩 3개의 군으로 나누었다: 5마리의 마우스를 4회의 염수 주사로 처리하였다; 5마리의 마우스를 3 mg/kg의 CM-vc-MMAE를 사용하여 Q4Dx4로 처리하였다; 마지막 5마리를 3 mg/kg의 이소타입 항체 대조군-vc-MMAE를 사용하여 Q4Dx4로 처리하였다. 결과는, 35일의 마지막에 염수 주사한 마우스 및 이소타입 대조군 주사한 마우스 둘 다가 1000 mm3의 평균 크기의 종양을 가졌음을 보여준다. 접합된 CM-918 항체 약물 처리된 마우스에서, 5마리의 마우스 중 4마리가 검출가능한 종양을 가지지 않았고, 1마리의 마우스는 약 25 mm3의 종양을 가졌다. CM-918과 같이 MMAE와 접합된 이소타입 대조군 항체는 1000 mm3의 평균 종양 크기를 갖는 염수 처리된 마우스와 유사한 결과를 나타냈다. 결과는, CM-919-vc-MMAE 항체 약물 접합체가 마우스에서 다능성 암 줄기 세포 종양을 치료하는 데 효과적이고 영향이 강함을 보여준다.

)
CM-918 항체 및 다능성 암 NCCIT 세포를 사용한 ADCC 분석은 12 내지 15%의 세포독성 및 CM-918 ADCC 활성, 및 TRA-포도칼릭신을 발현하지 않는 대조군 세포가 2%의 세포독성을 가짐을 보여주었다. 대조군 IgG 항체는 NCCIT 세포의 사용시 2% 세포독성을 가졌다. 결과는, CM-918 항체가 TRA-포도칼릭신을 발현하는 다능성 암 세포를 사용시 ADCC 활성을 가짐을 보여준다.
인간 PBMC를 피콜(Ficoll) 구배를 사용하여 건강한 공여체로부터 정제하였다. 표적 세포를 96-웰 플레이트에 1 × 104/웰로 시딩하고 막 단리한 PBMC와 함께 25:1 또는 50:1의 E:T로 4시간 동안 7℃에서 항온처리하였다. 시험 항체 및 IgG-Fc 대조군 항체의 농도를 190 nM로 고정시켰다. 공동-항온처리 후에, 세포 세포독성을 LDH 분석에 의해 측정하였다. 표적 세포 세포독성의 백분율을 효과기 및 표적 세포(항체 없음) 대조군에 의한 흡광도 값을 감한 배경에 의해 계산하였고, 표적 세포 단독(자발적 용해)을 0%로, 용해된 표적 세포 단독(최대 용해)을 100%로 비율로 정했다.

)
CDC 분석을 수행하였고, 이는 NCCIT 다능성 암 줄기 세포가 100 nM CM-918 항체의 사용시 18% 세포독성 및 10 nM CM-918 항체의 사용시 10% 세포독성을 가졌음을 나타냈다. 100 nM 대조군 항체를 사용한 CDC 분석에서 NCCIT 세포 세포독성의 결과는 1% 미만의 세포독성을 가졌다. 이들 결과는, CM-918 항체가 원형질막 세포 표면 상에 TRA-포도칼릭신을 발현하는 다능성 암 줄기 세포에 대해 CDC 활성을 유도할 수 있음을 보여준다.
인간 보체를 96-웰 포맷에서 1:4 부피 비로 104개의 표적 세포와 함께 항온처리하였다. 시험 항체를 190 nM에서 출발하는 10-포인트 반-로그 희석 시리즈로 적재하였다. IgG-Fc 대조군 항체만을 190 nM에서 첨가하였다. 항체를 표적 세포 및 보체와 함께 4시간 동안 37℃에서 공동-항온처리하였다. 공동-항온처리 후에, 세포 세포독성을 LDH 분석(로슈(Roche))에 의해 측정하였다. 표적 세포 세포독성 백분율을 보체 및 표적 세포(항체 없음) 대조군에 의한 흡광도 값을 감한 배경에 의해 계산하였고, 표적 세포 단독(자발적 용해)을 0%로, 용해된 표적 세포 단독(최대 용해)을 100%로 비율로 정했다.
