Abstract
인간 조직·장기 제조법으로 의료 응용을 목표로 하고, 1 세포당의 기능을 대폭 개선하여, 비용의 대규모 삭감을 실현하는 개량 수법을 제공한다. 혈액 세포의 존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내에서 배양하는 것을 포함하는, 기관아의 제조 방법. 상기 방법에 의해 제조된 기관아, 그 기관아를 사용한, 조직 또는 장기의 제조 방법, 기관아의 이식 방법, 조직 또는 장기의 재생 또는 기능 회복 방법, 비인간 키메라 동물의 제조 방법 및 약제를 평가하는 방법도 제공한다.
Description
본 발명은, 혈액 세포를 사용한 조직·장기의 제조 방법에 관한 것이다.
최근, 다양한 기능 세포로 분화되는 능력을 갖는 iPS 세포 등의 다능성 줄기 세포를 이용하여, 창약 (創藥) 스크리닝이나 재생 의료에 유익한 인간 기능 세포를 분화 유도에 의해 창출하는 방법이 주목받고 있다. 지금까지 본 발명자들은, 성체 조직에서 보이는 혈관 내피 세포, 간엽계 세포로 이루어지는 질서적인 입체 구조를 갖는 인간 조직·장기의 재구축법을 확립시켰다 (비특허문헌 1 : Takebe T. et. al., Nature (2013), 특허문헌 1 : 인간 조직·장기의 제조 방법 WO2013/047639 A1).
한편, 지금까지의 조혈 세포와 간 세포의 성숙에 관한 지견으로서 마우스의 발생 과정에 있어서 조혈 세포로부터 산생되는 사이토카인이 간 세포의 성숙을 촉진한다는 보고가 이루어져 있다 (비특허문헌 2 : Kamiya A., et al., EMBO J. 1999 18 : 2127-3, 비특허문헌 3 : Kinoshita T., et al., Proc Natl Acad Sci U S A. 1999 96 : 7265-70 (1999)). 또한, 조혈계 세포와 미성숙 간 세포를 혼합하여 면역 부전 동물에게 이식하여 인간 간 세포와 인간 조혈 세포를 동시에 생착 (生着) 하는 것을 나타낸 연구는 보고되어 있지만, 조혈 세포와의 동시 이식에 의한 간 세포의 성숙도에 관한 지견은 보고되어 있지 않다 (비특허문헌 4 : Bility MT., et al., Nat Protoc. 7 : 1608-17 (2012), 비특허문헌 5 : Washburn ML., et al., Gastroenterology. 140 : 1334-44. (2011)). 또한, 지금까지 시험관내 (in vitro) 에서 재구축된 입체 조직 중에 조혈 세포를 도입함으로써, 목적으로 하는 기능 세포의 성숙에 있어서의 중요성을 나타낸 지견은 없다.

종래의 인간 조직·장기 제조법으로 의료 응용을 목표로 하는 경우에는, 성인 1 명당을 대상으로 한 경우의 제조 비용이 약 1,000 만엔 정도로 매우 고액인 점이 기술의 보급 장벽이 되었다. 현 단계에서 적게 견적해 보아도 2 만명 이상의 절대 적응 환자 (간부전 대기 중인 적응 제외·사망 예의 국제 시산) 가 존재하는 현상황을 감안하면, 범용 기술로서 이와 같은 치료 기술을 널리 환자에게 적응시켜 가기 위해서는, 1 세포당의 기능을 대폭 개선하여, 비용의 대규모 삭감을 실현하는 개량 수법의 개발이 요망되고 있는 상황이었다.
본 발명은, 그와 같은 개량 수법을 제공하는 것을 목적으로 한다.
본 발명자들은, 간내 배엽 세포, 혈관 내피 세포 및 간엽계 세포를 공 (共) 배양하는 배양계에 조혈 세포를 첨가함으로써, 형성되는 기관아 (器官芽, Organ Bud) 의 더나은 기능 향상에 성공하였다. 지금까지 인간 조직·장기의 3 차원적인 재구축에 있어서의 조혈 세포의 효과를 나타낸 기술은 없어, 신규성/진보성이 매우 높은 방법인 것으로 생각된다.
본 발명의 요지는 이하와 같다.
(1) 혈액 세포의 존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내에서 배양하는 것을 포함하는, 기관아의 제조 방법.
(2) 기관아는, 성숙함으로써 기관 (器官) 으로 분화될 수 있는 구조체인 (1) 에 기재된 방법.
(3) 혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 기관아와 비교하여, 기능이 향상된 기관아가 제조되는 (1) 또는 (2) 에 기재된 방법.
(4) 족장 (scaffold) 재료를 사용하지 않고, 세포가 배양되는 (1) ∼ (3) 중 어느 하나에 기재된 방법.
(5) 혈액 세포가, 미 (未) 분화 조혈 세포를 포함하는 (1) ∼ (4) 중 어느 하나에 기재된 방법.
(6) 미분화 조혈 세포가, 조혈 전구 세포 및/또는 조혈 줄기 세포인 (5) 에 기재된 방법.
(7) 혈액 세포가, 제대혈에서 유래되는 (1) ∼ (6) 중 어느 하나에 기재된 방법.
(8) 혈액 세포가, 제대혈의 단핵구 분획의 세포인 (7) 에 기재된 방법.
(9) 조직 또는 장기 세포가, 다능성 줄기 세포로부터 유도된 미분화 세포인 (1) ∼ (8) 중 어느 하나에 기재된 방법.
(10) 다능성 줄기 세포가 인공 다능성 줄기 세포인 (9) 에 기재된 방법.
(11) 다능성 줄기 세포가 인간 유래인 (9) 또는 (10) 에 기재된 방법.
(12) 기관아가 간아 (肝芽, Liver Bud) 인 (1) ∼ (11) 중 어느 하나에 기재된 방법.
(13) 혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 간아와 비교하여, 알부민의 분비능이 향상된 간아가 제조되는 (12) 에 기재된 방법.
(14) 혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 간아와 비교하여, 간 세포 분화 마커 유전자의 발현이 상승된 간아가 제조되는 (12) 또는 (13) 에 기재된 방법.
(15) 간 세포 분화 마커 유전자가, α 페토프로테인, 알부민, CYP3A7, 트립토판 대사 효소 TDO2, 및 나트륨·타우로콜산 공 (共) 수송체로 이루어지는 군에서 선택되는 적어도 1 개의 마커인 (14) 에 기재된 방법.
(16) (1) ∼ (15) 중 어느 하나에 기재된 방법에 의해 제조된 기관아.
(17) (16) 에 기재된 기관아를 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 조직 또는 장기의 제조 방법.
(18) (16) 에 기재된 기관아를 인간 또는 비인간 동물에게 이식하는 것을 포함하는, 기관아의 이식 방법.
(19) (16) 에 기재된 기관아를 인간 또는 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 조직 또는 장기의 재생 또는 기능 회복 방법.
(20) (16) 에 기재된 기관아를 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 비인간 키메라 동물의 제조 방법.
(21) (16) 에 기재된 기관아, (17) 에 기재된 방법에 의해 제조된 조직 및 장기, 그리고 (20) 에 기재된 방법에 의해 제조된 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나를 사용하여 약제를 평가하는 방법.
지금까지의 조혈 세포와 간 세포의 성숙에 관한 지견으로서, 마우스의 발생 과정에 있어서 조혈 세포로부터 산생되는 사이토카인 (OSM) 이 간 세포의 성숙을 촉진한다는 보고가 이루어져 있다 (비특허문헌 2 : Kamiya A., et al., EMBO J. 1999 18 : 2127-3, 비특허문헌 3 : Kinoshita T., et al., Proc Natl Acad Sci U S A. 1999 96 : 7265-70 (1999)). 그러나, 종래의 평면 배양에 있어서는, 이미 OSM 을 첨가하고 있음에도 불구하고 완전히 분화 유도가 불충분하였다. 한편, 본 발명자들의 특허 (특허문헌 1 : 인간 조직·장기의 제조 방법 WO2013/047639 A1) 는 3 종 세포로부터 기관아를 제조하는 것이지만, 조혈 세포를 포함하는 4 종류 이상의 세포로부터 기관아를 형성할 수 있고, 또한 그 성숙을 촉진시킨다는 방법론에 대해서는 전혀 기재나 시사도 없다. 따라서, 지금까지 시험관내에서 재구축된 기관아 중에 제 4 세포 (조혈 세포) 를 도입하는 것, 나아가서는, 목적으로 하는 기능 세포의 성숙에 있어서의 중요성을 나타낸 지견에는 큰 신규성·진보성이 있다.
지금까지 본 발명자들은, 종래의 기술로는 전혀 달성되지 못한, 혈관계가 적절히 배치된 인간 조직·장기를 제조하기 위한 기반 기술의 구축에 성공하였다 (인간 조직·장기의 제조 방법). 상기 기술을 비약적으로 발전시키기 위해, 새롭게 조혈계 세포라고 하는 플레이어를 부가함으로써, 직접 내지 간접적인 세포 작용을 통해서 목적으로 하는 기능 세포의 성숙을 훨씬 촉진시키는 수법을 실현하였다. 지금까지 충분치 않았던 대사 기능 등, 시험관내에서의 기능 성숙을 달성하는 완전히 새로운 기술이다. 구체적으로는, 본 기술에 의해 간 세포에 대해서 약 3 - 4 배 (알부민 분비를 지표) 의 기능 향상이 보였다. 이로써, 종래에 필요한 세포수의 1/3 ∼ 4 량으로 동등한 기능을 발휘할 것으로 기대되고, 그 결과, 제조 비용을 1/3-4 로 억제할 수 있게 된다. 현 단계에서의 성인 말기 간부전 환자 1 명당의 치료 비용으로부터 환산하면, 대략 600 만엔 정도의 저감이 달성될 것으로 기대된다.
또한, 본 발명에 의한 기능 향상의 결과로서, 종래에 평가가 어려웠던 약물 대사 기능 등의 검출이 기대되는 점에서, 시험관내에서의 창약 스크리닝에 대한 응용도 가능해진다.
본 명세서는, 본원의 우선권 기초인 일본국 특허출원, 특허출원 제2015-251140호의 명세서 및/또는 도면에 기재된 내용을 포함한다.

이하, 본 발명을 상세하게 설명한다.
본 발명은, 혈액 세포의 존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내에서 배양하는 것을 포함하는, 기관아의 제조 방법을 제공한다.
본 발명에 있어서 「기관아」란, 성숙함으로써 기관으로 분화될 수 있는 구조체로서, 조직 또는 장기 세포, 혈관 내피 세포, 간엽계 세포 (미분화 간엽계 세포 혹은 그것으로부터 분화된 세포), 및 혈액 세포의 4 종류 세포를 포함하는 구조체를 말한다. 어느 구조체가 기관아인지의 여부는, 예를 들어, 그 구조체를 생체에 이식하여, 목적하는 기관으로 분화될 수 있는지의 여부를 조사하는 것 (목적하는 기관으로 분화되어 있으면 기관아인 것으로 판단할 수 있다.), 및/또는 그 구조체가 상기 서술한 4 종류의 세포를 모두 포함하고 있는지의 여부를 조사하는 것 (4 종류의 세포를 모두 포함하고 있으면 기관아인 것으로 판단할 수 있다.) 에 의해 확인할 수 있다. 기관아는, 예를 들어, 신장, 심장, 폐장, 비장, 식도, 위, 갑상선, 부갑상선, 흉선, 생식선, 뇌, 척수 등의 기관으로 분화되는 기관아 등이어도 되지만, 간장으로 분화되는 기관아 (간아), 췌장으로 분화되는 기관아 (췌아 (膵芽)), 및 장관으로 분화되는 기관아 등 내배엽성 기관으로 분화되는 기관아가 바람직하다. 어느 구조체가 내배엽성 기관으로 분화되는 기관아인지의 여부는, 마커가 되는 단백질의 발현을 조사함으로써 확인할 수 있다 (후술하는 마커 단백질 중 어느 하나 혹은 복수가 발현되어 있으면 기관아인 것으로 판단할 수 있다.). 예를 들어, 간아에서는, HHEX, SOX2, HNF4A, AFP, ALB 등이 마커가 되고, 췌아에서는, PDX1, SOX17, SOX9 등이 마커가 되고, 장관으로 분화되는 기관아에서는, CDX2, SOX9 등이 마커가 된다. 당업자 사이에서 사용되고 있는 용어 중, 간아 (liver bud), 간 게실 (liver diverticula), 간 오르가노이드 (liver organoid), 췌장 (등측 또는 배측) 아 (pancreatic (dorsal or ventral) bud), 췌장 게실 (pancreatic diverticula), 췌장 오르가노이드 (pancreatic organoid), 장아 (intestinal bud), 장 게실 (intestinal diverticula), 장 오르가노이드 (intestinal organoid) (K. Matsumoto, et al. Science. 19 ; 294(5542) : 559-63. (2001)) 등은 본 발명에 있어서의 기관아에 포함된다.
본 발명에 있어서, 「혈액 세포」란, 생체내로부터 분리된 세포 혹은 ES 세포나 iPS 세포 등의 줄기 세포로부터 분화 유도된 세포, 또는, 분화 세포에 유전자 도입함으로써 직접 리프로그램한 세포에서 하기 혈액 세포의 특징을 갖는 (예를 들어, CD34, CD2, CD3, CD4, CD7, CD8, CD10, CD14, CD16, CD19, CD20, CD24, CD41, CD45, CD56, CD66b, 또는, CD235a, CD38, CD90, CD49f, VEGFR2, CD43, CD71, GPA (글리코포린 A (Glycophorin A)), CD42b, c-kit, CD150, Sca-1, Ter119 등 중 어느 것을 발현시킨다) 세포를 말한다. 어느 세포가 혈액 세포인지의 여부는, CD34, CD2, CD3, CD4, CD7, CD8, CD10, CD14, CD16, CD19, CD20, CD24, CD41, CD45, CD56, CD66b, 또는, CD235a, CD38, CD90, CD49f, VEGFR2, CD43, CD71, GPA (글리코포린 A), CD42b, c-kit, CD150, Sca-1, Ter119 등의 발현을 조사함으로써 확인할 수 있다. 본 발명에서 사용하는 혈액 세포는, 분화된 것이어도 되고, 미분화된 것이어도 되지만, 조혈 전구 세포, 조혈 줄기 세포 등의 미분화된 조혈 세포를 포함하는 것이 바람직하다. 미분화 조혈 세포로는, 인공 다능성 줄기 세포 (iPS 세포), 배성 줄기 세포 (ES 세포) 등의 다능성 줄기 세포에서 유래되는 것, 혈액 (제대혈, 골수혈, 말초혈 등) 으로부터 채취되는 것, 기타 분화 세포로부터 직접 분화 전환 (다이렉토리 프로그래밍) 된 혈액 세포 등을 예시할 수 있다. 후술하는 실시예에서는, 제대혈의 단핵구 (monocyte) 분획을 취함으로써, 조혈 전구 세포, 조혈 줄기 세포를 농축시킬 수 있는 것으로 생각된다. 당업자 사이에서 사용되고 있는 용어 중, 조혈 줄기 세포, 혈액 줄기 세포, 조혈 전구 세포, 혈액 전구 세포, 골수계 전구 세포, 과립구·단구계 전구 세포, 과립구계 전구 세포, 과립구, 골수아구, 전골수구, 골수구, 후골수구, 간상핵구, 분엽핵구 (호중구), 단구계 전구 세포, 단구, 매크로퍼지, 조직구, 쿠퍼 세포, 폐포 매크로 퍼지, 소교 세포, 파골 세포, 유상피 세포, 거대 세포 (랑그한스 거대 세포, 이물질 거대 세포, 토톤형 거대 세포), 수상 세포, 랑그한스 세포, 골수성 단구, 골수아구, 호염기성 전골수구, 호염기성 골수구, 호염기성 후골수구, 호염기구, 골수아구, 호산성 전골수구, 호산성 골수구, 호산성 후골수구, 호산성 간상핵구, 호산성 분엽핵구 (호산), 거핵아구, 거핵구, 적아구, 망적혈구, 비만 세포 전구체 등은 본 발명에 있어서의 혈액 세포에 포함된다. 혈액 세포는, 주로 인간 유래의 것을 사용하지만, 인간 이외의 동물 (예를 들어, 실험 동물, 애완 동물, 사역 동물, 경주마, 투견 등에 이용되는 동물, 구체적으로는, 마우스, 래트, 토끼, 돼지, 개, 원숭이, 소, 말, 양, 닭, 상어, 가오리, 은상어, 연어, 새우, 게 등) 유래의 혈액 세포를 사용해도 된다.
본 발명에 있어서 「혈관 내피 세포」란, 혈관 내피를 구성하는 세포, 또는 그와 같은 세포로 분화될 수 있는 세포를 말한다. 어느 세포가 혈관 내피 세포인지의 여부는, 마커 단백질, 예를 들어, TIE2, VEGFR-1, VEGFR-2, VEGFR-3, CD41 이 발현되어 있는지의 여부를 조사함으로써 확인할 수 있다 (상기 마커 단백질의 어느 하나 혹은 복수가 발현되어 있으면 혈관 내피 세포인 것으로 판단할 수 있다.). 본 발명에 있어서 사용하는 혈관 내피 세포는, 분화된 것이어도 되고, 미분화된 것이어도 된다. 혈관 내피 세포가, 분화된 세포인지의 여부는, CD31, CD144 에 의해 확인할 수 있다. 당업자 사이에서 사용되고 있는 용어 중, 내피 세포 (endothelial cell), 제정맥 내피 세포 (umbilical vein endothelial cell), 내피 전구체 세포 (endothelial progenitor cell), 내피 선구체 세포 (endothelial precursor cell), 혈관형성 전구체 (vasculogenic progenitor), 혈액모세포 (hemangioblast) (HJ. joo, et al. Blood. 25 ; 118(8) : 2094-104. (2011)) 등은 본 발명에 있어서의 혈관 내피 세포에 포함된다. 바람직한 혈관 내피 세포는, 제대 정맥 유래의 혈관 내피 세포이다. 혈관 내피 세포는, 혈관으로부터 채취하거나 또는 인공 다능성 줄기 세포 (iPS 세포), 배성 줄기 세포 (ES 세포) 등의 다능성 줄기 세포로부터 공지된 방법에 따라 제조할 수 있다. 혈관 내피 세포는, 주로 인간 유래의 것을 사용하지만, 인간 이외의 동물 (예를 들어, 실험 동물, 애완 동물, 사역 동물, 경주마, 투견 등에 이용되는 동물, 구체적으로는, 마우스, 래트, 토끼, 돼지, 개, 원숭이, 소, 말, 양, 닭, 상어, 가오리, 은상어, 연어, 새우, 게 등) 유래의 혈관 내피 세포를 사용해도 된다.
본 발명에 있어서 「간엽계 세포」란, 주로 중배엽에서 유래되는 결합직 (結合織) 에 존재하고, 조직에서 기능하는 세포의 지지 구조를 형성하는 결합직 세포이지만, 간엽계 세포로의 분화 운명이 결정되어 있지만, 아직 간엽계 세포로 분화되어 있지 않은 세포도 포함하는 개념이다. 본 발명에 있어서 사용하는 간엽계 세포는, 분화된 것이어도 되고, 미분화된 것이어도 된다. 어느 세포가 미분화 간엽계 세포인지의 여부는, 마커 단백질, 예를 들어 Stro-1, CD29, CD44, CD73, CD90, CD105, CD133, CD271, 네스틴 (Nestin) 이 발현되어 있는지의 여부를 조사함으로써 확인할 수 있다 (상기 마커 단백질의 어느 하나 혹은 복수가 발현되어 있으면 미분화 간엽계 세포인 것으로 판단할 수 있다.). 또한, 전항의 마커 모두 발현되어 있지 않은 간엽계 세포는 분화 간엽계 세포로 판단할 수 있다. 당업자 사이에서 사용되고 있는 용어 중, 중간엽 줄기 세포 (mesenchymal stem cell), 중간엽 전구체 세포 (mesenchymal progenitor cell), 중간엽 세포 (mesenchymal cell) (R. Peters, et al. PLoS One. 30 ; 5(12) : e15689. (2010)) 등은 본 발명에 있어서의 간엽계 세포에 포함된다. 바람직한 간엽계 세포는, 골수 유래의 간엽계 세포 (특히, 간엽계 줄기 세포) 이다. 간엽계 세포는, 골수, 지방 조직, 태반 조직, 제대 조직, 치수 (齒髓) 등의 조직으로부터 채취하거나, 또는 인공 다능성 줄기 세포 (iPS 세포), 배성 줄기 세포 (ES 세포) 등의 다능성 줄기 세포로부터 공지된 방법에 따라 제조할 수 있다. 간엽계 세포는, 주로 인간 유래의 것을 사용하지만, 인간 이외의 동물 (예를 들어, 실험 동물, 애완 동물, 사역 동물, 경주마, 투견 등에 이용되는 동물, 구체적으로는, 마우스, 래트, 토끼, 돼지, 개, 원숭이, 소, 말, 양, 닭, 상어, 가오리, 은상어, 연어, 새우, 게 등) 유래의 미분화 간엽계 세포를 사용해도 된다.
본 발명에 있어서 「조직 또는 장기 세포」란, 조직 또는 장기를 구성하는 기능 세포, 또는 기능 세포로 분화되는 미분화 세포를 말한다. 「미분화된 조직 또는 장기 세포」로는, 예를 들어, 신장, 심장, 폐장, 비장, 식도, 위, 갑상선, 부갑상선, 흉선, 생식선, 뇌, 척수 등의 기관으로 분화 가능한 세포 등이어도 되고, 뇌, 척수, 부신수질, 표피, 모발·손톱·피부선, 감각기, 말초 신경, 수정체 등의 외배엽성 기관으로 분화 가능한 세포, 신장, 요관, 심장, 혈액, 생식선, 부신피질, 근육, 골격, 진피, 결합 조직, 중피 등의 중배엽성 기관으로 분화 가능한 세포, 간장, 췌장, 장관, 폐, 갑상선, 부갑상선, 요로 등의 내배엽성 기관으로 분화 가능한 세포 등을 들 수 있다. 어느 세포가 외배엽성 기관, 중배엽성 기관 또는 내배엽성 기관으로 분화 가능한 세포인지의 여부는, 마커가 되는 단백질의 발현을 조사함으로써 확인할 수 있다 (마커 단백질 중 어느 하나 혹은 복수가 발현되어 있으면 내배엽성 기관으로 분화 가능한 세포인 것으로 판단할 수 있다). 예를 들어, 간장으로 분화 가능한 세포에서는, HHEX, SOX2, HNF4A, AFP, ALB 등이 마커가 되고, 췌장으로 분화 가능한 세포에서는, PDX1, SOX17, SOX9 등이 마커가 되고, 장관으로 분화 가능한 세포에서는, CDX2, SOX9 등이 마커가 되고, 신장으로 분화 가능한 세포에서는, SIX2, SALL1, 심장으로 분화 가능한 세포에서는, NKX2-5 MYH6, ACTN2, MYL7, HPPA, 혈액으로 분화 가능한 세포에서는, C-KIT, SCA1, TER119, HOXB4, 뇌나 척수로 분화 가능한 세포에서는, HNK1, AP2, 네스틴 (NESTIN) 등이 마커가 된다. 당업자 사이에서 사용되고 있는 용어 중, 간모세포 (hepatoblast), 간 전구체 세포 (hepatic progenitor cell), 췌장모세포 (pancreatoblast), 간 선구체 세포 (hepatic precursor cell), 췌장모세포 (pancreatoblast), 췌장 전구체 (pancreatic progenitor), 췌장 전구체 세포 (pancreatic progenitor cell), 췌장 선구체 세포 (pancreatic precursor cell), 내분비 선구체 (endocrine precursor), 장 전구체 세포 (intestinal progenitor cell), 장 선구체 세포 (intestinal precursor cell), 중간 중배엽 (intermediate mesoderm), 후신 간엽 선구체 세포 (metanephric mesenchymal precursor cell), 다능성 네프론 전구체 (multipotent nephron progenitor), 신장 전구체 세포 (renal progenitor cell), 심장 중배엽 (cardiac mesoderm), 심혈관 전구체 세포 (cardiovascular progenitor cell), 심장 전구체 세포 (cardiac progenitor cell) (JR. Nature Biotechnology 25, 1468 - 1475 (2007)) 등은 본 발명에 있어서의 미분화된 조직 또는 장기 세포에 포함된다. 미분화된 조직 또는 장기 세포는, 조직 또는 장기로부터 채취하거나, 또는 인공 다능성 줄기 세포 (iPS 세포), 배성 줄기 세포 (ES 세포) 등의 다능성 줄기 세포로부터 공지된 방법에 따라 제조할 수 있다. 또한, 미분화된 조직 또는 장기 세포는, iPS 세포 등의 다능성 줄기 세포로부터 조직 또는 장기로의 분화가 중도 단계에 있는 세포, 예를 들어, 원시 장내 배엽 세포 (Primitive Gut Endoderm Cells : PGECs) (일본 특허 제5777127호) 등이어도 된다. PGECs 는, 간 세포, 췌 세포 및 장 세포로의 분화가 가능하고 (분화 기능이 높다), 암의 악성도에 관계된 마커를 발현시키지 않고 (안전성이 높다), iPS 세포로부터 비 (非) 피더 환경하에서 분화 유도하여 조제할 수 있으므로, 임상 응용도 가능하게 한다는 이점을 갖는다. 게다가, PGECs 는 대량 조제가 가능하다. PGECs 는, 일본 특허 제5777127호에 기재된 방법으로 조제할 수 있다. 또는, 무혈청 배지에 액티빈을 첨가하여 iPS 세포 등의 다능성 줄기 세포를 배양함으로써, CXCR4 및 E-카데린 (E-cadherin) 두 양성의 내배엽계 세포로 유도한 것이나, 또한 얻어진 내배엽계 세포를 BMP4, FGF2 를 첨가하여 2 일간 배양함으로써 얻어지는, CXCR4 음성 HNF4α 양성의 간장 내배엽 집단이어도 된다. 그 밖에, 예를 들어 간장으로 분화 가능한 장기 세포는, K.Si-Taiyeb, et al. (2009) 에 따라 제조할 수 있고, 장관으로 분화 가능한 장기 세포는, J. Cai, et al. J Mol Cell Biol. ; 2(1) : 50-60 (2010), R. Spence, et al. Nature. ; 470(7332) : 105-9. (2011) 에 따라 제조할 수 있고, 심장으로 분화 가능한 장기 세포는, J. Zhang, et al. Circulation Research. ; 104 : e30-e41 (2009) 에 따라 제조할 수 있고, 뇌나 척수로 분화 가능한 세포는, G. Lee, et al. Nature Biotechnology 25, 1468-1475 (2007) 에 따라 제조할 수 있다. 「분화된 조직 또는 장기 세포」로는, 췌장의 내분비 세포, 췌장의 췌관 상피 세포, 간장의 간 세포, 장관의 상피 세포, 신장의 뇨세관 상피 세포, 신장의 사구체 상피 세포, 심장의 심근 세포, 혈액의 임파구나 과립구, 적혈구, 뇌의 신경 세포나 그리아 세포, 척수의 신경 세포나 슈원 세포 등을 예시할 수 있다. 조직 또는 장기 세포는, 주로 인간 유래의 것을 사용하지만, 인간 이외의 동물 (예를 들어, 실험 동물, 애완 동물, 사역 동물, 경주마, 투견 등에 이용되는 동물, 구체적으로는, 마우스, 래트, 토끼, 돼지, 개, 원숭이, 소, 말, 양, 닭, 상어, 가오리, 은상어, 연어, 새우, 게 등) 유래의 조직 또는 장기 세포를 사용해도 된다.
공배양에 있어서의 4 종류 세포의 배양비는 기관아를 형성할 수 있는 범위 내이면 특별히 한정되지 않지만, 바람직한 세포의 수비는, 조직 또는 장기 세포 : 혈관 내피 세포 : 간엽계 세포 : 혈액 세포 = 10 : 10 ∼ 5 : 2 ∼ 0.1 : 2 ∼ 50이다. 조직 또는 장기 세포 25 만개 정도, 혈관 내피 세포 17 만개 정도, 간엽계 세포 2.5 만개 정도, 혈액 세포 10 만개 정도를 공배양하여, 크기가 50 ∼ 250 마이크로미터 정도인 기관아를 형성시킬 수 있다.
배양시에 사용하는 배지는, 기관아가 형성되는 것이면 어떠한 것이어도 되지만, 혈관 내피 세포 배양용 배지, 조직 또는 장기 세포 배양용 배지, 상기 두 가지 배지를 혼합한 것 등을 사용하는 것이 바람직하다. 혈관 내피 세포 배양용 배지는 어떠한 것을 사용해도 되지만, hEGF (재조합 인간 상피 세포 성장 인자), VEGF (혈관 내피 세포 성장 인자), 하이드로코르티손, bFGF, 아스코르브산, IGF1, FBS, 항생제 (예를 들어, 겐타마이신, 안포테리신 B 등), 헤파린 (Heparin), L-글루타민 (L-Glutamine), 페놀레드 (Phenolred), BBE 중 적어도 1 종을 함유하는 것을 사용하는 것이 바람직하다. 혈관 내피 세포 배양용 배지로는, EGM-2 BulletKit (Lonza 사 제조), EGM BulletKit (Lonza 사 제조), VascuLife EnGS Comp Kit (LCT 사 제조), Human Endothelial-SFM Basal Growth Medium (Invitrogen 사 제조), 인간 미소 혈관 내피 세포 증식 배지 (TOYOBO 사 제조) 등을 사용할 수 있다. 조직 또는 장기 세포 배양용 배지는 어떠한 것을 사용해도 되지만, 장기 세포가 간 세포인 경우, 아스코르브산 (ascorbic acid), BSA- FAF, 인슐린 (insulin), 히드로코르티손 (hydrocortisone), GA-1000 중 적어도 1 종을 함유하는 것을 사용하는 것이 바람직하다. 간 세포 배양용 배지로는, HCM BulletKit (Lonza 사 제조) 로부터 hEGF (재조합 인간 상피 세포 성장 인자) 를 제거한 것, RPMI1640 (Sigma-Aldrich 사 제조) 에 1 % B27 보충물 (Supplements) (GIBCO 사 제조) 와 10 ng/㎖ hHGF (Sigma-Aldrich 사 제조) 등을 사용할 수 있다. 인간 간아의 형성에 관해서는, GM BulletKit (Lonza 사 제조) 와 HCM BulletKit (Lonza 사 제조) 로부터 hEGF (재조합 인간 상피 세포 성장 인자) 를 제거한 것을 1 : 1 로 혼합한 것에, 덱사메타손 (Dexamethasone), 온코스타틴 M (Oncostatin M), HGF 를 첨가하면, 간아의 성숙에 효과가 있음이 판명되었다.
세포의 배양에 있어서는, 족장 재료를 사용할 필요는 없지만, 4 종류 세포의 혼합물을 간엽계 세포가 수축 가능한 겔상 지지체 상에서 배양하면 된다.
간엽계 세포의 수축은, (현미경 내지 육안으로) 형태학적으로 입체 조직 형성을 확인하는 것이나, 약숟가락 등에 의한 회수에 수반되어 조직의 형상이 유지되는 강도를 갖는 것을 나타내는 등 (Takebe et al. Nature 499(7459), 481-484, 2013)) 과 같이 하여 확인할 수 있다.
지지체는, 적정한 경도 (예를 들어, 영률 200 ㎪ 이하 (마트리겔을 코트한 형상이 평탄한 겔의 경우 등) 이지만, 지지체의 적정한 경도는 코팅과 형상에 따라 변화될 수 있다) 를 갖는 겔상 기재이면 되고, 그와 같은 기재로는, 하이드로겔 (예를 들어, 아크릴아미드겔, 젤라틴, 마트리겔 등) 등을 예시할 수 있지만, 그것들에 한정되는 일은 없다. 또, 목적으로 하는 집합체의 형태·사이즈·양에 따라, 지지체의 경도는 균일할 필연성은 없고, 경도에 공간적·시간적인 구배를 설정하는 것이나 패턴화하는 것이 가능하다. 지지체의 경도가 균일한 경우에는, 지지체의 경도는, 바람직하게는 100 ㎪ 이하, 보다 바람직하게는 1 ∼ 50 ㎪ 이다. 겔상 지지체는, 평면이어도 되고, 혹은 겔상 지지체의 배양하는 측의 단면이 U 또는 V 자 형상이면 된다. 겔상 지지체의 배양하는 측의 단면이 U 또는 V 자 형상임으로써, 지지체의 배양면에 세포가 모이게 되어, 보다 적은 수의 세포 및/또는 조직으로 세포 집합체가 형성되므로 유리하다. 또한, 지지체에, 화학적·물리적인 수식을 실시해도 된다. 수식 물질로는, 마트리겔, 라미닌, 엔탁틴, 콜라겐, 피브로넥틴, 비트로넥틴 등을 예시할 수 있다.
겔상 배양 지지체의 경도에 공간적인 구배를 설정한 일례는, 중심부의 경도가 주변부의 경도보다 단단한 겔상 배양 지지체이다. 중심부의 경도는, 200 ㎪ 이하가 적정하며, 주변부의 경도는, 중심부보다 부드러우면 되지만, 지지체의 중심부와 주변부의 적정한 경도는 코팅과 형상에 따라 변화될 수 있다. 겔상 배양 지지체의 경도에 공간적인 구배를 설정한 다른 일례는, 주변부의 경도가 중심부의 경도보다 단단한 겔상 배양 지지체이다.
패턴화된 겔상 배양 지지체의 일례는, 중심부의 경도가 주변부의 경도보다 단단하다는 패턴을 1 개 이상 갖는 겔상 배양 지지체이다. 중심부의 경도는, 200 ㎪ 이하가 적정하며, 주변부의 경도는, 중심부보다 부드러우면 되지만, 지지체의 중심부와 주변부의 적정한 경도는 코팅과 형상에 따라 변화될 수 있다. 패턴화된 겔상 배양 지지체의 다른 일례는, 주변부의 경도가 중심부의 경도보다 단단하다는 패턴을 1 개 이상 갖는 겔상 배양 지지체이다. 주변부의 경도는, 200 ㎪ 이하가 적정하며, 중심부의 경도는, 주변부보다 부드러우면 되지만, 지지체의 중심부와 주변부의 적정한 경도는 코팅과 형상에 따라 변화될 수 있다.
배양시의 온도는 특별히 한정되지 않지만, 30 ∼ 40 ℃ 로 하는 것이 바람직하고, 37 ℃ 로 하는 것이 더욱 바람직하다.
배양 기간은 특별히 한정되지 않지만, 3 ∼ 10 일로 하는 것이 바람직하고, 6 일로 하는 것이 더욱 바람직하다.
본 발명의 방법으로 제조한 기관아는, 혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 기관아와 비교해서 기능이 향상될 수 있다.
예를 들어, 기관아가 간아인 경우, 본 발명의 방법으로 제조한 간아는, 혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 간아와 비교하여 알부민의 분비능이 향상될 수 있다. 또한, 본 발명의 방법으로 제조한 간아는, 혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 간아와 비교하여, 간 세포 분화 마커 유전자의 발현이 상승될 수 있다. 간 세포 분화 마커 유전자로는, α 페토프로테인, 알부민, CYP3A7, 트립토판 대사 효소 TDO2, 나트륨·타우로콜산 공수송체 등을 예시할 수 있지만, 이것들에 한정되는 것은 아니다.
본 발명은, 상기 방법에 의해 제조한 기관아도 제공한다.
상기 방법에 의해 제조된 기관아를 비인간 동물에게 이식하여, 그 비인간 동물내에서 성숙시킴으로써, 조직 또는 장기를 제조할 수 있다. 즉, 본 발명은, 상기 방법에 의해 제조한 기관아를 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 조직 또는 장기의 제조 방법도 제공한다. 사용하는 비인간 동물로서는, 실험 동물, 애완 동물, 사역 동물, 경주마, 투견 등에 이용되는 동물, 구체적으로는, 마우스, 래트, 토끼, 돼지, 개, 원숭이, 소, 말, 양, 닭, 상어, 가오리, 은상어, 연어, 새우, 게 등을 들 수 있다. 또한, 사용하는 비인간 동물은, 면역 거절 반응을 회피하기 위해서, 면역 부전 동물인 것이 바람직하다.
따라서, 본 발명은, 상기 방법에 의해 제조된 기관아를 인간 또는 비인간 동물에게 이식하는 것을 포함하는, 기관아의 이식 방법도 제공한다. 기관아의 이식 부위는, 이식 가능하면 어느 부위여도 되지만, 두개 내, 장간막, 간장, 비장, 신장, 신피막하, 문맥 상 등을 예시할 수 있다. 두개 내에 이식할 경우에는, 시험관내에서 제조한 1 ∼ 3 개 정도의 5 ㎜ 크기의 기관아를 이식하면 되고, 장간막에 이식할 경우에는, 시험관내에서 제조한 1 ∼ 6 개 정도의 5 ㎜ 크기의 기관아를 이식하면 되고, 문맥 상에 이식할 경우에는, 시험관내에서 제조한 1 ∼ 20 개 정도의 5 ㎜ 크기의 기관아를 이식하면 된다. 신피막에 이식할 경우에는, 시험관내에서 제조한 1 ∼ 5 개 정도의 5 ㎜ 크기의 기관아를 이식하면 되고, 간장·비장·신장·에 이식할 경우에는, 시험관내에서 제조한 100 ∼ 200 개 정도의 100 ㎛ 크기의 기관아를 이식하면 된다.
이상과 같이 해서 제조한 조직 및 장기는, 창약 스크리닝이나 재생 의료 등에 사용할 수 있다.
따라서, 본 발명은, 상기 방법에 의해 제조된 기관아를 인간 또는 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 조직 또는 장기의 재생 또는 기능 회복 방법을 제공한다. 비인간 동물로서는, 실험 동물, 애완 동물, 사역 동물, 경주마, 투견 등에 이용되는 동물, 구체적으로는, 마우스, 래트, 토끼, 돼지, 개, 원숭이, 소, 말, 양, 닭, 상어, 가오리, 은상어, 연어, 새우, 게 등을 들 수 있다.
본 발명의 방법에 의해 제조된 기관아는, 제제화되어 재생 의료용 조성물이라는 형태로 사용될 수 있다. 본 발명의 조성물을 생체내에 이식하여, 조직 또는 장기를 제조할 수 있다. 또한, 본 발명의 조성물을 생체내에 이식하여, 조직 또는 장기의 재생 또는 기능 회복을 실시할 수 있다.
본 발명의 조성물이 생체에 이식된 후, 기관아는 혈관망을 갖는 조직 또는 장기로 분화될 수 있다. 그 혈관망에는 혈관 관류가 발생할 수 있다. 혈관망에 혈관 관류가 발생함으로써, 성체 조직과 동등 또는 그것에 가까운 고도로 질서있는 조직 구조를 갖는 조직·장기를 창출할 수 있게 될 것으로 생각된다.
본 발명의 조성물에는, FGF2, HGF, VEGF 등의 조직 혈관화 촉진제, 이식에 수반되는 지혈용 젤라틴 스펀지 (상품명 : 스폰젤, 아스테라스 주식회사), 및 이식 조직의 고정에 사용하는 볼힐 (테이진 파마 주식회사)·베리프라스트 (CSL 베링 주식회사)·타코콤브 (CSL 베링 주식회사) 등의 조직 접착제 등을 첨가해도 된다.
또한, 본 발명은, 상기 방법에 의해 제조된 기관아를 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 비인간 키메라 동물의 제조 방법도 제공한다. 기관아를 이식한 비인간 동물 (예를 들어, 마우스) 은, 기관아의 제조에 사용한 조직 또는 장기 세포의 유래 생물종 (예를 들어, 인간) 의 생리 기능을 모방할 수 있다.
또한, 본 발명은, 상기 방법에 의해 제조된 기관아, 조직 및 장기, 및 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나를 사용하여 약제를 평가하는 방법도 제공한다. 약제의 평가로서는, 예를 들어 약물 대사의 평가 (예를 들어, 약물 대사 프로파일의 예측), 약효 평가 (예를 들어, 의약품으로서 유효한 약제를 스크리닝하는 것 등), 독성 평가, 약물 상호 작용 평가 등을 들 수 있다.
약물 대사의 평가는, 상기 방법에 의해 제조된 기관아, 조직 및 장기, 및 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나에 있어서, 의약품의 후보 화합물을 투여한 후의 생물 시료를 채취·해석함으로써, 인간형의 약물 대사 프로파일을 취득할 수 있다. 이로써 종래의 기술로는 달성이 매우 어려웠던 인간에서의 의약품의 분포·대사·배설 과정을 예측할 수 있게 되어, 안전하고 효과가 있는 의약품의 개발을 비약적으로 가속할 수 있는 것으로 기대된다.
의약품으로서 유효한 약제의 스크리닝은, 질병 환자로부터 수립한 세포로부터 상기 방법에 의해 제조된 기관아, 조직 및 장기, 및 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나에, 신규 의약품 후보 화합물을 투여함으로써 해석할 수 있게 된다. 이로써, 종래의 시험관내 시험에서 불충분했던, 실제 인간에 투여했을 때의 약효의 예측 정밀도를 대폭 개선할 수 있는 가능성이 기대된다.
독성 평가는, 상기 방법에 의해 제조된 기관아, 조직 및 장기, 및 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나에, 피험 물질을 투여한 후에, 조직 장해 마커 등을 측정함으로써, 장해 예측의 정밀도 향상이 가능해진다.
약물 상호 작용 평가는, 상기 방법에 의해 제조된 기관아, 조직 및 장기, 및 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나를 사용하여 복수의 약제를 투여한 후에, 그 후의 각 약제의 분포·대사·배설 과정 등의 약물 동태나 독성 평가, 약효 평가를 행함으로써 실시할 수 있다.
또한, 본 발명의 방법에 의해 제조된 조직 및 장기로부터 조직 줄기 세포를 창출할 수도 있고, 본 발명은 인간 조직 세포나 장기 세포의 대량 창출의 세포 조작 기술에 대한 응용이 가능하다.
실시예
이하, 실시예에 의해 본 발명을 더욱 상세하게 설명한다.
〔실시예 1〕
[방법]
1, 혈액 세포의 조제
<제대혈의 취득>
제대 (요코하마 시립대학 부속 병원에서 제왕 절개에 의해 출산된 환자로부터 제공) 로부터 50 ㎖ 시린지, 18 G 바늘을 사용하여 채혈을 실시하였다. 혈액은 대략 40 ㎖ ― 50 ㎖ 채취하였다.
<용혈 처리>
50 ㎖ 튜브에 40 ㎖ 의 용해 (Lysis) 버퍼를 넣고, 10 ㎖ 의 제대혈을 부가하였다.
교반하고 실온에서 10 분간 정치 (靜置) 시켰다.
200 G 로 10 분 원심 분리하였다.
한번 더, 용해 버퍼 10 ㎖ 에 현탁하고, 5 개분 합쳐서 50 ㎖ 로 하였다.
다시 200 G 로 10 분 원심 분리하였다.
침전을 10 ㎖ DMEM+10% FBS 로 현탁하고, 200 G 로 5 분 원심 분리하였다.
재차 10 ㎖ DMEM+10% FBS 로 현탁하고 세포수를 계측하였다.
재료
1) 10×용해 버퍼의 제조 (×10 액 ; 1/10 희석하여 사용하였다.)
·NH4Cl 82.6 g
·NaHCO3 11.9 g
·EDTA2Na 0.378 g
/ 1 ℓ milliQ (pH 7.3 으로 조정)
2) 1×용해 버퍼의 제조
PBS (4 ℃ 로 식혀져 있는 것이 좋다. 2 mM EDTA 를 넣어도 된다) 로 ×1 로 희석하여 사용하였다.
<PI-/CD235- 세포의 분리>
혈액 세포에 대하여 프로피듐 요오드화물 (Propidium Iodide) (PI) 를 사용하여 죽은 세포를 염색하고, 형광 표지 CD235 항체를 사용하여 적혈구/적아구를 염색하고, 셀 소터를 사용하여 각각에서 염색되지 않은 세포 (생세포이고, 적혈구/적아구가 아닌 세포) 를 분리하였다.
<단핵구 (monocyte) 분획 (MNC) 세포의 분리>
셀 소터를 사용하여 PI-/CD235- 세포를 FSC (forward scatter : 전방 산란광), 및 SSC (side scatter : 측방 산란광) 로 전개하고 단핵구 분획 (MNC) 세포에 해당되는 세포 집단을 분리하였다.
2, 간내 배엽 세포의 제조
방법 :
미분화 iPS 세포 (TkDA3 (토쿄 대학에서 제공), 및 제대로부터 독자적으로 수립한 iPS 세포) 를 박리하고 단세포화한 후, 플라스틱 디쉬에 5×104 세포/㎠ 의 세포 밀도로 RPMI+1%B27+10uM Rockinhibitor+50ng/㎖ Wnt3a+100ng/㎖ ActivinA 존재하에서 1 일간 배양하였다. 계속해서, RPMI+1%B27+50ng/㎖ Wnt3a+100ng/㎖ ActivinA 존재하에서 6 일간 배양하여 간내 배엽 세포를 얻었다.
[결과]
1 (도 1, 2, 3)
간내 배엽 세포 (2.5×105 세포) 와 제대 유래 혈관 내피 세포 (1.7×105 세포) (Lonza, Basel, Switzerland) 및 간엽계 세포 (2.5×104 세포) (Lonza, Basel, Switzerland) (10 : 7 : 1) 에 대하여 셀 소터를 사용하여 분리한 PI- 세포 및 조혈 줄기 세포/조혈 전구 세포가 농축되어 있는, 단핵구 분획 (MNC) 세포 (도 1) 를 1×105 세포 현탁하고, 쿠라레 마이크로 웰 플레이트 상에서 간아 (liver bud) 를 제조하였다 (크기 : 50 ∼ 250 마이크로미터). 15 일간 배양 후, 각각의 조건에서 배양한 간아의 배양 상청 중에 24 시간 동안 분비된 인간 알부민 단백질량을 ELISA 법에 의해 분석하였다. 그 결과 PI- 세포 혹은 조혈 줄기 세포/조혈 전구 세포가 농축되어 있는 단핵구 분획 (MNC) 세포를 첨가한 군에서는, 종래법인 혈액 세포 비첨가군에 비해서 알부민 분비량이 증가되었다 (도 2). 다음으로 상기 조건에서 배양한 배양 15 일째의 간아를 회수하고 qPCR 에 의해 간 세포 분화 마커 (α 페토프로테인 (AFP), 알부민 (ALB), CYP3A7, 트립토판 대사 효소 (TDO2), 나트륨·타우로콜산 공수송체 (NTCP)) 의 발현을 검토한 바, PI- 세포 혹은 단핵구 분획 (MNC) 세포를 첨가한 군에서는, 종래법인 혈액 세포 비첨가군에 비해서 유전자 발현량이 증가되었다 (도 3).
2 (도 4, 5)
다음으로 첨가하는 혈액 세포의 용량 의존성을 검토하기 위해, 간내 배엽 세포 (2.5×105 세포) 와 제대 유래 혈관 내피 세포 (1.7×105 세포) 및 간엽계 세포 (2.5×104 세포) (10 : 7 : 1) 에 대하여 혈액 세포 첨가 제로, 혈액 세포 (PI- 세포) 첨가 1×105 세포, 2.5×105 세포, 5×105 세포 첨가군에서 간아 제조 15 일째에서의 배양액 (도 4) 중의 24 시간 인간 알부민 단백질 분비량을 검토하였다. 그 결과, 종래법에 비해서 혈액 세포의 세포수 의존적으로 인간 알부민 분비량이 증강되는 것이 명확해졌다 (도 5).
본 명세서에서 인용한 모든 간행물, 특허 및 특허출원을 그대로 참고로서 본 명세서에 도입하는 것으로 한다.
본 발명은, 인간 기능 세포의 공업적 제조를 위한 중요한 기반 기술이 된다. 이로써, 난치성 질환을 대상으로 한 재생 의료 기술로서 이식용 인간 조직·장기의 제조에 응용할 수 있다. 종래법으로부터 대폭 비용 삭감을 실현할 수 있는 것이 기대될 뿐만 아니라, 본 기술에 의해 시험관내에서의 기능 성숙을 향상시킬 수도 있을 것으로 생각되기 때문에, 종래 기술로는 치료가 어려웠던 급성·아급성의 간부전증에 대해서도 적용할 수 있게 될 것으로 기대된다.
또한, 본 발명에 의해 인위적으로 제조된 인간 간장 조직을 사용한 약제 평가계를 확립하면, 창약 개발에 있어서 필요한 인간 성숙 간 세포를 대량으로 제조할 수 있게 된다.
Claims (21)
- 혈액 세포의 존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내 (in vitro) 에서 배양하는 것을 포함하는, 기관아 (器官芽, Organ Bud) 가 제조되는 방법.
- 제 1 항에 있어서,
기관아는, 성숙함으로써 기관으로 분화될 수 있는 구조체인 방법. - 제 1 항 또는 제 2 항에 있어서,
혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포, 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 기관아와 비교하여, 기능이 향상된 기관아가 제조되는 방법. - 제 1 항 내지 제 3 항 중 어느 한 항에 있어서,
족장 (scaffold) 재료를 사용하지 않고, 세포가 배양되는 방법. - 제 1 항 내지 제 4 항 중 어느 한 항에 있어서,
혈액 세포가, 미분화 조혈 세포를 포함하는 방법. - 제 5 항에 있어서,
미분화 조혈 세포가, 조혈 전구 세포 및/또는 조혈 줄기 세포인 방법. - 제 1 항 내지 제 6 항 중 어느 한 항에 있어서,
혈액 세포가, 제대혈에서 유래되는 방법. - 제 7 항에 있어서,
혈액 세포가, 제대혈의 단핵구 분획의 세포인 방법. - 제 1 항 내지 제 8 항 중 어느 한 항에 있어서,
조직 또는 장기 세포가, 다능성 줄기 세포로부터 유도된 미분화 세포인 방법. - 제 9 항에 있어서,
다능성 줄기 세포가 인공 다능성 줄기 세포인 방법. - 제 9 항 또는 제 10 항에 있어서,
다능성 줄기 세포가 인간 유래인 방법. - 제 1 항 내지 제 11 항 중 어느 한 항에 있어서,
기관아가 간아 (肝芽, Liver Bud) 인 방법. - 제 12 항에 있어서,
혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 간아와 비교하여, 알부민의 분비능이 향상된 간아가 제조되는 방법. - 제 12 항 또는 제 13 항에 있어서,
혈액 세포의 비존재하에서, 혈관 내피 세포, 간엽계 세포 및 조직 또는 장기 세포를 시험관내에서 배양하여 제조한 간아와 비교하여, 간 세포 분화 마커 유전자의 발현이 상승된 간아가 제조되는 방법. - 제 14 항에 있어서,
간 세포 분화 마커 유전자가, α 페토프로테인, 알부민, CYP3A7, 트립토판 대사 효소 TDO2, 및 나트륨·타우로콜산 공수송체로 이루어지는 군에서 선택되는 적어도 1 개의 마커인 방법. - 제 1 항 내지 제 15 항 중 어느 한 항에 기재된 방법에 의해 제조된 기관아.
- 제 16 항에 기재된 기관아를 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 조직 또는 장기의 제조 방법.
- 제 16 항에 기재된 기관아를 인간 또는 비인간 동물에게 이식하는 것을 포함하는, 기관아의 이식 방법.
- 제 16 항에 기재된 기관아를 인간 또는 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 조직 또는 장기의 재생 또는 기능 회복 방법.
- 제 16 항에 기재된 기관아를 비인간 동물에게 이식하여, 조직 또는 장기로 분화시키는 것을 포함하는, 비인간 키메라 동물의 제조 방법.
- 제 16 항에 기재된 기관아, 제 17 항에 기재된 방법에 의해 제조된 조직 및 장기, 그리고 제 20 항에 기재된 제조 방법에 의해 제조된 비인간 키메라 동물로 이루어지는 군에서 선택되는 적어도 하나를 사용하여 약제를 평가하는 방법.
