Example 1. Preparation of Natural Copper Extract

In order to prepare natural copper extracts, hot water extraction was carried out by the following method.

Specifically, 50 g of natural copper was placed in a Daewoong water tank (DWP-5000M) together with 1 L of bottled water (White, Co., Ltd., Murakami Sanseong Spring, Kyungnam Sancheong-gun) and immersed for 1 hour and then heated for 3 hours to obtain natural copper extract . The natural copper extract was filtered through a filter sieve (Test sieve 106 μm (No. 140), Retsch, Germany), frozen in a cryogenic freezer and lyophilized to prepare a natural copper extract sample.

Example 2. Measurement of whitening activity of natural copper extracts

In order to measure the whitening activity of the natural copper extract, the tyrosinase inhibitory activity of the natural copper extract was confirmed.

Specifically, the extract was diluted with PBS to a final concentration of 0.5, 1.25, 2.5, 5, and 12.5% by weight of the natural copper extract, centrifuged at 3000 rpm for 5 minutes, and the resulting supernatant was taken. 50 μl of the supernatant was mixed with 10 μl of 0.2 M sodium phosphate buffer (pH 6.8), 50 μl of 10 mM L-tyrosine and 40 μl of mushroom tyrosinase (1000 U / ml) Min. ≪ / RTI > After the reaction, the absorbance at 510 nm was measured using an ELISA reader, and the tyrosinase inhibitory activity (%) was calculated using the following equation:

As a result of the experiment, it was confirmed that the tyrosinase inhibitory activity of the natural copper extract supernatant was increased in a concentration-dependent manner (Fig. 1). In particular, it was confirmed that when the concentration of the supernatant of natural copper extract was 5 / / mL or more, it showed a high tyrosinase inhibitory activity of 90% or more similar to the positive control (Fig. 1 and Table 1).

Therefore, it was found that the natural copper extract has a whitening activity through the tyrosinase inhibitory activity of the natural copper extract.

Example  3. Measurement of wrinkle-improving activity of natural copper extract

In order to measure the wrinkle improving activity of the natural copper extract, the elastase inhibitory activity of the natural copper extract was confirmed.

Specifically, the final concentration of the natural copper extract was diluted to 0.4, 4, 20 and 40% by weight using a 0.2 M Tris-Cl solution (pH 8.0). 100 μl of the natural copper extract diluted to various concentrations was mixed with 100 μl of the substrate solution and 50 μl of the enzyme solution, followed by reaction at 37 ° C for 30 minutes. At this time, the substrate solution was added to 1 mL of 50 Mm Tris-Cl solution (pH 8.6), and 50 mM N-succinyl- (Ala) 3-p-Nitroanilide, which is an elastase substrate, (Ala) 3-pnitroanilide). In addition, the enzyme solution was diluted to a concentration of 0.6 units / mL of porcine pancreatic elastase (PPE) using a 50 mM Tris-Cl solution (pH 8.6).

Next, the reactants were measured for absorbance at 410 nm using an ELISA reader, and the inhibitory activity (%) of elastase was calculated using the following equation:

Elastase inhibitory activity (%) = {1- (absorbance of sample addition group / absorbance of no addition group)} X 100.

At this time, distilled water was used instead of the natural copper extract, and adenosine was used as a positive control group.

As a result of the experiment, it was confirmed that the activity of inhibiting elastase of natural copper extract was increased in a concentration dependent manner. In particular, it was confirmed that when the concentration of the natural copper extract was 40 ㎍ / mL or more, it exhibited a high inhibitory activity of elastase of 90% or more similar to the positive control (Fig. 2).

Therefore, it has been found that the natural copper extract has wrinkle improving activity through the inhibition of elastase of natural copper extract.

Example  4. Antioxidant activity measurement of natural copper extract

In order to measure the antioxidative activity of natural copper extracts, the free radical scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) Respectively.

Specifically, methanol was used to dilute the natural copper extract to a concentration of 100 / / mL. 150 [mu] l of the diluted natural copper extract was dispensed into 96 well plates with 150 [mu] l of DPPH solution dissolved in 0.4 mM using methanol, and reacted at room temperature for 30 minutes. Next, the reactants were measured for absorbance at 517 nm using an ELISA reader, and the DPPH free radical scavenging activity (%) was calculated using the following equation:

At this time, distilled water was used instead of natural copper extract, and ascorbic acid was used as a positive control group.

Experimental results showed that DPPH free radical scavenging activity was enhanced when the natural copper extract was added. Specifically, it was confirmed that the natural copper extract-added group at a concentration of 100 ug / mL showed a DPPH free radical scavenging activity of 80% or more similar to the positive control group (FIG. 3).

Therefore, it was found that the natural copper extract has antioxidative activity through the high DPPH free radical scavenging activity of the natural copper extract.

Example  5. Natural copper extract Photoaging  Suppression or protection effect

In order to measure the photoaging inhibition or protective effect of the natural copper extract, the inhibition or protective effect of the natural copper extract on cell death induced by UV-B irradiation was confirmed.

Specifically, human fibroblasts in Hs27 and human skin keratinocytes in HaCat cells, 5% CO 2, 37 ℃ and in keeping incubator for at least 95% humidity 10% heat-inactivated fetal bovine serum , 100 ㎍ / mL streptomycin, and And cultured in DMEM (Dulbeccos modified Eagle's medium) containing 100 units / mL penicillin. The human Hs27 and HaCat cells were treated with a natural copper extract at a concentration of 10 μg / mL, washed with phosphate-buffered saline for 2 hours, and exposed to UV-B 8 mJ / cm 2 . As a result, the cell viability of human Hs27 cells was reduced to 100% or less when UV-B alone was irradiated, but the cell survival rate was increased to 100% or more when UV-B was irradiated after treatment with natural copper extract (Fig. 4). In addition, when human HaCaT cells were irradiated with UV-B alone, the cell viability was reduced to 100% or less, whereas when the natural copper extract was treated with UV-B, the cell viability was increased in a concentration-dependent manner 5).

Therefore, it was found that the natural copper extract exhibits photoaging inhibition or protection effect because the natural copper extract increases the cell survival rate of human Hs27 and HaCat cells irradiated with UV-B.

Example  6. Measurement of anti-wrinkle activity of artificial skin of natural copper extract

The wrinkle inhibitory activity of the natural copper extract was evaluated in 3D human skin. Specifically, the natural copper hydrothermal extract was treated with artificial skin for 1 hour and then irradiated with UVB at 50 mJ / cm 2 so as to have a final concentration of 50, 100% by weight. After incubation for 2 days in an incubator maintained at 5% CO 2 , 37 ° C and 95% humidity, the artificial skin was cut and fixed in 4% paraformaldehyde and processed into a paraffin block through tissue processing . Example  7. Measurement of whitening activity of artificial skin of natural copper extract

The whitening effect of natural copper extracts was evaluated in 3D human skin. Specifically, the natural copper hydrothermal extract was treated with artificial skin containing melanin at a final concentration of 50, 100% by weight, cultured in an incubator maintained at a humidity of 5% CO 2 , 37 ° C and 95% And observed with a microscope to confirm the inhibition of melanin production. Therefore, it was found that the natural copper extract showed the whitening activity because the natural copper extract reduces the melanin production of the artificial skin.
