Abstract

Description
본 발명은 높은 고체 함량의 바이오매스로부터 지질이 풍부한 분쇄된 미세조류 가루를 제조하는 방법으로, 이러한 미세조류는 클로렐라 속, 더욱 구체적으로는 클로렐라 프로토테코이데스(Chlorella protothecoides)인 것인 방법에 관한 것이다.
클로렐라 속의 미세조류는 단백질 및 기타 필수 영양소가 풍부하기 때문에, 그것들은 잠재적인 식품 공급원이라는 점은 당업자에게 잘 알려져 있다.

특히, 높은 고체 함량의, 지질이 풍부한 바이오매스 (오일)로부터 생산될 때, 용해된 세포의 현탁액은 상 분리를 겪을 자연스러운 경향을 나타낼 것이다.
사용된 출발 물질이 25%를 초과하고, 특히 28%를 초과하는 고체 함량의 바이오매스라면, 지질이 풍부한 미세조류 가루를 수득하는 것이 정말 어렵거나, 바이오매스의 고체 함량이 35%를 초과한다면 심지어 불가능하다.
특히, 35%를 초과하는 고체 함량에서, 지질이 풍부한 미세조류 바이오매스의 밀링된 물질이 취급될 때 가루 제조를 위해 분무화 단계 전에 사용되는 증발 장치(로타베이퍼(Rotavapor®)와 같은 장치)에 유감스러운 오일 방울 군집이 형성된다.

바이오매스의 생산은 발효조(또는 생물 반응기)에서 수행된다. 생물 반응기, 배양 조건 및 종속 영양 성장 및 증식 방법의 특정 예는 미생물 성장 및 지질의 효율을 개선하기 위해 임의의 적절한 방식으로 조합될 수 있다.
일 특정 구현예에서, 발효는 글루코스 흐름 속도가 3 내지 10 g/l의 잔류 글루코스 농도를 유지하도록 조정된 유가 배치 방식으로 수행된다.

본 발명에 따르면, 단계 (b)에서, 미세조류 가루의 생산에 사용되는 바이오매스 세포는 그의 오일 또는 지질을 방출하도록 용해된다.
세포벽 및 세포 내 성분들은 예를 들어, 비드 밀을 이용하여, 응집되지 않은 세포 입자 또는 잔해까지 분쇄되거나 줄어든다.

조건 하의
- 33% 이하의 고체 함량의 바이오매스를 위한 강하막 증발기,
- 20 내지 45%의 고체 함량의 바이오매스를 위한 강제 유동 증발기이다.
본 발명에 따르면, 단계 (d)에서, 농축된 용해물은 열 처리된다. 이러한 열 처리는 특히, 높은 고체 함량의 용해물의 탈산소화/탈취를 가능하게 한다.
바람직하게는, 단계 (d)는 짧은 시간 동안 높은 온도에서(HTST, 또는 초고온, UHT), 예를 들어, 6초 동안 140℃에서 수행된다.

- 제1 단계에서 150 내지 170 바, 바람직하게는 160 바의 압력, 및

두 단계로 이루어진 장치, 예를 들어, APV 회사가 판매한 가울린(Gaulin) 균질기에서 수행된다.
본 발명에 따르면, 최종 단계(단계 f)는 에멀젼을 건조시켜 미세조류 가루를 수득하는 데 있다.
건조는 바람직하게는 분무화에 의해 수행된다. 물이 제거되는 이 단계 종료 시, 세포 잔해와 지질을 함유하는 건조 분말이 수득된다.
건조 후, 분말의 물 함량 또는 수분 함량은 일반적으로 10% 미만, 우선적으로는 5% 미만이다.
선택적으로, 열 처리 단계 전에 용해물의 pH 조정이 수행될 수 있다.

Claims (6)
- 다음 단계들을 포함하는, 지질이 풍부한 미세조류 가루를 제조하는 방법:
(a) 바이오매스의 건조 중량 기준 50%를 초과하는 지질을 포함하는 미세조류 바이오매스를 제공하는 단계;
(b) 미세조류를 용해시키는 단계,
(c) 미세조류 용해물을 중량 기준 25%를 초과하는 고체 함량까지, 바람직하게는 중량 기준 35% 내지 50%의 고체 함량까지 농축하는 단계,
(d) 이렇게 농축된 용해물에 열 처리를 적용하는 단계,
(e) 단계 (d)에서 수득된 용해물을 고압에서 균질화하여 안정적인 에멀젼을 수득하는 단계,
(f) 상기 에멀젼을 건조시켜 미세조류 가루를 수득하는 단계. - 제1항에 있어서, 미세조류는 클로렐라 속, 더욱 구체적으로는 클로렐라 프로토테코이데스(Chlorella protothecoides)인 것을 특징으로 하는 방법.
- 제1항 또는 제2항에 있어서, 미세조류 용해물의 농축은 증발에 의해 수행되는 것을 특징으로 하는 방법.
- 제3항에 있어서, 용해물의 농축은 다음 조건 하의 증발기에서 높은 온도로 수행되는 것을 특징으로 하는 방법:
- 플래시 유입구 온도: 60 내지 75℃, 바람직하게는 68℃
