세포 호흡 과정은 생물학적 거대분자 가령 단백질, DNA 및 지질과 함께 이를 손상시키면서 반응할 수 있는 중요한 및 잠재적으로 위험한 부산물: 수퍼옥사이드 라디칼 O2 ㆍ-를 발생시키고, 세포 시스템에 대해 잠재적으로 공격적인 기타 매우 반응성 종 (ROS) 가령 ㆍOH, RO2 ㆍ, ROOH, ROㆍ, H2O2 및 ONOOㆍ을 발생시킬 수 있다. 모든 호기성 유기체는 O2 ㆍ- 라디칼에 대한 방어 매카니즘을 가지고 있고, 그 중에서 주요한 것은, O2 ㆍ-의 O2 및 H2O2로의 부동변화를 촉매화하는 디스뮤타제 수퍼옥사이드 (SOD) 효소이다. 따라서 생성된 과산화수소는, 결국, 카탈라제 및 글루타치온 퍼옥시다제에 의해 H2O로 전환되고, 따라서 Fenton 반응 (Fe(II)+)+ H2O2 → Fe(III)+ㆍOH+-OH)에 의해 발생되는 ㆍOH 라디칼의 형성을 저해한다. 특정 병리학적 병태 (염증 공정, 재관류 손상, 심근경색, 알츠하이머병, ALS, 등) 하에서, SOD 효소는 생성된 과량의 O2 ㆍ-를 제거하기에 불충분할 수 있고, 이러한 경우 생물학적 조직은 추가의 손상을 격을 수 있다. 더 높은 수준의 산화적 대사를 특징으로 하는 일부 척추동물 조직, 가령 근육 또는 뇌에서, 산화적 스트레스 공정에 대한 보호를 강화하는 기능을 가지고 있는 항산화제 활성을 가지는 소분자 가령 글루타치온 및 기타 펩티드 (카르노신, 호모카르노신, 안세린, 등)이 높은 수준으로 발견되었다. 특히, 그의 항-라디칼 및 항-과산화 특성과 관련하여, 카르노신에 대한 예비-임상 연구로 백내장을 예방하고 및 일부 경우 회복시키는 그의 능력이 입증되었다 (Biochem. Int. 15, 1105, 1987; Biochim. Biophys. 반면 본발명의 목적인 식 (1)을 가지는 화합물은, HA의 카르복실 기 및 카르노신의 아민 기 사이의 아미드 결합의 형성을 통해 히알루론산과 카르노신의 공유결합성 컨쥬게이션에 의해 수득된다.
히알루론산은, 글루쿠론산 (β-D-글루코피라누론산)의 아노머 탄소 원자와 아세틸 글루코자민(2-아세틸아미노-2-데옥시-β-D-루코피라노시움)의 C-3 사이의 글리코시드 결합에 의해 서로 결합된, 글루쿠론산 및 아세틸 글루코자민으로 구성된 반복적 2당 단위로 구성된 중합체이다. 2당 단위는, 결국, 2당 단위의 아세틸 글루코자민의 아노머 탄소와 다음 2당 단위에 속하는 글루쿠론산 잔기의 C-3 사이의 글리코시드 결합에 의해 서로 결합되어, 가변적 길이의 선형 사슬을 형성한다. 매우 놀랍게도, 본출원인은 이 결합이 예상되는 바와 같이 카르노신의 방출을 지연시켜 이전에 기술된 결합보다 덜 가수분해가능하도록 할 뿐만 아니라, 극히 높은 항산화제 활성을 식 (1) 화합물에 부여한다는 것을 발견하고 입증하였다. 본명세서에서 청구된 바와 같이 아미드를 통한 HA 및 카르노신의 컨쥬게이션은 효소적 가수분해로부터의 단순한 보호로 인한 효과보다 확실히 더 높은 정도로 카르노신의 생물학적 효과를 증가시키고, 이는 HA가 이와 같이 항산화제 활성을 가지지 않는 경우 완전히 예측불가하였다.
상기한 점에서, 본발명의 목적인 컨쥬게이트 내 히알루론산 존재는 카르노신 내에 이미 존재하는 산화적 공정에 대한 작용을 상승적으로 향상시키고, 후반에 혈청 카르노시나제에 대한 상당한 안정성을 보장하고 따라서 그와 같이 또는 아세틸-유도체로서 카르노신에 대한 더 높은 활성을 부여한다.
카르노신과의 아미드 결합을 통해 공액화된 히알루론산의 카르복실 기의 퍼센트는 HA의 총 카르복실 기의 2 내지 25%, 바람직하게는 5 내지 20% 범위이고, 더욱 바람직하게는 7%와 동일하다.
본특허출원의 목적은 또한 식 (1)을 가지는 컨쥬게이트의 Cu (II) 복합체에 관한 것이다.
식 (1)을 가지는 컨쥬게이트의 Cu (II) 복합체의 SOD-형 항산화제 활성은 사실 평가되었고, 유리 카르노신의 Cu (II) 복합체 및 비-공액화된 히알루론산의 Cu (II) 복합체와 인비트로에서 비교에 의해 검증하였다. 그 결과는 식 (1)을 가지는 Cu (II) 복합체의 활성은 카르노신 또는 히알루론산의 상응하는 복합체의 활성에 비해 매우 더 높다는 것을 나타내었다.
본발명의 목적은 또한 식 (1)을 가지는 화합물의 제조를 위한 공정에 관한 것이고, 여기서 히알루론산의 유도체는 카르복실 기에서 보호된 카르노신으로 공유결합적으로 공액화되어 있다. 본발명에 따라 식 (1)을 가지는 화합물은, 사실 적절히 활성화된 히알루론산 및 카르복실 기에서 보호된 카르노신으로부터 시작하여 합성되었다. 히알루론산의 활성 유도체는 바람직하게는 에스테르이고 더욱 바람직하게는 3-히드록시-1,2,3,-벤조트리아진-4(H)-온의 에스테르 (HOOBT)이고, 반면 카르노신 디펩티드는 바람직하게는 메틸 에스테르의 형태로 사용된다.
카르노신의 기타 에스테르 또는 HOOBT의 에스테르와 동등한 히알루론산의 기타 활성 유도체가 이 목적으로 사용될 수 있다.
식 (1)을 가지는 화합물은 비-공액화된 카르노신에 대해 인간 혈청 카르노시나제의 작용에 더 큰 저항성을 나타내었다.
본발명의 목적은 또한, 활성 성분으로서 식 (1)을 가지는 화합물을 포함하는 제약학적, 화장품 또는 건강기능성 조성물에 관한 것이다. 항산화제, 항당화, 항혈소판 및 전이금속에 대한 킬레이팅 특성을 고려하여, 식 (1)을 가지는 화합물을 함유하는 조성물은 당화반응 및 기타 원인 모두로 인한, 유리 라디칼 형성 또는 단백질의 손상된 배열이 병리적 역할을 하는 병태의 치료 또는 예방을 위해 사용될 수 있다. 이들 병태의 예시는 다음을 포함한다: 백내장, 안구건조증, 피부의 노화, 상처, 위장 상처, 당뇨병, 손상된 면역조절 반응, 신장 질환, 간 질환, 종양 및 신경학적 병인 (중추신경계 노화, 알츠하이머병), 국소빈혈/재관류로 인한 손상.
본발명의 목적은 따라서, 단백질 배열 장애 및 병태 가령 백내장, 안구건조증, 피부의 노화, 상처, 위장 상처, 당뇨병, 손상된 면역조절 반응, 신장 질환, 간 질환, 종양 및 신경학적 질환, 손상으로 인한 국소빈혈/재관류의 치료 및/또는 예방에서의 사용을 위한, 활성 성분으로서 식 (1)을 가지는 화합물을 포함하는 제약학적, 화장품 또는 건강기능성 조성물에 관한 것이다. 이들 조성물은, 사실, 바람직하게는 Zn2 +와의 복합체로서, 상처치료, 위장보호, 당조절, 면역조절, 항종양, 유전독성억제, 신경보호 활성, 중추신경계의 항-노화, 국소빈혈/재관류로 인한 손상에 대한 보호 활성, 신장보호 및 간장-보호 활성을 가진다.
투여량 및 투여경로는 병태에 따라 다르고 일반적으로 임상 전문가에 의해 결정되지만, 통상 경구로 100 내지 5000 mg/다이, 및 외용 또는 전신 용도 (즉 주사)에 대해 제제에 대한 중량으로 0.1 내지 10% 범위이다.
제제의 가능한 예시는 다음을 포함한다: 캡슐, 정제, 과립제, 산제, 액제, 연고제, 겔제, 점안제 및 제제 전문가에게 공지된 기타 제제.
가능한 투여경로는 치료될 질병과 당연히 관련되고, 경구, 외용, 국소-지역적, 근육내, 피내, 안내, 관절내 또는 주변, 주입 등을 포함할 수 있다.

본발명을 예시하는 예시를 이하에 제공한다.
다르게 나타내지 않는다면, 필요시 다음과 같이 상업적으로 이용가능한 시약을 사용하고 처리하였다: 카르노신 메틸-에스테르의 합성을 위해 사용된 무수 메탄올 (SIGMA, MeOH)은 사용전 4Å 분자체 상에서, 24 시간 동안 유지시켰다.
0℃에서 메탄올 내에서 카르노신 (Sigma)을 처리하여 카르노신 메틸-에스테르 (Car-OMe)을 합성하였다. 아세틸 클로라이드를 HCl 공급원으로서 사용하였다.
기준으로서 HOD 주파수를 사용하여 500 MHz에서 Varian 장비 (Inova 500) 상에서 1H-NMR 스펙트럼을 기록하였다.
Malvern 3중 검출기 (ViscoteckTDmax)를 구비한 CPG/SEC 크로마토그래피 시스템 상에서 공액화된 HyCar의 분자량 분포 (MWD) 및 고유 점도를 결정하기 위한 실험을 실행하였다.
SHIMADZU RF-5301 PC 분광형광광도계 상에서 형광 측정을 실행하고, SHIMADZU UV-1800 또는 Beckman DU 650 분광광도계 상에서 UV-vis 흡수 측정을 실행하였다.
공액화된 HyCar의 Cu(II) 복합체 및 카르노신 또는 HA의 Cu(II) 복합체의 SOD-형 활성을 Fridovich 시험에 의해 결정하였다 (Anal. Biochem . 44, 276, 1971).
상기 Cu(II) 복합체를 제조하기 위해, 다음 농도에서 수성 용액 내 초산제2동을 사용하였다: [L] = [Cu(II)] = 10-6 -10-7; 사실 카르노신은 쉽게 전이금속과 킬레이트를 형성한다는 것이 본업계에서의 숙련가에게 공지되어 있다 (Free Red Res. Commun.; 1991, 12-13 pt.1, 179-185). 사용된 모든 화학 생성물을 Sigma 로부터 구입하였다.

실시예:
2-(3- 아미노프로판아미드 )-3-(1H- 이미다졸 -4-일) 프로판산의 공액화된 3- N - 히알루로닐 (HyCar)의 합성
대표적인 합성 절차에서, 1그램의 소듐 히알루로네이트 (195 KDa)를, 5℃에서 20 ml의 차가운 테트라하이드로푸란 (THF)에 교반 하에서 부가하였다. 다음 생성물을 순서대로 결과로서 얻어진 현탁액에 부가하였다: 20 ml의 0.5 mmoles의 HOOBT를 함유하는 H2O/THF (1:1 v/v) 용액, 0.2 mmoles의 트리스[2-(2-메톡시에톡시)에틸]아민을 함유하는10 ml의 H2O/THF (1:1 v/v) 용액, 및 0.125 mmoles의 L-카노신 메틸에스테르를 함유하는5 ml의 메탄올 용액. 5℃에서 30 분 후, 10 ml의 N-(3-Di메틸아미노프로필)-N′-에틸-카보디이미드 하이드로클로라이드 (EDC.HCl; 0.25 mmoles) 용액을 상기 혼합물에 부가하고 20 시간 동안 5℃에서 연속 교반 하에서 전체 반응 부피를 두었다. 이 기간 이후, 반응 혼합물을 100 ml의 NaOH 0.1N 용액으로 처리하고 추가의 3.5 시간 동안 5℃에서 두었다. 이 시점에서, 상기 혼합물의 pH를 2N HCl로 7.0로 만들고, 800 ml의 아세톤의 부가에 의해 컨쥬게이트를 침전시켰다. 목적 생성물을 이후 제거하고 원심분리에 의해 상청액으로부터 제거하고 이후 60 시간 동안 물에 대해 투석시켰다. 컨쥬게이트를 이후 수집하고 동결건조시켰다 (수율 70%).
1H NMR (D20, 500 MHz) (ppm): 8.66 (단일항; 이미다졸 링의 H-2), 7.34 (단일항; 이미다졸 링의 H-5), 4.61-4.52 (넓은 다중항; 글루쿠론산의 잔기의 H-6, N-아세틸글루코자민의 잔기의 H-1 및 프로판산의 사슬의 H-2), 3.90-3.23 (넓은 다중항; 글루쿠론산의 잔기의 H-2, H-3, H-4 및 H-5, N-아세틸 글루코자민 잔기 및 프로판아미드 기의 C-3 메틸렌의 H-2, H-3, H-4, H-5, 2H-6), 3.17 (다중항; 프로판산의 사슬의 2H-3, 2.44 (다중항; 프로판아미드 기의 메틸렌 C-2), 2.06 (N-아세틸글루코자민의 잔기의 넓은 단일항 CH3). 주요 할당을 가지는 공액화된 HyCar의 1H NMR 스펙트럼을 도 1에 나타내는데, 도 1은 카르노신 단위에 아미드 다리로 연결된 카르복실 기의 퍼센트 (로딩 %)를 계산하기 위해 사용된 공명을 나타낸다.
컨쥬게이트 내에 존재하는 카르노신의 양은 2.06 ppm에서의 신호 (HA의 아세틸 기와 관련된)의 적분값과 2.44 ppm 에서의 신호 (프로판아미드 기의 C-3와 관련된), 또는 이미다졸 링의 H-2 또는 H-5 신호 중 하나 (각각 8.66 및 7.34 ppm에서)의 적분값 사이의 비로부터 결정된다.

시리즈로 설치된 두 개의 TSK-GEL GMPWXL 칼럼 (7.8 mm ID x 30 cm; Viscotek- TOSOH BIOSCIENCE)를 구비한 크로마토그래피 시스템 GPCmax VE 2001 (Malvern) 상에서 수행된 크기 선별 크로마토그래피에 의해, 위에서 나타낸 파라미터들을 결정하였다. 상기 시스템을, 시리즈로 배치된 굴절률 검출기, 광산란 검출기 및 4-모세관 편차 점도계:를 포함하는 세 개의 검출기의 시스템과 커플링시킨다. 40℃에서 0.6 ml/min 흐름으로 0.5 g/L의 소듐 아지드를 함유하는 소듐 니트레이트의0.1M 수성 용액을 사용하여 상기 샘플을 용리하였다. 취득 및 데이터 분석을 위해 소프트웨어 Omnisec 4.1를 사용하였다.
도 2는 TSK-GEL GMPWXL 칼럼 상 HyCar (7% 로딩) 샘플의 크로마토그래피 거동을 나타내는 HyCar (로딩 7%)에 대해 수득된 대표적인 크로마토그램을 나타낸다. 3중-검출기 검출을 Viscotek TDA 302 (Malvern) 상에서 실행하였다.
3중 검출기와 커플링된 크기 선별 크로마토그래피에 의해 결정된 상응하는 히알루론산과 비교하여, 히알루론산-카르노신의 컨쥬게이트에 대해 상이한 농도에서 결정된 고유 점도 및 분자량 (MW)에 관한 물리적 파라미터를 아래의 표 1에 나타낸다.
도 3는, 예시로서, 비-공액화된 히알루론산과 비교하여 히알루론산-카르노신의 컨쥬게이트 (로딩 7%)의 Mark-Howink 그래프 (분자량 log에 대한 점도 log, MW)을 나타낸다: 더욱 상세하게는, 상기 그래프는 상응하는 비-공액화된 히알루론산의 동일한 농도에서의 용액과 비교하여 HyCar (7% 로딩)의 용액 (0.5 mg/ml)에 대해 수득되었다.

SOD-형 항산화제 활성
Fridovich의 직접 방법 (Anal. Biochem . 44, 276, 1971)을 사용하여 HyCar 컨쥬게이트의 Cu(II) 복합체의 SOD-형 활성을 결정하였다. 크산틴-크산틴 옥시다제 시스템에 의해 수퍼옥사이드 음이온을 효소적으로 발생시키고 이후 560 nm에서 니트로 블루 테트라졸륨 (NBT) 감소를 분광학적으로 모니터링하였다. 반응 혼합물은 다음을 함유하였다: pH 7.4에서 인산염 완충액 (10 mM) 내 사이토크롬 c (30 μM) 또는 NBT (250 μM), 크산틴 (50 μM). 적절한 양의 크산틴-옥시다제를 2 ml의 이 혼합물에 부가하여 0.024의 ΔA min- 1를 생성하였다. 이는 1.1 μM min- 1와 같은 O2 ㆍ1- 생산속도에 해당한다. 발색 분자의 감소 속도를 600 초 동안 검사 하에서 복합체의 존재하 및 부재하에서 측정하였다. 1 cm의 광학 경로를 가지고, 온도조절장치-조절되고 자기 교반을 구비한 큐벳을 사용하여 25±0.2 ℃에서 모든 측정을 실행하였다. 크산틴-옥시다제 활성의 가능한 저해를 배제하기 위해, 별도의 실험에서 크산틴 옥시다제의 부분에 대한 요산 생성을 295 nm에서 분광학적으로 따랐다.
pH 7.4에서 결정된 검사 하 금속 복합체의 I50 값 (즉 NBT의 감소의 50% 저해를 생성하는 농도)을 도 4 및 아래의 표 2에 나타낸다.
따라서 표 2는 NBT의 감소의 50%를 저해할 수 있는 농도 (I50)로서 표시된, HyCar, Car 및 HA의 Cu(II) 복합체의 SOD-형 활성을 나타낸다.

카르노신 + HA 또는 카르노신 단독의 혼합물의 안정성과 비교하여, 인간 혈청 카르노시나제에 대한 HyCar의 시간-의존적 안정성을, 이전에 나타낸 바와 같이 안정적으로 형질감염된 Hela Cells의 배지에 의해 정제된, 상기 효소 (CN1)와 함께 50 mM 트리스/HCl (pH 8.0) 내에서 37℃에서 이들 물질 (900 μM) 각각을 배양함으로써 결정하였다 (Antioxid. Claims (10)
