The absorbance of the reactant was confirmed at 475 nm, and the inhibition rate of dopa oxidation was calculated therefrom, as shown in FIG. 4 (b). The tyrosinase inhibition rate shown in FIG. 4 (a) was determined from the absorbance obtained in each experimental sample and the absorbance reduction rate in the experimental group to which no sample was added.

As a result, as shown in Fig. 4 (a), it was confirmed that the ground-band fermented product (ESR-F) inhibited the oxidation of tyrosinase and dopa related to melanin production in a concentration-dependent manner. In addition, it was confirmed that the effect of ESR-F was much better than that of ESR-E.

[ Experimental Example  5] Groundbreaker  Fermented product ( ESR -F) MITF  Expression Inhibition  evaluation

B16F10 cells (Korean Cell Line Bank) were cultured in a 5% CO 2 incubator for 24 hours in a cell culture dish, and then α-MSH (α-Methylene-stimulating hormone, α-M ) 100 [mu] g / ml to each cell to induce melanogenesis. Then, each cell was treated with an experimental sample and incubated for 24 hours. RNA was extracted from each cell and the expression pattern of MITF gene related to melanogenesis was observed by RT-PCR. The ground band extract (ESR-E) prepared in Comparative Example 1 and the ground-band fermented product (ESR-F) obtained in Example 1 were treated at a concentration of 50 μg / ml and 100 μg / ml, respectively. The sequences of the primers used in the RT-PCR were as follows.

As a result, as shown in FIG. 5, it was confirmed that the ground-band fermented product (ESR-F) inhibits melanogenesis by inhibiting the expression of MITF related to melanin production. On the other hand, it was also confirmed that the effect of the ground-fed fermented product (ESR-F) was far superior to that of the ground-band extract (ESR-E).

[ Experimental Example  6] Groundbreaker  Fermented product ( ESR -F) is cytotoxic

Mouse-derived B16 / F10 cells (Korean Cell Line Bank) and human-derived CCD-986sk cells (Korean Cell Line Bank) were used to confirm cytotoxicity. Experimental samples were treated with DMEM (Lonza) medium containing 1% penicillin / streptomycin (GIBCO) and 10% fetal bovine serum (GIBCO) 2 < / RTI > The ground-band fermented product prepared in Example 1 and the ground-band extract (ESR-E) prepared in Comparative Example 1 were used as test samples at concentrations of 100 μl / ml, 50 μl / ml, 25 μl / ml and 12.5 μl / . After that, cytotoxicity was confirmed using MTT (3- (4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide) assay in each cultured cell. MTT reacts with mitochondrial enzymes in living cells with a light yellow matrix to form Formazan. The formazan is dissolved in DMSO (Dimethyl sulfoxide), and the cell viability can be measured by measuring the absorbance at 570 nm.

As a result, as shown in FIG. 6, the groundnut fermented product as an active ingredient of the composition according to the present invention showed no cytotoxicity at the total experimental concentration, while the ground band extract (ESR -E) was found to have cytotoxicity as it was confirmed that the cell survival rate was rapidly lowered at some concentrations.

Hereinafter, a formulation example of the composition according to the present invention will be described, but it can be applied to various formulations, which are not intended to limit the present invention but to describe it specifically.

[Formulation Example 1] Soft capsule

The soft capsules were prepared by mixing 150 mg of fermented ground wax, 2 mg of palm oil, 8 mg of palm kernel oil, 4 mg of yellow radish oil, and 6 mg of lecithin and 400 mg per capsule according to a conventional method.

[Formulation Example 2] Tablets

The granules were prepared by mixing 150 mg of fermented ground wax, 100 mg of glucose, 50 mg of red ginseng extract, 96 mg of starch and 4 mg of magnesium stearate and 40 mg of 30% ethanol, followed by drying at 60 ° C. And the tablet was tableted.

[Formulation Example 3] Granules

The granules were formed by mixing 150 mg of fermented ground wax, 100 mg of glucose, 50 mg of red ginseng extract and 600 mg of starch, and adding 100 mg of 30% ethanol, followed by drying at 60 ° C to form granules, . The final weight of the contents was 1 g.
