Claims (14)
Among the wild type CPC (Cephalosporin C) acylase sequence consisting of the alpha-subunit represented by the amino acid sequence of SEQ ID NO: 3 and the beta-subunit represented by the amino acid sequence of SEQ ID NO: 4, the F58? Amino acid is substituted with Valine Mutation CPC acylase.
The mutant CPC acylase according to claim 1, wherein the mutant CPC acylase further comprises a mutation selected from the group consisting of Y153? T, F177? L and Y153? T + F177? L.
3. The mutant CPC acylase according to claim 2, wherein the mutant CPC acylase further comprises a mutation in which the amino acid I45 [beta] or V382 [beta] is substituted with another amino acid.
4. The method of claim 3, wherein said mutation is selected from the group consisting of I45 beta replaced by an amino acid selected from the group consisting of Methionine, Valine, Alanine, Cysteine, and Leucine, Leucine) or isoleucine (Isoleucine).
5. The mutant CPC acylase according to claim 4, wherein the mutant CPC acylase comprises an alpha-subunit represented by the amino acid sequence of SEQ ID NO: 6 and a beta-subunit represented by the amino acid sequence of SEQ ID NO:
7. A gene encoding a mutant CPC acylase according to any one of claims 1 to 5.
7. The gene according to claim 6, wherein the gene is represented by the nucleotide sequence of SEQ ID NO: 5.
A recombinant expression vector comprising the gene of claim 6.
9. The recombinant expression vector according to claim 8, wherein the recombinant expression vector comprises the gene of claim 7.
A host cell transformed by the expression vector of claim 8.
A microorganism transformed by the expression vector of claim 8.
12. The microorganism according to claim 11, wherein the microorganism is Escherichia coli MC1061 / pBC-PM2 (accession number: KCTC12344BP) transformed by the recombinant expression vector of claim 9.
A composition for the preparation of 7-ACA (7-Aminocephalosporanic acid) comprising a mutant CPC acylase according to any one of claims 1 to 5.
